The present review summarizes the literature data regarding the application of Triticum aestivum assay as an alternative method for toxicity assessment of environmental pollutants or potential therapeutic agents. Plant bioassays present several advantages among other biological assays (simplicity, low cost, rapid test activation, a wide array of assessment endpoints). They present a good correlation with animal and human cells models, and are a reliable tool for genotoxicity assessment. Furthermore, in the context of toxicology guidelines that promote the substitution of assays using animal models with other bioassays, genotoxicity assays using higher plants models have gained in popularity. The present review focuses on three major aspects regarding Triticum aestivum assay - its utility in environmental pollution monitoring, its application in genotoxicity assessment studies, and its application in phytotoxicity evaluation of nanomaterials.   ********* In press - Online First. Abstract The fern Nephrolepis exaltata (L.) Schott, fam. Nephrolepidaceae, has little traditional medicinal use. In order to estimate its potential safety, in the present study we have investigated the phytotoxicity (on Triticum aestivum L.) and toxicity on brine shrimp of extracts from plants grown hydroponically. The species identity was confirmed by macroscopic and microscopic examinations on rhizomes, rachises, pinnae and runners, using bright field and fluorescent microscopy. Biological assays were performed on aqueous and ethanol solutions of the fronds. The brine shrimp lethality assay was performed on Artemia franciscana Kellog and a phytobiological bioassay on Triticum aestivum L. Lethality, root elongation and karyokinetic film modifications were evaluated, and LC50 and IC50 values were calculated. The microscopic analysis revealed the main histo-anatomic elements: polystelic structure and hypodermis (rhizome, rachis, runners), trichomes (rachis), homogenous structure, trichomes and diacytic/anisocytic stomata (leaves). The ethanol and aqueous extracts showed low cytotoxic effects on both Triticum aestivum roots and Artemia franciscana nauplii.

Khorasan wheat (Triticum turgidum ssp. turanicum (Jakubz.)) is an ancient tetraploid spring wheat variety originating from northeast parts of Central Asia. This variety can serve as a full-fledged alternative to modern wheat but has a lower yield than modern varieties. It is commonly known that wheat growth is influenced by soil tillage technology (among other things). However, it is not known how soil tillage technology affects ancient varieties. Therefore, the main objective of this study was to evaluate the influence of different soil tillage technologies on the growth of the ancient Khorasan wheat variety in comparison to the modern Kabot spring wheat (Triticum aestivum) variety. The trial was arranged in six small plots, one half of which was sown by the Khorasan wheat variety and the other half of which was sown by the Kabot wheat variety. Three soil tillage methods were used for each cultivar: conventional tillage (CT) (20–25 cm), minimum tillage (MTC) with a coulter cultivator (15 cm), and minimization tillage (MTD) with a disc cultivator (12 cm). The soil surface of all of the variants were leveled after tillage (harrows & levelling bars). An unmanned aerial vehicle with multispectral and thermal cameras was used to monitor growth during the vegetation season. The flight missions were supplemented by measurements using the GreenSeeker hand-held sensor and plant and soil analysis. The results showed that the Khorasan ancient wheat was better suited the conditions of conventional tillage, with low values of bulk density and highvalues of total soil porosity, which generally increased the nutritional value of the yield in this experimental plot. At the same time, it was found that this ancient wheat does not deplete the soil. The results also showed that the trend of developmental growing curves derived from different sensors was very similar regardless of measurement method. The sensors used in this study can be good indicators of micronutrient content in the plant as well as in the grains. A low-cost RGB camera can provide relevant results, especially in cases where equipment that is more accurate is not available.

Pollutant gases, such as CO, NO2, O3, and SO2 affect human health, and low-cost sensors are an important complement to regulatory-grade instruments in pollutant monitoring. Previous studies focused on one or several species, while comprehensive assessments of multiple sensors remain limited. We conducted a 12-month field evaluation of four Alphasense sensors in Beijing and used single linear regression (SLR), multiple linear regression (MLR), random forest regressor (RFR), and neural network (long short-term memory (LSTM)) methods to calibrate and validate the measurements with nearby reference measurements from national monitoring stations. For performances, CO > O3 > NO2 > SO2 for the coefficient of determination (R2) and root mean square error (RMSE). The MLR did not increase the R2 after considering the temperature and relative humidity influences compared with the SLR (with R2 remaining at approximately 0.6 for O3 and 0.4 for NO2). However, the RFR and LSTM models significantly increased the O3, NO2, and SO2 performances, with the R2 increasing from 0.3–0.5 to >0.7 for O3 and NO2, and the RMSE decreasing from 20.4 to 13.2 ppb for NO2. For the SLR, there were relatively larger biases, while the LSTMs maintained a close mean relative bias of approximately zero (e.g., <5% for O3 and NO2), indicating that these sensors combined with the LSTMs are suitable for hot spot detection. We highlight that the performance of LSTM is better than that of random forest and linear methods. This study assessed four electrochemical air quality sensors and different calibration models, and the methodology and results can benefit assessments of other low-cost sensors.

Abstract Background Fecal calprotectin is a non-invasive test of colonic inflammation used for monitoring inflammatory bowel disease activity and for risk stratifying non-specific colonic symptoms. Calprotectin is a leukocyte specific enzyme. A similar test, leukocyte esterase is used to detect leukocytes in urine and is widely available as a low-cost point-of-care test strip. We hypothesize that an unmodified version of the urine test strip would be highly accurate in predicting a positive fecal calprotectin test in a real world sample of patients. Aims To explore a low cost, rapid alternative to the fecal calprotectin test Methods All inpatient and outpatient stool samples tested for calprotectin by the Vancouver General Hospital laboratory from February 2020 to November 2020 were included prospectively. Samples were simultaneously tested for fecal leukocyte esterase using an unmodified Roche Cobas Chemstrip urinalysis test strip by central lab personnel. An identical aliquot was sent to LifeLabs for calprotectin as per standard protocol. All samples were suspended in buffer using established laboratory protocols prior to testing. Fecal leukocyte esterase results were reported as 0–4+ based on visual interpretation, calprotectin results were reported as mcg/g of stool. REB review and approval was obtained prior to data collection. Sensitivity, Specificity and AUROC were calculated using Microsoft Excel and JROCFIT. Results 26 samples were collected. Using a fecal calprotectin greater than 120 mcg/g as a gold standard an AUROC of 0.89 (SE= .06) was calculated. A leukocyte esterase reading of 2+ or greater had the best test characteristics based on ROC curve analysis. Using this cutoff, 21/26 samples were concordant, giving an accuracy of 80.8%, sensitivity of 90.9% and specificity of 73.3%. Positive likelihood ratio was 8.07 and negative likelihood ratio was 0.29. Assuming an AUROC of 0.8, the sample size N=26 is 90% powered (β=0.9) to predict the true AUROC within 0.1 with a type I error rate of .05 (α&lt;.05). Conclusions This study suggests application of a prepared stool sample to a urinalysis test strip gives a result highly predictive of a positive fecal calprotectin test. Further results are being collected prospectively to improve the robustness of these preliminary data. Secondary outcomes including comparison to endoscopy and biopsy results where available are planned if an adequate sample size can be accrued. Future studies justifying independent clinical use of leukocyte esterase would require a common gold standard comparator such as endoscopy. Fecal calprotectin testing is not universally insured and is not available as a rapid test strip. Use of fecal leukocyte esterase may reduce costs and shorten time to results if proven to be independently reliable. Funding Agencies None

Abstract Exposure to environmental pollutants like chemicals or air pollution is major health concern for the human population. Especially the nervous system is a sensitive target for environmental toxins with exposures leading to life stage-dependent neurotoxicity. Developmental and adult neurotoxicity are characterized by specific adverse outcomes ranging from neurodevelopmental disorders to neurodegenerative diseases like Alzheimer’s and Parkinson’s disease. The risk assessment process for human health protection is currently undergoing a paradigm change toward new approach methods that allow mechanism-based toxicity assessment. As a flagship project, an in vitro battery of test methods for developmental neurotoxicity evaluation is currently supported by the Organization for Economic Co-operation and Development (OECD). A plethora of stem cell-based methods including brain spheres and organoids are currently further developed to achieve time- and cost-saving tools for linking MoA-based hazards to adverse health effects observed in humans.

Adsorption is a widely used technique for wastewater remediation. The process is effective and economical for the removal of various pollutants from wastewater, including dyes. Moreover, Besides commercial activated carbon, different low-cost materials such as agricultural and industrial wastes are now used as adsorbents. The present review focused on the removal of a teratogenic and carcinogenic dye, orange G (OG) via adsorption using several adsorbents, together with the experimental conditions and their adsorption capacities. Based on the information compiled, various adsorbents have shown promising potential for OG removal.
