The aim of this study was to investigate the hydrolysis conditionsof virgin coconut oil (VCO) by immobilized lipase (Lypozyme TL IM). Then the hydrolyzed products wereevaluated with their antibacterial activityagainst Gram – negative and Gram – positive bacteria (Escherichia coli (ATCC 25922), Salmonella enteritidis (ATCC 13076), Staphylococcus aureus (ATCC 25923) and Bacillus cereus (ATCC 25924)).The effect of four factors on hydrolysis degree of VCO was investigated: the ratio of VCO to buffer, the ratio of enzyme to substrate, pH condition, and temperature.The best reaction conditions in the case of using Lypozyme TL IM as the catalyst was determined: the ratio of VCO to buffer of 1.3 (g/g); the ratio of enzyme to substrate of 0.5 (%),  pH of 8.0 and temperature of 65 oC, VCO was hydrolyzed in 28 hours, acid value achieved to 188.05 (mg/g). The products of hydrolyzed VCO (HVCO) showed activity against Salmonella enteritidis (ATCC 13076) at the concentrations of 5 % and against Escherichia coli (ATCC 25922), Staphylococcus aureus (ATCC 25923) and Bacillus cereus (ATCC 25924) at the concentrations of 20 %.

ABSTRACTObjective: The purpose of this study was to evaluate the antibacterial activity of chitosan, hydrolyzed coconut oil and their combination againstBacillus cereus and Escherichia coli.Methods: The materials used in this study were powder of chitosan (obtained from prawn shell produced by Laboratory of Research Centre FMIPAUniversity of Sumatera Utara) and virgin coconut oil (VCO) product of Siti Nurbaya-Indonesia. VCO was partially hydrolyzed by Lipozyme TL IM(active at sn-1,3 position) and the result called hydrolyzed virgin coconut oil (HVCO). The bacteria used in this study were B. cereus and E. coli. Theantibacterial activity of chitosan in 1% acetic acid and HVCO in dimethylsulfoxide was tested by Kirby–Bauer agar diffusion method using paper discwith diameter of 6 mm.Results: The results showed that the minimum inhibitory concentration of chitosan against B. cereus and E. coli is at concentration of 0.05% withinhibition zone diameter of 6.86 mm and 7.56 mm, respectively. MIC of HVCO against B. cereus is at concentration of 0.25% with inhibition zonediameter of 6.40 mm, and against E. coli is at a concentration of 0.50% with inhibition zone diameter of 6.20 mm. The inhibition zone diameter ofchitosan 0.05% and HVCO 0.25% in combination against B. cereus is 8.33 mm which is higher than half the sum of chitosan 0.05% and HVCO 0.25%(6.63 mm). The inhibition zone diameter of chitosan 0.05% and 0.5% HVCO in combination against E. coli is 8.53 mm which is higher than half thesum of chitosan 0.05% and HVCO 0.5% (6.53 mm).Conclusion: The findings of this study indicate that chitosan is more antibacterial than HVCO, and the interaction between chitosan and HVCO incombination demonstrated to be synergistic against B. cereus and E. coli.Keywords: Antibacterial, Chitosan, Coconut oil, Combination, Bacillus cereus, Escherichia coli.®

Introdução: A participação em ensaios de proficiência (EP) é utilizada para avaliar a competência de laboratórios de ensaio e calibração. Objetivo: Avaliar a viabilidade da técnica de liofilização na produção de seis lotes de itens de EP, dois contendo Escherichia coli, dois contendo Bacillus cereus e Staphylococcus aureus concomitantemente, e dois contendo Salmonella Enteritidis em matriz frango desfiado cozido. Método: Foram realizados testes de homogeneidade segundo o protocolo harmonizado e de estabilidade em longo prazo pelo modelo clássico e curto prazo pelo modelo isócrono segundo a ISO Guide 35. Resultados: Todos os lotes produzidos foram considerados suficientemente homogêneos. No estudo de estabilidade em longo prazo, todos os lotes se apresentaram suficientemente estáveis nas temperaturas de -80 ± 10ºC e -20 ± 4ºC, exceto o lote contendo B. cereus e S. aureus. Os outros lotes apresentaram estabilidade por pelo menos 126 dias a -80 ± 10ºC e 84 dias a -20 ± 4ºC. Na avaliação da estabilidade em curto prazo, foram analisados apenas os lotes suficientemente estáveis no estudo em longo prazo. Os lotes foram suficientemente estáveis nas temperaturas de 5 ± 3ºC e 35 ± 2ºC, com exceção do lote contendo Salmonella Enteritidis a 35 ± 2ºC, devido ao decréscimo significativo da concentração celular. Conclusões: A técnica de liofilização foi satisfatória para produção de itens de ensaio contendo E. coli e Salmonella Enteritidis em matriz frango viáveis para utilização em um EP, sendo que o lote contendo Salmonella Enteritidis deve ser transportado aos laboratórios participantes em temperatura ≤ 8ºC por até quatro dias. Lotes contendo S. aureus e B. cereus, simultaneamente, apresentaram estabilidade insuficiente, indicando que a produção de lotes individuais contendo cada bactéria individualmente é necessária.

Members of the genus Centaurea (Asteraceae) have been used in traditional plant-based medicine. The methanol extracts of twelve Centaurea species, of which five are endemic to Turkey flora, were screened for antibacterial activity against four bacteria (Escherichia coli, Bacillus cereus, Salmonella enteritidis, Staphylococcus aureus). The antibacterial activity was evaluated by the microdilution method and the minimum inhibition concentrations (MIC) of the extracts were determined. C. cariensis subsp. microlepis exhibited an antimicrobial effect on all tested microorganisms. The extracts from eight Centaurea species (C. balsamita, C. calolepis, C. cariensis subsp. maculiceps, C. cariensis subsp. microlepis, C. kotschyi var. kotschyi, C. solstitialis subsp. solstitialis, C. urvillei subsp. urvillei and C. virgata) possessed antibacterial activity against several of the tested microorganisms.

Abstract The antibacterial activity of coumarin per se and other 45 coumarin derivatives was tested against strains of Bacillus cereus MIP 96016, Escherichia coli ATCC 25922, Pseudomonas aeruginosa ATCC 27853, and Staphylococcus aureus ATCC 25923. The inhibitory effects of coumarins were affected by their substitution patterns. Osthenol (44) showed the most effective antibacterial activity against Gram-positive bacteria with MIC values ranging between 125 and 62.5 μg/ml. These results suggested that the prenyl chain of 44 at position 8 and the presence of OH at position 7 of the benzenic ring are required for the antibacterial activity against these strains.

Celem niniejszej pracy było określenie poziomu wybranych mikroorganizmów, porównanie zawartości polifenoli i antocyjanów oraz zdolności przeciwutleniającej napojów kombucha dostępnych w handlu detalicznym z napojem wytworzonym doświadczalnie metodą tradycyjną. Badania objęły 12 napojów komercyjnych oraz jeden uzyskany doświadczalnie, którego bazą była zielona herbata z 10-procentowym dodatkiem sacharozy. W badanych produktach oznaczono: liczbę bakterii Acetobacter sp., Gluconobacter sp., LAB i drożdży, pH, ogólną zawartość antocyjanów, ogólną zawartość polifenoli oraz aktywność przeciwutleniającą. Aktywność przeciwdrobnoustrojową napojów kombucha oceniono metodą dyfuzyjną, z zastosowaniem kolumienek, w stosunku do szczepów wzorcowych: Escherichia coli ATCC 35218, Listeria monocytogenes serotyp 1/2a (KMSiFŻC), Yersinia entercolitica (PCM 1919), Salmonella enteritidis (ATCC 130764), Bacillus cereus (PCM 2019), Enterococcus faecalis (ATTC 29212), Staphylococcus aureus (ATTC 25923), Saccharomyces cerevisiae (ŁOCK 14), Candida albicans (KMSiFŻC). Stwierdzono, że napoje deklarowane jako pasteryzowane (K7 - K12) były pozbawione mikroorganizmów z analizowanych grup. W pozostałych produktach komercyjnych dominującym mikroorganizmem były bakterie Acetobacter sp., których liczba zawierała się w przedziale 6 × 106 ÷ 1,1 × 107 jtk/ml. W napoju doświadczalnym wykazano drożdże w ilości 6,5 × 105 jtk/ml. Odczyn pH produktów był charakterystyczny i nie przekraczał 4,3. Zawartość antocyjanów we wszystkich napojach pochodzących z handlu wahała się między 0,2 ÷ 0,3 mg/dl, natomiast w próbie doświadczalnej było ich ponad dwa razy więcej. Zawartość polifenoli oraz zdolność przeciwutleniająca prób były zróżnicowane i nie stwierdzono ich korelacji z pochodzeniem produktu. Głównym czynnikiem warunkującym zdolność napojów kombucha do inhibicji bakterii potencjalnie chorobotwórczych dla człowieka jest obecność kwasów organicznych. Żaden z badanych napojów nie wykazywał antagonizmu w stosunku do testowanych drożdży.

Các loài nấm Linh chi được sử dụng nhiều trong y học cổ truyền và việc xác định các hoạt tính sinh học của các loài nấm Linh chi mới là cần thiết cho sự phát triển các sản phẩm từ dược liệu. Năm 2016, nấm quế Linh chi, Humphreya endertii, thuộc họ Ganodermataceae, đã được phát hiện ở Vườn quốc gia Phước Bình, tỉnh Ninh Thuận, Việt Nam. Nghiên cứu này đã khảo sát khả năng gây độc tế bào ung thư phổi NCI H460 và tế bào ung thư gan HepG2 của dịch chiết hai loài nấm Linh chi Ganoderma lucidum và H. endertii có nguồn gốc từ Vườn quốc gia Phước Bình, IC50 tương ứng cho dịch chiết G. lucidum và H. endertii là 1,78 ± 0,35 và 2,25 ± 0,28 mg/ml trên tế bào NCI H460; và 4,53 ± 0,48 và 4,13 ± 1,05 mg/ml trên tế bào HepG2. Bên cạnh đó, dịch chiết hai loài nấm này cũng thể hiện khả năng kháng 5 chủng vi khuẩn gây bệnh Bacillus cereus, Staphylococcus aureus, Pseudomonas aeruginosa, Salmonella enteritidis và Salmonella typhimurium; dịch chiết nấm G. lucidum còn thể hiện sự kìm hãm với Escherichia coli trong khi dịch chiết H. endertii thì không có hoạt tính này

Dentre as inúmeras espécies da flora nativa brasileira, encontra-se a Ilex paraguariensis, conhecida popularmente como erva-mate. Estudos atribuem diversas atividades biológicas para essa espécie. O trabalho teve por objetivo avaliar a atividade antifúngica e antibacteriana do óleo volátil, extrato das folhas e dos ramos da I. paraguariensis. O material vegetal foi coletado na URI (28°16’41.4”S 54°16’13.0”W) em Santo Ângelo – RS/Brasil, sendo moído e filtrado e o óleo volátil obtido por hidrodestilação. A atividade antimicrobiana foi avaliada empregando a técnica de difusão em meio sólido Brain heart infusion (BHI). As bactérias utilizadas foram Corynebacterium fimi, Listeria monocytogenes, Bacillus cereus, Staphylococcus aureus, Salmonella enteritidis e Escherichia coli. Para a atividade antifúngica, foi empregado o método de difusão em meio sólido ágar Patato dextrose (PDA), e os fungos utilizados foram Aspergillus phoenicis isolado, Aspergillus niger, Fusarium sp. e Fusarium sp. isolado clínico. As alíquotas empregadas foram as mesmas, tanto para as bactérias quanto para os fungos, de 25 e 50 µL nas concentrações de 50 e 100 mg/mL do óleo e extratos. Os resultados obtidos, quando comparados aos resultados de inibição do antifúngico comercial Fox®, usados como padrão comparativo, comprovaram a acentuada atividade antimicrobiana apresentada pelo óleo de I. paraguariensis. Entre os diferentes extratos testados, o de folhas apresentou melhor resultado para atividade antimicrobiana, com a concentração de 100 mg/mL do óleo. O óleo volátil de I. paraguariensis, por apresentar halos de inibição superiores a 10 mm, indica uma boa atividade antimicrobiana.

Free fatty acids (FFAs) were obtained from hydrolyzed virgin coconut oil (VCO) by Candida rugosa lipase (CRL). Four factors’ influence on hydrolysis degree (HD) was examined. The best hydrolysis conditions in order to get the highest HD value were determined at VCO to buffer ratio 1 : 5 (w/w), CRL concentration 1.5% (w/w oil), pH 7, and temperature 40°C. After 16 hours’ reaction, the HD value achieved 79.64%. FFAs and residual hydrolyzed virgin coconut oil (HVCO) were isolated from the hydrolysis products. They were tested for their antibacterial activity against Gram-negative and Gram-positive bacteria, which can be found in contaminated food and cause food poisoning. FFAs showed their inhibition against Bacillus subtilis (ATCC 11774), Escherichia coli (ATCC 25922), Salmonella enteritidis (ATCC 13076), and Staphylococcus aureus (ATCC 25923) at minimum inhibitory concentration (MIC) of 50%, 60%, 20%, and 40%, respectively. However, VCO and HVCO did not show their antibacterial activity against these tested bacteria.

Background: [1, 3, 4]oxadiazolenone core containing chalcones and nucleosides were synthesized by Claisen-Schmidt condensation of a variety of benzaldehyde derivatives, obtained from oxidation of substituted 5-(3/6 substituted-4-Methylphenyl)-1, 3, 4-oxadiazole-2(3H)-one and various substituted acetophenone. The resultant chalcones were coupled with penta-O-acetylglucopyranose followed by deacetylation to get [1, 3, 4] oxadiazolenone core containing chalcones and nucleosides. Various analytical techniques viz IR, NMR, LC-MS and elemental analysis were used to confirm the structure of the synthesised compounds.The compounds were targeted against Bacillus subtilis, Staphylococcus aureus and Escherichia coli for antibacterial activity and Aspergillus flavus, Aspergillus niger and Fusarium oxysporum for antifungal activity. Methods: A mixture of Acid hydrazides (3.0 mmol) and N, Nʹ- carbonyl diimidazole (3.3 mmol) in 15 mL of dioxane was refluxed to afford substituted [1, 3, 4]-oxadiazole-2(3H)-one. The resulted [1, 3, 4]- oxadiazole-2(3H)-one (1.42 mmol) was oxidized with Chromyl chloride (1.5 mL) in 20 mL of carbon tetra chloride and condensed with acetophenones (1.42 mmol) to get chalcones 4. The equimolar ratio of obtained chalcones 4 and β -D-1,2,3,4,6- penta-O-acetylglucopyranose in presence of iodine was refluxed to get nucleosides 5. The [1, 3, 4] oxadiazolenone core containing chalcones 4 and nucleosides 5 were tested to determined minimum inhibitory concentration (MIC) value with the experimental procedure of Benson using disc-diffusion method. All compounds were tested at concentration of 5 mg/mL, 2.5 mg/mL, 1.25 mg/mL, 0.62 mg/mL, 0.31 mg/mL and 0.15 mg/mL for antifungal activity against three strains of pathogenic fungi Aspergillus flavus (A. flavus), Aspergillus niger (A. niger) and Fusarium oxysporum (F. oxysporum) and for antibacterial activity against Gram-negative bacterium: Escherichia coli (E. coli), and two Gram-positive bacteria: Staphylococcus aureus (S. aureus) and Bacillus subtilis(B. subtilis). Result: The chalcones 4 and nucleosides 5 were screened for antibacterial activity against E. coli, S. aureus and B. subtilis whereas antifungal activity against A. flavus, A. niger and F. oxysporum. Compounds 4a-t showed good antibacterial activity whereas compounds 5a-t containing glucose moiety showed better activity against fungi. The glucose moiety of compounds 5 helps to enter into the cell wall of fungi and control the cell growth. Conclusion: Chalcones 4 and nucleosides 5 incorporating [1, 3, 4] oxadiazolenone core were synthesized and characterized by various spectral techniques and elemental analysis. These compounds were evaluated for their antifungal activity against three fungi; viz. A. flavus, A. niger and F. oxysporum. In addition to this, synthesized compounds were evaluated for their antibacterial activity against gram negative bacteria E. Coli and gram positive bacteria S. aureus, B. subtilis. Compounds 4a-t showed good antibacterial activity whereas 5a-t showed better activity against fungi.

Two types of lipase, Candida rugosa lipase (CRL) and porcine pancreas lipase (PPL), were used to hydrolyze virgin coconut oil (VCO). The hydrolysis process was carried out under four parameters, VCO to buffer ratio, lipase concentration, pH, and temperature, which have a significant effect on hydrolysis of lipase. CRL obtained the best hydrolysis condition at 1 : 5 of VCO to buffer ratio, 1.5% of CRL concentration, pH 7, and temperature of 40°C. Meanwhile, PPL gave different results at 1 : 4 of VCO to buffer ratio, 2% of lipase concentration, pH 7.5, and 40°C. The highest hydrolysis degree of CRL and PPL was obtained after 16 hours and 26 hours, reaching 79.64% and 27.94%, respectively. Besides, the hydrolysis process was controlled at different time course (every half an hour) at the first 4 hours of reaction to compare the initial hydrolysis degree of these two lipase types. FFAs from hydrolyzed products were isolated and determined the percentage of each fatty acid which contributes to the FFAs mixture. As a result, medium chain fatty acids (MCFAs) made up the main contribution in composition of FFAs and lauric acid (C12) was the largest segment (47.23% for CRL and 44.23% for PPL).
