Abstract Trypanosoma (Duttonella) vivax is an important cause of economic losses among feedlot cattle. These losses are related to the morbidity, mortality, reproductive issues and decreased production. It is known that the clinical signs observed in infections by this protozoon are similar to other hemoparasitosis, which difficult the diagnosis. Therefore, the aim of this study was to detect and molecularly characterize an outbreak of trypanosomiasis caused by T. (D.) vivax in dairy cattle in the municipality of São Miguel Aleixo, state of Sergipe, Brazil. Blood samples from cattle (n = 15) presenting clinical signs compatible with trypanosomiasis were collected and parasitological and molecular evaluated. Among the samples analyzed, 34% (5/15) were positive from blood smears, 60% (9/15) from the buffy coat method and 80% (12/15) from the molecular method. The DNA sequence obtained (659 bp) showed 99% similarity to T. (D.) vivax sequences that are available in the GenBank database. The presence of this protozoon in cattle herds is a problem for producers. Diagnosing trypanosomiasis is problematic because its evolution is similar to that of other parasitic blood diseases. In addition, this is the first report of infection by T. (D.) vivax in cattle in the state of Sergipe, northeastern Brazil.

Mycoplasma bovis is an important bovine pathogen causing pneumonia, mastitis, and arthritis and is responsible for major economic losses worldwide. In the absence of an efficient vaccine, control of M. bovis infections mainly relies on antimicrobial treatments, but resistance is reported in an increasing number of countries. To address the situation in Spain, M. bovis was searched in 436 samples collected from beef and dairy cattle (2016–2019) and 28% were positive. Single-locus typing using polC sequences further revealed that two subtypes ST2 and ST3, circulate in Spain both in beef and dairy cattle, regardless of the regions or the clinical signs. Monitoring of ST2 and ST3 isolates minimum inhibitory concentration (MIC) to a panel of antimicrobials revealed one major difference when using fluoroquinolones (FQL): ST2 is more susceptible than ST3. Accordingly, whole-genome sequencing (WGS) further identified mutations in the gyrA and parC regions, encoding quinolone resistance-determining regions (QRDR) only in ST3 isolates. This situation shows the capacity of ST3 to accumulate mutations in QRDR and might reflect the selective pressure imposed by the extensive use of these antimicrobials. MIC values and detection of mutations by WGS also showed that most Spanish isolates are resistant to macrolides, lincosamides, and tetracyclines. Valnemulin was the only one effective, at least in vitro, against both STs.

In the present study we investigated the presence of infections by vaccinia-like viruses in dairy cattle from 12 counties in the state of Rio de Janeiro in the last 9 years. Clinical specimens were collected from adult animals with vesicular/pustular lesions mainly in the udder and teats, and from calves with lesions around the nose and mouth. A plaque reduction neutralization test (PRNT) was applied to search for antibodies to Orthopoxvirus; the vesicular/pustular fluids and scabs were examined by PCR, electron microscopy (EM) and by inoculation in VERO cells for virus isolation. Antibodies to Orthopoxvirus were detected in most cases. The PCR test indicated a high nucleotide homology among the isolates and the vaccinia viruses (VACV) used as controls. By EM, typical orthopoxvirus particles were observed in some specimens. The agents isolated in tissue culture were confirmed as vaccinia-like viruses by EM and PCR. The HA gene of the vaccinia-like Cantagalo/IOC virus isolated in our laboratory was sequenced and compared with other vaccinia-like isolates, showing high homology with the original Cantagalo strain, both strains isolated in 1999 from dairy cattle. Antibodies to Orthopoxvirus were detected in one wild rodent (genus Akodon sp.) collected in the northwestern region of the state, indicating the circulation of poxvirus in this area. Nonetheless, PCR applied to tissue samples collected from the wild rodents were negative. Vesicular/pustular lesions in people in close contact with animals have been also recorded. Thus, the vaccinia-like virus infections in cattle and humans in the state seem to be an expanding condition, resulting in economic losses to dairy herds and leading to transient incapacitating human disease. Therefore, a possible immunization of the dairy cattle in the state should be carefully evaluated.

Bovine herpesvirus 1 (BoHV-1) causes major losses in worldwide livestock, affecting the respiratory and reproductive tracts of bovine. In the past decades, the number of cases in Brazil has been gradually increasing. Therefore, it is important to assess the distribution of infection in different regions of the country. In the state of Espírito Santo (ES) the BoHV 1 infection rate in dairy cattle herds is unknown. Thus, the aim of this study was to detect neutralizing antibodies against BoHV-1 in serum samples from 1,161 non-vaccinated cows from 59 dairy cattle herds in 23 municipalities of the Metropolitan, North, Northwest and South macro-regions. The identification of seropositive cows was evaluated by the virus neutralization test. The results showed that of all serum samples evaluated 775 (66.75%) had neutralizing antibodies against BoHV-1. Moreover, all herds were found positive; however, the percentage of positive cows varied among regions; 49.06%, 62.15%, 67.21% and 80.04% for the Metropolitan, South, North and Northwest macro-regions, respectively. In this study, the results clearly indicate the dissemination of the viral agent in dairy cattle in the ES state, requiring the monitoring and control of diseases related to BoHV-1 infection.

Background: Contagious agalactia is an infectious disease caused by Mycoplasma agalactiae (M. agalactiae) that occurs in small ruminants leading to productive and economic losses. Due to the similarity of clinical signs presented in Caprine Arthritis Encephalitis (CAE), which is a viral disease, a differential diagnosis is important. Therefore, this study aimed to investigate the presence of anti-Mycoplasma agalactiae antibodies in dairy goat flocks in Ceará State and possible correlation with CAE.Materials, Methods & Results: The research was performed in four mesoregions in Ceará State (Metropolitan Region of Fortaleza-MRF; Northeast Ceará - NeC; North Ceará - NC; Sertões in Ceará - SC), from which 16 productions located in 10 cities with the highest representativeness for goat milk production within the State or mesoregion were sampled. A total of 417 females and 69 males (486 animals) of breeds with dairy production aptitude, pure or crossbreed, maintained in semi-intensive or intensive systems, were tested. Blood serum was obtained by venipuncture of the jugular vein with vacuum pressure syringe followed by centrifugation at 1,500 g for 10min. Antibodies against the caprine arthritis encephalitis virus (CAEV) were detected with micro technique of agarose gel immunodiffusion (AGID) and Western Blot (WB). The anti-Mycoplasma agalactiae antibodies were detected with commercial kit of enzymatic immunoassay (IDEXX Laboratories™). Seroprevalence of M. agalactiae in dairy goat flocks in Ceará State was 0.62% (3/486). From the total of 16 visited productions, 18.75% (3/16) had seropositive animals for M. agalactiae located in MRF, NC and SC mesoregions. CAE was diagnosed in 56.25% (9/16) of productions with AGID and in 81.25% (13/16) with WB. In addition, 5.2% (25/486) of animals were seropositive for CAE with AGID and 16.6% (80/486) with WB. Animals that reacted positive for M. agalactiae were all females of pure breed with milk production aptitude in distinct mesoregions submitted to intensive rearing system. None of these animals was positive in neither test (AGID or WB) for CAE. Therefore, no correlation of results obtained in diagnosis of M. agalactiae by ELISA and CAEV by AGID or WB (P < 0.05) was identified. However, two out of three productions that were positive for M. agalactiae presented positive results for CAEV with frequencies of 10% and 20%.Discussion: Seroprevalence of M. agalactiae in Ceará State was low in comparison with other Brazilian states and even other countries. However, the presence of the pathogen in more than one mesoregion indicates that the disease occurs in different locations within the State. Therefore, flocks in Ceará are susceptible to the infection, which may be favored by uncontrolled commerce that occurs with deficient surveillance, associated with the importation of animals to improve flock genetic quality. The presence of the pathogen in dairy goats may contribute to significant losses in the local production. On the other hand, CAE was diagnosedin nearly all productions proving the dissemination of this lentivirus infection among dairy goat flocks in Ceará State. Although an association between these diseases was not identified, the presence of a retrovirus in the organism may favor co-infection with another micro-organism, promoting the deficiency in the immune system of the host. In conclusion, M. agalactiae is present in different mesoregions of the Ceará State and control measures should be adopted in short term to prevent pathogen dissemination and, consequently reduce economic and productive losses in the local dairy goat production. No correlation was identified between the prevalence of infection by CAEV and M. agalactiae in this study.Keywords: correlation, diagnosis, caprine lentivirus, mycoplasmosis.

The state of Paraíba is part of the semi-arid region of Brazil, where severe droughts have occurred in recent years, resulting in significant socio-economic losses associated with climate variability. Thus, understanding to what extent precipitation can be influenced by sea surface temperature (SST) patterns in the tropical region can help, along with a monitoring system, to set up an early warning system, the first pillar in drought management. In this study, Generalized Additive Models for Location, Scale and Shape (GAMLSS) were used to filter climatic indices with higher predictive efficiency and, as a result, to perform rainfall predictions. The results show the persistent influence of tropical SST patterns in Paraíba rainfall, the tropical Atlantic Ocean impacting the rainfall distribution more effectively than the tropical Pacific Ocean. The GAMLSS model showed predictive capability during summer and southern autumn in Paraíba, highlighting the JFM (January, February and March), FMA (February, March and April), MAM (March, April and May), and AMJ (April, May and June) trimesters as those with the highest predictive potential. The methodology demonstrates the ability to be integrated with regional forecasting models (ensemble). Such information has the potential to inform decisions in multiple sectors, such as agriculture and water resources, aiming at the sustainable management of water resources and resilience to climate risk.

Background: The occurrence of economic losses in buffaloes may be related to reproductive problems such as chlamydiosis caused by the bacteria Chlamydia abortus considered as a zoonotic agent; and digestive problems highlighting the infection by Mycobacterium avium subsp. paratuberculosis (Map), responsible for paratuberculosis. There is a little information about these diseases in buffaloes, therefore the aim of this study was to determine the occurrence of anti-Chlamydia abortus and anti- Mycobacterium avium subsp. paratuberculosis (Map) antibodies in water buffaloes in the state of Pernambuco, Brazil.Materials, Methods & Results: The 262 bubaline sera belonging to the serum bank of the Infectious Diseases Laboratory (LIDIC) of the Federal Rural University of Pernambuco (UFRPE) were analyzed. The samples were from nine properties distributed in the municipalities of Agreste and Zona da Mata of the state of Pernambuco. For the detection of anti-Chlamydia abortus and anti-Map antibodies was used a technique of Enzyme immunoabsorption Assay (ELISA) of the IDEXX® by following the manufacturer's instructions. Regarding the detection of anti-Chlamydia abortus antibodies, it was observed that 47.70% (125/262) of the samples were positive. All properties showed at least one positive animal for the investigation of anti-Chlamydia abortus antibodies. It was also verified the occurrence of 7.25% of suspected animals for the investigation of anti-C. abortus antibodies. No positives animals were observed for Map in the bubaline analyzed.Discussion: The occurrence of anti-C. abortus antibodies in buffaloes in the region can be explained by the fact that properties with buffalo breeding has sanitary management that allows the contact between animals, thus increasing the risk of transmission of the agent. In addition, the variation found from 35% to 68.75% may be associated with divergences to the type of management and breeding system used in each property. The percentage of suspected animals may suggest that the number of positive animals is higher or that there were non-specific reactions with other species of Chlamydia, but it is not possible to determine the seroconversion without the accomplishment of paired serology. Because it is a zoonosis, C. abortus may be a risk to the health of the population involved, since the transmission of the bacteria to humans can occur by contact with secretions and excretions of these animals. No positive animals were found for the occurrence of anti-Map antibodies, however, there are reports of the infection identifying animals with clinical signs or properties with a history of the disease, and this may be related to the differences in each property in relation to the history and sanitary management. Moreover, the ELISA test may not be sensitive to the agent depending on the phase of the infection, because if the response is predominantly cellular, the number of antibodies is diminished, making the serological diagnosis difficult. The use of more sensitive tests for the bubaline species can also favor the diagnosis of the infection. Although the occurrence of anti-Map antibodies in the analyzed samples was not detected, it is important to carry out routine epidemiological studies, since the disease has already been registered in water buffaloes in the state of Pernambuco. Considering the occurrence of anti-Chlamydia abortus antibodies it is suggested to conduct studies in order to isolate the agent and verify its real importance in the reproductive disorders of the bubaline species, since this agent causes reproductive losses and has a zoonotic character.

Paratuberculosis (PTB) is a disease which causes considerable economic losses to producers of livestock, particularly dairy cows. Nowadays PTB is one of the most prevailing and costly infectious diseases of dairy cattle. The purpose of the present study was to review economic losses, which may be caused by Mycobacterium avium subsp. paratuberculosis (MAP) above all in herds of dairy cattle. The most important losses caused by the presence of clinically ill animals have been thoroughly described: loss of milk production and poor body condition followed by death or culling. In contrast, losses arising from a subclinical disease have not been well documented and contradictory results have been published to date. The calculation of losses caused by PTB depends to a certain degree on the production system in a herd, efficiency level, herd management system and other factors. Direct economic losses are above all caused by decreased milk production concurrent to increased incidence of mastitis, changes in milk parameters and increased somatic cell counts, reproductive dysfunctions, poor feed conversion, shortened production age and increased predisposition to other diseases etc. Indirect economic losses are caused by premature culling of animals and their unrealized future income, expenses for non-active production, herd replacement, diagnostic testing, &ldquo;unnecessary&rdquo; veterinary care and establishing disease control programmes. Genetic value of animals and their progeny is lost. Last but not least, the reputation of the farm where MAP infected animals are kept is lost for a long-time, which is also important.

Bovine anaplasmosis is one of the most important tick borne diseases of ruminants worldwide causing significant economic losses in the livestock industries due to the high morbidity and mortality in susceptible cattle herds. Bovine anaplasmosis, caused by Anaplasma marginale, is an infectious but non-contagious disease. The mode of transmission of bovine anaplasmosis includes mechanical (blood contaminated fomites (needles, ear tagging, dehorning and castration equipment), biological (tick bites) and transplacental (mother to fetus). Bovine Anaplasmosis occurs in tropical and subtropical regions worldwide. Cattle of all ages are susceptible to infection with A. marginale, but the severity of disease increases with age. The common clinical sign of bovine anaplsmosis includes; fever, anorexia, rapid loss of body condition, severe decrease in milk production, pale and icteric mucous membranes, increased heart and respiratory rates, muscle weakness and depression. Diagnosis of bovine anaplasmosis can be made by demonstration of A. marginale on stained blood smears from clinically infected animals during the acute phase of the disease, but it is not reliable for detecting infection in pre-symptomatic or carrier animals. Instead, serological demonstration of antibodies and confirmation of antigen with molecular detection tools are used for diagnosis. Anaplasmosis can be treated by administration of oxytetracyclines, however oxytetracyclines do not clear the parasite from carrier animals. Control measures for bovine anaplasmosis vary with geographical location and include maintenance of Anaplasma free herds, vector control, administration of antibiotics and vaccination. Intensive acaridae application to control ticks has a number of limitations, therefore, immunization together with strategic tick control is recommended for exotic and crossbred cattle. Further studies on epidemiology of bovine anaplasmosis is needed

In Brazil, Trypanosoma vivax is present in several states. The disease is endemic in Pantanal and Minas Gerais. In Paraná there is still no report of the parasite, but due to the state borders with Mato Grosso do Sul, São Paulo, Paraguay and Argentina, it is believed that the protozoan circulates in the region without diagnosis. The objective of this study was to investigate the prevalence of T. vivax in dairy cattle in the western region of Paraná. For this purpose, 600 blood samples and 400 serum samples were collected from dairy cattle, distributed in 60 and 40 properties, respectively. While buffy coat smears were performed on blood samples, serum samples were used in Indirect Immunofluorescence Reaction. All samples, in both techniques, presented negative results for T. vivax. These results indicate that the studied hemoprotozoan is not circulating among the cattle in the western region of Paraná. However, future work evolving beef cattle must be carried out and preventive measures should be adopted in order to avoid the entry of the parasite in the State of Paraná.

Poiretia punctata (Willd.) Desv. was associated with cattle and sheep poisoning on nine farms in the State of Sergipe, northeastern Brazil. The animals were found dead or died later after showing clinical signs for up to 18 hours. Two sheep that ingested 40g/kg body weight (g/kg) of fresh P. punctata died three and eight hours after ingestion, respectively. Another sheep that ingested 40g/kg five days after plant collection showed mild clinical signs and recovered after 24 hours. Two sheep that received 20g/kg and another that ingested three daily doses of 20g/kg showed clinical signs, but recovered. Two cattle that ingested 20g/kg of the fresh plant exhibited clinical signs and recovered. The clinical observations of poisoning were depression, ataxia, loss of equilibrium, broad-based stance, head down, falls, mandibular trismus, opisthotonous, nystagmus, and recumbence. Significant gross and histologic lesions were not observed. Samples of P. punctata were analyzed for nitrates, cyanogenic glycosides, and sodium monofluouracetate with negative results. It is concluded that P. punctata is a toxic plant that caused death in cattle and sheep in the State of Sergipe.
