Anti-inflammatory, anti-diarrheal, thrombolytic and cytotoxic activities of an ornamental medicinal plant: Persicaria orientalis

AbstractBackground:, an ornamental medicinal plant, has been used in traditional medicine for the treatment of various diseases. Although the plant is reported to have some important pharmacological effects, many medicinal values remain unidentified. Our objective was to evaluate the anti-inflammatory, anti-diarrheal, thrombolytic, and cytotoxic properties of the methanol extract ofMethods:Anti-inflammatory activity was measured by the inhibition of hypotonicity-induced human red blood cell hemolysis and albumin denaturation technique in vitro of Po-MeOH. Diarrheal episodes were examined in mice with castor oil-induced diarrhea. The clot lysis and brine shrimp lethality bioassay in vitro were used to evaluate the thrombolytic and cytotoxic activities of the plant extract, respectively.Results:Using in vitro anti-inflammatory models, the results demonstrated that Po-MeOH at the five different dose ranges from 31.25 to 500 μg/mL significantly (p<0.05) protected (0.98%–50.71%) the erythrocyte membrane against lysis induced by hypotonic medium solution and protein denaturation (38.27%–79.22%) of bovine albumin, respectively. The extract exhibited a significant reduction of severity (75.17%) of castor oil-induced diarrhea in mice at the highest dose of 400 mg/kg compared to loperamide (82.06%) at 5 mg/kg. Po-MeOH also showed 33.14% clot lytic activity in the thrombolytic test and cytotoxicity with LCConclusions:These findings suggest that Po-MeOH has significant anti-inflammatory and anti-diarrheal effects along with moderate thrombolytic and lower cytotoxic properties that may warrant the further exploration.

Objective: The objective of this study was to determine the anti-inflammatory activity of methanol extract of Niebuhria apetala and its possible mechanism of action. Methods: Methanol extract of Niebuhria apetala leaf (NAL) was assessed for its anti-inflammatory activity by in vitro methods. Using albumin denaturation assay, proteinase inhibitory activity, membrane stabilization, and antilipoxygenase activity at different concentrations, in vitro anti-inflammatory activity was estimated. The standard drug used for this purpose was aspirin. Results: Methanol extract NAL at a concentration range of 100–500 μg/ml significant (p<0.01) protects the heat-induced protein denaturation. At the concentration of 500 mg/ml, NAL showed significant (p<0.01) inhibition of protease inhibitory action. Heat-induced hemolysis of erythrocyte, hypotonicity-induced hemolysis, and lipooxygenase activity were significant (p<0.01) inhibited at the concentration of 500 μg/ml. Conclusion: Finally, the present study indicates that methanol extract of Niebuhria apetala can be a potential source of anti-inflammatory agent.

AbstractBackgroundAsparagus horridus is an edible plant known as “Ayrelli” in North Cyprus. The scientific literature has not yet submitted a report about the antioxidant, anti-inflammatory and anti-cancer activities of A. horridus plant from North Cyprus until now. The purpose of the research was to determine the antioxidant, anti-inflammatory and anti-cancer activities of A. horridus.Materials and methodsSoxhlet extraction of A. horridus was performed using methanol. Antioxidant activity was determined by DPPH, TFC, FRAP and TPC assays. Protein-denaturation assay was performed to determine the anti-inflammatory effect. The anti-cancer effects of the extract on HepG2 and B-CPAP cell lines were determined with MTT assay.ResultsAntioxidant activity for A. horridus extract was determined by DPPH (50%), TFC (266.26 μg QUE/mg extract), FRAP (1.27 μg FeSO4/mg extract) and TPC (167.613 μg GAE/mg extract) assays at 25 mg/mL. Inhibition of protein-denaturation activity was found as 29.42% at 25 mg/mL. After 24 h of the extract treatment, cell proliferation of HepG2 and B-CPAP cancer cells were inhibited at IC50 values 63.24 μg/mL and 101.24 μg/mL, respectively.ConclusionThese results have shown that the methanol extract of A. horridus grows in North Cyprus has antioxidant, anti-inflammatory and anti-cancer activities.

Objective: Sarcostemma brunonianum Wight and Arn is a potential medicinal plant belonging to Asclepiadaceae. Bioactive constituents of the plant support the application of treating various ailments in the traditional system of medicine. The study aims to determine the presence of various phytoconstituents in stem, root, and flowers. Methods: Hot percolation method was carried out to obtain crude extracts using different solvent systems from low polar-high polar solvents ranging from petroleum ether, chloroform, (mid-polar) ethyl acetate, methanol and water. Estimation of total phenols, tannins and In vitro antioxidant, anti-inflammatory activities were evaluated for the determination of potential pharmaceutical drugs. Results: The results revealed the presence of some phytoconstituents such as phenols, tannins, glycosides, gums and mucilages. Ascorbic acid, BSA, Rutin and Gallic acid were used as the reference standard. The total phenolic content was found to be high in stem methanol extract 440.84±69.99 mg/g Gallic acid equivalent, whereas the tannin content was 291.78±4.68 mg/g GAE. The result proves that the S. brunonianum stem methanol extract possesses antioxidant and anti-inflammatory activities when compared to reference standards. In vitro, Nitric oxide scavenging activity of stem showed a maximum % of inhibition in methanol stem extract (24.39µg/ml) and anti-inflammatory activity maximum inhibition was found to be (55.56 %) in stem methanol and flower(53.62 %). The IC50 (concentration required for 50% inhibition) was also calculated for the DPPH radical model. Conclusion: This study results proclaims and justifies the role of folklore medicinal plant S. brunonianum in the treatment of inflammatory-related ailments and can be recommended for an effective drug.

Abstract Background Campsis radicans L. is a flowering plant in Bangladesh, traditionally used for the treatment of several human diseases. In this study, in vitro antioxidant, thrombolytic, membrane stabilizing and in vivo analgesic, hypoglycemic, anti-diarrheal and CNS antidepressant activities of organic soluble fractions of crude methanol extract of C. radicans leaf were investigated using appropriate experimental models. Methods The leaves of C. radicans were collected, authenticated, dried and extracted with methanol at room temperature for 30 days. The concentrated methanol extract was partitioned to petroleum-ether (PESF), dichloromethane (DMSF) and ethyl acetate (EASF) soluble fractions. The antioxidant activity of these fractions was determined by DPPH free radical scavenging method. Total phenolic content was determined by the Folin-Ciocalteau’s spectrophotometric method. The thrombolytic activity was assessed by measuring clot lysis ability whereas the membrane stabilizing activity was evaluated by heat- and hypotonic solution-induced hemolysis assay. Tail immersion procedure and acetic acid- induced writhing model were used to measure the analgesic activity of C. radicans. The hypoglycemic, anti-diarrheal and CNS antidepressant activities were determined by oral glucose tolerance test, castor oil-induced diarrheal model and thiopental-sodium induced sleeping time test in mice, respectively. Results All the organic soluble fractions of C. radicans contained phenolic compounds varying from 6.38 to 60.13 mg of GAE/gm of extractive, while in DPPH assay, EASF showed the highest free radical scavenging activity with IC50 is 4.69 μg/ml. The PESF exhibited highest thrombolytic activity (57.14% clot lysis) and the DMSF showed maximum 53.95% inhibition of heat-induced hemolysis of human RBCs. In both tail immersion and acetic acid induced writhing models, the PESF, DMSF, EASF at the doses of 200 and 400 mg/kg body weight, induced a significant (P < 0.001) decrease in the painful sensation in mice. Substantial (P < 0.05) anti-hyperglycemic activity of test samples was found in mice loaded with glucose at the same doses mentioned earlier. Castor oil induced diarrheal test of the plant extractives has shown significant effect in comparison to control group. In CNS antidepressant activity assay, the test samples were able to reduce the duration of sleep in mice caused by thiopental administration. Conclusion All these findings revealed that C. radicans possess significant antioxidant, thrombolytic, membrane stabilizing, analgesic, hypoglycemic, anti-diarrheal and CNS antidepressant activities.

Pani heloch (Antidesma montanum) is traditionally used to treat innumerable diseases and is a source of wild vegetables for the management of different pathological conditions. The present study explored the qualitative phytochemicals; quantitative phenol and flavonoid contents; in vitro antioxidant, anti-inflammatory, and thrombolytic effects; and in vivo antipyretic and analgesic properties of the methanol extract of A. montanum leaves in different experimental models. The extract exhibited secondary metabolites including alkaloids, flavonoids, flavanols, phytosterols, cholesterols, phenols, terpenoids, glycosides, fixed oils, emodines, coumarins, resins, and tannins. Besides, Pani heloch showed strong antioxidant activity (IC50 = 99.00 µg/mL), while a moderate percentage of clot lysis (31.56%) in human blood and significant anti-inflammatory activity (p < 0.001) was achieved with the standard. Moreover, the analgesic and antipyretic properties appeared to trigger a significant response (p < 0.001) relative to in the control group. Besides, an in silico study of carpusin revealed favorable protein-binding affinities. Furthermore, the absorption, distribution, metabolism, excretion, and toxicity analysis and toxicological properties of all isolated compounds adopted Lipinski’s rule of five for drug-like potential and level of toxicity. Our research unveiled that the methanol extract of A. montanum leaves exhibited secondary metabolites that are a good source for managing inflammation, pyrexia, pain, and cellular toxicity. Computational approaches and further studies are required to identify the possible mechanism which responsible for the biological effects.

The present study is carried out to evaluate anti-inflammatory and anti-arthritic potential of Justicia gendarussa. Leaves and stem extracted with ethanol and chloroform. These extract is tested against in-vitro (HRBC membrane stabilization method and Inhibition of protein denaturation method) and in-vivo (Carrageenan induced paw edema model) anti-inflammatory models. To assess the anti-arthritic activity, FCA induced arthritis model is used. Assessment of arthritis is done by paw volume, joint diameter, body weight, biochemical parameters, hematological parameters (Hb, RBC, WBC, ESR). The effect of in-vitro anti-inflammatory model depends on concentration. Both test extract and standard Diclofenac sodium has been shown concentration dependent effect. The maximum anti-inflammatory effect of the test extract achieved at 2000μg/ml. The test (JGLE, JGLC, JGSE) has been shown inhibition of paw edema induced by carrageenan at 50mg/kg body weight. The extract JGLE, JGLC, JGSE at 50mg/kg body weight and 100mg/kg body weight orally showed the significant (P< 0.05) and dose dependent inhibitory effect against FCA induced arthritis model. Diclofenac sodium 20mg/kg body weight orally is used as a standard. JGLE exhibit more significant and most promising anti-arthritic and anti-inflammatory effect than other extracts these effects support the traditional role of J. gendarussa in arthritis and other inflammatory condition.

Abstract Abstract In the treatment of cancer, patients that receive anti-cancer drugs such as Vincristine develop peripheral neuropathic pain. Scyphocephalione A is a new bioactive compound isolated from Scyphocephalium ochocoa (Myristicaceae), a medicinal plant traditionally used in African countries. Recently, an in vitro study has shown its anti-inflammatory and cytotoxic activities on MCF-7 cell line of mammary carcinoma. The purpose of the present study was to assess the in vitro anti-inflammatory and in vivo anti-nociceptive activities of Scyphocephalione A. In vitro tests were carried out on cyclooxygenase and 5-lipoxygenase activities, and on protein denaturation; while in vivo tests were performed on acute and chronic pain models. It was noticed that, Scyphocephalione A (1000 µg/ml), inhibits proteins denaturation, cyclooxygenase and 5-lipoxygenase activities respectively by 74.21%, 75.80% and 64.43%. The dose 50 mg/kg of Scyphocephalione A, inhibits acetic acid (63.43%, p<0.001) and formalin (42.12% (p<0.001) within first phase and 67.53% (p<0.001) within second phase)-induced pains. At the same dose, Scyphocephalione A significantly inhibited mechanical and heat hyperalgesia, as well as cold allodynia induced by vincristine. In addition, the compound restored haematological, biochemical and oxidative stress parameters which were altered following Vincristine administration. These results suggest that Scyphocephalione A is endowed with anti-inflammatory potential and antinociceptive properties. Therefore, Scyphocephalione A can be classified as a promising molecule for the management of peripheral neuropathic pain triggered by anti-cancer drug.

The present study was conducted to evaluate the phytochemical constituents, anti-inflammatory, antipyretic, thrombolytic and CNS modulatory activities of the methanol, chloroform and n-hexane extracts of Plumbago indica L. and Alpinia conchigera Griff. The plants are used in the treatment of inflammations and fever in traditional systems of medicine in Bangladesh. Qualitative tests revealed that the crude extracts of the plants contain reducing sugars, steroids, alkaloids and flavonoids. The anti-inflammatory activity was evaluated by means of protein denaturation and membrane stabilization assay. The methanol extract of P. indica and A. conchigera demonstrated inhibition of protein denaturation by 34.55 and 29.55% while their n-hexane soluble extract exhibited 46.87 and 37.21% inhibition of lysis of erythrocyte membrane, respectively which were considerable as compared to the standard drug acetyl salicylic acid. The methanol extract of P. indica and A. conchigera prominently reduced Brewer’s yeast-induced pyrexia which were close to the standard drug, paracetamol. A considerable thrombolytic activity was exhibited by the chloroform soluble extract of A. conchigera (48.27%) compared to the reference drug, streptokinase (72.13%) in red blood cell clot lysis test. The chloroform soluble materials of P. indica extract demonstrated noticeable reduction of CNS depression activity (63.52 % inhibition of locomotion), compared to reference drug diazepam (80.69 %) in open field experiment. The study justifies the medicinal applications of P. indica and A. conchigera in traditional systems and reveals the bioactivity of the plants which could be suitable for isolation and identification of the bioactive compounds.Dhaka Univ. Satureja cuneifolia Ten. (wild savoury) belongs to the Lamiaceae family and is used to produce essential oil and aromatic water. This plant is also used as a condiment and herbal tea due to its stimulating, tonic and carminative effects. The in vitro antioxidant, anti-urease, anticholinesterase and cytotoxic activities of the different extracts from the plant’s aerial parts were examined. Besides, the in vivo anti-inflammatory activities of the fraction and direct methanol extracts were determined comparatively. In the current study, fraction methanol extract exhibited the strongest ABTS (52.34 mM trolox/mg extract) radical scavenging and ferric reduction (17.22 mM Fe2+/mg extract) activity. It was also found that the fraction methanol extract had stronger anti-urease (12.52%) and anticholinesterase (69.02%) activity than other extracts.  The XTT results showed that fraction methanol extract had the most cytotoxic activity on MCF-7 cell lines (39.92%).  According to the results of in vivo anti-inflammatory activity, it was found that both fraction and direct methanol extracts exhibited close and significant anti-inflammatory activity. The fact that methanol extracts have significant biological activity suggests that these may be used as a natural source in the future.

Objective: This study aims to uncover the anti-diarrheal, antioxidant, thrombolytic, and anthelmintic activities of methanol extract of A. solanacea (ASME) and its soluble n-hexane fraction in methanol (ASNH).Materials and Methods: The phytochemical assessment of this plant was performed by using the standard method.
