Among laboratory researches in animals the most widespread is the common blood test that can determine hidden changes in organs and tissues, monitor the effectiveness of therapeutic and preventive measures to predict outcome of disease. The question of the legality comparing the results of blood analysis from central veins with rates that are developed from capillary and peripheral vessels blood is studied in the article. Researches were conducted in seven pigs of ukrainian white breed in age 2–4 months. Blood was collected in the morning before feeding by puncture of ear vein, orbital sinus and cranial vena cava. From each animal were picked out 3 blood samples. For taking blood were applied the vacuum blood collection systems. In blood were determined the number of erythrocytes, leukocytes and hemoglobin by conventional methods (Kibkalo et al., 2016). From the results of leukocyte profile the major differences were found in the number of eosinophils whose content was significantly higher (r≤ 0.001) in the blood of ear veins. Should be noted that this index was much higher than the norm, which is possible associated with the pathological process. But their level in blood of the cranial vena cava and eye sinus was normal. Therefore, this question needs further study. Also found significantly lower (r ≤ 0.01) level of lymphocytes in the blood from the ear vein. In the vena cava were not found monocytes and basophils unlike eye sinus and ear veins, in the last their detected more. Based on the foregoing, it can be noted that in the blood of peripheral veins are registered larger number of granulocytes, which in future will be tissue macrophages. Neutrophils, monocytes, basophils and eosinophils have the ability to attach to the capillaries and small vessels walls. So they are providing the cellular immunity. In blood of central vessels are more lymphocytes that provides humoral immunity in the bloodstream. Perspectives of the next studies will be comparing the hematological results that are realized on automatic hematology analyzer from ear vein, eye sinus and cranial vena cava from the same piglets.

A frequently debated question for studies involving the measurement of stress hormones in rodents is the optimal method for collecting blood with minimal stress to the animal. Some investigators prefer the implantation of indwelling catheters to allow for frequent sampling. Others argue that the implantation of a catheter creates a chronic stress to the animal that confounds stress hormone measures and therefore rely on tail vein sampling. Moreover, some investigators measure hormones in trunk blood samples obtained after anesthesia, a practice that may itself raise hormone levels. To address these controversies, we 1) compared plasma ACTH and corticosterone (Cort) concentrations in pre- and poststress rat blood samples obtained via previously implanted vena cava catheters, tail vein nicks, or clipping the tip off the tail and 2) compared plasma ACTH and Cort in rat blood samples obtained by decapitation with and without anesthesia. Rats sampled via indwelling catheters displayed lower prestress ACTH levels than those sampled by tail vein nick if the time to acquire samples was not limited; however, elevated basal ACTH was not observed in samples obtained by tail clip or tail nick when the samples were obtained within 3 min. Baseline Cort levels were similar in all groups. After restraint stress, the profile of the plasma ACTH and Cort responses was not affected by sampling method. Decapitation with prior administration of CO2 or pentobarbital sodium increased plasma ACTH levels ∼13- and 2-fold, respectively, when compared with decapitation without anesthesia. These data indicate that tail vein nicking, tail clipping, or indwelling venous catheters can be used for obtaining plasma for ACTH and Cort during acute stress studies without confounding the measurements. However, the elevation in basal ACTH seen in the tail vein nick group at baseline suggests that sampling needs to be completed rapidly (<3 min) to avoid the initiation of the pituitary stress response. Death by CO2 and pentobarbital sodium injection before trunk blood collection cause significant stress to animals, as reflected in the elevated plasma ACTH levels. These results support the use of either chronic vascular cannulas or sampling from a tail vein. However, collection of blood under pentobarbital sodium or CO2 anesthesia is likely to confound the results of stress studies when ACTH is an important endpoint.

Blood collection methods in guinea pigs are limited due to the animals' compact neck, short limbs, and lack of a tail. Gingival venipuncture is a recently described blood sampling technique that is minimally traumatic with no significant alterations in hematologic parameters when multiple blood samples were collected weekly for 6 wk. The purpose of this study was to determine whether the gingival vein can be used as an alternative blood collection site in guinea pigs, such that: (1) hematologic parameters would be consistent with samples collected from the cranial vena cava; and (2) no contaminants from the oral cavity would be introduced into the sample. Blood samples were obtained from both the gingival vein and cranial vena cava of anesthetized Dunkin Hartley guinea pigs for CBC (n = 9) and aerobic blood cultures (n = 10). Only MCV was significantly different between sampling sites. Bland–Altman analyses calculated a small mean bias for all hematologic parameters, indicating clinical interpretation is unlikely to be affected by the sampling site. Bacterial growth occurred in all 5 gingival vein blood samples prepared by using saline and 2 of the 5 prepared with dilute chlorhexidine. Bacteria did not grow from any cranial vena caval blood samples prepared with dilute chlorhexidine. No clinical signs of hemorrhage or trauma were detected at either site. These results provide evidence that gingival venipuncture can be used as an alternative blood collection method for guinea pigs for hematologic analysis but should not be used for blood culture.

Abstract. To determine optimal conditions for blood collection during clinical trials, where sample handling logistics might preclude prompt separation of erythrocytes from plasma, healthy subjects (n=8, 6 M/2F) were recruited and non-fasting blood samples were collected into tubes containing different anticoagulants (ethylenediaminetetra-acetic acid (EDTA), Li-heparin or Na-heparin). We hypothesized that heparin, but not EDTA, would effectively protect plasma tocopherols, ascorbic acid, and vitamin E catabolites (α- and γ-CEHC) from oxidative damage. To test this hypothesis, one set of tubes was processed immediately and plasma samples were stored at −80°C, while the other set was stored at 4°C and processed the following morning (~30 hours) and analyzed, or the samples were analyzed after 6 months of storage. Plasma ascorbic acid, as measured using HPLC with electrochemical detection (LC-ECD) decreased by 75% with overnight storage using EDTA as an anticoagulant, but was unchanged when heparin was used. Neither time prior to processing, nor anticoagulant, had any significant effects upon plasma α- or γ-tocopherols or α- or γ-CEHC concentrations. α- and γ-tocopherol concentrations remained unchanged after 6 months of storage at −80°C, when measured using either LC-ECD or LC/mass spectrometry. Thus, refrigeration of whole blood at 4°C overnight does not change plasma α- or γ-tocopherol concentrations or their catabolites. Ascorbic acid is unstable in whole blood when EDTA is used as an anticoagulant, but when whole blood is collected with heparin, it can be stored overnight and subsequently processed.

SummaryWe have investigated the effects of 1-desamino-8-D-arginine vasopressin (DDAVP) infusion on platelet count and bleeding time in 4 patients with type IIB von Willebrand’s disease (vWd). Three of four patients showed a normalization of the bleeding time within 1 h after the infusion, while bleeding time was not modified in the fourth. In accordance with the literature, thrombocytopenia was observed after DDAVP infusion, but this thrombocytopenia was due to the anticoagulants used for blood collection. In two patients (F. I., G. F.) no thrombocytopenia was observed when platelets were counted by fingerstick method but there was a 20% platelet decrease in blood samples collected in sodium citrate and a 50% decrease in samples collected in EDTA. Dramatic falls in platelet counts (70–95%) were observed in the additional two patients (C. A., D.Z.) after DDAVP infusion, when both sodium citrate or EDTA were used as anticoagulants. In the latter two patients there was also a 50% decrease in platelet count when the fingerstick method was used. The decrease in the patient’s platelet count in EDTA samples after DDAVP infusion could be prevented, in part, by the previous additions of an anti GPIb monoclonal antibody and an anti GPIIb-IIIa monoclonal antibody.Thus, the thrombocytopenia observed in the four IIB vWd patients studied after DDAVP infusion seems to be, at least partially, a pseudothrombocytopenia depending on the calcium concentration in the blood samples and the availability of GPIb and GPIIb-IIIa receptors. These findings and the normalization of the bleeding time observed in three of the four patients has led us to reconsider the possible use of DDAVP in the treatment of our IIB vWd patients.

Aim: The aim of the study is to determine the factors that may affect the demands of patients who come with the request to have a blood test without any chronic disease or a planned examination check. Methods: The data of this descriptive, cross-sectional study, were collected with a questionnaire that was prepared to examine the opinions of the patients who claim to have a blood test by coming to the family health center without any complaints. Patients over 18 years of age, who did not have any chronic disease and had no scheduled examination appointments were included in the study. Results: A total of 278 patients who wanted to have a blood test within the 6-months period were included in the study. Female patients who wanted to have a blood test were significantly more than male patients. When we look at the causes of patients who wanted to have a blood test; 61.2% (n=170) patients stated that they are concerned about their health and 6.1% (n=17) stated that they were affected by media warnings. There was no significant relationship between the frequency of blood test requests of patients and their age, gender, education, and general health status. Conclusion: Patients with high expectations and anxiety may be more willing to perform blood tests at inappropriate intervals. Family physicians, whose primary role is preventive medicine, have consultancy and information duties in order to protect their patients from the risk of over-examination and diagnosis. ABSTRACT We have described a novel technique for collecting pituitary venous effluent in the horse by placing a cannula in the intercavernous sinus close to the outlet of the pituitary veins using a venous pathway unique to equids. Cannula placement and blood collection are carried out painlessly in fully conscious, ambulatory, unstressed animals. There is no interference to hypothalamic, pituitary or target organ function. The blood collected contains readily measurable concentrations of gonadotrophin-releasing hormone, and LH concentrations which can be up to 40 times those in concurrent peripheral blood samples. Four millilitre blood samples, a quantity which permits simultaneous measurement of many hypothalamic and pituitary hormones, can be collected at 2-min intervals for several days. Intercavernous sinus blood flow can be calculated allowing secretion rates of hypothalamic and pituitary hormones to be determined for any time-period. This model is uniquely useful for investigating the normal functional characteristics of several neuroendocrine and endocrine systems. J. Endocr. (1987) 113, 183–192

Sodium bicarbonate and other alkalinising solutions (‘milkshakes’) have been given to horses surreptitiously before exercise to provide exogenous buffering effects. After an initial positive blood test, some accused horse trainers claim that their horses ‘naturally test high’, so some jurisdictions allow a secured quarantine in which the horse is tested multiple times. The objective of this experiment was to determine the intra- and inter-day variability of plasma total CO2 (tCO2) and other plasma strong ions in a group of sedentary horses housed similarly to a quarantine period. The hypothesis was that plasma tCO2 would not remain constant over a multi-day monitoring interval, but would vary measurably during that interval. Eight sedentary (unconditioned) horses were studied for 2 weeks. Horses were acclimated to a climate-controlled indoor environment and an alfalfa-only diet for a minimum of 10 days prior to sampling. Horses were sampled 3 times daily for 5 consecutive days at 7:00, 11:00 and 15:00 h. Blood samples were collected directly into 10 ml heparinised evacuated glass tubes by jugular venipuncture using a double-ended 0.91 mm needle. Samples were chilled until concentrations of plasma tCO2, Na+, K+, and Cl-, were determined within 1-3 h of sampling using an automated serum chemistry analyzer which was calibrated daily using commercial reagents obtained from the manufacturer as well as externally-obtained NIST-traceable calibrating solutions. Mean results documented mild variations in mean plasma tCO2 (range 28.9-31.6 mmol/l), but individual horses’ plasma tCO2 ranged over 4-7 units. Results showed that there was considerable intra- and inter-individual variability in plasma tCO2. Mean pooled tCO2 and measured strong ion difference (SIDm) differed by time-of-day, with both late morning and early afternoon values lower than early morning values (P<0.001). There was a strong positive linear relationship between plasma SIDm and tCO2 (r=0.75, P<0.001).

The values of haematological and selected blood plasma biochemical parameters of juvenile common carp (Cyprinus carpio Linnaeus, 1758) were compared between blood samples taken from caudal vein and heart to evaluate the influence of blood sampling body site on the obtained results in two groups of fish of different blood sampling order: I – first by caudal and then by cardiac puncture, II – first by cardiac and then by caudal puncture. The obtained results revealed statistically significant (p<0.05) differences only in group I where red blood cell (RBC) count was higher in caudal vein blood, while haematocrit (Ht) value, mean corpuscular volume (MCV), mean corpuscular haemoglobin (MCH), total protein (TP) concentration, and magnesium (Mg) level were higher in cardiac blood samples. No statistically significant differences occurred in white blood cell (WBC) count, differential leukocyte count or erythrocyte morphology based on stained blood smears. The obtained results showed that blood sampling body site may affect the results of haematological and plasma biochemical analyses.

The bacteremia level as well as the administration of antibiotics before blood collection may significantly affect the recovery of bacterial pathogens from pediatric blood cultures in BacT/Alert Virtuo or Bactec FX BC systems, which remain the common techniques to diagnose bacteremia in pediatric patients. We simulated pediatric blood cultures with low or intermediate bacteremia level to evaluate BacT/Alert PF Plus and Bactec Peds Plus blood culture bottles for resin-based inactivation of 16 antibiotic–bacterium combinations. Overall, 105/192 (54.7%) of BacT/Alert PF Plus bottles and 69/192 (36.0%) of Bactec Peds Plus bottles allowed organisms to grow when exposed to antibiotics. In particular, both BacT/Alert PF Plus and Bactec Peds Plus bottles proved to be effective with piperacillin/tazobactam and Pseudomonas aeruginosa or with oxacillin and methicillin-susceptible Staphylococcus aureus (100% growth), whereas no effectiveness was apparent with ceftriaxone and Escherichia coli, Streptococcus agalactiae, or Streptococcus pneumoniae or with cefepime and E. coli (0% growth). In some relevant instances (e.g., with vancomycin and methicillin-resistant S. aureus or Streptococcus pneumoniae), BacT/Alert PF Plus bottles were superior to Bactec Peds Plus bottles. Together, these findings underscore the potentiality of resin-containing bottles to enhance diagnosis of bacteremia in pediatric patients on antimicrobial therapy. This is particularly true with one of the evaluated BC systems and with simulated intermediate bacteremia level only.
