Abstract The need for wastewater treatment is progressively rising as the release of copious amounts of industrial wastewater is increasing. Likewise, there is an urgent requirement for renewable energy sources because of the growing energy demand and depletion of fossil fuels. The use of microalgae to convert toxic phenolic wastewater to lipid-enriched biofuel has recently been proposed. Here, we report a new strategy for coupling N-doped TiO2-coated photocatalytic optical fibers and a microalgal biofilm to degrade 4-chlorophenol (4-CP) and produce biomass. In the combined photocatalysis and biodegradation system, the photocatalytic products were directly biodegraded by the heterotroph-enriched (Salinarimonas and Pseudomonas) biofilm, promoting biomass production; O2 produced by the phototrophs (Scenedesmus obliquus) promoted the generation of hydroxyl free radicals using N-doped TiO2. Thus, the combined photocatalysis and biodegradation system rapidly and sustainably degraded 4-CP while maintaining the growth of the microalgal biomass. The 4-CP removal, dechlorination, and biofilm growth rates reached ~78 µM/h, ~41 µM/h, and 1.8 g/h/m2, respectively. Overall, we present a useful synergy between an optical catalyst and a bioreactor that has implications for both wastewater remediation and sustainable microalgal biomass production.

Media device (MD) use is increasing worldwide among children. Adolescents and young children spend a lot of time using MD, Internet, and social networks. The age of initial use is getting lower to 12 years old. The aim of this research is to study children’s use and perception of MD. The Italian Paediatric Society (SIP) conducted a Survey on Italian children in collaboration with Skuola.net using an online questionnaire. A total of 10,000 questionnaires were completed. Children admitted they spend more than 3 h (41%), more than 2 h (29%), more than 1 h (21%) and less than 1 h (9%) daily. Problematic MD use has been found with children using MD before sleeping (38%), during school (24%), and at wake up in the morning (21%). Addiction was documented in 14% of adolescents. Among the reported consequences, low academic outcomes, and reduced concentration (24%), neck and back pain (12%), insomnia (10%), and mood disturbances (7%) were referred. Adolescents may have a low perception of the risks related to excessive MD. The duration of time spent using media devices is a main risk factor. In this context, parents should strongly discourage excessive MD use, mainly during school, at bedtime, and wake-up. Additionally, parents should be informed and start conversations with their children on the potential negative effects of prolonged MD use.

The aim of this study was to examine the exercise intensity during the swimming, cycling, and running legs of nondraft legal, Olympic-distance triathlons in well-trained, age-group triathletes. Seventeen male triathletes completed incremental swimming, cycling, and running tests to exhaustion. Heart rate (HR) and workload corresponding to aerobic and anaerobic thresholds, maximal workloads, and maximal HR (HRmax) in each exercise mode were analyzed. HR and workload were monitored throughout the race. The intensity distributions in three HR zones for each discipline and five workload zones in cycling and running were quantified. The subjects were then assigned to a fast or slow group based on the total race time (range, 2 h 07 min–2 h 41 min). The mean percentages of HRmax in the swimming, cycling, and running legs were 89.8% ± 3.7%, 91.1% ± 4.4%, and 90.7% ± 5.1%, respectively, for all participants. The mean percentage of HRmax and intensity distributions during the swimming and cycling legs were similar between groups. In the running leg, the faster group spent relatively more time above HR at anaerobic threshold (AnT) and between workload at AnT and maximal workload. In conclusion, well-trained male triathletes performed at very high intensity throughout a nondraft legal, Olympic-distance triathlon race, and sustaining higher intensity during running might play a role in the success of these athletes.

e13545 Background: SLC3A2 (solute carrier family member 2) 3 is a member of the SLC3 family Ⅱ transmembrane glycoprotein, which is a key component of heteromeric amino acid transporters (HATs). The association between SLC3A2 and cancer was established, given that the amino acid supply by SLC3A2 is required for the resistance of oxidative stress and the maintenance of cell proliferation. However, whether SLC3A2 mutation plays a role in carcinogenesis remains unclear. Methods: This study enrolled 1000 colorectal cancer patients. Tumor tissues and paired peripheral blood mononuclear cells (PBMC) were collected for the sequencing of cancer-related genes by target capture and next-generation sequencing. Results: p.Lys301Argfs*31 mutation in SLC3A2 were detected in 40(40/1000, 4.0%) patients. low TMB and MSS were reported in 2 cases while the remaining 38 cases exhibited high TMB and MSI-H. 41(41/1000, 4.1%) DNA mismatch repair (dMMR) were identified in 41(41/1000, 4.1%) tumor samples, 27(27/41, 65.9%) of which had p.Lys301Argfs*31 mutation. Moreover, this frameshift mutation (COSM253097) was observed in 28 cases of large intestine cancer, 1 case of peritoneum cancer, 1 case of Stomach cancer, 1 case of biliary tract cancer and 1 case of ovary cancer in the COSMIC database. Conclusions: p.Lys301Argfs*31 mutation of SLC3A2 was identified in the majority of colorectal cancer cases with dMMR events, suggesting that this mutation may be a biomarker of dMMR.

Abstract Background Low-intensity shockwave therapy (LI-SWT) is suggested as a therapy for promoting tissue regeneration. In pigs, it was recently found that LI-SWT improved renal function after ischaemic injury. Our objectives were to study glomerular filtration rate (GFR) and albuminuria in diabetic nephropathy (DN) after treatment with LI-SWT. The present pilot study reports on the clinical safety of LI-SWT in DN. Methods A total of 14 patients with diabetes mellitus and Stage 3 chronic kidney disease were recruited for this prospective, one-arm Phase 1 study. The patients were treated with six sessions of LI-SWT during a 3-week period. At each session, 3000 shockwaves were applied to each kidney with 0.265 mJ/mm2, extended focal size and 4 Hz. Follow-up visits were performed at 1, 3 and 6 months. Results In general, the treatment was well tolerated. Transient macroscopic haematuria was observed in three patients immediately after LI-SWT. The majority of patients experienced lower back tenderness lasting up to 2 days after treatment. There was no need for analgesic treatment. LI-SWT showed no negative effect on GFR and albuminuria. At baseline, median (interquartile range) GFR was 33.5 mL/min/1.73 m2 (27.8–43.8) compared with 36.0 mL/min/1.73 m2 (27.5–52.0) at 6 months follow-up. In parallel, median albuminuria was 256 mg/24 h (79–619) at baseline and tended to decrease to 137 mg/24 h (41–404) 6 months after LI-SWT. There was no statistical difference between baseline and follow-up results. Conclusions LI-SWT is a safe treatment for DN. Inclusion of more patients is needed to determine whether LI-SWT can improve renal functional outcomes.

It’s tropical climate make the air conditioning system use for cooling system only (AC). The centralized air conditioning system in high rise building uses ducting system with rectangular cross-sectional area which often depends on a given blue print. Some specific area of the blue print may lead to adjustment of the ducting system, e.g. the gradual step of the ducting system. This gradual base of the ducting may cause additional consequences, a recirculation flow at the gradation area (the expansion zone). As already known, the recirculation flow leads to flow losses. This paper discuss an numerical analysis with CFD simulation of a backward-facing step (BFS) geometry. The BFS geometry used in this has total length (L) = 4050 mm, step height (h) = 41 mm, upstream height (H) = 81 mm, expansion ratio = 1.5, and width (t) = 20h. The CFD simulation conducted three dimensionaly with RNG k-? turbulence model, and 36.806 triangular nodes. Recirculation vortices represented by recirculation zone with x-velocity and reattachment point. The result of CFD simulation that is conducted to Reynolds number Re = 7.315,79; Re = 21.947,37; dan Re = 29.263,16 shows that recirculation vortices occur at X/h = 29,2 to X/h = 35 for tested Reynolds number. Reattachment point at Reynolds number of 7.315.79 occur at X/h > 35, which farther than that is achieved at larger Reynolds number.

ABSTRACTPharmacokinetic (PK) data describing a prolonged time course of antiretrovirals in plasma and peripheral blood mononuclear cells (PBMCs) are important for understanding and managing late or missed doses and to assess the appropriateness of compounds for preexposure prophylaxis (PrEP). This study aimed to evaluate the PK of coformulated tenofovir disoproxil fumarate (DF), emtricitabine, and rilpivirine in plasma and of the intracellular (IC) anabolites tenofovir diphosphate (TFV-DP) and emtricitabine triphosphate (FTC-TP) in healthy volunteers up to 9 days after drug cessation. Individuals received daily tenofovir DF-emtricitabine-rilpivirine (245/200/25 mg) for 14 days. Drug intake was stopped, and serial sampling occurred prior to the final dose and up to 216 h (9 days) after stopping drug intake. Concentrations were quantified and PK parameters calculated. Eighteen volunteers completed the study. The terminal elimination plasma half-lives for tenofovir and emtricitabine over 216 h (geometric mean [90% confidence interval]) were higher than those seen over 0 to 24 h (for tenofovir, 31 h [27 to 40 h] versus 13.3 h [12.5 to 15.1 h]; for emtricitabine, 41 h [36 to 54 h] versus 6.4 h (5.9 to 7.6 h]). Model-predicted IC half-lives (0 to 168 h) were 116 h (TFV-DP) and 37 h (FTC-TP). The plasma rilpivirine concentration at 216 h was 4.5 ng/ml (4.2 to 6.2 ng/ml), and half-lives over 0 to 216 h and 0 to 24 h were 47 h (41 to 59 h) and 35 h (28 to 46 h), respectively. These data contribute to our understanding of drug behavior following treatment interruption; however, adherence to therapy should be promoted. Validated plasma and IC target concentrations are necessary to allow interpretation with respect to sustained virus suppression or HIV prevention. (The trial was conducted in accordance with the Declaration of Helsinki [EudraCT 2012-002781-13].)
