Modulatory effect of Polyalthia longifolia leaves against cadmium-induced oxidative stress and hepatotoxicity in rats

AbstractBackgroundCadmium serves as a major pollutant in the environment and it has been documented for its widespread harmful effects. This study sought to investigate the prophylactic and the curative effects of aqueous and methanolic leaf extracts of Polyalthia longifolia against cadmium-induced hepatotoxicity in rats.MethodsAnimals in group I served as the normal control and administered distilled water only for 14 days, group II was administered cadmium (4 mg/kg/body weight) for 7 days, groups III and IV rats served as the prophylactic group and were pre-treated with P. longifolia aqueous and methanolic leaf extract for 7 days and then exposed to cadmium for another 7 days, serving as pre-treatment group, groups V, VI, VII, and VIII served as curative groups and were first exposed to cadmium for 7 days and then post-treated with 100 and 200 mg/kg body weight of aqueous extract and 100 and 200 mg/kg body weight of methanolic extract P. longifolia for another 7 days.ResultsPre- and post-treatment with both extracts of P. longifolia revealed a significant hepatoprotective ability by decreasing the alanine transaminase, alkaline phosphatase, aspartate transaminase, acid phosphatase, gamma glutamyl transferase enzymatic activities were elevated due to cadmium intoxication. Pre- and post-treatment with aqueous and methanolic extract of P. longifolia extract significantly decreased hepatic malondialdehyde levels, together with an improvement in the antioxidant status of superoxide dismutase, glutathione peroxidase, catalase, glutathione S-transferase, and reduced glutathione of rats exposed to cadmium. Histopathology examinations also confirm the above biochemical findings.ConclusionThe findings from this work suggested that P. longifolia may be beneficial in ameliorating the cadmium-induced hepatotoxicity and oxidative stress in rats.

The objective of this study was to determine the effect of three monthly doses of 60 mg/kg/day PZQ on schistosome egg count, liver and renal function during the treatment of urinary schistosomiasis in Ghana. Methods A nested case-control study was designed from a cohort screened for schistosomiasis; 28 schistosomiasis positive cases by microscopy matched with 53 healthy controls by age and gender. The study population was urban dwellers from the Asokwa sub-metropolitan area, Kumasi in Ghana. Participants were within the age range of 6 to 30 years. We assessed Schistosoma haematobium egg counts in urine and its associated impact on liver and renal function at baseline, treatment and post-treatment phases using serum. Results Of the 28 cases and 53 controls, 78.6% and (81.1% were males respectively. Globulin levels before treatment was higher in cases [36.7 (32.8, 40.1) vrs 30.5 (22.4, 33.8) , p=0.005] at pre-treatment but not at post-treatment [35.8 (31.2, 39.1)vrs 37.4 (29.7, 43.0), p= 0.767]. Estimated cure rate was 42.9%, 46.4% and 96.4% after first, second and third dose respectively. Schistosome egg counts dropped significantly (p= 0.001) from before second dose to post-treatment. Similarly, levels of alanine aminotransferase (p=0.001), aspartate aminotransferase (p=0.028) and gamma glutamyl transferase (p=0.001) significantly declined towards post-treatment. Estimated glomerular filtration rate significantly improved from before second dose to post-treatment using both the Chronic Kidney Disease Epidemiology Program (p=0.001) and 4-variable Modification of Diet in Renal Disease (p=0.002) equations. Conclusion Treatment of urinary Schistosoma hematobium infections with a repeated high monthly dose of 60 mg/kg of praziquantel for 3 months is safe and effective.

Estimated cure rate was 42.9%, 46.4% and 96.4% after first, second and third dose respectively. Schistosome egg counts dropped significantly (p= 0.001) from before second dose to post-treatment. Similarly, levels of alanine aminotransferase (p=0.001), aspartate aminotransferase (p=0.028) and gamma glutamyl transferase (p=0.001) significantly declined towards post-treatment. Estimated glomerular filtration rate significantly improved from before second dose to post-treatment using both the Chronic Kidney Disease Epidemiology Program (p=0.001) and 4-variable Modification of Diet in Renal Disease (p=0.002) equations. Conclusion: Treatment of urinary Schistosoma hematobium infections with a repeated high monthly dose of 60 mg/kg of praziquantel for 3 months could be an effective and safe alternative for the current treatment option 40 mg/kg body weight.

Bicalutamide (BCT) is a potent anti-androgen chemotherapeutic drug indicated for prostate cancer. However, BCT is known to cause oxidative stress and impairment of male reproductive function. Whereas Morin (MOR), a flavonoid has been found to be a potent antioxidant, with free radical scavenging capacity. This study investigated the protective effect of MOR on BCT-induced testicular toxicity in Wistar rats. Twenty-four male albino rats were randomized into four groups (n=6/group). Group I which served as control received distilled water. Group II, received 3 mg/kg body weight (bwt) BCT orally (per os); group III received 3 mg/kg/day BCT p.o. plus 100 mg/kg/d MOR p.o. and group IV received 100 mg/kg/d MOR p.o. All treatments lasted for 14 days, thereafter, animals were sacrificed and epididymis and testis were collected for sperm and biochemical analyses. The result revealed that BCT treatment caused a significant increase in abnormal sperm morphology. Sperm production, sperm count, motility and viability were significantly reduced when compared with control (p<0.05). Similarly, testicular superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx),glutathione S-transferase (GST) and acid phosphatase (ACP) activities, as well as ascorbic acid and GSH levels were significantly reduced in the BCT- treated animals when compared to control (p<0.05). Conversely, testicular alkaline phosphatase (ALP), gamma glutamyl transferase (GGT) and lactate dehydrogenese (LDH) activities as well as malondialdehyde (MDA) levels of BCT-treated animals  increased significantly relative to control (p<0.05). However, co-treatment with Morin ameliorated BCT-induced alterations in sperm parameters, ascorbic acid, GSH and MDA levels, as well as LDH, SOD, CAT, GST, GPX, ACP, ALP and GGT activities. Data obtained from this study suggest that Morin protected against altered sperm parameters and testicular oxidative stress caused by BCT. This in vitro study analyses the biochemical interaction between saliva and three types of dental composite resins (a direct resin, an indirect resin and a dual-cure resin used for cementation of indirect dental restorations). The resin samples were obtained following a specific protocol and in line with the producers� recommendations; the resin samples were incubated with saliva samples collected from 19 healthy volunteers. The obtained results showed that the tested composite resins did not produce significant changes in oxidative stress parameters that were analysed (albumin, uric acid, GGT / gamma glutamyl transferase, OXSR-1 / oxidative stress responsive kinase 1) and do not influence the inflammatory salivary status reflected by the levels of IL-6 - an inflammatory marker.

The aim of this study was was to determine the nutritional requirements of digestible methionine+cystine (M+C) for white-egg layers aged one to six weeks. A completely randomized design with five methionine+cystine levels, six replicates, and 30 birds per experimental unit was adopted. Dietary treatments consisted of five diets supplemented with DL-Methionine with resulted in five levels of digestible methionine + cystine, 80% (0.516%), 90% (0.578%), 100% (0.640%), 110% (0.702%), and 120% (0.764%), based on Brazilian tables of nutritional requirements. Performance, serological blood, and histological data were evaluated. Feed intake, feed conversion, hepatic glycogen deposition, enzymatic activity of alanine aminotransferase and gamma-glutamyl transferase, and serum creatinine and albumin levels had showed a quadratic response to the levels of digestible M+C, with the respective requirements: 89.78% (0.575%), 114.33% (0.732%), 86.50% (0.554%), 100% (0.640%), 100.40% (0.643%), 104.30% (0.668%), and 111.88% (0.716%). Increasing levels of methionine+cystine elevated the relative liver weight and the deposition of hepatic glycogen, in addition to promote higher growth in pullets, with better body weight and body weight gain and feed conversion ratio. Our findings suggest that 0.732% digestible methionine+cystine is recommended, which corresponds to an intake of 151.20 mg/bird/d and a Met+Cys:Lys  ratio 83%, for light pullets from one to six weeks.

Erucaria pinnata (Viv.) is a wild annual plant growing in North-Western Coastal Region in Egypt. This study reports for the first time the cytotoxic activity of different extracts of Erucaria pinnata plant against HEP-G2 cell line. The 70% methanolic extract (E1) recorded the best potent cytotoxic activity (IC50=13.6 µg/ml), so we analysis the flavonoids constituent of this extract using HPLC, which show that our extract is rich with important flavonoids compounds (rutin, quercetin, leutolin, etc.). We evaluated its antitumor activity against hepatocellular carcinoma (HCC) induced by diethylnitrosamine (DEN) (200mg/Kg. b.wt., i.p, single dose) after two weeks, animals received carbontetrachloride (CCl4) (3ml/Kg. b.wt., SC, once a week for 6 weeks) and the experiment continued for 44 weeks in rats. After the experimental period, the administration of DEN/CCl4 showed significant increase in the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), total bilirubin, gamma-glutamyl transferase (γGT) and significant decrease in the levels of total proteins and albumin content in the serum with reduction in the liver antioxidants, including superoxide dismutase (SOD) and catalase (CAT). This was accompanied by increases in serum specific tumor markers (AFP). The 70% methanolic plant extract (E1) was orally administrated (400mg/kg/day respectively) for the whole study period, and it showed a significant improvement at the different biological liver functions, remodeled the antioxidant enzymes activity and down-regulated the serum AFP. All these findings were confirmed by histopathological studies of the liver samples obtained from all groups. In addition, we evaluated its antiangeogenic activity by docking study against VEGFR-2 tyrosine kinase after it showed an ability to inhibit the VEGFR-2 expression in vitro and inhibit the concentration of VEGF-A in vivo. The hepatoprotective effect of our extract was attributed to its antioxidant and antiangeogenic activity.

BACKGROUND: Mastitis is an inflammation of the breast tissue, usually caused by bacteria. Mastitis stimulates pro-inflammatory cytokines. The cytokine interleukin-1β (IL-1β) is a crucial mediator of the inflammatory response. This cytokine has adverse effects of hosting immunity that mediates resistance to pathogens and also exacerbates damage during chronic disease and acute tissue injury. Scaevola taccada (Gaertn.) Roxb. has been used as an ethnomedicine for healing sores in several provinces in Indonesia. AIM: This study aimed to assess the profile of pro-inflammatory cytokine IL-1β through the treatment effect of leaf extracts of S. taccada (Gaertn.) Roxb. as adjuvant for healing mastitis. METHODS: This study was a true control group experiment using the pre-test-post-test control design aimed to measure the effect of hydroalcoholic compounds in leaf extracts of S. taccada on the systemic pro-inflammatory activity of interleukin-1β (IL-1β). The treated animals were 18 mice of Sprague Dawley strain induced by Staphylococcus aureus. These treated mice were divided into three groups in which each group consisted of six mice. The mice in the Group I (negative control) were given 1 ml aquabides/250 g body weight, those in the Group II (positive control) were delivered with 9.6 mg/ml amoxicillin/250 g body weight, and those in the Group III (experimental) were given 9.6 mg amoxicillin/250 g body weight + 400 mg/ml leaf extracts of S. taccada/g body weight for 5 days, respectively. Pathological examinations were carried out from the inflamed tissues to prove the healing process of the treated mice. IL-1Β levels were determined using the enzyme-linked immunosorbent assay method. Data were analyzed using ANOVA and post hoc tests. RESULTS: There were statistically significant differences of IL-1β levels after the administration of leaf extracts of S. taccada among all the treated mice groups at p < 0.05. The Group III had the lowest IL-1β level with the mean value ± 1.45 pg/ml compared to the IL-1β level in the Group II (positive control) with the mean value ± 3.82 pg/ml and the IL-1β level in the Group I (negative control) with the with mean value ± 5.22 pg/ml. The pathological analysis of breast tissues of the treated mice proved that leaf extracts of S. taccada (Gaertn Roxb.) could reduce damaged tissues, cellular infiltration, and subcutaneous edema induced by this pathogenic microorganism. CONCLUSION: Leaf extracts of S. taccada had a significant function as adjuvant for healing mastitis by reducing pro-inflammatory cytokine IL-1β.

<p>Oxidative stress is related to pregnancy complications that could increase maternal and infant mortality. This study aimed to determine the effect of propolis extract supplementation during pregnancy on oxidative stress level and pregnancy outcomes utilizing Malonedealdehyde (MDA) and 8-Oxo-2′-Deoxogunosine (8-OHdG) levels, maternal body weight, and the average number of fetuses as the parameters. The study was conducted by using a posttest only control group design on 24 pregnant Wistar rats, which were divided into four groups. Group I was control, Group II-IV were the treatment groups given propolis extract of 1.8mg, 3.6mg, and 7.2mg/200gBW/day, respectively. The standard feed given was AIN93G dose of 20g/day and distilled water ad libitum. Propolis extract was given using a gastric feeding tube every morning for 20 days. At the end of the treatment, body weight was meisured and blood collected for assessed MDA and 8-OHdG levels  by ELISA method  and then we performed abdominal surgery to count number of fetuses. The result are there were decreasing level of MDA and 8-OHDG by administration of propolis significantly (p&lt;0.05) group: I: 2,04±0,091, II: 1,55±0,067, III: 1,05±0,176, IV: 0,73±0,075 (mmol/mL) (p=0.001); 8 OHdG level (ng/mL) group I: 10,02±0,403, II: 8,60±0,078, III: 7,89±0,051, IV: 7,53±0,063 (p=0,001). Average of maternal body weight (g) were increased: group I: 228,33±3,93, II: 237,17±4,36, III: 244,83±4,02, IV: 248,00±5,76 (p=0,001) and Average number of fetuses tend to increased as well, group I : 8,5±0,05, II: 7,8±0,41, III: 9,5±1,05, IV: 9,6±0,52 (p=0,02). The conclusion of this research are supplementation of propolis extract in pregnant rats can reduce oxidative stress and improve pregnancy outcomes.</p>

Aim: To determine and compare gamma glutamyl transferase (GGT) and fibrinogen among obese males with and without obstructive sleep apnea (OSA). Second objective was to investigate correlation between blood pressure and GGT. Methodology: Sixty-four obese males aged 20-45 years with BMI > 25kg/m2 were included by convenience sampling. The study was conducted, after obtaining ethical approval from IRB, at the Department of Physiology, Post Graduate Medical Institute, Lahore from August 2014 to May 2015. Participants having acute or chronic inflammatory conditions were excluded. Participants were screened for OSA by Berlin and STOP BANG questionnaires. Diagnosis of OSA was made by overnight portable pulse oximetry. The participants were divided into two groups. Group I had 32 obese males with OSA. Group II contained 32 obese males without OSA. After an overnight fasting of 10-12 hours blood samples were drawn. Serum fibrinogen and GGT were measured by spectrophotometer. The data was analyzed using SPSS-22. Quantitative variables were compared between the two groups by Mann-Whitney U test. Spearman correlation was used to correlate blood pressure and GGT among the participants. Results: Fibrinogen was significantly raised (p=0.015) in obese males with OSA. Systolic blood pressure (p=0.003), diastolic blood pressure (p=0.001) and mean arterial blood pressure (p<0.001) showed strong positive correlation with GGT in obese males with OSA. Conclusion: Proinflammatory, procoagulant and proatherogenic marker fibrinogen levels were significantly raised in obese otherwise healthy males with OSA. Oxidative stress marker GGT showed strong positive correlation with blood pressure in obese males with OSA.
