Abstract Environmental factors can alter exopolysaccharide biosynthesis in LAB. To further clarify this potential relationship, the mRNA expression of exopolysaccharide-related genes such as glmU, pgmB1, cps4E, cps4F, cps4J and cps4H genes in Lactiplantibacillus plantarum VAL6 under different conditions including high temperature, acidic or alkaline pH, sodium chloride (NaCl) and carbon dioxide (CO2) intensification culture was studied. The transcriptomic data revealed that the exposure of L. plantarum VAL6 to acid at pH 3 increased the expression level of cps4H but decreased the expression levels of pgmB1 and cps4E. Under pH 8 stress, cps4F and cps4E were significantly upregulated, whereas pgmB1 was downregulated. Similarly, the expression levels of cps4F, cps4E and cps4J increased sharply under stresses at 42 or 47oC. In the case of NaCl stress, glmU, pgmB1, cps4J and cps4H were downregulated in exposure to NaCl at 7-10% concentrations while cps4E and cps4F were upregulated at 1 h of 10% NaCl treatment and at 5 h of 4% NaCl treatment. Remarkably, CO2 intensification culture stimulated the expression of all tested genes. In addition, simultaneous changes in expression of cps4E and cps4F under environmental challenges may elicit the possibility of an association between the two genes. These findings indicated that the expression level of exopolysaccharide-related genes is responsible for changes in the yield and monosaccharide composition of exopolysaccharides under environmental stresses.

Background: Converging evidence indicates that glutamatergic system and glia are directly implicated in the pathophysiology of depression. Clinical studies indicate that electroacupuncture (EA) has antidepressant-like effect with low side effects for depression. However, the underlying antidepressant mechanism of acupuncture remains obscure. Methods: Chronic unpredictable mild stress (CUMS)-induced depressive rats were used to induce depressive-like behavior, and evaluated by the weight change, open field test, sucrose preference test, and novelty suppressed feeding test. EA, NMDA receptor subunit 2A antagonist (NR2A RA) or NMDA receptor subunit 2B antagonist (NR2B RA) was used for comparison. High performance liquid chromatography (HPLC) was performed to detect the content of hippocampal glutamate, while western blot for the hippocampal protein expression levels of calcium/calmodulin-dependent protein kinase II (CaMKII), Bax, caspase 3 and B-cell lymphoma-2 (Bcl-2). The distribution of glutamate ionotropic receptor NMDA type subunit 2A (NR2A), neuronal nuclear protein (NeuN), glutamate ionotropic receptor NMDA type subunit 2B (NR2B) and glial fibrillary acidic protein (GFAP) were detected by immunofluorescence. Results: Significant depression behavior (reduced body weight and sucrose preference, increased feeding and immobility time) was produced in CUMS-induced depressive rats, which was reversed significantly by EA. EA decreased hippocampal glutamate level. EA led to a significant decrease in expression levels of Bax, caspase 3 and CaMKⅡ accompanied by increased Bcl-2 expression level. Furthermore, EA significantly increased NR2A expression level as well as decreased NR2B expression level in hippocampus. Conclusion: EA ameliorated depression-like behavior in CUMS rats, which might be mediated, at least in part, by regulating the glutamate, NMDA receptors and apoptosis in the hippocampus.

AbstractThe LOX genes have been identified and characterized in many plant species, but studies on the banana LOX genes are very limited. In this study, we respectively identified 18 MaLOX, 11 MbLOX, and 12 MiLOX genes from the Musa acuminata, M. balbisiana and M. itinerans genome data, investigated their gene structures and characterized the physicochemical properties of their encoded proteins. Banana LOXs showed a preference for using and ending with G/C and their encoded proteins can be classified into 9-LOX, Type I 13-LOX and Type II 13-LOX subfamilies. The expansion of the MaLOXs might result from the combined actions of genome-wide, tandem, and segmental duplications. However, tandem and segmental duplications contribute to the expansion of MbLOXs. Transcriptome data based gene expression analysis showed that MaLOX1, 4, and 7 were highly expressed in fruit and their expression levels were significantly regulated by ethylene. And 11, 12 and 7 MaLOXs were found to be low temperature-, high temperature-, and Fusarium oxysporum f. sp. Cubense tropical race 4 (FocTR4)-responsive, respectively. MaLOX8, 9 and 13 are responsive to all the three stresses, MaLOX4 and MaLOX12 are high temperature- and FocTR4-responsive; MaLOX6 and MaLOX17 are significantly induced by low temperature and FocTR4; and the expression of MaLOX7 and MaLOX16 are only affected by high temperature. Quantitative real-time PCR (qRT-PCR) analysis revealed that the expression levels of several MaLOXs are regulated by MeJA and FocTR4, indicating that they can increase the resistance of banana by regulating the JA pathway. Additionally, the weighted gene co-expression network analysis (WGCNA) of MaLOXs revealed 3 models respectively for 5 (MaLOX7-11), 3 (MaLOX6, 13, and 17), and 1 (MaLOX12) MaLOX genes. Our findings can provide valuable information for the characterization, evolution, diversity and functionality of MaLOX, MbLOX and MiLOX genes and are helpful for understanding the roles of LOXs in banana growth and development and adaptations to different stresses.

The BIRC (baculoviral IAP repeat-containing; BIRC) family genes encode for Inhibitor of Apoptosis (IAP) proteins. The dysregulation of the expression levels of the genes in question in cancer tissue as compared to normal tissue suggests that the apoptosis process in cancer cells was disturbed, which may be associated with the development and chemoresistance of triple negative breast cancer (TNBC). In our study, we determined the expression level of eight genes from the BIRC family using the Real-Time PCR method in patients with TNBC and compared the obtained results with clinical data. Additionally, using bioinformatics tools (Ualcan and The Breast Cancer Gene-Expression Miner v4.5 (bc-GenExMiner v4.5)), we compared our data with the data in the Cancer Genome Atlas (TCGA) database. We observed diverse expression pattern among the studied genes in breast cancer tissue. Comparing the expression level of the studied genes with the clinical data, we found that in patients diagnosed with breast cancer under the age of 50, the expression levels of all studied genes were higher compared to patients diagnosed after the age of 50. We observed that in patients with invasion of neoplastic cells into lymphatic vessels and fat tissue, the expression levels of BIRC family genes were lower compared to patients in whom these features were not noted. Statistically significant differences in gene expression were also noted in patients classified into three groups depending on the basis of the Scarff-Bloom and Richardson (SBR) Grading System.

A method for the estimation of urea in milk using ammonia electrode is described. Urea is first degraded by urease enzyme into ammonium ion and carbon dioxide at neutral pH. The ammonium ion is then converted into ammonia at alkaline pH. A linear inverse relationship was observed between logarithmic concentration of ammonia or urea and electrode response. Repeatability, expressed as a coefficient of variation, was 1·77% at a level of 8·92 mm-urea in milk. The method was validated in milk samples spiked with between 2×10−3 and 10×10−3 m-urea and recovery of added urea was quantitative. Whereas, preservative sodium azide at 0·5 g/l or 2 g/l level did not affect results, lower values of urea concentration in presence of Bronopol at 0·5 g/l were observed. Urea levels in milk samples estimated by this method were comparable to standard enzymatic method. The method is simple, fast and is not prone to interference from other milk constituents.

In China, as popularizing the technology of fully mechanized top-coal mining and increasing the strength and depth of mining, the gutter-up gob can be formed. But the work of fire preventing and extinguishing for its coal spontaneous combustion is more complexity and difficulty. In this paper, based on geology parameters and mining practice for 93up12 fully mechanized top-coal caving face in Nantun coalmine, the form and character of gutter-up gob are analyzed. According to the hidden danger of high temperature for spontaneous combustion in the gob, we adopt the comprehension technologies of fire extinguishing and preventing which include sealing air-leakage, grouting, and injecting compound gel with fly-ash, foam of retarding oxidation, liquid carbon dioxide, and gas of nitrogen. By putting in practice the pre-controlling technology in gutter-up gob and monitoring data of target gases, it obtains a good effect and ensures the safety in production of the fully-mechanized top-coal caving face.
