Background: Methionine (Met) requirements have not been clearly established for fattening pigs due to their metabolic interrelationships and its bioavailability for protein synthesis. Objective: To determine the optimum level of regular crystalline or protected Met in pig diets from nursery to finishing. Methods: A total of 48 crossbred pigs (11.74±1.72 kg of initial body weight) were used. The treatments consisted of adding four levels (0.00, 0.05, 0.10, 0.15% in addition to dietary content) and two types of Met (regular and protected) to pig diets. Results: Nursery, Finishing I, and II pigs fed protected Met increased daily feed intake (DFI; p≤0.10). Protected Met raised daily weight gain (DWG) in nursery pigs and increased backfat thickness (BT) in nursery and grower pigs (p≤0.10). In Finishing I pigs, protected Met increased DWG and improved carcass characteristics (p≤0.10). In nursery and grower pigs, an extra 0.15% Met decreased feed:gain ratio (FGR; p≤0.10). In grower and Finishing II pigs fed extra 0.05% Met improved DWG and extra 0.10% Met reduced plasma urea concentration (p≤0.10). Conclusions: Feeding protected Met in pig diets increases DWG, DFI and BT. Increasing 0.05-0.15% Met level improves FGR, DWG, potentially reducing nitrogen excretion to the environment.

The nitrogen excretion was studied in the one-humped camel, Camelus dromedarius. When a growing camel was maintained on a low N intake (dates and hay) the amount of N excreted in the form of urea, NH3 and creatinine decreased to 2–3 gm/day. This decrease was caused by a drop in urea excretion from 13 gm to 0.2–0.5 gm/day. Urea given intravenously during low N intake was not excreted but was retained. (The camel like other ruminants can utilize urea for microbial synthesis of protein.) The renal mechanism for urea excretion was investigated by measuring urea clearance and glomerular filtration rate during a period of 7 months. During normal N intake about 40% of the urea filtered in the glomeruli were excreted in the urine while during low N intake only 1–2% were excreted. The variations in urea clearance were independent of the plasma urea concentration and of glomerular filtration rate, but were related to N intake and rate of growth. No evidence of active tubular reabsorption of urea was found since the urine urea concentration at all times remained higher than the simultaneous plasma urea concentration. The findings are not in agreement with the current concept for the mechanism of urea excretion in mammals. It is concluded that the renal tubules must either vary their permeability to urea in a highly selective manner or secrete urea actively.

The objective of this study was to evaluate performance, quantitative characteristics of carcass, and visceral responses of barrows maintained on heat stress enviroment and fed diets supplemented with ractopamine. It was used 48 animals with initial weight of 67.3 ± 3.8 kg, distributed in a randomized block design with four levels of ractopamine (0, 5, 10 and 20 mg/kg), each one with six replicates with two animals each. The experimental period lasted 28 days. Air temperature was 31.8 ± 2.0ºC, air humidity was 72.6 ± 10.2% and BGHI was 82.7 ± 2.8. Daily feed intake was not affect by the levels of ractopamine but they improved feed conversion and increased daily weight gain and final weight of the animals. The carcass quantitative characteristics were not affected by levels of ractopamine. The weights of the liver and kidneys showed linear increases accordingly to increase of ractopamine levels in the diet whereas weight of other organs and length of small intestine were not affected by supplementation with ractopamine. The optimum level of ractopamine for the best performance of barrows under heat stress is 20 mg/kg and it does not affect the quantitative characteristics of carcass.

Supplementation of 5 ppm of ractopamine, associated or not to feed restriction in diets with a high total lysine content (1.04%) was evaluated on performance and nitrogen balance. In experiment 1, 60 hybrid castrated male swine (76.2 ± 2.3 kg) were housed in pairs according to a randomized complete block design in a factorial treatment arrangement (2 ×2 + 1) with or without ractopamine supplementation, two forms of feeding, ad libitum and feed restriction with 1.04% lysine, and an additional treatment (control), with ad libitum feed without ractopamine with 0.8% lysine, amounting to five treatments and six replications. The final live weight, daily weight gain, daily feed intake and feed conversion for 28 days were evaluated. There were improvements in supplementation with ractopamine for final live weight, daily weight gain and feed conversion. The ad libitum feeding improved both final live weight and daily weight gain compared with ractopamine. Compared with the control, there was a decrease in feed conversion with ractopamine supplementation and improvement for final weight (FW) in the treatment with the ractopamine and ad libitum feed. In experiment 2, 30 hybrid castrated male swine (74.1 ± 2.5 kg) were housed in metabolism cages and fed the same experimental treatments to evaluate the percentages of absorbed nitrogen (Nabs), retained nitrogen (Nret), nitrogen retained from the absorbed nitrogen (Nret/Nabs) and the plasma urea concentration at 14 and 28 days. At 14 days, there was an increase in the Nret/Nabs and plasma urea concentration with ractopamine supplementation. Compared with the control, there was a decrease in the plasma urea concentration for feed restriction with ractopamine. At 28 days, there was an increase in the Nret/Nabs with ractopamine supplementation. Thus supplementation with 5 ppm ractopamine improved the performance and efficiency of swine nitrogen use. Feed restriction interfered negatively in weight gain, but it improved the efficiency of the nitrogen use of barrows.

Two experiments were carried out with the objective of evaluating the performance and carcass traits of growing and finishing pigs fed rations with sticky coffee hull silage. In experiment 1, the coffee hulls were ground through a 4-mm screen and ensiled with 30% water and enzymatic-bacterial inoculant and evaluated in digestibility trial with 15 crossbred pigs distributed in a completely randomised design. Overall, the ensiling process did not improve the digestibility of the sticky coffee hulls. In experiment 2, it was used 60 pigs (32.52 to 59.58 kg) in the growing phase and 55 pigs (61.70 to 90.27 kg) in the finishing phase, distributed in a completely randomised design with five diets (0, 4, 8, 12 and 16% of sticky coffee hull silage) and six replicates. In the growing and finishing phase, inclusion of levels of sticky coffee hull silage did not affect feed intake, weight gain, and plasma urea nitrogen. However, in the finishing phase, feed conversion improved as the levels of sticky coffee hull silage increased. Responses by backfat thickness and marbling were quadratic, whereas empty stomach weight increased linearly when sticky coffee hull silage was included in the diet. Sticky coffee hull silage has good nutritional value and if used in levels up to 16% of the diet, it does not impair performance of pigs in the growing and finishing phases and it results in leaner carcasses. However, the economic feasibility of its use depends on the price relationship of this by-product with the other feedstuffs.

Abstract Background: The influence of PAD4 concentration and its polymorphisms in SAKI development are poorly evaluated. Thus, the aim of this study is to evaluate the PAD 4 concentration and PADI4 polymorphisms, as predictors of AKI development, need for renal replacement therapy (RRT), and mortality in patients with septic shock. Methods: We included all individuals aged ≥ 18 years, with the diagnosis of septic shock at ICU admission. Blood samples were taken within the first 24 hours of the patient’s admission to determine serum PAD4 concentration and its polymorphism PADI4 (rs11203367) and (rs874881). Patients were followed during their ICU stay and the development of SAKI was evaluated. Among the patients in whom SAKI developed, mortality and need for RRT were also evaluated. Results: 99 patients were included in the analysis. SAKI developed in approximately 51.5% of patients during the ICU stay; of these, 21.5% required RRT and 80% died. There was no difference between PAD4 concentration (p = 0.116) and its polymorphisms rs11203367 (p = 0.910) and rs874881 (p = 0.769) in patients in whom SAKI did or did not develop. However, PAD4 had a positive correlation with plasma urea concentration (r = 0.269 and p = 0.007) and creatinine (r = 0.284 and p = 0.004). The PAD4 concentration and PADI4 polymorphisms were also not associated with RRT and with mortality in patients with SAKI. Conclusion: PAD4 concentration and its polymorphisms were not associated with SAKI development, the need for RRT, or mortality in patients with septic shock. However, PAD4 concentrations were associated with creatinine and urea levels in these patients.

SUMMARYThe effect of rumen fermentation on the relationship between urea and glucose kinetics was examined in sheep fed chopped lucerne hay with intraruminal infusions of water, urea, sucrose, or urea plus sucrose at Palmerston North, New Zealand in 1986. Sheep were fed hourly and infused intraruminally with water (1200 m1/day), or a similar volume containing either urea alone (13·7g/day), sucrose alone (178·2 g/day) or urea (14·6 g/day) plus sucrose (175·0 g/day). The added sucrose resulted in a lower rumen ammonia concentration (P< 0·05), lower plasma urea concentration (P< 0·05) and reduced urinary urea excretion (P< 0·05). Urea recycled to the gut tended to increase with the sucrose, urea or sucrose plus urea treatments compared with the water treatment. The fermentation of sucrose in the rumen resulted in decreases in ruminal pH (P< 0·05) and in the ratio of acetate to propionate (A:P) (P< 0·05). The infusion of sucrose also increased the concentration of propionate in rumen fluid (P< 0·05), tended to increase the plasma glucose level and increased plasma glucose irreversible loss (P< 0·05). The infusion of urea resulted in an increase in the plasma urea level (P< 0·05), urea pool size (P< 0·05) and urea irreversible loss (P< 0·01). However, urea infusion did not affect glucose metabolism or volatile fatty acid (VFA) fermentation. The effects of sucrose infusion on glucose and urea kinetics were broadly similar when given alone or with urea, apart from changes in the urea degradation rate. It was concluded that the additional fermentative activity resulting from sucrose increased propionate production which, in turn, was available for glucose production, thus ‘sparing’ amino acids for tissue protein utilization and reducing urea excretion.

Abstract Objective The insecticidal activity of Bacillus thuringiensis directly depends on the yield of delta-endotoxins. In this study, various nutritional and cultural parameters influencing delta-endotoxin synthesis by a local isolate of B. thuringiensis Se13 were investigated using Taguchi methods. Methods In the first experiment, four factors, incubation period, incubation temperature, initial pH and medium, each at four levels, were selected and an orthogonal array layout of L16 was carried out. In the second experiment, Taguchi’s orthogonal array method of L27 was used to evaluate the effects of the different concentration of medium components. Taguchi’s signal–noise ratio and variance analysis were applied to determine the effect of the factors. After each experiment, verification studies were carried out using determined optimum conditions. Results The optimum conditions for incubation period, incubation temperature, initial pH, and medium determined as 72 h, 30°C, pH 9, and M4 medium, respectively. In the second experiment, soybean flour (5%), glucose (5%), KH2PO4 (0.3%), K2HPO4 (0.1%), MgSO4 (0.4%) were determined as the optimum conditions. The delta-endotoxin yield was elevated to 1559.25 μg mL−1 when the factors were adjusted to optimum level. Conclusion Optimization using the Taguchi method appeared to be a good choice for the overproduction of delta-endotoxin.
