In an in vivo experiment the 2 × 2 Latin square method and Cr as a marker were used to determine ruminal degradation and postruminal digestion of nutrients from a feed ration containing 3 kg of crushed or ground maize (CM vs. GM). The experiment included 4 cows with rumen fistulae and duodenal T-cannulae. Amino acid passage to the duodenum was also determined. The mean concentration of ammonia nitrogen in the rumen fluid was 11.6 ± 6.22 and 11.5 ± 6.41 mg/100 ml in the CM and GM ration, respectively. Crushing considerably decreased starch degradation in the rumen where starch passage reached 33.5% with CM and only 21.5% with GM. Both post-ruminal (82.2% vs. 85.5%) and total digestibility of starch (94.0% vs. 96.8%) was high. The passage of other nutrients was not influenced by crushing. With both feed rations higher amounts of crude protein and amino acids passed to the duodenum than were ingested (127.8% vs. 130.4% and 118.1% vs. 124.5%, respectively). In comparison with intake the highest increase was observed in essential lysine (156.5% and 165.0%, respectively) and glycine (261.1% and 280.2%, respectively). Apparent crude protein digestibility was at the level of 68.5% (CM) and 67.2% (GM).

This study aimed to evaluate the effect of intraruminal infusion of propionic acid (RPA) associated to two (80:20 and 60:40) forage:concentrate ratios (FCR), for young bulls. Four Holstein × Zebu crossbred young bulls with average age of 7 months and 160 kg of body weight (BW) were distributed to a 4 × 4 Latin square design. A total mixed ration, isonitrogenous diets, was ad libitum fed to animals. The propionic acid was obtained by fermentation of reconstituted milk whey by the consortium of bacteria Enterococcus sp. and Veillonella sp.. Daily and along with the supply of solid feeds, 2 L of the compound containing propionic acid were intrainfused in the rumen. There was no RPA × FCR interaction for the intake of nutrients, except for total digestible nutrients (TDN). The organic matter (OM), crude protein (CP), ether extract (EE), neutral detergent fiber (NDF) and TDN intakes were influenced by the FCR. There was no RPA × FCR interaction on the total digestibility of DM, OM, CP, EE, NDF and TC. However, the digestibility of DM, OM, EE and CT were influenced by the FCR. There was no RPA × FCR interaction for the mean N-NH3 concentration and pH of the rumen fluid; however, RPA provided effect on the pH measured 4 hours after meal. There was no interaction or effect of the main factors on volatile fatty acids concentrations in the rumen fluid, or on glucose levels and serum urea nitrogen.

This study aimed to evaluate the inclusion of 0.0%, 0.4%, 0.8%, and 1.2% slow release urea (SRU) in sheep feed on the intake and total digestibility coefficient (DC) of nutrients and ruminal parameters of sheep living in tropical regions. Four sheep with a body weight (BW) average of 30.8 ± 1.7 kg were assigned to a Latin square design. The animals were placed in metabolic cages and received two meals a day. Intake data and DC of nutrients of the sheep were subjected to analysis of variance and differences were tested using a regression equation to 5% probability. The inclusion of 0.0%, 0.4%, 0.8%, and 1.2% SRU in sheep feed did not change (p > 0.05) the intake of dry matter (DM), organic matter (OM), crude protein (CP), ether extract (EE), neutral or acid detergent fiber (NDF, ADF), total carbohydrates (TC), and non-fibrous carbohydrate (NFC) expressed in g animal-1 day-1, g (kg0.75)-1, and % BW. The mean values of 3.04%, 2.82%, 0.41%, 0.07%, 1.36%, 0.87%, 2.31%, and 1.00% of BW were obtained for the intake of DM, OM, CP, EE, ANF, ADF, TC, and NFC, respectively. The inclusion of 0.0%, 0.4%, 0.8%, and 1.2% of SRU in sheep rations had a quadratic manner (p < 0.05) the DC of DM and CP, with maximum values of 69.67% and 63.59%, respectively, for 0.51% and 0.66% levels of SRU inclusion in the experimental diets. The pH of the rumen fluid did not vary (p > 0.05) for different levels of inclusion of SRU in the feed. However, the pH of the rumen fluid did differ (p < 0.05) in relation to time (T) after feeding for all experimental diets. The inclusion levels of SRU in sheep feed did not affect (p > 0.05) the concentration of ammonia nitrogen (NH3-N) of the rumen fluid, but there was a quadratic effect (p < 0.05) with relation to T for all experimental diets. Thus, we conclude that the inclusion of 0.66% SRU provides the best digestibility value of CP and is the appropriate value for the digestion of DM. The inclusion of up to 1.2% of SRU in sheep rations did not change the intake of nutrients and ruminal parameters.

Abstract Eight rumen-cannulated Nellore steers (327 ± 30 Kg BW) were used in a 4x4 duplicate Latin square to evaluate the effects of two feed additives (MON - Sodium Monensin 26 mg/kg DM vs. Crina® RumistarTM - combination of blend of essential oils - 90 mg/kg DM + exogenous α-amylase - 560 mg/kg DM, respectively; DSM Produtos Nutricionais Brasil S.A.) and two roughage sources at 10% of peNDF (CS - corn silage vs SB - sugarcane bagasse, on DM basis). Final diets contained 19.5% CS or 12% SB, corn grain (70.7 and 78.2% for CS and SB diets, respectively), 5.5% soybean meal, 4% mineral-vitamin mixture and 0.3% urea, on DM basis. The experimental periods had 14-d for adaptation and 5-d of collections. Data were analyzed with PROC MIXED of SAS and means compared by Tukey test. No interaction between additives and roughage sources was observed. Animals fed Crina® RumistarTM had 0.600 kg increase (P = 0.03) on DM intake and decreased (P = 0.01) apparent crude protein digestibility (CP; 75.3 vs. 77.5%, respectively) compared to MON. Animals fed SB presented greater apparent CP digestibility (77.6 vs 75.2%, respectively; P = 0.003) compared to CS. No difference in apparent NDF digestibility for additives and roughage source was observed. Animals fed Crina® RumistarTM had lower acetate concentration (P = 0.01) compared to MON on time 9, 12, 15, 18 and 21 hours. Animals fed CS had higher acetate:propionate (P = 0.03) compared to SB at all collection times–24 hours. Ruminal ammonia-nitrogen was lower (P = 0.01) in animals fed Crina® RumistarTM compared to MON. To use Crina® RumistarTM increased DM intake and decreased ruminal ammonia-nitrogen compared to monensin, suggesting a higher CP by-passing the rumen. Crina® RumistarTM increased DM intake without negative impacts on nutrients digestibility, regardless roughage source, thus a higher amount of nutrients reaches the intestine to be absorbed and animal performance can be increased.

AbstractRuminal bacterial counts and in vivo digestibility were determined on four Mediterranean buffalo bulls and four Friesian bulls, all fistulated at the rumen, and given at maintenance level (50 g/kg M0·75 per day of dry matter) four different diets with the same crude protein content (N ✕ 6·25 = 140 g/kg dry matter) and with forage: concentrate ratios as follows: diet D12·5 = 0·875: 0·125; diet D25·0 = 0·75: 0·25; diet D37·5 = 0·625: 0·375; diet D50·0 = 0·5: 0·5. All the animals received the diets during four consecutive periods in a Latin-square design. Buffaloes had higher total microbial counts (10·78 v. 10·08 log10cells per g dry rumen content, P < 0·01) as compared with cattle; differences in total ruminal bacterial counts among the diets were only observed within the buffalo species (diet D12·5 v. diets D25·0, D37·5, D50·0: 10·04 v. 10·92, 10·98, 11·17 log10cells per g dry rumen content, P 0·01) and when comparing the two species for each diet, significantly higher values for bacterial counts in buffaloes were found for diets D25·0: 10·92 v. 10·28 (P 0·05), D37·5: 10·98 v. 10·08 (P 0·01) and D50·0: 11·17 v. 9·76 (P 0·01) log10cells per g dry rumen content. Cattle showed significantly higher digestibility values for: organic matter (0·696 v. 0·676, P 0·05), neutral-detergent fibre (NDF; 0·548 v. 0·511, P 0·05) and cellulose (0·621 v. 0 · 509, P 0·01), while the crude protein digestibility (CPD) values were similar (0·667 and 0·671). Comparing the two species for each diet, cattle showed significantly higher digestibility values for organic matter in diet D50·0 only (0·714 v. 0·688, P 0·01), for NDF in diet D12·5 only (0·578 v. 0·531, P 0·05) and for cellulose in all diets (0·660 v. 0·546, 0·630 v. 0·525, 0·605 v. 0·505, 0·588 v. 0·460, P 0·01); in contrast buffaloes showed higher values of the CPD for diet D12·5 (0·662 v. 0·632, P 0·05).

ABSTRACT The objective of this study was to evaluate the effect of replacing Tifton 85 hay with maniçoba hay in diets based on spineless cactus on the nutrient intake and digestibility, ingestive behaviour and ruminal parameters of confined sheep. In order to do this, eight male sheep with ruminal cannulae were used, distributed across four levels (0, 333, 666 and 1.000 g kg-1 of dry matter) of replacement of Tifton 85 hay with maniçoba hay in a double Latin square experimental design. The animals were confined for 60 days divided into four periods of 15 days. The replacement of Tifton 85 hay with maniçoba hay did not influence the dry matter intake or digestibility. However, it influenced in a positive linear fashion the intake of non-fibrous carbohydrates and in a linear negative fashion the digestibility of insoluble fibre in neutral detergent and crude protein. The ruminal ammonia-nitrogen and the crude protein ruminal content decreased linearly with the replacement of Tifton 85 hay with maniçoba hay. There was no effect of hay replacement on the production of volatile fatty acids or the microbial biofilm. The total replacement Tifton 85 by maniçoba hay in spineless cactus diets for sheep did not influence total digestible nutrient intake, volatile fatty acid production or biofilm, but did affect crude protein digestibility, ammonia-nitrogen and crude protein content in the rumen. Therefore, maniçoba hay can replace 300 g kg-1 grass hay in spineless cactus diets for sheep without compromising intake and ruminal parameters.

Tarramba leucaena (Leucaena leucocephala cv. Tarramba) foliage had per kilogram dry matter, 169 g protein and 29.8 g condensed tannins. Its value as a supplement, given either with or without urea, to sheep given a low-quality Callide Rhodes grass (Chloris gayana cv. Callide) hay was studied. Six rumen fistulated sheep (mean s.d. liveweight, 34 1.4 kg) were used to compare 6 dietary treatments in an incomplete latin square design. Rhodes grass hay was given ad libitum either alone, or with urea 7 g/day (U), or with leucaena 150 g/day (L150), or leucaena with urea (L150U), or leucaena 300 g/day (L300), or leucaena with urea (L300U). Digestible organic matter intake was increased significantly by leucaena supplementation although digestibility of the whole diet did not alter. Rumen fluid ammonia-N was not altered by leucaena supplementation, but was increased by urea. This suggests that Tarramba foliage protein has some resistance to ruminal degradation. Liquid and solids passage rates were not affected by the treatments. Microbial nitrogen supply to the intestine (g/day), and the efficiency of microbial nitrogen synthesis (g/kg organic matter apparently digested in the rumen), were increased by leucaena supplementation (P<0.01). Microbial protein synthesis was the only response in which 300 g/day air-dry Tarramba foliage gave improved results over 150 g/day.

Abstract Binding of 63Ni(Il) to ultrafiltrable constituents of rabbit serum was studied (a) after in vitro incubation (2 h, 37 °C) of rabbit serum with 63NiCl2 (10-100 µmol/liter), and (b) at intervals (0.25-2 h) after in vivo administration of 63NiCl2 (40-160 µmol/kg body wt, i.v.). Serum ultrafiltrates were fractionated by thin-layer chromatography, and the separated compounds made visible by autoradiography and by ninhydrin staining. Several (≃5) ultrafiltrable 63Ni-complexes were demonstrable as distinct radiodense 63Ni-bands with chromatographic mobilities corresponding to those of ninhydrin-positive bands. Unbound 63Ni(II) was not detected in serum ultrafiltrates in either the in vitro or in vivo experiments. In sera (n = 10) incubated in vitro with 63Ni(II) (10 µmol/ liter), the mean percentage of ultrafiltrable 63Ni was 36% (range = 33-38) of total serum 63Ni. In contrast, in sera (n = 10) obtained 2 h after i.v. injection of 63Ni(II) (40 µmol/kg), the mean concentration of total serum 63Ni was 10.8 µmol/liter (range = 6-14), and the mean percentage of ultrafiltrable 63Ni was 15% (range = 9-21) of total serum 63Ni. The disparity between the percentages of ultrafiltrable 63Ni obtained in vitro and in vivo was obviated when the in vivo experiments were performed in rabbits bilaterally nephrectomized, with ligated common bile ducts. This investigation confirms the existence of several nickel receptors in serum ultrafiltrates and substantiates the role of ultrafiltrable complexes in the excretion of nickel.

The effects of glycerol with fat sources as a feed alternative were investigated in the ration for Nellore steers (Bos indicus). Eight cannulated steers at 30 months of age with initial bodyweight of 554 ± 36.0 kg were used in a double 4 by 4 Latin square design with four consecutive 16-day periods. Treatments were three different diets with lipid sources (soybean, soybean oil and calcium salts of fatty acids) and one diet control without lipid sources. All diets formulated contained 10% crude glycerol and 5% ether extract with a forage:concentrate ratio of 35:65; corn silage was used as forage. The ruminal liquid was sampled for 24 h and ruminal fermentations were monitored by measuring pH, concentrations of ammonia nitrogenand volatile fatty acids in rumen fluid. Urine samples were obtained from the total collection for 24 h for estimation of rumen microbial protein supply using urinary purine derivatives. Our results showed that the use of lipid sources combined with glycerol did not induce significant changes in rumen pH, acetate molar proportion, ruminal microbial protein or dry matter intake. Although the acetate molar proportion was kept constant within normal parameters, the propionate molar proportion was increased by the diet containing lipid sources. Moreover, we found that there was a negative effect of lipid sources on crude protein and neutral detergent fibre corrected for ash and protein intake, and one positive effect on ether extract intake. We believe that association between fat and glycerol may affect rumen fermentation parameters through reducing fibre intake and increasing propionate production and ammonia nitrogen.

AbstractAnimal by-product meals have large variability in crude protein (CP) content and digestibility. In vivo digestibility procedures are precise but laborious, and in vitro methods could be an alternative to evaluate and classify these ingredients. The present study reports prediction equations to estimate the CP digestibility of meat and bone meal (MBM) and poultry by-product meal (PM) using the protein solubility in pepsin method (PSP). Total tract CP digestibility of eight MBM and eight PM samples was determined in dogs by the substitution method. A basal diet was formulated for dog maintenance, and sixteen diets were produced by mixing 70 % of the basal diet and 30 % of each tested meal. Six dogs per diet were used to determine ingredient digestibility. In addition, PSP of the MBM and PM samples was determined using three pepsin concentrations: 0·02, 0·002 and 0·0002 %. The CP content of MBM and PM ranged from 39 to 46 % and 57 to 69 %, respectively, and their mean CP digestibility by dogs was 76 (2·4) and 85 (2·6) %, respectively. The pepsin concentration with higher Pearson correlation coefficients with the in vivo results were 0·0002 % for MBM (r 0·380; P = 0·008) and 0·02 % for PM (r 0·482; P = 0·005). The relationship between the in vivo and in vitro results was better explained by the following equations: CP digestibility of MBM = 61·7 + 0·2644 × PSP at 0·0002 % (P = 0·008; R2 0·126); and CP digestibility of PM = 54·1 + 0·3833 × PSP at 0·02 % (P = 0·005; R2 0·216). Although significant, the coefficients of determination were low, indicating that the models were weak and need to be used with caution.

In vitro protein digestibility of protein sources has been correlated with in vivo digestibility values. However, factors like protein origin, enzyme used and its concentration, pH and processing have been related with the significance of the correlation between the estimates. To address only the enzyme concentration factor, this paper had the objective of testing pepsin at 0.2, 0.02, 0.002 and 0.0002% using the standard AOAC (1995) procedure. Two meat and bone meals (MBM) with low and high crude protein (CP) content were used to determine the coefficient of solubility of CP in pepsin and HCl (CSCPPEPH). Centrifugation was used to establish the nitrogen (N) in the soluble phase, instead of filtration and analysis of N in the residue. The variance analysis and a non-linear asymptotic model were adjusted. The CSCPPEPH under different pepsin concentrations for the two MBM showed higher solubility discrimination with low pepsin concentration. The level of 0.0002% pepsin is better to predict the CP soluble in MBM. This finding implies the assumption that 0.2% pepsin found in the AOAC is not correct for the purpose of determining the range of solubility in high and low CP content in MBM.
