Background: RGL mandarin is one of the important mandarin citrus varieties in Indonesia. The tolerance of RGL mandarin citrus to water deficit can be induced by the rootstock. This study aimed to characterize the physiological responses and transcriptional gene expression of RGL mandarin citrus grafted onto three rootstock genotypes during the dry and the rainy seasons.Methods: Three-years-old mandarin citrus trees cv. Rimau Gerga Lebong (RGL) grafted onto three citruses (JC, Cit and K) were planted at the experimental field of the ICISFRI. The experiment was conducted with a randomized block design and each scion-rootstock combination contained five replications. All of the physiological and relative gene expression parameters observed were conducted at the last of the dry season (September 2019) and the mid of the rainy season (January 2020). Water deficit was induced by no watering plants during the dry season.Result: The results of physiological responses and relative gene expression analyses showed that RGL-Cit combination had better than RGL-JC combination and RGL-JC combination had better than RGL-K combination. RGL-Cit combination showed less wilt than RGL-JC and RGL-K combinations. The RGL-Cit combination had the highest stomatal density, stomatal aperture, stomatal conductance and photosynthetic rate in the dry season. Different plant combinations showed different gene expressions. RGL-JC and RGL-K combinations were upregulated in almost all the primers related to drought responses tested in the dry season. RGL-Cit combination only upregulated the PIP1, PIP2 and ACS2 and unregulated the others in the dry season. Therefore, citrumelo seems to be a valuable type of rootstock.

Purpose. To study the effect of duodenal-jejunal omega switch (DJOS) in combination with different dietary patterns on the retinol-binding protein (RBP4), fetuin-A, and fibroblast growth factor 21 (FGF21) plasma levels and their hepatic gene expressions in rats. Methods. A high-fat diet (HF) was given to 28 rats and 28 more were fed with a control diet (CD) for 2 months. After that, half of each group underwent either DJOS or SHAM surgery. For the next 2 months, half of the animals in each operation group were kept on the same diet as before and half of them had the diet changed. After 16 weeks of the experiment RBP4, fetuin-A, and FGF21 plasma levels as well as liver Rbp4, Ahsg, and Fgf21 gene expressions were measured. Results. DJOS had a reductive impact on plasma levels of RBP4, fetuin-A, and FGF21 and Rbp4, Ahsg, and Fgf21 relative gene expression in the liver when compared to SHAM. The HF/HF group expressed significantly higher RBP4 and fetuin-A plasma levels in comparison to the control. The HF diet used before and/or after surgery led to upregulation of Rbp4, Ahsg, and Fgf21 relative gene expression. The lowest levels of analyzed parameters were observed in the CD/CD group. Conclusions. The efficiency of DJOS surgery, measured by hepatokines’ plasma levels and their gene expressions in the liver, depends on the type of diet applied before and after surgery. Manipulation of dietary patterns can lead to marked improvements in metabolic profile after DJOS surgery.

Abstract Background: Androgen-Deprivation Therapy (ADT) is a standard treatment for advanced prostate cancer (PCa). However, there is a high recurrence or progression rate during ADT. Until now, there is no evidence on when the progression starts. This study would like to evaluate the early response of intraprostatic androgen receptor (AR) and steroidogenic enzyme gene expressions in ADT.Methods: Prostate tissue samples were taken from PCa patients with urinary retention, who had ADT (ADT- PCa; n=10), and further grouped into ≤12 months (n=4) and ADT >12 months (n=6). ADT-PCa group were then compared with BPH (n=12) and primary (no treatment) PCa tissues (n=16). AR and steroidogenic enzyme genes were extracted from Formalin Fixed Paraffin embedded (FFPE) tissues and analysed using rtPCR. Protein expressions were evaluated by immunohistochemistry of specific antibodies. Results: AR gene expression was found higher in ADT-PCa group compared to BPH and primary PCa. Both ADT ≤12 and > 12 months subgroups had significantly higher relative gene expression of AR (p 0.01 and 0.03) compared to primary PCa. AR protein expression in ADT-PCa group showed an increase trend in ADT ≤12 months subgroup and a significantly elevated expression AR protein in ADT >12 months subgroup compared with PCa (100%; p <0.01). Half (50%) of ADT ≤12 months patients were found to have upregulation of AR, and one undergone upregulation from only 3 months of ADT. A trend of elevating relative gene expression of SRD5A3 were also found within the groups given ADT. Conclusion: There are upregulation of AR and steroidogenic enzymes in ADT-PCa patients, as early as 3 months without showing PSA elevation. Steroidogenic enzyme, especially SRD5A3 expression was also showing upregulation before PSA rises.

Aim. To determine the levels of PTEN relative expression (RE) and the possible effects of its loss on some tumor-associated genes and histological characteristics of human prostate tumors. Methods. PTEN RE and 46 genes were identified in 37 pairs of adenocarcinoma and conventionally normal tissues (CNT) and 20 adenomas by real time PCR. Results. We are shown PTEN RE decreasing in the adenocarcinoma group as well as in CNTs compared with adenomas. 54 % of adenocarcinomas, 48.6 % of CNT and 25 % of adenomas were detected with low PTEN RE. We have found 9 of 46 differentially expressed genes in groups with different levels of PTEN RE (ESR1, GCR, KRT18, MMP2, MMP9, SRD5A2, VIM, PCA3, HOTAIR). It was revealed some histological characteristics correlated with PTEN RE in prostate adenocarcinomas. Conclusions. PTEN aberrations in prostate tumors indicate that PTEN associated changes already have been occurred at the benign tumors. We have shown some tumor histological characteristics associated with PTEN expression changes.Keywords: prostate tumors, relative gene expression, PTEN, EMT, histological characteristics.

Increasing evidence implicates intervertebral disc (IVD) degeneration as a major contributor to low back pain. In addition to a series of pathogenic processes, degenerated IVDs become vascularized in contrast to healthy IVDs. In this context, angiopoietin (Ang) plays a crucial role and is involved in cytokine recruitment, and anabolic and catabolic reactions within the extracellular matrix (ECM). Over the last decade, a progenitor cell population has been described in the nucleus pulposus (NP) of the IVD to be positive for the Tie2 marker (also known as Ang-1 receptor). In this study, we investigated the influence of Ang-1 and Ang-2 on human NP cell (Tie2+, Tie2- or mixed) populations isolated from trauma patients during 7 days in normoxia (21% O2) or hypoxia (≤ 5% O2). At the end of the process, the proliferation and metabolic activity of the NP cells were analyzed. Additionally, the relative gene expression of NP-related markers was evaluated. NP cells showed a higher proliferation depending on the Ang treatment. Moreover, the study revealed higher NP cell metabolism when cultured in hypoxia. Additionally, the relative gene expression followed, with an increase linked to the oxygen level and Ang concentration. Our study comparing different NP cell populations may be the start of new approaches for the treatment of IVD degeneration.
