Evaluation of biological activities of Barbarea integrifolia and isolation of a new glucosinolate derivated compound

Abstract The aim of the present study is to determine the potent biological activities and carry out isolation studies on Barbarea integrifolia. The antioxidant capacity of the species was evaluated by total phenolic content, FRAP, CUPRAC, and DPPH radical scavenging activity. Anticancer activity studies were performed by MTT assay in MDA-MB-231, MCF-7, Hep3B, PC-3, A549, HCT116, L-929 cell lines. It was observed that the remaining aqueous fraction has higher total phenolic content while higher activity in the CUPRAC and FRAP assays was displayed for the methanolic extract and chloroform fraction. The extracts showed anticancer activity as compared with vincristine. It was observed that chloroform fraction has the highest anticancer activity on MCF-7 cell line, while ethyl acetate fraction has the highest anticancer activity on Hep-3B and A549 cell lines. Methanolic extract has the highest anticancer activity on HCT116 and MDA-MB-23 cell lines. The isolation studies have been performed using several chromatographic methods. The chemical structures of compounds have been identified by means of 1H NMR, 13C NMR, 2D-NMR, and MS. Five major compounds, one steroid (β-Sitosterol), one phenolic acid (Rosmarinic acid), one flavonol heteroside (kaempferol 7-O-α-l-rhamnoside-3-O-β-d-(2-O-β- d -glucosyl)-β-d-glucoside), and two glucosinolates (Gluconasturtiin, Gluconasturtiin choline salt) have been isolated.

The in vitro antiproliferative and antioxidant studies of the leaf extract and fractions of Conyza sumatrensis was investigated by applying the Sulforhodamine-B and 2, 2-diphenyl-1-picrylhydrazyl assays (DPPH-RSA) respectively. While the antiproliferative activity was carried out at 1-250 and 1-100 μg/ mL for the extract and fractions against breast (MCF-7) and lung (NCI-H460) cancer cell lines, the antioxidant study was conducted using DPPH at 31.25 -500 μg/ mL with the total phenolic and flavonoid contents calculated as well with reference to quercetin and gallic acid respectively. The extract and fractions were observed to elicit cytotoxic and growth inhibitory effects against breast (MCF-7) and lung cancer cell lines (NCI-H460) respectively. At 250 μg/mL, the extract of C. sumatrensis gave cytotoxicity of –1.76 ± 0.20 % against MCF-7 cell lines and inhibited growth of NCI-H460 at +94.40 ± 1.0 % respectively. While the chloroform fraction at 100 μg/mL gave -5.38 ± 0.33 % and 91 ± 1.61 % against MCF-7 and NCI-H460 cell lines, the aqueous fraction was observed to be inactive. For the DPPH-RSA activity, the chloroform fraction demonstrated an IC50 value of 125.5 μg/ mL compare to quercetin at 62.5 μg/ mL. The bioactivities were more pronounced in the chloroform fraction. This work has shown that C.  sumatrensis has antiproliferative and antioxidant activities which could be tied to the secondary metabolites present in the plant.

<p class="Default"><strong>Objective: </strong>The present work was aimed to investigate the <em>in vitro </em>antioxidant and anti-cancer activities of methanolic extract of <em>Stephania elegans</em>, an unexplored species from <em>Menispermaceae</em> family.</p><p class="Default"><strong>Methods: </strong>The methanolic extract of <em>S. elegans </em>tubers was prepared and phytochemical screening and total phenolic content were analyzed by using standard methods. <em>In </em><em>vitro,</em><em> </em>antioxidant potential of methanolic extract was determined by 2-2’-azinobis (3-ethyl-benzothiazoline-6-sulfonic acid) (ABTS) and ferric reducing antioxidant power (FRAP) assays. Cytotoxicity against human breast cancer cell line, Michigan Cancer Foundation-7 type (MCF-7) was evaluated by 3-(4, 5 dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide) (MTT) assay.</p><p class="Default"><strong>Results: </strong>Preliminary phytochemical screening revealed the presence of alkaloids, flavonoids, carbohydrates, tannins, terpenoids, steroids, and saponins in the methanolic extract of <em>S. elegans </em>tubers. The total phenolic content in the methanolic extract was 23.0±0.06 mg GAE/g (dry mass). As revealed by ABTS assay, the methanolic extract of plant tubers showed significant radical scavenging activity (IC<sub>50</sub> 41.66±0.015 μg/ml). The reducing power activity of the extract increased with the concentration of the extract. MTT assay indicated that <em>S. elegans </em>has potent cytotoxic activity towards MCF-7 cells (IC<sub>50 </sub>158.7±0.13 μg/ml).</p><p class="Default"><strong>Conclusion: </strong>This is the first study demonstrating the antioxidant and anticancer capabilities of the methanolic extract of <em>S. elegans</em>. This study also provides a significant basis for further isolation and characterization of bioactive compounds from <em>S. elegans</em>.</p>

Current treatments for complex diseases have remarkable side effects that negatively impact patients’ quality of life. Thus, natural compounds with fewer side effects represent a promising source for safe drugs. The genus Senecio is widely used in folk medicine due to its various pharmacological properties. In the present study, the total phenolic content of Senecio glaucus, which is grown in Saudi Arabia, was assessed using the Folin-Ciocalteau colorimetric method. Scavenging DPPH and ABTS assays were utilized to determine the antioxidant properties of S. glaucus fractions, and MTT assay was used to screen the cytotoxic activity of S. glaucus against various cancer cells. In addition, HPLC-UV was utilized to detect the presence of two phenolic acids, namely, vanillic acid (VA) and gallic acid (GA). Among all fractions tested, S. glaucus chloroform fraction (SGCF) yielded the highest value (125.3 mg·GA/g) in terms of total phenolic content. SGCF also exhibited the highest scavenging activities (76.7 and 74.1%) on both DPPH and ABTS assays, respectively. Similarly, SGCF also possessed the most potent cytotoxic activity against the MCF-7 cell line, with an IC50 value of 41.8 μg/ml. The validated HPLC method confirmed the presence of VA (4.8 μg/mg DW) and GA (3.9 μg/mg DW) in SGCF. Overall, our data show that S. glaucus had antioxidant and cytotoxic properties. A developed validated HPLC method which could be helpful for quantifying phenolic compounds in S. glaucus was established.

AbstractObjective: Scientists are looking for new resources which have biological activities. The present study was planned to evaluate the antioxidant and antimicrobial activity of ethanol and acetone extracts, as well as the volatile compounds of two freshwater green algal species Spirogyra spp. and Zygnema stellinum (Vaucher) C. Agardh belonging to Zygnemaphyceae (Conjugatophyceae) obtained from Ordu University Campus wetlands.Methods: The extracts were tested in vitro for their antimicrobial effects using disc diffusion method. Total phenolic content and the antioxidative activity according to FRAP and DPPH methods of the extracts were also determined. The secondary metabolites from the investigated extracts were identified using GC-MS.Results: The extracts dramatically inhibited almost all tested microorganisms. The antimicrobial activity of the acetone extract of the Zygnema against C. albicans was found as more higher than positive control Nystatin. In accordance with antimicrobial activity, the highest total phenolic content was also determined in the presence of the acetone extract of the Zygnema. Furthermore the highest FRAP value and the lowest EC50 (mg/mL) value were calculated for the same extract. The main components of the all consisted of dimethyl and diethyl ester of 1,2-benzenedicarboxylic acid. However, the abundance of these metabolites in the extracts was not associated with antimicrobial or antioxidant activity. Biological activities of these algal species could be attributed to chemicals such as 1-Pentadecene and 1-Tetradecene which were present in smaller amounts.Conclusion: Investigated algal species can be evaluated to use in biotechnological applications such as food industry and medicine.

Background: The investigation of total flavonoids and antioxidant activity of polar extracts of Corchorus depressus is the major aim of this study. As observed from ancient literatures and folkloric claims the plant Corchorus depressus worshipped by the married women of Odisha, India, in the rituals called as “Jama Jutia”, possesses different biological activities including antioxidant property. Methods: The diphenyl picryl hydrazine, hydroxyl radical and nitric oxide radical scavenging methods were performed for measurement of the antioxidant activity at different extracts. The flavonoid and phenolic content of the extracts were determined by using aluminium chloride and Folin-Ciocalteau’s reagent (FCR) methods respectively. Results: The results for estimation of total phenolic content (mg/ 100 g) expressed as gallic acid equivalent (GAE) and total flavonoid (mg/ 100 g) in weight of quercetin equivalent (QE) was highest in methanolic extract 78.46 and  21.2 respectively, followed by 18.18 mg/100 g in GAE and 1.80 mg/100 g in QE for aqueous extract. Conclusion: The methanolic extract of C. depressus at 100µg/ml showed highest DPPH, hydroxyl and nitric oxide radical scavenging activity and this activity may be attributed to the presence of saponins and flavonoids as detected in the extract.

Xylaria species associated with termite nests or soil have been considered rare species in nature and the few which have been reported upon have been found to act as a rich source of bioactive metabolites. This study evaluated 10 ethyl acetate extracts of five new Xylaria species associated with termite nests or soil for their antioxidant activity, and cytotoxicity against different cancer and normal cell lines. DPPH and ABTS radical scavenging activities of the extracts demonstrated strong capacity with low IC50 values. The highest observed activities belonged to X. vinacea SWUF18-2.3 having IC50 values of 0.194 ± 0.031 mg/mL for DPPH assay and 0.020 ± 0.004 mg/mL for ABTS assay. Total phenolic content ranged from 0.826 ± 0.123 to 3.629 ± 0.381 g GAE/g crude extract which correlated with antioxidant activities. The high total phenolic content could contribute to the high antioxidant activities. Cytotoxicity was recorded against A549, HepG2, HeLa and PNT2 and resulted in broad spectrum to specific activity depending on the cell lines. The highest activities were observed with X. subintraflava SWUF16-11.1 which resulted in 11.15 ± 0.32 to 13.17 ± 2.37% cell viability at a concentration of 100 µg/mL. Moreover, LC-MS fingerprints indicated over 61 peaks from all isolates. There were 18 identified and 43 unidentified compounds compared to mass databases. The identified compounds were from various groups of diterpenoids, diterpenes, cytochalasin, flavones, flavonoids, polyphenols, steroids and derivatives, triterpenoids and tropones. These results indicate that Xylaria spp. has abundant secondary metabolites that could be further explored for their therapeutic properties.
