Among the principles used for the evaluation of seed vigor, two approaches are predominant: seed resistance to stresses and the direct or indirect identification of the "current" state of the seeds, e.g., enzymatic activity, integrity of the cellular membranes, growth parameters. In this presentation recent innovations are discussed in procedures for seed vigor evaluation emphasizing the methodologies of accelerated aging with salt saturated solutions, controlled deterioration and potassium leachate tests. These tests, performed at seed the laboratory of ESALQ/USP, in function of their simplicity and efficiency, deserve the attention of seed technologists.

International research on vegetable seed vigor is not at the same level attained for grain crops species. This study was conducted to identify reliable procedures for the accelerated aging and controlled deterioration tests to rank onion (Allium cepa L.) seed lots according to their physiological potential. Six seed lots of the cultivars Aurora and Petroline were evaluated in the laboratory for germination, first count, seedling vigor classification, traditional and saturated salt accelerated aging (41ºC / 48 and 72 h), controlled deterioration (24% of water / 45ºC / 24 h) and seedling emergence tests. Seed moisture content after the saturated salt accelerated aging test was lower and uniform, which is considered an important advantage in comparison to the traditional procedure. The saturated salt accelerated aging (41ºC / 48 and 72 h) and controlled deterioration (moisture content adjusted to 24% / 45ºC / 24 h) tests were the best procedures to assess the physiological potential of onion seeds, and are indicated for use in quality control programs.

The efficiency of vigor tests in assessing the physiological potential of seeds depends on their standardization for different species. In this context, the research aimed to study specific methodologies to evaluate the vigor of spinach seeds (Tetragonia tetragonoides). For this purpose, five lots of spinach seed cv. New Zealand were submitted to germination, first count of germination, seedling emergence in the field, accelerated aging (with and without use of saturated solution of NaCl at 41and 45 ºC for 24,48 and 72 hours), controlled deterioration (18,21 and 24% seed water content adjustments for 24 hours at 45 ºC) and electrical conductivity tests with variations inwater volume (25, 50 and 75 mL), seed quantity (25 and 50) and soaking period (1, 2, 4, 8,12, 16 and 24 hours) at 25 ºC. The first count test allowed obtaining preliminary information about seed vigor, and other tests in general showed similarity with seedling emergence in the field. Thus, it was concluded that traditional accelerated aging test 24 h /41 ºC and accelerated aging test with saturated salt solution for 24 h at 41 ºC and 45 ºC, and controlled deterioration test with 21% seed water content / 24 h / 45 ºC were all efficient for evaluating the physiological potential of spinach seeds.

The controlled deterioration test is efficient for seed vigor evaluation of several species. However, the procedures conditions are incipient, especially in respect to tropical forages such as sunn hemp, which still does not have a standardized procedure. The objective of this research was to adjust the procedures for a controlled deterioration test in Crotalaria juncea L. seeds. For this, five commercial seed lots were evaluated for water content and initial physiological potential (germination test, first germination count, germination speed index, saturated salt accelerated aging with NaCl, electrical conductivity and seedling field emergence). For controlled deterioration test, the initial seed moisture content was adjusted to 18, 21 and 24% and, subsequently, the samples were exposed at 45 °C during 24 hours. Seed germination test was conducted with the use of sand as substrate. Evaluations were performed on the fourth, fifth and sixth day after sowing. The controlled deterioration test for sunn hemp seeds should be performed with 24% seed moisture content, at 45 °C during 24 hours, and the test evaluation on the fourth day after sowing.

1-aminocyclopropane-1-carboxylic acid (ACC) was used to measure seed vigor of lettuce, cabbage, tomato, snap bean and sweet corn seeds. Accelerated aging at 40C and 93% RH over saturated solution of KH2PO4 and natural aging under ambient storage conditions (5-7C, 28 to 60% RH) were used to obtain seeds of differing vigor levels. Depending on the type of seeds, the amount of ACC needed for maximal ethylene production (saturating dose) ranged from 0.25 to 2mM. Seeds produced much larger amounts of ethylene in the presence than in absence of ACC, the ACC-derived ethylene was detected prior to germination, and ACC had no adverse effect on germination. ACC-derived ethylene production paralleled vigor loss as determined by percentage germination, mean germination time (for lettuce only) and seedling growth (for snap bean only). Second degree polynomial and logarithmic equations generated for the relationship of ACC-derived ethylene production to germination or growth parameters following seed aging, provided good to excellent fit. As a vigor test, the ACC-ethylene procedure has several advantages over the non-ACC ethylene procedure: it greatly improves the sensitivity of the test by enhancing ethylene production, it permits detection of small differences in vigor levels and it allows detection of ethylene prior to germination within a few hours of soaking.

The assessment of seed physiological performance is essential to provide consistent information to compose quality control programs conducted by seed companies and allow prompt decisions to reduce post-harvest and commercialization problems. This research was carried to evaluate the efficiency of different procedures to determine cauliflower seed vigor. Six lots of the cultivar Sharon were initially characterized by germination and seedling emergence, and subsequently subjected to traditional and saturated salt accelerated aging, controlled deterioration, electrical conductivity, and potassium leachate tests, in two evaluation times. The traditional accelerated aging (41ºC/48 h), the saturated salt procedure (45ºC/72 h), the controlled deterioration (moisture content adjusted to 20% or 22% and aging at 45ºC/24 h), the electrical conductivity (50 seeds/75 mL water and evaluation after 4 h) and the potassium leakage (25 seeds/50 mL of water and evaluation after 30 min) laboratory tests are sensitive to detect differences in the physiological potential of cauliflower seeds. However, those results did not relate to the percentage seedling emergence in greenhouse controlled conditions, as a consequence of the favorable conditions for emergence. However, the differences identified in the tests may be related to the ability of seed lots to perform in less favorable environments.

Vigor of soybean [Glycine max (L.) Merrill] seeds can be evaluated by measuring the electrical conductivity (EC) of the seed soaking solution, which has shown a satisfactory relationship with field seedling emergence, but has not had aproper definition of range yet. This work studies the relationship between EC and soybean seedling emergence both in the field and laboratory conditions, using twenty two seed lots. Seed water content, standard germination and vigor (EC, accelerated aging and cold tests) were evaluated under laboratory conditions using -0.03; -0.20; -0.40 and -0.60 MPa matric potentials, and field seedling emergence was also observed. There was direct relationship between EC and field seedling emergence (FE). Under laboratory conditions, a decreasing relationship was found between EC and FE as water content in the substrate decreased. Relationships between these two parameters were also found when -0.03; -0.20 and -0.40 MPa matric potentials were used. EC tests can be used successfully to evaluate soybean seed vigor and identify lots with higher or lower field emergence potential.

Seed vigor testing is an important component of quality control programs adopted by seed industry. The software Seed Vigor Imaging System (SVIS) has been successfully used for seed vigor assessment in different species. The objective of this research was to verify the SVIS efficiency to assess okra seed vigor in comparison to other vigor tests used for this species. Five seed lots of 'Clemson Americano' and four of 'Santa Cruz' were submitted to germination (speed and percentage), cold germination (speed and percentage), traditional and saturated salt accelerated aging (41 ºC/72 and 96 h) and seedling emergence tests during 12 months storage. Vigor index, uniformity of growth and seedling length were determined by the software Seed Vigor Imaging System (SVIS). Results showed that the vigor index and seedling length determined by the SVIS analyses are efficient to determine okra seed vigor as well as the accelerated aging test (41 ºC/72 h).

Research on pea seed storage and the identification of efficient seed vigor tests for this species is still insufficient, especially for cultivars that produce wrinkled seeds used for consumption in natura. The objective of this study was to verify the accuracy of tests to assess the physiological potential of pea seed lots stored in different environments. Four seed lots of two cultivars, 'Telefone Alta' (indeterminate growth habit) and 'Itapuã' (determinate growth habit) were stored under different environmental conditions for 8 months: a) laboratory uncontrolled b) dry and cold chamber (10 ºC and 30% RH), c) controlled environment (20 ºC and 70% RH). Seed vigor (accelerated aging, electrical conductivity, seedling length and seedling emergence) was evaluated initially and after four and eight months storage. Results showed that the use of seed lots of high initial physiological potential is fundamental for guaranteeing the desired performance of pea seeds during storage. Wrinkled pea seeds should be stored between harvest and sowing in an environment in which the sum of the relative humidity (%) and temperature (°C) does not exceed 70. Physiological potential of pea seeds should be evaluated by at least two vigor tests; consideration to accelerated aging and electrical conductivity tests is recommended.

<p>ABSTRACT<br />Information of seed quality during storage can be determined through the actual storage and storability vigor estimation. This study aimed at comparing effective accelerated aging method<br />between physical and chemical, and studying the seed deterioration during storage in ambient (T =28-30 0C, RH=75-78%) and AC (T =18-20 0C, RH =51-60%) condition with three levels of initial moisture content (8-10%, 10-12%, and 12-14%) for 20 weeks. The final objective of this research<br />was to develop model for storability vigor of papaya seed. Two experiments, accelerated aging and seed storage were conducted at Seed Laboratory, Department of Agronomy and Horticulture, Bogor Agricultural University from October 2015 to May 2016. A completely randomized design with nested factors and four replications was applied to both experiments. The results showed that physical accelerated aging using IPB 77-1 MMM machine was more effective than chemical accelerated aging using IPB 77-1 MM machine for papaya seed. The viability of seed stored in AC condition remained high until the end of the storage period, whereas it declined at 16 week storage period in the ambient condition. The viability of seed with initial moisture content of 12-14% declined faster than that of initial moisture content of 8-10% after 18 week storage periode. The model used to estimate the storability vigor of papaya seed accurately was the equation y = a + b expcx where y : storability vigor estimation, x : aging time and a,b,c : constant value. Penelitian ini bertujuan untuk membandingkan metode<br />pengusangan cepat yang efektif antara fisik dengan kimia serta mempelajari pola penurunan viabilitas benih selama penyimpanan aktual pada kondisi simpan kamar (suhu =28-30 0C, RH =75-78%) dan AC (suhu =18-20 0C, RH =51-60%) dengan tiga tingkat kadar air awal (8-10%, 10-12%, dan 12-14%) selama 20 minggu. Tujuan akhirnya adalah membangun model vigor daya simpan benih pepaya. Penelitian pengusangan cepat dan penyimpanan dilakukan pada bulan Oktober 2015 sampai Mei 2016 di Laboratorium Benih, Departemen Agronomi dan Hortikultura, Institut Pertanian<br />Bogor. Kedua penelitian menggunakan rancangan acak lengkap tersarang dengan empat ulangan. Hasil penelitian menunjukkan bahwa pengusangan cepat secara fisik dengan alat IPB 77-1 MMM lebih efektif daripada pengusangan kimia dengan alat IPB 77-1 MM untuk benih pepaya. Viabilitas benih yang disimpan pada kondisi AC tetap tinggi hingga akhir periode simpan, sedangkan pada kondisi kamar penurunan viabilitas dimulai pada periode simpan 16 minggu. Benih yang disimpan dengan tingkat KA awal sebesar 12-14% lebih cepat mengalami penurunan viabilitas mulai periode simpan 18 minggu dibandingkan dengan benih dengan KA awal 8-10%. Hasil penelitian juga menunjukkan terdapat korelasi yang erat antara pola kemunduran benih pada pengusangan cepat dan penyimpanan aktual, sehingga model pendugaan vigor daya simpan (y) berdasarkan waktu pengusangan (x) dapat disusun dengan persamaan y = a + b expcx. Abstract: Initial seed vigor and sowing density interact in establishment of plants in the field, and can thus affect expression of the plasticity of the wheat crop and the yield and physiological quality of the seeds produced. The aim of this study was to evaluate the effect of seed vigor levels in combination with sowing densities on the yield and physiological potential of wheat seeds. The cultivars BRS Gralha-Azul and BRS Sabiá were used in a randomized block experimental design in a 2 × 4 factorial arrangement, with four replications. The treatments consisted of seeds of high and low vigor and four sowing densities (150, 250, 350, and 450 seeds.m-2). The following evaluations were made: seed yield, germination, first germination count, seedling length, seedling dry matter, accelerated aging, emergence speed index, and seedling emergence in sand. An increase in sowing density favors the seed yield of both cultivars; however, it reduces the vigor of the seeds produced by the cultivar BRS Gralha-Azul, especially of the seeds produced by plants originating from high vigor seeds. For the cultivar BRS Sabiá, an increase in sowing density decreases the germination performance of seeds produced by plants originating from low vigor seeds.
