BACKGROUND Ameloblastoma is a rare, benign odontogenic neoplasm which is locally aggressive and mostly present with a painless swelling. The enigma about the diagnosis of proliferative ameloblastoma and ameloblastic carcinoma is still a debate because the diagnostic criteria is not standardized or quantified which has a direct correlation on its biological behaviour and prognosis. Despite numerous studies, correlation between the histological patterns of ameloblastoma and tumour behaviour has not been consistently established. The present study was done to compare the expression levels of Ki-67 between conventional ameloblastoma, proliferative ameloblastoma and ameloblastic carcinoma and to assess the usefulness of these markers for diagnostic differentiation. METHODS A retrospective study of total of 18 cases of ameloblastoma were retrieved from the archives of Department of Oral and Maxillofacial Pathology, Saveetha Dental College from 2012 till 2019, which included conventional ameloblastoma, proliferative ameloblastoma and ameloblastic carcinoma. Immunohistochemical (IHC) analysis was done using the marker Ki-67 and labelling index were determined for the same. RESULTS The results of the current study showed that the cellular proliferative activity assessed using Ki-67 in follicular ameloblastoma was (55 %), 4 cases of plexiform ameloblastoma (22 %), 17 % of proliferative ameloblastoma and 6 % of ameloblastic carcinoma showed negative expression. CONCLUSIONS Immunophenotyping using the marker Ki-67 may be a useful tool for histological typing of ameloblastoma. KEY WORDS Ameloblastoma, Ki-67, IHC, Immunophenotyping

Abstract Purpose: Epidermal growth factor receptor tyrosine kinase inhibitors (TKIs) are beneficial in patients with lung cancer. We explored the clinical value of 99mTc-Galacto-RGD2 single-photon emission computed tomography (SPECT/CT) in patients with lung cancer, Integrin αvβ3 expression, and neovascularization in lung cancer subtypes was also addressed. Methods: A total of 185 patients with lung cancer and 25 patients with benign lung diseases were enrolled in this prostective study from January 2013 to December 2016. All patients underwent 99mTc-Galacto-RGD2 imaging. The region of interest was drawn around each primary lesion, and tumour uptake of 99mTc-Galacto-RGD2 was measured as the tumour/normal tissue ratio(T/N). The diagnostic efficacy was evaluated by receiver operating characteristic curve analysis. Tumour tissues were obtained from 66 patients with malignant diseases and seven with benign disease. Tumour expression levels of αvβ3, CD31, Ki-67, and CXCR4 were analysed to determine their value for phenotyping and metastasis potential evaluation. Results: The lung cancer patients included 22 cases of small cell lung cancer (SCLC), 48 squamous cell carcinoma (LSC), 97 adenocarcinoma (LAC), and 18 other types of lung cancer. The sensitivity, specificity, and accuracy of 99mTc-Galacto-RGD2 SPECT/CT using a cut-off value of 2.5 were 91.89 %, 48.0 %, and 86.67 %, respectively. Integrin αvβ3 expression was higher in non-SCLC compared with SCLC, while LSC showed denser neovascularization and higher integrin αvβ3 expression. Integrin αvβ3 expression levels were significantly higher in advanced (Ⅲ, Ⅳ) than early stages (Ⅰ, Ⅱ). However, there was no significant correlation between tumour uptake and αvβ3 expression. Conclusion: 99mTc-Galacto-RGD2 SPECT/CT has high sensitivity but limited specificity for detecting primary lung cancer. RGD imaging may help evaluate the biological behaviour and phenotyping, and thus aid management in lung cancer.

Objective To investigate the association between microRNA-3615 (miR-3615) expression and the prognosis and clinicopathological features in patients with hepatocellular carcinoma (HCC). Methods We obtained clinicopathological and genomic data and prognostic information on HCC patients from The Cancer Genome Atlas (TCGA) database. We then analyzed differences in miR-3615 expression levels between HCC and adjacent tissues using SPSS software, and examined the relationships between miR-3615 expression levels and clinicopathological characteristics. We also explored the influence of miR-3615 expression levels on the prognosis of HCC patients using Kaplan–Meier survival curve analysis. Results Based on data for 345 HCC and 50 adjacent normal tissue samples, expression levels of miR-3615 were significantly higher in HCC tissues compared with adjacent tissues. MiR-3615 expression levels in HCC patients were negatively correlated with overall survival time and positively correlated with high TNM stage, serum Ki-67 expression level, and serum alpha-fetoprotein level. There were no significant correlations between miR-3615 expression and age, sex, and pathological grade. Conclusion MiR-3615 may be a promising new biomarker and prognostic factor for HCC.

Using anti-p53 (PAb1801 and PAb240), anti-DNA polymerase α and Ki-67 monoclonal antibodies, the expression of p53 was studied in 11 normal endometria, 14 endometrial hyperplasias and 27 endometrial carcinomas and its relationship to the proliferative activity of the tumors was examined. Normal endometria and simple hyperplasias were completely negative for p53. The PAb1801 indices of complex hyperplasias and complex atypical hyperplasias were 2.5±1.8% and 5.0±3.2%, respectively. The PAb1801 indices of grade 1, grade 2 and grade 3 endometrial carcinomas were 10.2±14.2%, 44.4±29/0% and 45.0±32.5%, respectively. These results indicate a progressively enhanced p53 expression in the sequence from normal endometrium, through hyperplasia to carcinoma. A significant correlation between p53 expression and labeling indices of Ki-67 and DNA polymerase α was observed in endometrial carcinomas. The endo-metrial carcinomas with p53 overexpression developed mainly in post-menopausal patients and were frequently high-grade tumors with deep myometrial invasion. These findings may indicate that overexpression of p53 protein contributes to the proliferative activity of the tumor cells.

AimBreast carcinoma proliferative activity, histological grade and commercial molecular tests are all important in prognostication and treatment. There is a particular need for improved, standardised techniques for subclassification of grade 2 breast cancers into low-risk and high-risk prognostic groups. In this study we investigated whether gene expression profiling of five proliferation genes was feasible using breast cancer tissue in a clinical setting and whether these profiles could enhance pathological assessment.MethodsExpression of five proliferation gene mRNAs; Ki-67, STK 15, CCNB1, CCND1 and MYBL2, was quantified in 27 breast carcinomas and compared with Ki-67 proliferation index (PI) and Nottingham mitotic score.ResultsExpression of Ki-67, STK15 and MYBL2 mRNA showed moderate Spearman's correlation with Ki-67 PI (p<0.01), but CCND1 and CCNB1 showed weak, non-significant correlation. Individual gene expression did not associate with mitotic score but combined mRNA expression correlated with both Ki-67 PI (p=0.018) and mitotic score (p=0.03; 0.007).ConclusionsThis study confirms mRNA analysis in breast carcinoma formalin-fixed, paraffin-embedded samples is feasible and suggests gene expression profiling, using a small set of five proliferation genes, has potential in aiding histological grading or assessment of proliferative activity of breast cancers. To fully evaluate the clinical applicability of this approach, a larger cohort study with long-term follow-up data is required.

In a retrospective study of lymphomas in animals, tumors in 72 dogs, 81 cats and 90 cows were classified on the basis of cell size (small, medium and large), nuclear cleavage (follicular center cells), and histologic architecture (nodular or diffuse). Each subtype was classified by age of animal at death, number of metastases, breed, and sex. As in man, nodular cleaved tumors are rare in animals, the cow having the most varied tumor types. There was one cleaved-cell tumor in 72 lymphomas in dogs, 23 of 81 in cats, and 33 of 90 in cows. There were six nodular tumors of 72 in dogs, two of 81 in cats, and eight of 90 in cows. Fifteen of 16 nodular lymphomas had noncleaved cells and twelve had small or predominantly small cells. Cats with nodular lymphomas were older at death than cats with diffuse lymphomas. Nodularity was not associated with greater age at death in dogs and cows. Animals with cleaved-cell lymphomas were older at death than those with noncleaved tumors; this difference was highly significant in cows. The number of metastases was greater with nodular tumors in all three species, and was equal in cleaved and noncleaved tumors. The biological behaviour of lymphoid tumors in animals is similar to those in man when the same criteria of classification are used.

A morphological and immunohistochemical study of 24 gums biopsies was conducted in 19 patients aged 35-60 years with a diagnosis of partial secondary edentulous, chronic generalized periodontitis of moderate and severe degree (14 patients), and also without pathological changes in the periodontal disease (5 patients), who underwent dental implantation. Immunohistochemical reactions with antibodies to Ki-67, VEGF, SMA were performed on serial paraffin sections. It has been established that chronic periodontitis is characterized by a higher proliferative activity of the epithelium, which reflects its hyperplastic changes, as well as a lower content of SMA positive cells and the practical absence of the formation of privascular couplings from SMA-positive cells that are associated in tissues with growth zones, which indirectly indicates reduced tissue regenerative capacity. Therefore, in the case of the operation of dental implantation requires additional treatment aimed at anti-inflammatory and pro-regenerative effects.

Abstract Background: Sepsis is a highly complex and fatal syndrome. It is the main cause of death in the intensive care unit. Early diagnosis is beneficial to reduce the mortality of sepsis and improve the prognosis of patients. Therefore, we look forward to finding cheap and fast diagnostic criteria to quickly assess the patient's condition.Methods: This is a retrospective study. The study enrolled 499 patients in the First Affiliated Hospital of Xinjiang Medical University from January 1, 2018 to June 22, 2020, and 96 healthy cases in the same period. Using the diagnostic criteria of bacterial infection, SIRS criteria and Sepsis-2 consensus criteria, 499 patients and 96 healthy cases were divided into 4 groups: sepsis group (n=300), SIRS group (n=151), infection group (n= 48), the control group (n=96). We collected the results of routine laboratory tests, inflammation indicators and blood culture results of these patients. Results: The sepsis group compared with the control group, MCV, NE, WBC, PLT, HB, D-Dimer, PT, CRP, PCT, IL-6, ALB, TBIL, Cr, LAC, CysC and BNP were statistically significant. D-dimer, CRP and PCT have higher diagnostic efficiency. Compared with the difference between the infection group and the SIRS group, PLT and IL-6 are statistically significant, and have a certain diagnostic value. Sepsis group VS infection group, WBC, IL-6, NE and TBIL showed statistical differences in the comparison. The AUC of NE was 67.6, which was the largest among the three. The specificity (95.8%) was the highest, but the sensitivity (49%) was low.Conclusions: This retrospective study shows that NE, WBC, and D-dimer can help in the early diagnosis of sepsis. D-dimer performs best. WBC and NE may have a differential diagnosis significance between the sepsis group and the infection group. This result can provide a timely and convenient assessment tool for early diagnosis of sepsis.

BackgroundGrowth arrest and DNA-damage-inducible protein 45 alpha (GADD45A) was previously found to be associated with risk of several kinds of human tumors. Here, we studied the expression and clinical significance of GADD45A in breast cancer.MethodsWe performed an immunohistochemical study of GADD45A protein from 419 breast cancer tissues and 116 adjacent non-neoplastic tissues.ResultsSignificantly high GADD45A expression were observed in breast cancer tissues compared with adjacent non-neoplastic tissues (P < 0.001) and were independently correlative with estrogen receptor negative (P = 0.028) and high Ki-67 index (P < 0.001). Kaplan–Meier survival analysis revealed that patients with high GADD45A expression levels had a worse long-term prognosis in triple negative breast cancer (P = 0.041), but it was not an independent prognostic factor in multivariate analysis (P = 0.058).ConclusionsGADD45A expression levels are significantly correlative with estrogen receptor status and Ki-67 index in human breast cancer. Patients with triple negative breast cancer might be stratified into high risk and low risk groups based on the GADD45A expression levels.
