AbstractThe effect of temperature, dose and storage period on the infectivity and development of Heterorhabditis megidis (strain NLH-E 87.3) infective juveniles (IJ) was studied in the laboratory. IJ were stored at 5, 10, 15 and 20°C for a period of up to 70 days (10 weeks). Every second week, mortality, infectivity, reproductive capacity, time to emergence and length of IJ were examined. Galleria mellonella larvae were infested with doses of one and 30 IJ. As the period of storage increased, the number of active nematodes, the infectivity and the reproduction of IJ decreased both at low and high temperatures. Time to first emergence and body length of IJ were significantly affected by time, storage temperature and inoculum level. Independent of the dose and storage periods, highest infectivity and optimal development were observed when IJ were stored at 10 and 15°C. Effets de la durée du stockage et de la température sur le pouvoir infestant, la reproduction et le développement d'Heterorhabditis megidis parasitant Galleria mellonella - Les effets de la température, de la dose d'inoculum et de la durée du stockage sur le pouvoir infestant et le développement des juvéniles infestants (JI) d'Heterorhabditis megidis ont été étudiés au laboratoire. Les JI ont été stockés à 5, 7, 10, 15 et 20°C pendant une période allant jusqu'à 70 jours (10 semaines). Toutes les 2 semaines ont été examinées: la mortalité, le pouvoir infestant, la capacité de reproduction, la durée de l'émergence et la longueur de ces JI. Les larves de Galleria mellonella ont été infestées à deux doses: un et 30 JI. Lorsque la durée du stockage augmente, le nombre de nématodes actifs, le pouvoir infestant et la reproduction des JI diminuent, aussi bien à basse qu'à haute température. La durée de la première émergence et la longueur du corps des JI ont été significativement affectées par la durée et la température du stockage, ainsi que par la valeur de l'inoculum. De façon indépendante de cette dernière valeur et de la durée de stockage, le pouvoir infestant la plus élevée et le développement optimal ont été observés lorsque les JI sont stockés à 10 et 15°C.

The electrical conductivity test has been used to evaluate seed vigour in many species, especially legumes, such as peas, beans and soybeans. The objective of this research was to evaluate the effect of temperature and storage period on the results of the electrical conductivity test and the mineral composition of the imbibition solution of bean seeds (Phaseolus vulgaris L.). Three seed lots of the cultivar IAPAR - 81 and cultivar IPR - TIZIU were stored at 10, 25 and 25-10º C (six months at 10º C and six months at 25º C) The following evaluations were made every three months for a year: seed water content, germination and vigour (accelerated ageing and electrical conductivity) and the potassium, calcium and magnesium contents of the imbibition solution. The cultivars showed similar behavior in storage. The reduction in the vigour of bean seeds stored at 10º C was verified by accelerated ageing and electrical conductivity tests. The bean seed storage at low temperature (10º C) does not influence the results of the evaluation of seed vigour by electrical conductivity test and the quantification of calcium, magnesium and potassium ions. The evaluation of vigour by the electrical conductivity test is not recommended for bean seed stored by long periods (above 9 months).

Abstract Pekmez (molasses) is a traditional food commonly produced from grape and other kind of fruit juices by evaporation processes. In this study, 5-Hydroxymethylfurfural (HMF) level of various pekmez samples was investigated during storage at different temperatures. HMF content of apricot, mulberry, carob, grape, Juniperus communis pekmez changed from 133.0 ppm to 1060.5 ppm, from 88.2 to 1921.5 ppm, from 11.1 to 1153.6 ppm, from 75.5 to 2077.0 ppm, from 19.9 to 280.1 ppm throughout eight months storage period, respectively. Samples of pekmez from the Juniperus communis had the minimum k values for each temperature that means HMF formation in these samples were slower than other pekmez types. The kinetic data analysis for HMF formation during storage was performed and an Arrhenius equation was used to determine the effect of temperature on reaction kinetics of 5-HMF formation in pekmez samples. Ea values were found between 10.58–37.73 (kcal/mol). Apricot pekmez was found as the least sensitive sample to HMF formation resulted from temperature changes.

The aim of this work is to study the effect of temperature fluctuations on spoilage microbial flora behaviour of a semi-preserved seafood product in modified atmosphere packaging (MAP) as well as to find correct interpretation criteria for simulating temperature fluctuations during storage tests. The study concerned 54 packages of “Octopus carpaccio” that were grouped in three batches and stored at 3 different temperature profiles: the first (16 packages - Group 4°C) was stored at 4±0.5°C; the second (16 packages - Group 8°C) was stored at 8±0.5°C; the third (16 packages - Group F) was stored under a fluctuating temperature regime between 2°C and 14°C. Spoilage microflora, pH and AW has been monitored, at regular intervals, along the storage period (44 days). A predictive model was constructed according to the accredited scientific literature and validated against the observed growth curves of the above three groups. Afterwards, the predictive model has been used setting the temperature at the mean value of fluctuations (6.72°C), at the kinetic mean value of fluctuations (7.80°C) and at the 75th percentile value of fluctuations (11.14°C). The best fitting to the observed data was obtained with the kinetic mean temperature value and this result shows that this parameter can be proposed to reproduce the temperature fluctuation along the distribution and the domestic storage when a storage test has to be carried out.

Dried fig is susceptible to infection by Aspergillus flavus, the major producer of the carcinogenic mycotoxins. This fruit may be contaminated by the fungus throughout the entire chain production, especially during natural sun-drying, post-harvest, industrial processing, storage, and fruit retailing. Correct management of such critical stages is necessary to prevent mould growth and mycotoxin accumulation, with temperature being one of the main factors associated with these problems. The effect of different temperatures (5, 16, 25, 30, and 37 °C) related to dried-fig processing on growth, one of the regulatory genes of aflatoxin pathway (aflR) and mycotoxin production by A. flavus, was assessed. Firstly, growth and aflatoxin production of 11 A. flavus strains were checked before selecting two strains (M30 and M144) for in-depth studies. Findings showed that there were enormous differences in aflatoxin amounts and related-gene expression between the two selected strains. Based on the results, mild temperatures, and changes in temperature during drying and storage of dried figs should be avoided. Drying should be conducted at temperatures >30 °C and close to 37 °C, while industry processing, storage, and retailing of dried figs are advisable to perform at refrigeration temperatures (<10 °C) to avoid mycotoxin production.

The effect of testing temperature and storage period on the polarization fatigue properties of poly (vinylidene fluoride-trifluoroethylene) (P(VDF–TrFE)) ultrathin film devices were investigated. The experimental results show that, even after stored in air for 150 days, the relative remanent polarization (Pr/Pr(0)) of P(VDF–TrFE) of ultrathin films can keep at a relatively high level of 0.80 at 25 °C and 0.70 at 60 °C. To account for this result, a hydrogen fluoride (HF) formation inhibition mechanism was proposed, which correlated the testing temperature and the storage period with the microstructure of P(VDF–TrFE) molecular chain. Moreover, a theoretical model was constructed to describe the polarization fatigue evolution of P(VDF–TrFE) samples.

Age gelation is a major quality defect in ultra-high-temperature (UHT) pasteurized milk during extended storage. Changes in plasmin (PL)-induced sedimentation were investigated during storage (23 °C and 37 °C, four weeks) of UHT skim milk treated with PL (2.5, 10, and 15 U/L). The increase in particle size and broadening of the particle size distribution of samples during storage were dependent on the PL concentration, storage period, and storage temperature. Sediment analysis indicated that elevated storage temperature accelerated protein sedimentation. The initial PL concentration was positively correlated with the amount of protein sediment in samples stored at 23 °C for four weeks (r = 0.615; p < 0.01), whereas this correlation was negative in samples stored at 37 °C for the same time (r = −0.358; p < 0.01) due to extensive proteolysis. SDS-PAGE revealed that whey proteins remained soluble over storage at 23 °C for four weeks, but they mostly disappeared from the soluble phase of PL-added samples after two weeks’ storage at 37 °C. Transmission electron micrographs of PL-containing UHT skim milk during storage at different temperatures supported the trend of sediment analysis well. Based on the Fourier transform infrared spectra of UHT skim milk stored at 23 °C for three weeks, PL-induced particle size enlargement was due to protein aggregation and the formation of intermolecular β-sheet structures, which contributed to casein destabilization, leading to sediment formation.
