<em>Temanggung is one of the centers of Robusta coffee production in Central Java, with a variety of potential local coffee clones. The exploration found that several numbers of local Robusta coffee clones had the potential to have high productivity and were resistant to pests/diseases. However, their level of genetic similarity to the superior clones that have been released has not been clearly identified. This study aimed to investigate the genetic variability of Temanggung local Robusta coffee clones based on SSR markers. The study was conducted at the Molecular Laboratory of the Integrated Laboratory, Indonesian Industrial and Beverage Crops Research Institute from February to December 2018. A total of 29 local coffee clones derived from Temanggung along with two control coffee clones (BP 42 and BP 358) were used. All the 14 SSR markers used in the present study were polymorphic and could cluster those local coffee clones into 5 major groups at a genetic similarity coefficient of 0.57. Four local coffee clones (Putih Daun Lebar, Lokal, Tugusari Hijau and Tugusari Kuning) were in the same group with control clones in group I. One local clone (Tugusari Hijau) had a genetic similarity with the control clone BP 358 at 0.91. Meanwhile, the other 25 local clones were in different groups from the control clones. These local clones, which showed genetic distance far from the control clones, can be selected as candidates for local superior clones in coffee breeding program.</em>

Introduction. The main direction of increasing the productivity of milk is to increase the proportion of heredity of the Holstein breed in the genotype of cows. Industrial breeds in Ukraine are improving due to the increase in the Holstein inheritance in the genotype of cows. The "holsteinization" of the most widespread domestic Black-and-White diary breed is intensively conducted. Currently, the percentage of heredity from Holstein is 90% or more. The negative effect of "holsteinization" appeared in reducing the resistance of animals to diseases that led to the spread of necrobacterial pathology. The control of the spread of necrobacteriosis can be based on genetic markers. Important markers can be the allele of the BoLA-DRB3.2 gene responsible for the formation of adaptive immunity. Due to the ambiguity of the results of "holsteinization", the following tasks were solved: To study the genetic structure of the herd for the BoLA-DRB3.2 gene at the beginning of the "holsteinization" and now. To compare the detected genetic structures with the alleles spectrum of North American Holstein and identify quantitative and qualitative changes in the structure of the herd genotype. To determine the effect of "holsteinization" on the dynamics of milk production and the state of morbidity by necrobacteriosis. Materials and methods of research. Comparison of alleles of population of the Ukrainian Black-Pied Dairy (UBPD) breed and Holstein breed was conducted to detect the consequences of "holsteinization" on milk yield and incidence of necrobacteriosis. The data of the allelic polymorphism of the BoLA-DRB3.2 gene of the UBPD10 (2010, n = 162), UBPD15 (2015, n = 114) and two Holstein populations of the USA and Canada were collected. The allelic spectrum was determined by the PCR-RFLP method. The amplification of the BoLA-DRB3.2 gene was performed using 2-step PCR with the use of primers HLO-30, HLO-31 and HLO-32 and allele-specific PCR. Restriction analysis was performed with endonuclease RsaI, HaeIII, BstYI (XhoII). Restriction fragments were separated by electrophoresis in 4% agarose gel. Counting of allele frequencies was performed taking into account the number of homozygotes and heterozygotes found for the corresponding alleles. To determine the phylogenetic relationships between the populations of the studied herds, genetic distance and genetic similarity were determined by the M. Nei method. Individual dairy productivity of cows was estimated for all lactation (regardless of its duration). Average milk yields were determined as the total volume of milk produced divided by the number of dairy cows. Results and discussion. The breeding measures carried out led to the accumulation of alleles characteristic of the Holstein breed. For Holstein, there are eight alleles with a frequency of more than 4%. It is alleles *03, *07, *08, *11, *16, *22, *23, *24. A high degree of consolidation of weighty alleles can be outlined. In total they occupy 84,6% of allele spectrum of the population. Consolidation of such alleles in the herd of the Ukrainian Black-and-White diary breed is much lower - only 52.2%, although it increased by 6.2% over 5 years. Alleles *10, *13 and *28 are "weighty" for the Ukrainian Black-and-White diary breed, but they are almost non-existent in Holsteins. The genetic similarity of the herd UBPD15 and Holstein increased by ΔI = 0,085, and the genetic distance between the herds of the UBPD increased by ΔD = 0,085 for 5 years. The comparison of the allele spectrum of Holstein and the Ukrainian Black-and-White diary breed shows both the accumulation and the elimination of alleles associated with high productivity. The largest consolidation is typical for alleles *24 (+ 6.75%) and *16 (+ 4.65%). The frequency of "milk" alleles *22 and *08 decreased, respectively, by 4.14 and 1.27%. Alleys, which cause low milk productivity, have the following dynamics: * 23 + 2.53%, *11 – 0.67 and *28 – 0.26. The accumulation of alleles *16 and *23 (7.18%) was found that are associated with predisposition to necrobacteriosis and elimination of *03 and *22 alleles (4.75%) that influence on this disease. Conclusions. It is determined that the role of alleles characteristic for Holstein is increasing in the the Ukrainian Black-and-White diary herd. Breeding measures for holsteinization are conducted in the right direction. There is accumulation of alleles associated with high milk productivity and predisposition to necrobacteriosis. It positively affects the growth of milk production and negatively affects the incidence of necrobacteriosis.

Biodiversitas 21: 3811-3817. This study aimed to investigate the effects of genotypes and primary callus induction media on somatic embryogenesis of superior robusta coffee clones of Lampung. Leaf explants of clones Tugusari, Komari, Tugino, and Wanto were cultured on two types of primary callus induction media (PCIM). PCIM1 consisted of half-strength MS salts, 30 gL-1 sucrose, added with (mgL-1) 0.1 thiamine-HCl, 0.5 nicotinic acids, 0.5 pyridoxine-HCl, 100 Myo-inositol, 200 ascorbic acids, 150 citric acids, and 1 benzyl adenine. PCIM2 consisted of NPCM salts, 30 gL-1 sucrose, added with (mgL-1) 15 thiamine-HCl, 1 nicotinic acid, 1 pyridoxine-HCl, 2 glycines, 130 Myo-inositol, 200 ascorbic acids, 150 citric acids, 1 2,4-dichlorophenoxyacetic acid, and 2 thidiazuron. The highest percentage (100%) of primary callus formation was found in Komari and Wanto clones. PCIM2 resulted in more primary calli than PCIM1. When subcultured to embryogenic callus induction medium, primary calli of clone Komari and Wanto developed into a high percentage of embryogenic calli, while those of the other two turned brown and died. PCIM2-derived primary calli developed into more embryogenic calli. When subcultured on somatic embryo (SE) regeneration medium, these calli underwent the formation of SE of various stages. When subcultured to plant regeneration medium, these SEs developed into plantlets.

This paper presents the results of analysis of the genetic variability of seven Scots pine (Pinus sylvestris L.) populations in Serbia using SSR markers. Genomic DNA was isolated from seed tissue of all seven populations. The concentration of DNA samples was within the range of 1-4 mg/ml. Different PCR protocols were used depending on the type of SSR markers. The total number of fragments obtained by SSR analysis with 4 selected primers was 17 (only bands of strong and medium intensity were considered), of which 6 fragments were polymorphic (35.29%). In order to analyze the genetic similarity of the analyzed populations, graphs of correspondence analysis and UPGMA clusters were produced. By comparative analysis of the obtained dendrograms, the dependence of population genetic differentiation and spatial distance was observed, i.e. their isolation by natural barriers. The results indicate that in further research of interpopulation variability it is necessary, when graphically interpreting genetic distances, to use both methods of statistical analysis (UPGMA analysis and correspondence analysis).

Kolaka, which is located in Southeast Sulawesi, has long been known as one of cacao production centers in Indonesia. Therefore, many different cacao germplasms can be found in this region. The study aimed to evaluate genetic diversity and relationships of 12 cacao genotypes collected from Kolaka. Genomic DNA was extracted by using a modified CTAB method. Meanwhile, genetic diversity was analyzed based on 16 SSR markers, which then separated by 6% non-denaturing polyacryl-amide gel electrophoresis. The result showed that all of those markers, 14 markers exhibited polymorphism and subsequently used for data analysis using NTSYS and PowerMarker program. About 70 different alleles were generated from 12 cacao genotypes analyzed with an average of 5 alleles per locus. Average value of polymorphism information content (PIC) resulted in this study was 0.59. The cluster analysis using UPGMA method based on the genetic similarity coefficient revealed that all cacao genotypes were separated into three major groups. The first group consisted of five cacao genotypes, the second one held four cacao genotypes, whereas the third group contained three genotypes. This result indicates that three genotypes that clustered separately from the others could be used as a good clonal candidate for cacao breeding program. The information resulted from this present study would be useful for future cacao breeding program, especially in efforts to release a new variety.

This study aims to evaluate the dinamics of coffee production  in Mandang, Sucen Village, Gemawang District, Temanggung on 2018 and 2019. The research was carried out at  people coffee plantation in Mandang Hamlet, Sucen Village, Temanggung. Research using survey methods. Observation of performance with 30 samples taken by purposive sampling technique on 3 clones. Land suitability analysis was carried out at 3 observation points. The results obtained are: The vegetative characteristics  of robusta coffee BP 288 and BP 409 are better than  BP 358 clones, while the robusta coffee production is the same  on various clones and  plantation location.  The long dry season  in 2018 and 2019 has an effect on the decline of the number of leaves and coffee production in 2019 compared to 2018 in Mandang Hamlet, Sucen  Village, Gemawang district, Temanggung.

<em>Analysis of genetic diversity of cacao germplasm collections using molecular markers has an important role in the assembly of new superior clones. The availability of commercial and superior local clones could increase the success of new superior clones’ assembly. Hence, the genetic diversity analysis of these materials needs to be done. The study was aimed to analyze genetic diversity of 28 cacao collections based on SSR markers that would be useful for selection of parental lines. The research was conducted in the Integrated Laboratory, Indonesian Industrial and Beverage Crops Research Institute, Sukabumi, and Plant Molecular Biology laboratory, Bogor Agricultural University, from November 2015 to May 2016.</em> <em>Analysis of genetic diversity was conducted using 28 cacao clones (13 superior local clones and 15 commercial clones). DNA was extraction using CTAB method, which then amplified by PCR technique using 20 SSR primers. The result showed that all SSR markers used in this study were polymorphic with an average value of PIC was high (57%). Phylogenetic tree constructed using DARwin program version 6.05 is divided into 3 major groups, which placed commercial and superior local clones together in each group. Superior local clones observed herein might have close relationships with commercial clones that have long been cultivated in Indonesia. Furthermore, some cacao clones could potentially be parental lines because they had high genetic distance. The results showed that SSR markers are powerful tools to determine potential parental lines, which is expected to increase the chances of heterosis in their progenies.</em>

In this study, simple sequence repeat (SSR) and sequence-related amplified polymorphism (SRAP) markers were used to analyze the genetic diversity of 48 wild Vitis davidii accessions. A total of 78 distinct alleles were amplified by 11 SSR primers, and the average allele number was 8.8. The average observed heterozygosity (Ho) and expected heterozygosity (He) values were 0.785 and 0.814, respectively. The effective allele numbers ranged from 3.92 to 9.61. The average polymorphism information content (PIC) was 0.798. Twelve of 169 SRAP primer combinations were selected for SRAP analysis. A total of 188 bands were produced, and the average was 15.7 bands per primer combination; the average percentage of polymorphic bands was 84.0%. The average PIC was 0.76. The results of the clustering analysis based on SSR markers showed that the 48 wild V. davidii accessions could be classified into five main clusters and had a genetic similarity coefficient level of 0.68. The dendrogram obtained from the SRAP data showed that 48 wild V. davidii accessions could be classified into five main clusters and had a genetic similarity coefficient of 0.72. SSR and SRAP markers differentiated all accessions studied including those with a similar pedigree. We speculated on the origin of Ciputao 0941♀, Ciputao 0940♂, and Fu’an-ci-01 using SSR markers and used both SSR and SRAP markers to resolve homonymy. The result will be valuable for further management and protection of V. davidii germplasm resources.
