ABSTRAK<br />Penanganan benih yang baik sangat diperlukan untuk mempertahan-<br />kan viabilitas benih nilam yang dikirim ke daerah yang jauh dari kebun<br />induk. Benih yang tidak dikemas dengan baik akan berakibat menurunnya<br />viabilitas benih dengan cepat, sehingga pada saat ditanam benih tidak<br />dapat tumbuh (mati). Untuk itu telah dilaksanakan penelitian yang<br />bertujuan untuk mengetahui pengaruh jenis kemasan setek berakar<br />terhadap pertumbuhan nilam. Percobaan dilaksanakan di rumah kaca Balai<br />Penelitian Tanaman Obat dan Aromatik (Balittro) dari bulan Desember<br />2004 sampai dengan Maret 2005. Percobaan disusun dalam rancangan<br />acak lengkap (RAL) terdiri atas tujuh perlakuan dengan 3 ulangan.<br />Perlakuan yang diberikan adalah : (1) setek nilam dibalut tissue dan<br />dibungkus plastik transparan, (2) setek nilam dibalut koran dan dibungkus<br />plastik transparan, (3) setek nilam dibalut cocopeat dan dibungkus plastik<br />transparan, (4) setek nilam dibalut tissue dan dibungkus karung plastik, (5)<br />setek nilam dibalut koran dan dibungkus karung plastik, (6) setek nilam<br />dibalut cocopeat dan dibungkus karung plastik, (7) kontrol (setek tidak<br />dibalut dan tidak dibungkus). Hasil percobaan menunjukkan bahwa<br />persentase hidup setek berakar nilam yang disimpan selama 7 hari pada<br />semua kemasan kecuali kontrol masih tinggi yaitu &gt;90%. Hampir dari<br />seluruh parameter pertumbuhan (tinggi tanaman, jumlah tunas dan jumlah<br />daun) yang diamati menunjukkan pengemasan setek nilam dibalut tissue<br />dan dibungkus plastik transparan lebih baik dibandingkan dengan<br />pengemasan setek yang lain.<br />Kata kunci : Nilam, Pogostemon cablin, benih, setek berakar, jenis<br />kemasan, pertumbuhan, Jawa Barat<br />ABSTRACT<br />Effect of packing materials on the quality and growth of<br />patchouli rooted cuttings<br />Good seed handling should be carried out seriously to maintain the<br />cutting viability of pacthouli especially when the cutting distributed to<br />other region far away from the seed garden. Improper cutting packing will<br />decrease of viability the cutting very fast and when it is planted the cutting<br />will not grow.Therefore, the research was conducted to find out the effect<br />of packing material on the growth of rooted cutting. The experiment was<br />conducted in the green house of the Indonesian Research Institute for<br />Aromatic and Medicinal Crops Bogor from December 2004 to March<br />2005, and it was arranged completely in a randomized design (CRD) with<br />3 replications, consisted of 7 packing treatments. The treatments were : (1)<br />rooted cuttings wrapped using transparent plastic with tissue media, (2)<br />rooted cuttings wrapped using transparent plastic with newspaper media,<br />(3) rooted cuttings wrapped using transparent plastic with cocopeat media,<br />(4) rooted cuttings wrapped using plastic sack with tissue media, (5) rooted<br />cuttings wrapped using plastic sack with newspaper media, (6) rooted<br />cuttings wrapped using plastic sack with cocopeat media, (7) control<br />(unwrapped seedling). The results indicated that after 7 days of storage,<br />rooted cuttings of patchouli were still viable (&gt;90%) in all treatments<br />except control. Packing of rooted cuttings by wrapping it with transparent<br />plastic with tissue media showed the best growth with more number of<br />leaves and higher plant compared to other packing treatments.<br />Key words : Patchouli, Pogostemon cablin, seedling, rooted cutting,<br />packing material, growth, West Java

ABSTRAK<br />Dalam upaya pengembangan nilam (Pogostemon cablin) di daerah<br />yang jaraknya jauh dari kebun induk, pengadaan benih nilam yang berkua-<br />litas menjadi masalah yang serius, karena bibit akan cepat mengalami<br />penurunan kualitas selama transportasi. Untuk itu dilaksanakan penelitian<br />yang bertujuan untuk mengetahui pengaruh lama simpan setek berakar<br />nilam terhadap pertumbuhan. Percobaan dilaksanakan di rumah kaca Balai<br />Penelitian Tanaman Rempah dan Obat (Balittro) dari bulan April –<br />Agustus 2004. Percobaan disusun dalam rancangan petak terbagi (RPT)<br />dengan 3 ulangan. Petak utama (main plot) adalah 2 jenis setek nilam<br />yaitu: (1) setek berdaun dan (2) setek tidak berdaun. Anak petak (sub plot)<br />adalah lama penyimpanan setek yaitu: (1) setek langsung ditanam<br />(kontrol), (2) setek disimpan 1 hari, (3) setek disimpan 3 hari, (4)setek<br />disimpan 5 hari dan, (5) setek disimpan 7 hari. Pengamatan dilakukan<br />sejak tanaman berumur 2 minggu sampai tanaman berumur 8 minggu.<br />Parameter yang diamati meliputi pertumbuhan tanaman (tinggi, jumlah<br />daun dan jumlah tunas), bobot kering (batang, daun, akar). Hasil perco-<br />baan menunjukkan bahwa persentase hidup setek nilam berakar (setek<br />berdaun dan setek tidak berdaun) masih 100% setelah disimpan selama 7<br />hari. Hampir dari seluruh parameter pertumbuhan (tinggi tanaman, jumlah<br />cabang, jumlah daun) yang diamati menunjukkan bahwa pertumbuhan<br />bibit setek berdaun lebih baik dibandingkan dengan setek yang tidak<br />berdaun.<br />Kata kunci : Nilam, Pogostemon cablin Benth, bibit, setek berakar,<br />penyimpanan, pertumbuhan, Jawa Barat<br />ABSTRACT<br />Effect of storage periods of rooted cutting on the growth<br />of patchouli (Pogostemon cablin Benth)<br />Providing high quality of patchouli (Pogostemon cablin Benth)<br />seedlings is necessary to support the development of patchouli plants. In<br />the new developing area transportation become serious problems (high<br />cost transportation), therefore some alternative solution is reducing the<br />transportation cost without reducing the quality of the seedlings. Base on<br />the problems, this experiment was conducted. The objective of this<br />experiment was to study the effect of storage periods of rooted cuttings on<br />the growth of patchouli plant. The experiment was conducted in the green<br />house of Indonesian Spice and Medicinal Crops Research Institute<br />(ISMECRI), from April to August 2004. The experiment was arranged in<br />a split-plot design with 3 replications. The main plot was 2 kinds of cutting<br />there were: (1) leaf cutting and (2) non leaf cutting. The sub plot was 5<br />different storage periods, there were: (1) control (no storage), (2)1 day<br />storage, (3) 3 day storage, (4) 5 day storage, (5) 7 day storage. The<br />variables observed were plant growth (plant height, number of leaves and<br />number of shoots), dry weight of stems, leaves and roots. The results of<br />experiment indicated that after 7 day storage, rooted cuttings of patchouli<br />were still 100% viable. The growth of patchouli from leaf cutting was<br />significantly different from non leaf cutting. Patchouli plant from leaf<br />cutting produced higher plant height, number of leaves, number of shoots<br />and dry weight of plants compared to those of patchouli plants from non<br />leaf cuttings. Mindi besar (Melia dubia Cavanilles) is one of fast growing tree species from family Meliaceae which growing in tropical evergreen forests. The aim of this study was to determine the best combination of medium and growth regulator for vegetative propagation of mindi besar trees. The completely randomized factorial design was used two factors; medium (M) there were two types of media; zeolite (M1) and soil + rice husk (2:1 v/v) medium (M2), the growth regulator (H) had three levels; control (H0), pure coconut water (H1), and Auksin sintetis (H3). The study was conducted at two locations, first at green house with rooting room KOFFCO System and second at seedbed with rooting room containment model (MS). The research at greenhouse using rooting room KOFFCO System showed that interaction of media with growth regulator had a very significant effect on the percentage of fresh cuttings and rooted cuttings and had a significant effect on the number of primary roots. The averages of percentage of fresh cuttings and rooting were 83.3% and 66.7% for M1H2 and 76% and 60% for M2H0. The M2H1 reached 2.9 number of primary roots. At the MS model, the medium factor had significantly different only the number of secondary roots (18.11). The growth regulator factors had significantly different on the percentage of rooted cutting (3.33%), root lenght (10.5 cm) and root dry weight (0.18 g).Key words: containment model, KOFFCO system, Rootone-F, zeolite.

<p>Skrining fitokimia dimaksudkan untuk melakukan evaluasi pendahuluan tentang kandungan kimia pada teh (Camellia sinensis). Selain itu, teh mengandung katekin yang dapat digunakan sebagai petunjuk kualitas dari daun teh. Penelitian bertujuan mengetahui kandungan senyawa aktif dan kadar katekin pada teh. Penelitian dilaksanakan mulai bulan April sampai dengan Juni 2012 di laboratorium Pengujian Balai Penelitian Tanaman Rempah dan Obat, Bogor. Skrining fitokimia pucuk peko dengan dua daun (p+2) dilakukan berdasarkan prosedur dari Materia Medika Indonesia (MMI), sedangkan analisis katekin dengan menggunakan metode SNI gambir. Penelitian disusun berdasarkan rancangan acak lengkap (RAL), enam perlakuan dengan empat ulangan. Perlakuan yang digunakan adalah enam genotipe teh (Tbs 1, Tbs 2, Hibrid, Cin 143, Rb 3, dan Kiara 8). Hasil penelitian menunjukkan keenam genotipe yang diuji mengandung senyawa alkaloid, saponin, tanin, fenolik, flavanoid, steroid, dan glikosida. Genotipe Tbs 1, Hibrid, dan Kiara 8 positif mengandung senyawa triterpenoid, sedangkan Tbs 2, Rb 3, dan Cin 143 negatif. Genotipe Tbs 1 dan Tbs 2 memiliki kandungan katekin paling tinggi (kecuali bagian ruas+tangkai daun) dibandingkan dengan empat genotipe lainnya. Pucuk peko, daun pertama, dan daun kedua pada genotipe Tbs 1 memiliki kadar katekin masing-masing 17,92%, 11,73%, dan 14,67%, sedangkan pada genotipe Tbs 2 masing-masing 18,22%, 13,48%, dan 15,81%. Kadar katekin terendah dihasilkan oleh bagian ruas+tangkai daun pada genotipe Rb 3 (1,78%). Pucuk peko menghasilkan kandungan katekin bervariasi antara 8,36%-18,22%, lebih tinggi dibandingkan dengan daun pertama, daun kedua, dan bagian ruas + tangkai daun.</p><p>Kata kunci: Camellia sinensis, fitokimia, genotipe, katekin, pucuk peko</p><p>Phytochemical screening was intended for a preliminary evaluation of the chemical constituents of the tea (Camellia sinensis). In addition, tea also contains catechin that can be used as an indication of the quality of tea leaves. The objectives of this study were to determine the content of the active compounds and catechin in tea. The research was conducted from April to June 2012 in the Laboratory of the Research Institute for Spices and Medicinal Crops, Bogor. The phytochemical screening was performed based on the procedure of Materia Medika Indonesia (MMI), while the catechin analysis used the method of SNI gambir. The study was carried out in completely randomized design with six treatments and four replications. The treatments used are six tea genotypes namely Tbs 1, Tbs 2, Hibrid, Cin 143, Rb 3, and Kiara 8. The results showed that the six tea genotypes tested contained the compounds of alkaloid, saponin, tannin, phenolic, flavanoid, steroid, and glycoside. Positively triterpenoid compounds present in the genotype of Tbs1, Hybrids, and Kiara 8, and negative in Tbs 2, Rb 3, and Cin 143. The genotypes of Tbs 1 and Tbs 2 produced the highest catechin content compared to the other genotypes. Catechin content was lowest in the part of internodes+leaf stalk of Rb 3 (1.78%). Pecco shoots produce catechin content of about 8.36%-18.22%, higher than the first leaf, second leaf, and the parts of internodes+leaf stalk.</p>

ABSTRACT Grape orchards are highly affected by oviposition of fruit flies on grape berries, which compromises the productivity and quality of the grapes. The goal of this study was to evaluate the susceptibility of American, European, and hybrid grape genotypes to Anastrepha fraterculus and Ceratitis capitata (Diptera: Tephritidae) under laboratory conditions (25 ± 2ºC, 60 ± 10% RH, and 14-hour photophase). The assays were conducted by evaluating oviposition preference through choice and no-choice (antibiosis) tests. The choice test was set up using circular arenas (diameter: 300 mm) with 10 grapes per genotype. The no-choice test was set up using 150 grapes per genotype which were placed inside plastic boxes (417 x 297 x 289 mm). After exposure of the grapes to one or two couples of fruit flies per genotype in choice and no-choice tests, respectively, the grapes were transferred to transparent plastic containers (750 mL). The number of eggs per grape and its viability were evaluated. The no-choice test also evaluated the biological cycle of the fruit flies. The assays were conducted in a completely randomized design with 15 replicates. The most preferred grapes for oviposition by A. fraterculus were Cabernet Sauvignon, Niagara Rosada, and BRS Cora, while C. capitata mainly preferred Isabel Precoce grapes. We observed the complete development of fruit flies in the Moscato Embrapa grapes, but this only occurred with C. capitata. All genotypes evaluated were considered susceptible to A. fraterculus and C. capitata. However, the fruit flies expressed differences in preference for oviposition and host quality.

ABSTRACT Ectomycorrhizal fungi (EMF) may improve the adaptation of eucalypts saplings to field conditions and allow more efficient fertilizer use. The effectiveness of EMF inoculum application in promoting fungal colonization, plant growth, nutrient uptake, and the quality of rooted cuttings was evaluated forEucalyptus urophylla under commercial nursery conditions. For inoculated treatments, fertilization of the sapling substrate was reduced by 50 %. The experiment was carried out in a completely randomized design in a 4 × 4 factorial arrangement, wherein the factors were inoculum application rates of 0 (control), 5, 10, and 15 gel beads of calcium alginate containing the vegetative mycelium of Amanita muscaria, Elaphomyces antracinus, Pisolithus microcarpus, andScleroderma areolatum, plus a non-inoculated treatment without fertilization reduction in the substrate (commercial). Ectomycorrhizal fungi increased plant growth and fungal colonization as well as N and K uptake evenly. The best plant growth and fungal colonization were observed for the highest application rate. The greatest growth and fungal colonization and contents of P, N, and K were observed at the 10-bead rate. Plant inoculation with Amanita muscaria, Elaphomyces anthracinus, and Scleroderma areolatum increased P concentrations and contents in a differential manner. The Dickson Quality Index was not affected by the type of fungi or by inoculum application rates. Eucalypt rooted cuttings inoculated with ectomycorrhizal fungi and under half the amount of commercial fertilization had P, N, and K concentrations and contents greater than or equal to those of commercial plants and have high enough quality to be transplanted after 90 days.

Plant inoculation with ectomycorrhizal fungi (EMF) maximizes the productive potential of forest stands. Thus, the inoculation efficiency of calcium alginate gel entrapped EMF vegetative mycelium was evaluated in a commercial nursery using cuttings of Eucalyptus clones GG100 and GG680. The cuttings were inoculated with Pisolithus microcarpus G. Cunn. (Cooke & Massee), Hysterangium gardneri E. Fisch., and Scleroderma areolatum Ehrenb. The cuttings were cultivated under low phosphate fertilization and compared with uninoculated control treatments with reduced phosphate (low P control) and full phosphate (high P control) fertilization. Pisolithus microcarpus inoculation increased shoot height, root collar diameter, shoot dry mass, total dry mass, and frequency of maximum score for root ball formation of the two clones compared with the low P control treatment. Also, in relation to the low P control treatment, H. gardneri inoculation increased shoot dry mass in GG100 rooted cuttings. Scleroderma areolatum inoculation did not enhance any characteristic of Eucalyptus rooted cuttings. Inoculation of vegetative mycelium with EMF impregnated in calcium alginate gel intensified rooted cutting growth in a commercial Eucalyptus nursery and decreased the phosphate dose required. Based on the comparison of two Eucalyptus clones, efficiency of the inoculants in promoting benefits depends on the fungus and the Eucalyptus clone. Pisolithus microcarpus is most promising for inoculation in Eucalyptus cuttings.

ABSTRACT Propagation of “coquinho-azedo” (Butia capitata) is limited due to dormancy and high incidence of fungi in its seeds. The aim of this study was to evaluate the effect of treating “coquinho-azedo” seeds with different concentrations of bactericide and/or fungicide on the incidence of fungi and on the germination of seeds. Thus, B. capitata seeds were immersed in four doses of the bactericide kasugamycin (0, 1.5, 3.0 and 4.5 mL/L) and/or in four concentrations of the fungicide carbendazim+ thiram (0, 2.92, 5.84 e 8.75 mL/Kg), totaling 16 treatments. After the treatments, the operculum was removed from the seeds, which were placed on trays containing autoclaved sand, covered with transparent plastic film and kept in the nursery. In the germination test, treatments were distributed in randomized block design, factorial arrangement of 4x4 (bactericidal doses x fungicide doses), with four replicates of 50 seeds per plot. For the test of seed health quality, the Blotter test was employed in completely randomized design, using 20 replicates and 10 seeds. Health quality and germination were evaluated at 15 and 40 days after the treatment of seeds, respectively. There was no interaction between the bactericide and the fungicide on germination and control of fungi associated with seeds. However, there was an effect of fungicide concentrations on the evaluated parameters. We observed a negative correlation between the concentrations of the fungicide carbendazim + thiram and fungal incidence, and a positive correlation between the fungicide concentrations and seed germination. Therefore, fungicide treatment reduces the fungal incidence and provides better germination of B. capitata seeds.

Norway spruce cuttings were treated with phytohormone dusts, and nutrient solution was added to the sand in which some of the cuttings were planted. The nutrient treatment greatly increased the number of rooted cuttings and the number that developed new growth, and reduced the number that died. Although talc alone increased top growth, indolylacetic acid, present in three concentrations in talc, had no significant effect on the number of cuttings rooted or dead. However, the hormone dust treatment effected a significant reduction in the length of root per rooted cutting and the mean root length. The results indicate that nutrient salts may, under certain conditions, have a marked influence on the rooting and growth of Norway spruce cuttings.

Abstract Bougainvillea spectabilis is an ornamental plant grown in both temperate and tropical zones. They are primarily propagated by stem cuttings; however, difficulty in growth at early stages occurs routinely, causing barrier for its mass propagation. This study was conducted to study the effects of integrating compost with Trichoderma longibrachiatum (NGJ167), in improving the early growth of Bougainvillea spectabilis. Trichoderma longibrachiatum (NGJ167) used for this purpose was obtained from the Bioscience Center, International Institute of Tropical Agriculture (IITA), Ibadan. One hundred and twenty (120) pots were filled with 5 kg top soil each from an alfisol. Treatments included: Top soil as Control (C), Top soil inoculated with T. longibrachiatum (T), Top soil + compost at 5, 10 and 15 t ha-1 as C1, C2, C3 respectively, Top soil inoculated with T. longibrachiatum + compost at 5, 10 and 15 t ha-1 as C1T, C2T, C3T respectively. The eight treatments were arranged in a completely randomized design (CRD) and replicated three times. Results indicated that integrated use of compost with Trichoderma significantly (p < 0.05) had higher growth values than the control. Higher plant height, stem diameter, number of branches and number of leaves, were recorded with the use of compost at 10 t ha-1 compared to control. Compost with Trichoderma gave higher shoot dry weight compared to treatments of compost alone. In conclusion, use of compost alone or inoculated with Trichoderma had significant effect on early growth and development of Bougainvillea spectabilis.
