Antimicrobial activity of Centella asiatica and Gigantochloa apus

Abstract Objectives Antibiotic treatments can create multi-drug resistance among several pathogens. There is a need for an antibiotic alternative to overcome this problem. In Indonesia, Centella asiatica (Asiatic pennywort) and Gigantochloa apus (string bamboo) are two common medicinal plants used to treat tuberculosis, diarrhea, and other symptoms. This study was done to compare the antimicrobial activity of C. asiatica and G. apus against five pathogenic bacteria, i.e., Mycobacterium tuberculosis H37Rv strain, Escherichia coli, Staphylococcus aureus, Bacillus subtilis, and Salmonella typhi. Methods The ethanol extracts of C. asiatica, and G. apus shoot were obtained by using speed extractor, pressure, and temperature extraction. The phytochemical contents of each extract were screened. The ethanol extract’s antimycobacterial activity was determined using Lowenstein Jensen (LJ) medium and antibacterial activity was determined using Kirby–Bauer methods on Mueller Hinton agar (MHA). Results The phytochemical analysis showed that G. apus extract contains alkaloids and tannins, whereas C. asiatica extract contains flavonoids, alkaloids, saponins, and tannins. This study showed that G. apus inhibited the growth of M. tuberculosis H37Rv strain and S. typhi. C. asiatica showed antimicrobial activity against all pathogenic bacteria tested, except B. subtilis. Conclusions Both medicinal plants extract can inhibit the growth of five pathogenic bacteria tested, thus, have the potential as an alternative treatment, or complementary, to treat the pathogenic bacterial infection.

Context: Plant based antimicrobial represent a vast untapped source for medicines and further exploration of plant antimicrobial neeto occur. Evolvulus alsinoides (L) (Convolvulaceae) is a perennial herb is used in traditional medicine in East Asia, India, Africa and Philippines to cure fever, cough, cold, venereal diseases, azoospermia, adenitis and dementia.   Objective: The objective of this research was to evaluate the antimicrobial activity of the extracts of E. alsinoides on some clinical microbial isolates.   Materials and Methods: The ed thanolic and aqueous extracts of the whole plant (leaves and twigs) were analysed for alkanoids, tannins, glycosides, steroids, flavonoids, saponins, volatile oil and resins. The determination of antibacterial activity was done using the agar well diffusion technique. Pure cultures of pathogenic bacteria such as Bacillus cereus, Staphylococcus aureus, Micrococcus leutus, Klebsiella Pneumoniae, Pseudomonas aeruginosa, Escherichia coli and Salmonella typhi were used for antibacterial activity assay, minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC).   Results: The ethanolic extract of the plant had MIC values ranging from 16 mg/ml to 512.5 mg/ml. The least MIC was 16mg-ml against Salmonella typhi while Bacillus cereus and Staphylococcus aureus showed the highest MIC of 512.5 mg-ml. In the aqueous extract the MIC ranged between 512.5 to >1025 mg/ml. Salmonella typhi, Micrococcus luteus and Staphylococcus aureus were not inhibited by the water extract. Phytochemical result showed ethanol to be a better solvent for the extraction of the bioactive agents in this plant which include: glycosides, alkaloids, saponins, tannins, flavonoids and volatile oil.   Conclusion: In this study the gram-negative organisms had the lowest MICs and MBCs. This suggests their higher susceptibility to the extract of this plant. On the basis of the result obtained in this investigation it can be concluded that ethanol extract of Evolvulus alsinoides had significant in vitro broad spectrum antimicrobial activity.   Keywords: Evolvulus alsinoides; Phytochemical screening; Antibacterial activity. Comparative study of plant extracts crude and aqueous, methanolic and ethanolic with antibiotics, provide evidence that calotropis procera extracts has the similar antibacterial activity as these antibiotics against test pathogens i.e. Salmonella typhi and E.coli. The analysis of antimicrobial activity of aqueous, methanolic and ethanolic extract of leaves and flower of Calotropis procera was carried out in disc method and also determined MIC value at 600nm through optical density using spectrophotometer. The zone of inhibition produced by extracts was examined and compares it with zone produced by antibiotics. The effect exhibited by ethanolic extract of leaves and flower was significantly greater than the aqueous and methanolic extract of leaves and flower. Crude extracts i.e. latex, leaves, fruit and flower crude extracts. Among them, flower crude extracts shows similar zone of inhibition to test anitibiotics.while in MIC value, we made different concentration of extracts and antibiotics.i.e for crude we made 25%, 50%.75% and 100% concentration of crude juice and for the aqueous, methanolic and ethanolic we made 0.5mg/mL,1mg/mL,2mg/mL,3mg/mL,4mg/mL, 5mg/mL and 6mg/mL in Dimethyl salfoxide, and same antibiotics concentration. Aqueous leaves extracts show MIC at 0.5mg/ml against E.coli while against Salmonella it shows MIC at 1mg/ml... We also determine phytochemical analysis for presences of different compounds in crude extracts. The obtained results provide a support for the use of Calotropis procera, in traditional medicine and suggest its further advance investigation.

Background: Plants in traditional healthcare services in West Africa were selected based on ethnobotanical data for this study. Aqueous and ethanol extracts from these plants’ parts were comparatively screened for phytochemicals and in vitro antimicrobial activity. Methods: The antimicrobial activity of five medicinal plants’ extracts (aqueous and ethanol) were evaluated against Proteus mirabilis (LHC201), Pseudomonas aeruginosa (LHC181) and Aspergillus fumigates (LUML56) using the agar-well diffusion protocol. Retailed chloramphenicol and griseofulvin were used as positive controls respectively. Phytochemicals and percentage yield were determined by modified standard methods. Results: The target bacteria showed varied degrees of susceptibility to both aqueous and ethanol extracts. A. fumigates was insensitive to the treatments. The ethanol extracts of the sampled plants’ parts showed better inhibitory performance against the target bacteria compared to aqueous extracts. Aqueous and ethanol extracts of Aframomum melegueta, Moringa oleifera and Cola nitida showed marginal difference in inhibitory activity with higher inhibition zones observed for the ethanol extracts of A. melegueta seed and M. oleifera pod against the target bacteria. Phytochemicals composition and density observed in extractants and plants’ parts also varied. Phenols were detected in both the aqueous and ethanolic extracts of C. nitida and C. acuminata, but appeared relatively richer in extracts of A. melegueta seeds and C. albidium fruits. C. nitida, C. acuminate and A. melegueta extracts were positive for flavonoids which were undetected in C. albidium fruits, M. oleifera seeds and pod extracts. No single extract had all the phytochemicals assayed. Conclusions: Screened extracts of medicinal plants’ parts used for this study showed promise antibacterial and resource for developing safer pharmaceutics. Optimization of the antibacterial potential of the extracts for commercial exploitation requires further studies. This study has provided information on the antibacterial property of C. albidum fruits which was hitherto underutilized for traditional medicine purpose.

India is found to be a country with rich biodiversity and enormous treasure of herbal plants and consequently called as medicinal garden of the world. Plants are the richest source of natural antimicrobial agents. In recent years drug resistance to human pathogenic bacteria has been commonly reported from all over the world. Therefore, there is a need to develop alternative antimicrobial drugs for the treatment of infectious diseases; one approach is to screen local medicinal plants for possible antimicrobial properties. The present study was designed to evaluate the antimicrobial efficacy of hexane, chloroform and methanol crude extracts of the leaves of three important medicinal plants viz., Biophytum sensitivum (L.) DC, Bougainvillea spectabilis L. and Caesalpinia bonducella (L.) Fleming, collected from in and around Visakhapatnam District. The antimicrobial activity of the crude extracts was tested against three Gram Positive bacteria (Bacillus subtilis MTCC 441, Enterococcus faecalis MTCC 439, Staphylococcus aureus MTCC 737), Three Gram Negative bacteria (Escherichia coli MTCC 443, Proteus vulgaris MTCC 426 and Pseudomonas aeruginosa MTCC 1688) and three Fungal strains (Candida albicans MTCC 227, Epidermophyton floccosum MTCC 613 and Trichophyton mentagrophytes MTCC 7687) using agar well diffusion assay. Our results demonstrated that methanol extracts of these plants leaves have concentration dependent antibacterial activity against some of the tested organisms. Further studies should be undertaken to elucidate the exact mechanism of action of antimicrobial effect to identify the active ingredients which can be used for drug development program.

The antibacterial activity of Tokar sha; a local traditional medication widely used by many people in North-west zone of Nigeria especially Sokoto, Kebbi and Zamfara against enteric infections were examined against some clinical isolates of pathogenic bacteria (Staphylococcus aureus, Escherichia coli, Bacillus cereus and Salmonella typhi) using agar well diffusion method. The pattern of inhibition varied with the tokar sha concentrations and the organisms tested. The tokar sha was more effective on E. coli with a maximum zone of growth inhibition of 25mm at 35mg/ml followed by B. cereus (20mm). However, S. aureus and S. typhi were resistant to tokar sha at all concentrations tested. The minimum inhibitory concentrations (MIC) were found to be 35mg/ml for both E. coli and B. cereus. The antibacterial activities exhibited by tokar sha in this study could be attributed to the presence of its constituents which signifies the potential of the tokar sha as a therapeutic agent. These findings may justify the ethnomedicinal use of tokar sha as an antibacterial agent against enterobacteria

AbstractCystic Echinococossis is a serious zoonotic parasitic infection caused by Echinococcus granulosus species complex. The current study was designed to evaluate the in-vitro antiprotoscolices effect of alcoholic extracts of three selected medicinal plants including Buxus Wallichiana, Berberis vulgaris and Euphorbia heliscopia against Echinococcus granulosus. Fertile hydatid cysts were collected from livestock and viability of the protoscolices was confirmed by 0.1% eosin red stain method. Protoscolices were subjected to three different concentrations of alcoholic extracts (10mg/ml, 30mg/ ml and 50mg/ml) for 10, 20 and 30 min. The highest efficacy was shown by B. vulgaris (97.92%) followed by B. wallichiana (65.98%) and E. heliscopia (61.22%) respectively, after exposure of 30 minutes at 50mg/ml concentration, that lead to the significant reduction in the viability of protoscolices. Alkaloids, flavonoids, tannins and saponnins were identified qualitatively and weighted quantitatively, that might help in the identification of bioactive compounds involved in selective action on the tegument layer of protoscolices. Alcoholic extracts of all the three selected medicinal plants showed toxic activities against protoscolices of Echinococcus granulosus. These findings suggest that all the selected medicinal plants could be a promising source of potent antiprotoscolices effect. However, the mechanism by which plant extracts killed protoscolices and also their safety for living cells are unclear and need to be investigated further.

The aim of the present study was to investigate phytochemical and antibacterial potential of Calendula officinals L. and Catharanthus roseus L. G. Don leaves. The leaves of both plants were screened for physico-chemical analysis. Phytochemical screening was carried out to find phytoconstituents and subsequent antibacterial studies of extracts were carried out against the pathogenic bacterial strains Bacillus cereus, Bacillus subtilis, Escherichia coli, Staphylococcus aureus and Salmonella typhi using Kirby bauer method. The extract with maximum antibacterial activity was further processed for TLC studies. The results of the present investigation revealed superiority of C. roseus over C. officinalis in terms of ash value. Both plants showed the presence of alkaloids, saponnins, tannins, phenolic compounds, flavonoids and carbohydrates. The antibacterial studies revealed that the petroleum ether extract of C. officinalis has been active against all strains, whereas the same extract of C. roseus showed activity against S. typhi, S. aureus and B. subtilis. Chloroform extract of C. officinalis was active against only two strains i.e. E. coli and B. subtilis. Methanolic and aqueous extracts of both plants were also found to be active against all strains. Studies of TLC analysis revealed that petroleum ether extract of C. officinalis and methanolic extract of C. roseus showed distinct RF values. Further studies are required to separate characterize and screen these components for therapeutic purposes.
