We studied the effect of unstable ubiquitous endocrine disrupters bisphenol A (BPA) and triclosan (TCS) on the reproductive function of adult male Wistar rats. BPA and TCS concentrations were measured directly in testicular tissue by gas chromatography with mass spectrometry (GC-MS). Comparison groups were found to be statistically significantly different in the concentrations of bisphenol A and triclosan in testicular tissues. The study showed that bisphenol A and triclosan have a negative effect on the morphofunctional state of the male reproductive glands of sexually mature rats, causing a decrease in testicular mass, a decrease in parenchyma, the number and diameter of the seminiferous tubules, an increase in the number of convoluted seminiferous tubules with desquamated epithelium, and a decrease in sperm cells on the background of an increase in their atypical forms.

The aimof this study was to analyze the morphofunctional state of the testes of mature male rats treated with bisphenol A (BPA) and triclosan (TCS).Material and methods.The work was performed on mature male rats (n=28). Experimental animals were divided into three groups – control (intact) and two experimental ones. For two months, rats of the experimental groups received daily bisphenol A and triclosan (Sigma-Aldrich, USA) with food in the amount of 200 mg/kg. The total count of spermatozoa in 1 ml of sperm was determined with the calculation of their atypical forms, and morphometric measurements were made (the total number and area of Leydig cells with their nuclearcytoplasmic ratio). The concentrations of bisphenol A and testicular triclosan were measured by gas chromatography with mass spectrometry (GC-MS). The obtained data was subjected to statistical processing using IBM SPSS Statistics v.21 (IBM Corp., Armonk, NY, USA).Results.Differences between the comparison groups in the concentration of bisphenol A and triclosan in testicular tissues were statistically significant (p <0.001). The endocrine disruptors studied reduced the mass of the testes. A group of male rats exposed to bisphenol A showed a decrease in the total number of spermatozoa (p=0.004) with an increase in their atypical forms (p=0.014) compared with a group of intact animals. Bisphenol A and triclosan caused a decrease in the total number of Leydig cells (p=0.001; p=0.001) respectively, and a statistically significant change in the nuclear-cytoplasmic ratio. Moreover, bisphenol A led to a decrease in the nuclear-cytoplasmic ratio of Leydig cells, whereas triclosal, on the contrary, increased its value in comparison with a group of intact animals.Conclusion.Bisphenol A and triclosan have a negative effect on the morphofunctional state of the male testes of sexually mature rats (decrease in testicular mass, total count of spermatozoa on the background of an increase in their atypical forms, as well as a decrease in the total number of Leydig cells with a change in their nuclear-cytoplasmic ratio).

Cadmium disrupts the blood-testes barrier, interferes with various antioxidant levels thus enhancing lipid peroxidation and ultimately leading to apoptosis and necrosis of testicular tissue. Moringa oleifera is a medicinal plant and a rich source of essential phytochemicals possessing antioxidant properties. The effect of aqueous leaf extract of M. oleifera on reproductive function following cadmium chloride induced oxidative stress in male Wistar rats was investigated. Forty adult male Wistar rats were assigned into five groups of eight rats each. Treatment was administered orally daily as follows: Group 1 (control): animal feed and tap water ad libitum; Group 2: 5 mg kg-1 cadmium chloride for 21 days; Group 3: 500 mg kg-1 of M. oleifera and 5 mg kg-1 of cadmium chloride for 21 days; Group 4: 5 mg kg-1 cadmium chloride for 21 days followed by 500 mg kg-1 M. oleifera for the next 35 days; Group 5: 5 mg kg-1 cadmium chloride for 21 days followed by 750 mg kg-1 M. oleifera for the next 35 days. At the end of treatment, blood was obtained by direct cardiac puncture for fertility hormone assay and testicular tissue specimens were harvested for semen analysis and determination of antioxidant levels. Results obtained indicated that rats treated with the various extracts had significantly increased superoxide dismutase, malondialdehyde and catalase levels, increased serum concentrations of testosterone, follicle stimulating hormone and luteinizing hormone and increased percentage of viable and normal spermatozoa compared to control and only cadmium chloride treated rats (p < 0.05). The results obtained suggest that treatments with M. oleifera extract could ameliorate possible cellular damage caused by administration of cadmium chloride.

Abstract Introduction: AAzathioprine and Methotrexate are both used in the treatment of cancer; and are classified as cytotoxic drugs with reported adverse effects such as oxidative damage to the DNA/RNA, the testes and sperm cells. This study, therefore, tested the hypothesis that AAzathioprine and Methotrexate administrations impair the morphology and functions of the testes in adult male wistar rats. Methods: AAzathioprine (50-150mg per day) and Methotrexate (2.5mg per week) are used in the treatment of cancer in adult Man. We tested the hypothesis that AAzathioprine and Methotrexate impair the morphology and functions of testes in rats. Forty adult male wistar rats (150-230g) were employed in the study: Control Group I received physiological saline while Experimental Groups II - V received oral administrations of 5mg/kg/bodyweight of AAzathioprine per day, 15mg/kg/bodyweight of AAzathioprine per day, 8mg/kg/bodyweight of Methotrexate per week and 20mg/kg/bodyweight of Methotrexate per week respectively for 35 days. Results: Histological examinations of the testes of rats of Groups II - V showed dose-dependent morphological anomalies such as fewer collagen ibers of connective tissues, disrupted seminiferous tubules and scanty spermatozoa when compared to rats of Group I. Statistical analyses showed dose-dependent elevated levels (P≤0.05) of superoxide dismutase and malondialdehyde in testes homogenates of rats of Groups II - V when compared to rats of Group I. This implied increased oxidative stress in rats of Groups II - V. Evaluations of Follicle Stimulating Hormone and Testosterone showed dose-dependent significantly elevated levels (P≤0.05) in rats of Groups II - V when compared to rats of Group I. Conclusions: Our findings are consistent with the stated hypothesis.

Abstract Introduction Antipsychotics are drugs that are widely prescribed for mental disorders, such as schizophrenia and psychosis. Recent in vitro studies show antipsychotics play a role in the initiation of neuronal cell apoptosis. This study aims to determine the effect of haloperidol and risperidone on neuronal cell apoptosis in Wistar white rats. Methods Male wistar rats aged 8 weeks (n = 30) were used in this study. Wistar rats were randomized into 6 groups. Group A: 5 wistar rats as a control without induced schizophrenia, aquades and drugs. Group B: 5 Wistar-induced psychotic mice (using 30 mg / kgBB ketamine, intraperitoneal injection for 5 days) and aquadest. Group C: 5 rats were induced psychotic and were given haloperidol or 0.05 mg / kgBB orally, for 28 days. Group D: 5 mice were induced psychotic and were given haloperidol 0.1 mg / kg orally, for 28 days. Group E: 5 mice were induced psychotic and were given risperidone 0.05 mg / kgBB orally, for 28 days. Group F: 5 mice were induced psychotic and given risperidone 0.1 mg / kgBB orally, for 28 days. Apoptosis of neuronal cells in the ventral tegmental area was assessed by caspase-3 immunohistochemistry. The colored area will be calculated as a total percentage using the imageJ program. Results Risperidone and haloperidol increase caspase-3 activity, but haloperidol increases caspase-3 activity more than risperidone. Conclussion Risperidone and haloperidol induce apoptosis of neuronal cells and tardive dyskinesia in Wistar rats with psychotic models.

This study evaluated the ameliorative and prophylactic effects of 2 different doses of polyphenolic-rich fraction of Garcinia kola (PPRF Gk) seeds on the histology and hormones of pituitary–testicular axis of male Wistar rats. Thirty-five male Wistar rats (150-200 g) were divided into 7 groups of 5 rats each. Groups I and II were given distilled water (0.5 mL/day) for 8 days followed by propylene glycol (0.2 mL/d) and 600 mg/kg of PPRF Gk, respectively, for 21 days. Group III received sodium arsenate (8 days), left untreated for 21 days. Groups IV and V received sodium arsenate (20 mg/kg) for 8 days followed by PPRF Gk (300 and 600 mg/kg, respectively) for 21 days. Groups VI and VII received PPRF Gk (300 and 600 mg/kg, respectively) for 21 days followed by sodium arsenate (20 mg/kg) for 8 days. Rats were killed by cervical dislocation 24 hours after the last dose and their blood collected through cardiac puncture. Blood sera were assayed for the levels of follicle-stimulating hormone (FSH), luteinizing hormone (LH), and testosterone using immunoassay techniques. Histology of the pituitary gland and testes was carried out. A significant reduction was observed in the concentration of FSH in groups IV, V, VI, and VII in comparison with groups I and II. The concentrations of both LH and testosterone showed significant decreases in groups IV, V, VI, and VII in comparison with group I. Group III presented with the lowest serum hormonal concentrations. Photomicrographs of the pituitary gland revealed greatly reduced basophils in group III and mildly reduced basophils in groups IV, VI, and VII in comparison with groups I and II. Group V revealed hypercellularized and distorted basophils. Photomicrographs of the testes showed detachment of the seminiferous tubules from the basement membrane and disruption of the interstitial space which was worse in group III, moderate in groups V and VI, and mild in group VII. In conclusion, PPRF Gk effected a dose-dependent reversal and prevention of the perturbations caused by arsenate in rats.

To study the probable negative influence of the complex remedy �Tireoton� on the reproductive function of the laboratory animals the following tests have been carried out, namely the probable occurrence of embryo-toxicity, teratogenicity, foetotoxicity and its influence on the postnatal development of the laboratory animals progeny. The experiments have been carried out of the white Wistar rats. It has been established that the �Tireoton� at the doses of 75 and 750 mg/ kg has no embryotoxic, teratogenic and fetotoxical effect. The introduction of the �Tireoton� at the dose of 750 mg/kg since 6th to 19th day of pregnancy does not influence on the postnatal development of white rats progeny.

The use of herbs as an adjutant form of medical treatment is gaining popularity all over the world. This study is aimed at determining the effects of Indian liquorice on the fertility potentials of male Wistar rats. Fifty-five rats divided into seven groups were used in this study. Doses of 200 and 400mg/kg of the extract were used. The parameters considered were the weight of the testis, malondialdehyde concentration, hormonal levels, histological characteristics and sperm analysis. We observed a significant change in the weight of the testes, inhibition of testicular tissue peroxidation, testosterone levels, sperm concentration and motility were changed in comparison to the negative control. Obvious cellular changes were also observed in the liquorice-treated groups. These changes were brought back to almost pre-treatment values in the reversal groups. The changes observed in groups co-treated with vitamin E were minimal and statistically insignificant. The results suggest that the seed extract of Indian liquorice has a negative effect on the fertility potentials of the male rats and shows oxidative properties.
