The OvSynch program was formulated over 20 years ago (it consists in administering gonadoliberin on the day of starting the procedure, then injecting prostaglandin 7 days after the GnRH initial injection, and administration of GnRH two days after applying PGF2α. The procedure ends in the insemination of a female 16–20 hours after administering the final dose of the hormone) and it is commonly applied in cow reproduction management protocols. It has evolved into numerous modifications which has boosted its effectiveness. They have consisted in replacing GnRH with hCG or pLH, diversifying the time of the final injection of GnRH, which follows the prostaglandin injection, and postponing insemination after the final GnRH dose, as well as changing the dosage of the aforementioned hormones. To expedite the Ovsynch protocol, so-called PreSynch was used, which involved administering two prostaglandin injections with a 14 day interval before applying the initial GnRH dose in protocol OvSynch. The interval between the second prostaglandin injection in the PreSynch program varied (PreSynch-14, PreSynch-12, PreSynch-11, PreSynch-11 and PreSynch). The pregnancy rate can be also improved by introducing GnRH injection before starting the OvSynch program, administered 7, 6 or 4 days before the initial GnRH injection in OvSynch (G7G, G6G, G4G). Higher pregnancy rates, in comparison to the PreSynch-OvSynch program, can be obtained by applying the DoubleOvsynch protocol. In this protocol two OvSynch programs are administered in a 7-day interval. Lately, alternative programs such as DoubleSynch and EstraDoubleSynch have emerged. The DoubleSynch protocol consists in administering PGF2α in day 0, GnRH on the second day, another PGF application on the 9th day, and the second GnRH dose on the 11th day. Artificial insemination is performed in oriented time (TAI). This solution is used in order to achieve a higher percentage of pregnant cows in comparison to the PreSynch+OvSynch protocol. It also enables synchronisation of heat and ovulation, both after the first and the second hormonal treatment of GnRH. In EstraDoubleSynch protocol the second injection of GnRH at the end of the DoubleSynch program is replaced by estradiol benzoate (BE) administration on the 10th day, in order to acquire a higher level of follicle waves synchronisation and ovulation rate. Those programs applied in cow reproduction management can positively influence fertility and boost cow breeding profitability..

Artificial insemination results can be improved using fixed-timed AI (FTAI). Currently, progesterone release devices, indispensable in FTAI procedures, such as DIB® (Schering-Plough, Kenilworth, NJ, USA), are made of silicone, a nonbiodegradable biocompatible polymer. Recently, biodegradable progesterone (P4)-releasing devices made of poly(3-hydroxy)butyrate-valerate copolymer (PHB-V) and poly-ϵ-caprolactona (PCL) have been developed. To investigate the efficiency on pregnancy rate of FTAI programs using a new intravaginal device made of PHB-V (Progestar®, Innovare, São Paulo, Brazil), 240 Nellore (Bos indicus) cows, between 3 and 8 years of age, were randomly assigned to 2 experiments. In experiment 1, new P4 devices DIB (control group, DIB1; n = 60) were compared with PHB-V (Progestar1; n = 76); in experiment 2, previously used DIB (DIB2; n = 52) and PHB-V (Progestar2; n = 52) devices were compared. All groups were treated with 2 mg of estradiol benzoate i.m. (Innovare) and received the intravaginal device according to group (Day 0). Together with P4 device withdrawal (Day 8) all animals received 300IU of eCG i.m. (Novormon®, Schering-Plough) and 150 μg i.m. of D-cloprostenol (Croniben®, Biogenesis-Bagó, Buenos Aires, Argentina). On Day 9, all cows were treated with 1 mg of estradiol benzoate and the FTAI was performed 30 h later. The data were submitted to ANOVA (PROC GLM of SAS, SAS Institute, Cary, NC, USA), separating as causes of variance the effect of treatment, postpartum period (PPP1-45 until 70 days and PPP2-71 until 90 days), body condition score (BCS4, BCS5, and BCS6 using a 1 to 9 scale), cyclicity (cycling and noncycling) and clean-up bulls (5 bulls nominated with numbers 1 to 5). Significant differences were considered to have P < 0.05. In experiment 1, interactions were not significant, there was no effect of treatment on FTAI (DIB1: 66.7% v. Progestar1: 65.8%) or breeding season pregnancy rates (BSPR; DIB1: 95.0% v. Progestar1: 94.7%) and no effect of other variables on FTAI pregnancy rates (FTAIPR) and BSPR. In experiment 2 there was no effect of treatment on FTAIPR (DIB2: 61.6% v. Progestar2: 55.8%) and BSPR (DIB2: 94.2% v. Progestar2: 88.5%). As ANOVA indicated a significant effect of clean-up bulls on BSPR, Tukey test was used to split it up. A bull of the Progestar2 group had lower performance (41.7%) causing the difference (albeit nonsignificant) on BSPR. This experiment showed that Progestar and DIB had the same efficiency on pregnancy rate and both treatments have the potential to produce FTAIPR >50% and BSPR >90%. Hormonal treatment of cows at the coastal region of Barisal district of Bangladesh was performed to assess the improvement of pregnancy rate. A total of 100 cows and heifers with irregular history of cyclicity were selected randomly and divided into five treatment groups. The groups were A (treated with anthelmintic), B (treated with anthelmintic, vitamin ADE and multivitamin powder), C (treated with PGF2α), D (treated with GnRH) and E (treated with GnRH and PGF2α). Each group comprised of 20 animals. The age, breed and parity of experimental cows were considered during treatment. In the study, the cows treated with both GnRH and PGF2α (group E) showed significantly (p<0.05) higher estrus (80%) and pregnancy rate (60%) than that of group A, B, C and D. The overall estrus rates of local and crossbred cows were 64% and 70%, respectively and the pregnancy rates were 40 and 52%, respectively. The crossbred cows responded significantly (p<0.05) to hormonal treatment than that of local cows. Parity-2 cows showed higher estrus sign than that of other parities. However, the pregnancy rates were higher significantly (p<0.05) in parity-2 and parity ≥4 cows than that of parity-0, parity-1 and parity-3 cows. The pregnancy rate was also found higher in case of 4 to <5 years old cows than that of 2 to <3, 3 to <4, 5 to <6, and ≥6 years old. It may conclude that the hormonal regimen increases the pregnancy rate as well as decreases the undesired waiting of estrus and conception. Further study with more sample size will reveal the more effective treatment for cows at the coastal areas of Bangladesh.

The objective of the experiment was to evaluate the effects of recombinant bovine somatropin (rbST), and the reuse of the progesterone (P4) releasing devices in resynchronization, on the pregnancy rates of Nellore cows submitted to fixed-time artificial insemination. A group of 123 multiparae Nellore cows, was submitted to a resynchronization protocol: on day 0 a Bovine Intravaginal Device (DIB® ) with 1,0g of P4 was implanted, associated with intramuscular administration of 2,0mg of estradiol benzoate (IM); on day 8 DIB was removed; and 1,0mg of estradiol cypionate, 0,15mg of prostaglandin F2? and 300 UI of equine chorionic gonadotropin were administered; on day 10, fixed-time artificial insemination was conducted (FTAI). The cows were randomized into G1 (n=12) – without rbST / with used Bovine Intravaginal Device, G2 (n=50) – without rbST / with new DIB, G3 (n=11) - with rbST / with used DIB and G4 (n=50) – with rbST/ with new DIB. rbST was introduced on the eighth day of the protocol. Sixty days after TAI, pregnancy diagnoses were conducted, via rectal palpation. Blood samples were taken on day 0, 8 and 10 of the protocol, in order to assess P4 plasma concentrations. Pregnancy rates were statistically evaluated through Generalized Linear Models Theory and their significance was tested with Analysis of Deviance. Pregnancy rates were 58%, 40%, 81% and 48% for G1, G2, G3 and G4, respectively, with significant statistical difference for G3. Plasma concentrations of P4 were not statistically different among groups, or collections. In view of the results obtained, we concluded that the administration of rbST in association with P4 DIB, used for the third time, improves pregnancy rates. Estrus resynchronization and re-insemination positively impacted pregnancy rates.

Abstract The present study aimed to determine the effects of progesterone-based gonadotropin-releasing hormone (GnRH), prostaglandin F2α (PGF2α), and equine chorionic gonadotropin (eCG) injections on progesterone profiles and pregnancy rates in cows with no estrus symptoms within 60 days after parturition. A total of 80 cows were included in the study. All animals had the progesterone-releasing device PRID®Delta placed intravaginally for nine days with an injection of GnRH. On the eighth day, PGF2α was injected, and PRID®Delta was removed from the vagina on day nine. Artificial insemination was carried out 60 hours after PRID®Delta removal. In half of the animals (n = 40), 600 IU of eCG was injected when PRID®Delta was removed on the ninth day before artificial insemination 60 hours later. Blood samples were taken from the tail vein on days 0 and 8 to determine progesterone levels. The pregnancy rate in the group that received eCG was 37.5%, while it was 27.5% in those that did not (P = 0.4). While the dominant follicle diameter was 15.5 mm in cows injected with eCG during timed artificial insemination, the follicle diameter was 12.4 mm in cows with no eCG injection (P <0.001). There were no differences in serum progesterone values in blood samples taken until the time of artificial insemination. However, progesterone values in the blood taken during artificial insemination were 0.94 ng/ml in the eCG- group and 0.72 ng/ml in the eCG+ group (P <0.05). As a result, it was determined that eCG injections, in addition to progesterone-based GnRH and PGF2α applications, increased the pregnancy rates in cows without symptoms of estrus. The dominant follicle diameter was larger in cows treated with eCG during artificial insemination; however, follicle size did not increase the pregnancy rate.

The aim of this study was to investigate the effects of hormonal support on the pregnancy rate in repeat breeder cows. Prostaglandin F2α + Ovsynch oestrus synchronization protocol was applied to the cows. Following the fixed time insemination (day 0), the cows were divided into 4 groups. In Group 1 (n = 42), progesterone releasing intravaginal device (PRID) was placed vaginally at 84 h and removed on the 9th day after the artificial insemination. In Group 2 (n = 40), the cows were administered human chorionic gonadotropin (hCG) on the 7th day. Group 3 (n = 45) was applied a combination of progesterone and hCG. Group 4 (n = 42) was not given any treatment. Blood samples were collected from all cows 4 times on days 3.5, 7, 12, and 18 to evaluate progesterone concentrations. The pregnancy rates were 40.47%, 37.50%, 44.44%, and 30.95% in Group 1, 2, 3, and 4, respectively (P > 0.05). In addition, in cows with progesterone concentrations <2 ng/ml on day 3.5, the pregnancy rates were found to be lower than in the cows with progesterone concentrations >2 ng/ml in Group 4 (P < 0.05). Progesterone supplementation in cows with progesterone concentrations < 2 ng/ml appeared to increase pregnancy rates (P < 0.05) in Groups 1 and 3. As a result, post-insemination hormonal applications in the repeat breeder cows did not increase the pregnancy rate. However, it was concluded that determination of progesterone concentrations on day 3.5 following artificial insemination and then hormonal support in the cows with low concentrations would increased the pregnancy rate.

Evaluation of the reproductive tract development (RTD) is a criterion rarely used when heifers are included in estrous synchronization programs. The objective of the present study was to determine the effect of RTD (mature and immature) and hormonal treatment (EC and EB) on estrous expression rate and pregnancy rate in Bos indicus heifers under tropical conditions. RTD was evaluated with an ultrasound and classified as mature (n=99) or immature (n=101). Heifers received an intravaginal device (DIB®; day 0), 2 mg of estradiol benzoate (EB) and 250 µg of cloprostenol (PGF2α). The DIB was removed on day 7 post insertion, and 250 µg of PGF2α was applied and heifers were divided into two groups: One group of heifers received 0.5 mg of estradiol cypionate (EC group), and on day 8, a second group of heifers received 1 mg of estradiol benzoate (EB group). Estrus was detected by visual observation, and all heifers were inseminated at a fixed-time (FTAI). Fifteen days after insemination, bulls were introduced to the treatment groups. Data were analyzed using general modeling and binary logistic regression procedures. The percentage of estrus was similar for both mature and immature heifers (P>0.05), however, the heifers treated with EB had 17% animals in estrus, than the heifers that received EC (P <0.05). The pregnancy rate after FTAI (74.8%) and total pregnancy (FTAI + natural mating = 91.9%) was greater for the group of heifers with a mature reproductive tract (P<0.05). The hormone treatment had not effect (P>0.05) on the pregnancy rate. In conclusion, the heifers with mature reproductive tracts had the highest pregnancy rate after artificial insemination and total pregnancy. Hormonal treatment did not influence the pregnancy rate.

Pregnancy rates and litter sizes following insemination of sows with fresh-extended and frozen-thawed semen averaged 71 vs. 53% and 10.6 vs. 4.4, respectively, and was not affected by the addition of 12.5 μg relaxin to semen. These results indicate no relaxin mediated effect on either pregnancy rate or litter size when sows are artificially inseminated with relaxin-supplemented semen. In all domestic species, acceptable pregnancy rate with frozen-thawed semen is 20-30% through simply cervical and vaginal insemination. Researchers have proved that achievable high results can be expected only from intrauterine insemination in ewes. Unlike the cow, and goat, the cervix of ewes is a formidable barrier to penetrate for transcervical intrauterine insemination. It is not only due to the physical size of the canal and a narrow luminal diameter (even at estrus), but also due to its caudally facing eccentric series of four to eight funnel-like rings. The major aim of this study was to evaluate a comparison of transcervical and laparoscopic intrauterine artificial insemination techniques with frozen –thawed semen on the reproductive performance of ewes.58ewes of 3-4 years of age, so called Altanbulag subspecies of breed were selected and equally divided into 2 groups for the AI methods from October 15-20, 2013 in the breeding station of Research Institute of Animal Husbandry in Hongor sum, Darhan uul province. The estrus was synchronized with EAZI-BREED TMCIDR in which the progesterone content was 0.3 g. After 12 days, the CIDR were removed, PMSG of 400 and 200 IU were administrated intramuscularly per ewe. The total success of synchronization rate was 93.3%. The results has shown the possibilities of laparoscopic and transcervical AI techniques in 55.17%, 41.37% (p>0.05) of pregnancy rates respectively, with frozen-thawed semen in ewes by ultrasonography diagnosis.Mongolian Journal of Agricultural Sciences Vol.13(2) 2014: 43-47

ABSTRACTSix experiments involving the artificial insemination (AI) of a total of 300 female red deer (hinds) with frozen-thawed red deer semen (collected by electro-ejaculation) were conducted over 3 years. Insemination took place at fixed times following various oestrous synchronization procedures using progesterone withdrawal and treatment with pregnant mare's serum gonadotropin (PMSG). In the 1st year, the experiments evaluated basic AI techniques in which pregnancy rates were 45% in 20 hinds receiving two inseminations per vaginam (PV) and 56% in 27 hinds inseminated by the laparoscopic intrauterine method (IU). In the 2nd year, the experiments involved comparisons of the progesterone regime, one or two PV inseminations, and the timing of a single PV insemination. There was no effect of replacement of the progesterone device after 9 days and withdrawal 3 days later compared with the use of one device for the whole period in two experiments. The pregnancy rate for a double PV insemination was significantly higher than for a single PV insemination (58 and 34%; P < 0·05) and there was also a small effect of timing of insemination relative to the synchronization treatment. In the 3rd year all hinds were inseminated by the IU method. The experiments involved a comparison of various times of AI following progesterone withdrawal and a comparison of the progesterone regime. The overall pregnancy rate for 63 hinds inseminated was 56% with no difference between three times of insemination (48, 52 and 55 h). In the second experiment, the difference in pregnancy rate between treatment with progesterone for 15 days and 12 days (44 and 72% for 18 hinds per group) was not significant, but the interaction between the length of progesterone treatment and insemination time was significant (P < 0·05), with the 12-day progesterone/55 h insemination giving a much higher pregnancy rate than the 15-day/55 h insemination (89 and 20% respectively). Although no experiments involved direct comparisons of the routes of insemination, overall pregnancy rates were 56% for IU, 53% for double PV and 35% for single PV inseminations.
