The aim of ultrastructural investigation is to analyze cell architecture and relate a functional role(s) to cell components. It is known that aqueous chemical fixation requires seconds to minutes to penetrate and stabilize cell structure which may result in structural artifacts. The use of ultralow temperatures to fix and prepare specimens, however, leads to a much improved preservation of the cell’s living state. A critical limitation of conventional cryofixation methods (i.e., propane-jet freezing, cold-metal slamming, plunge-freezing) is that only a 10 to 40 μm thick surface layer of cells can be frozen without distorting ice crystal formation. This problem can be allayed by freezing samples under about 2100 bar of hydrostatic pressure which suppresses the formation of ice nuclei and their rate of growth. Thus, 0.6 mm thick samples with a total volume of 1 mm3 can be frozen without ice crystal damage. The purpose of this study is to describe the cellular details and identify potential artifacts in root tissue of barley (Hordeum vulgari L.) and leaf tissue of brome grass (Bromus mollis L.) fixed and prepared by high-pressure freezing (HPF) and freeze substitution (FS) techniques.

The article raises the problem of ensuring metrological traceability of the measurement results of indicators of quality and nutritional value for food products and food raw materials: water (moisture), nitrogen (protein, crude protein), fat, ash and carbohydrates. The problem under consideration can be solved by applying reference materials of food composition, traceable to state primary measurement standards GET 173-2017 and GET 176-2019 and primary reference measurement procedures (PRMP), for attestation of measurement procedures and accuracy checking of measurement results. The article discusses the results of the PRMP development of mass fraction of fat, ash and carbohydrates in food products and food raw materials, as well as mass fraction of crude fat (oil content) in oil crops seeds and products based on them. The paper also presents metrological characteristics of reference materials of composition of dry dairy products, grain-milk dry porridges for nutrition of babies, grain dry porridges for nutrition of babies, egg powder, freeze-dried meat products, animal feed. The results of the work allow for building a chain of metrological traceability from GET 173-2017, GET 176-2019 and PRMP to routine measurement procedures, thereby ensuring the uniformity of measurements of nutritional value of food products.

Problems of providing the population and agricultural production by qualitative potable and process water in the Central Yakutia are covered. This territory belongs to the region with acute shortage of water resources which is always a limiting factor of development of agricultural production. For the solution of this burning issue in the 80th years of the last century along the small rivers the systems of hydraulic engineering constructions providing requirements with process water practically of all settlements of the Central Yakutia were constructed. At a construction of all hydraulic engineering buildings the method of construction with preservation of soils of the basis of constructions in a frozen state was applied. When warming the climate which is observed in recent years hydraulic engineering constructions built in regions of a wide spread occurance of breeds of an ice complex and with the considerable volume of water weight, were especially vulnerable. On character and a design they experience continuous threat of damage and demand very attentive relation from the operating organizations. Taking this into account, safe operation of hydraulic engineering constructions in a zone of distribution of permafrost breeds demands new approaches. The article examines features of hydraulic engineering constructions’ operation of agricultural water supply objects in the Central Yakutia. Distinctiveness of hydraulic engineering constructions’ operation is that stability of constructions is intimately bound to temperature impact of a reservoir on ground dams’ body and the basis of constructions. The possibility of inclusion of ways for an intensification of a freezing of constructions in the structure of operational actions is studied. The new method on safe operation of hydraulic engineering constructions as prewinter abatement of the water level in a reservoir accounting volumes and norms of water consumption of the settlement is offered.

In recent years, the number of pasture species and cultivars commercially available to farmers in New Zealand has increased significantly. Reasons for this include increased commercial activity in the development and supply of proprietary cultivars, and more specialisation in New Zealand's pastoral agriculture. In particular, pasture cultivars are increasingly marketed as proprietary products with associated technical support. This year there are 109 certified cultivars available, belonging to 23 grass, legume and herb species. As a result, farmers are now able to develop improved pasture feed supply, but they are somewhat confused about pasture species and cultivars, and this frustrates their aims to produce high quality animal products from productive pastures. Accordingly, the herbage seeds industry needs to work from a clear reference list, to avoid any confusion when supplying farmers and overseas clients with their seed requirements. Our list of pasture species, types and cultivars available within New Zealand aims to be that reference. Flour of Anodonta woodiana meat may be added in the manufacture of food products such as granola bar. The aim of addition is to increase the nutritional value on snacks needed by children with special needs through fortification. This research method was done by making products granola bar in three formulas: without the addition of shells flour, the addition of 2.8 gram, and 5.6 gram. Granola bar were made for consumption by children (7-9 years old) and teenager (10-18 years old). Tests performed included organoleptic test. The results of organoleptic test between three formulas of produced granola bar have different characteristics in flavor parameter. The results of the analysis of parameters of color, aroma, and flavor tested by analysis of variance software showed that the most preferred formula is the second formula with the addition 2.8 gram of mussel meat flour. This granola bar snack is a healthy snack that is safe for consumption by children with special needs, because it is made from ingredients that do not contain flavorings, dyes and artificial preservatives.
