This work focused on pre-formulation studies for freeze-dried protein formulations, using albumin as a model protein. Two aminoacids and two sugars were selected as lyoprotectors and studied according a mixture design (simplex-lattice). Sample characterization included total protein content, collapse temperature and visual inspection. Sugar formulations collapsed, but not aminoacids, which indicate non sugar excipients as the best choice under the studied conditions.

A study was conducted at Summerland, BC, in 2000 and 2001 to investigate the effect of harvest period on the protein content of sea buckthorn leaves. Leaves of both male and female plants were collected bi-weekly during the growing season and analyzed with a LECO FP-528 nitrogen analyzer after air- and freeze-drying. A conversion factor of kA = 5.7 was applied to obtain the protein content. Total protein content in sea buckthorn leaves showed very small differences between male and female plants and from either freeze-dried or air-dried materials. There were significant fluctuations among harvest periods in both years. Based on the results of this experiment, sea buckthorn leaves should be harvested from late July to early August since leaf protein content peaked around this period of time and started to decline significantly by the middle of August. Probiotic «Evitaliya» is freeze–dried, specific strains of lactic acid microorganisms (Lactococcus lactis, Streptococcus thermophilus, Lactobacillus acidophilus, Lactobacillus helveticus, Propionibacterium fredenreichii ssp shernanii), they contain vitamins B1, B2, B6, B12, A, E, C iron minerals, calcium, magnesium.The purpose of the study was to clarify the effect of the probiotic ferment «Evitaliya» on nonspecific resistance of kids zaanovskoy breed microclimate conditions according VNTP (for sheep and goat breeding enterprises): the room temperature 12–14 °C, relative humidity of 55 – 70%, the carbon dioxide content – 1.5 – 1.8 l/m3 and ammonia – 10 – 15mg/m3 air bacterial contamination 15 – 20 tys. KOE/m3. Implementation of this goal was decided to use hygienic (temperature, relative humidity, air velocity); hematological (red blood cells, white blood cells, hemoglobin), biochemical (total protein, albumin, globulin); immune (humoral factors – lysozyme and bactericidal activity of blood serum, cell – the neutrophil phagocytic activity), statisticsGetting results and their novelty: for the first time tested the new probiotic yeast «Evitaliya», with its goats orally at a dose of – 200 ml/head in the morning and evening, 100 ml.It was found that the use of «Evitaliya» promotes activation of redox processes in the body kids, stimulation of erythropoiesis: increase in hemoglobin concentration of 0.56% (P ≤ 0.05), the number of red blood cells to – 10.2% (P ≤ 0.05).Use of the drug is moderately activates metabolic processes in the body of kids: increases total protein content by 7.14% (P ≤ 0.05), globulin – by 1.76% (P ≤ 0.05). Use of the drug «Evitaliya» at the indicated doses helps stimulate natural resistance olrganіzmu kids: humoral protection indicators (BASK) rose by 1.52% (LASK) by 1.81%, the cell indices (FA) compared to the control were higher – 4.97%.

Lyophilized vegetative mycelium of ectomycorrhizal fungi was subjected to various viability tests to confirm functional integrity. Physical integrity of freeze-dried cultures was comparable to that of non-lyophilized cultures. Inter- and intraspecific variations in morphology, physiology, and metabolic rate were maintained after lyophilization. Maintenance of total protein content confirmed metabolic stability. According to the assays of viability, a plating assay and determination of total biomass confirmed stable mitotic activity of the freeze-dried cultures.Key words: lyophilization, physical integrity, structural stability, total protein, total biomass.

Abstract The aim of the this study was to evaluate the effects of fresh, dried and freeze-dried Centaurea depressa M. Bieb. (Asteraceae) on the oxidant and antioxidant status of the model organism D. melanogaster Meigen (Diptera: Drosophilidae) experimentally. The study was carried out from 2016 to 2019, and plant leaf extracts (0-50 mg/l) were added to insect standard artificial diets. The total protein, protein carbonyl content and glutathione-S-transferase, superoxide dismutase and catalase activities were quantified at the insect’s third larval stage. Our data showed that protein carbonyl content varied from 2.70 nmol/mg protein in the control group to 59.11 nmol/mg protein in the group fed with fresh leaf extract signifying induction of oxidative stress. All extracts increased the levels of all antioxidant enzymes and decreased the amounts of total protein. Meanwhile, the group fed with the freeze-dried extract showed no significant difference in the levels of total protein and protein carbonyl content except at the 50 mg/l concentration of the extract. Moreover, this group had superoxide dismutase and catalase activities 4 to 5 times higher than in the control group. In conclusion, induction of oxidative stress indicates that the fresh form of C. depressa leaves may have potential as a natural pesticide, whereas induction of endogenous antioxidant enzymes by the freeze-dried extract suggest its potential as an antioxidant.

The color of a therapeutic monoclonal antibody solution is a critical quality attribute. Consistency of color is typically assessed at time of release and during stability studies against preset criteria for late stage clinical and commercial products. A therapeutic protein solution’s color may be determined by visual inspection or by more quantitative methods as per the different geographical area compendia. The nature and intensity of the color of a therapeutic protein solution is typically determined relative to calibrated standards. This review covers the analytical methodologies used for determining the color of a protein solution and presents an overview of protein variants and impurities known to contribute to colored recombinant therapeutic protein solutions.

Lipidomics provides an overview of lipid profiles in biological systems. Although blood is commonly used for lipid profiling, cerebrospinal fluid (CSF) is more suitable for exploring lipid homeostasis in brain diseases. However, whether an individual’s background affects the CSF lipid profile remains unclear, and the association between CSF and plasma lipid profiles in heathy individuals has not yet been defined. Herein, lipidomics approaches were employed to analyze CSF and plasma samples obtained from 114 healthy Japanese subjects. Results showed that the global lipid profiles differed significantly between CSF and plasma, with only 13 of 114 lipids found to be significantly correlated between the two matrices. Additionally, the CSF total protein content was the primary factor associated with CSF lipids. In the CSF, the levels of major lipids, namely, phosphatidylcholines, sphingomyelins, and cholesterolesters, correlated with CSF total protein levels. These findings indicate that CSF lipidomics can be applied to explore changes in lipid homeostasis in patients with brain diseases.

In our previously described Hep G2/24-hour/total protein assay, protein levels were measured by using the Lowry method. This assay was the best acute in vitro assay for the prediction of human toxicity within the Multicentre Evaluation of In Vitro Cytotoxicity (MEIC) study. In order to increase the MEIC data-base with a wider range of chemicals, we were interested in introducing the more practical 3-(4-carboxybenzoyl)-quinoline-2-carboxaldehyde (CBQCA) method for the quantification of the total protein content. Therefore, we investigated whether the same good results for the prediction of acute human toxicity would be obtained with the CBQCA method. The cells were treated for 24 hours, then cytotoxicity was determined by measuring the total protein content with CBQCA. The results were quantified by using the PI50c: the concentration (in mM) of test compound required to reduce the total protein content measured with the CBQCA-method by 50% as compared to the control cells. The results were compared with the PI50, the corresponding value when the Lowry method was used. A relatively low correlation was observed between PI50 and PI50c, reflecting the large and unexpected, differences when using the two protein assays. However, when comparing the log PI50c with the human toxicity, a correlation coefficient of r2 = 0.761 ( n = 44) was obtained for exactly the same series of MEIC chemicals. This value is clearly higher than that for the Lowry method ( r2 = 0.695). Compared to the Lowry method originally used, the Hep G2/24-hour/CBQCA total protein assay has the additional important advantage that it can be very easily adapted for large-scale analyses with robotic systems, including the on-line calculation of the results.
