An Easy Efficient Method of Veterinary Drug Residue Analysis in Raw Milk by RP-HPLC-UV with Application to Raw Milk

Background: This study reports an easy method of a veterinary drug investigation in raw milk, based on QuECHERS extraction followed by RP-HPLC-UV analysis. Use of this benchtop system was motivated by its availability and moderate cost relatively to other sophisticated methods such as LC-MS which are more efficient. Methods: This developed method has been optimized and then after validation according to EU legislation, it demonstrated good linearity with R²>0.997, acceptable peak resolution within a short time (<9.5 min) and good recovery of the analyzed drugs (OXY, ALZ and IVR, respectively 87.08, 99.02 and 92.01 %). Additionally, we applied the method to the analysis of cow milk, collected in Nouakchott, capital of Mauritania. Results: The obtained results indicated a mixed level of drug use according to targeted molecules. In 42% of sampled farms, the anti-parasitics IVR and ALZ were detected whereas the antibiotic OXY was detected in 50%. Conclusion: This investigation shows that 17% of the sampled farms exceeded European standards for IVR drug.

Milk quality is influenced by many factors: lactation, fat, protein, lactose, number of somatic cells. In order to process raw milk and compare with criteria of quality and food safety the Regulation of European Parliament and the council no. 853/2004. Analysing the total number of somatic cells (SCC) in the period July-August 2017 it is noted that in case of samples collected from first automatic milk dispenser exceed 2 times the maximum admissible values and the samples collected from second automatic milk dispenser are up to the maximum allowable values which show that milking hygiene and animal health are at the European standards required. Analysis of fat content for both cases indicates that it is within the standard values for cow's milk and fat variations for DM1 samples are very low at temperatures above 30 degrees Celsius which shows that high temperatures do not influence these parameters. The biological material study was represented analysed by 30 samples of milk from only two cow milk dispensers functional located in this period in Cluj-Napoca city. These samples were collected at the same time period during July-August months. The aim of present study is to determine whether milk marketed through dispensers under the high temperature conditions specific to this period is affected in terms of qualitative parameter analysis.

The clinical and bacteriological findings in two cows the udders of which became infected under natural conditions with hemolytic streptococci of the scarlet fever type are discussed. One of the cows was found in a herd supplying raw milk to a small town where a milkborne outbreak of scarlet fever had occurred a short time before. When small numbers of the streptococcus obtained from this case were injected into the udder of a normal cow severe mastitis accompanied by a well marked general reaction resulted. Evidence leads to the conclusion that a severe attack of mastitis due to this organism in one quarter does not sufficiently immunize the other quarters to protect them completely since the streptococcus can be readily implanted in them. The secondary infections were much milder than the original process.

Objectives: The aim of this study is to evaluate the microbial quality and associated health Methodology: An intensive study was carried out from January¬ to October 2019 at the Tangail district of Bangladesh to assess bacterial contamination of raw milk. A total of 60 samples were examined by following the standard bacteriological methods and the health impact was evaluated with the help of a semi-structured-based questionnaire. Molecular characterization of isolated bacteria was carried out by PCR. Results: Results revealed the mean values of TVC and TCC in raw milk, udder, oil, and utensil samples were 7.6×106, 8.8×106, 4.6×106, and 1.3×106 cfu/ml and 3.7×105, 1.4×106, 4.4×105, and 8.9×104 cfu/ml, respectively. The yeast and mold in raw milk, udder, and oil were 1.5×103, 1.8×103, and 1.3×102cfu/ml, respectively, and the mean values of E. coli in the above-mentioned samples were 1.9×103, 1.8×104, 2.1×103, and 1.6×103cfu/ml, respectively. On the other hand, the mean values of Salmonella spp. in raw milk, udder, oil, and utensil samples were 2.4×102, 7.9×102, 1.5×102, and 1.1×102cfu/ml, respectively. Some selected isolates confirmed by molecular identification were tested for their sensitivity against some common antibiotics used in Bangladesh. Escherichia coli showed 70% resistance to Amoxicillin and 90% sensitivity to Ciprofloxacin whereas Salmonella spp. And Staphylococcus aureus showed 90% resistance to Ampicillin and both were 80% and 90% sensitive to Ciprofloxacin, respectively. Conclusion, Significance, and Impact of Study: Milk producers should be appropriately trained to monitor the overall hygienic conditions surrounding the production and handling of milk until it reaches the consumer. Frequent monitoring of the milk production facility, occasional testing of raw milk, and minimal use of antibiotics will ensure the quality of milk. Consumption of quality milk and avoidance of raw milk consumption will reduce the health risk of the consumers.

A collection of lactic acid bacteria (38 Enterococcus and 41 Lactobacillus strains) was tested for the antilisterial activity against 15 Listeria spp. strains (two L. monocytogenes, one L. ivanovii and 12 L. innocua strains) using agar spot method. Out of all 79 bacteria only six Enterococcus strains (1/3A, 3/3A, 6/4D, 6/1A, 1282 and EN3) exhibited antilisterial activity against almost all used indicator strains, when their live cells were tested. When their cell free neutralised supernatants (CFNS) were tested against four selected indicator strains (L. innocua Ln-03, Ln-06, Ln-10 and L. monocytogenes CCM5576) only two Enterococcus spp. strains were active &ndash; E. faecalis 6/1A strain from raw cow milk of minor interest due to the activity of its CFNS only against L. innocua Ln-06 and thermolability of the compound and E. mundtii 1282 strain from goat raw milk with CFNS active against 13 Listeria spp. strains including L. monocytogenes. E. mundtii 1282 strain produced probably a bacteriocin, because it completely lost the activity after treatment CFNS with proteinase K.

In less than one century concrete has become the most widely used construction material over the world. In less than half of century it is difficult to imagine a concrete totally without polymers. An implantation of polymers into concrete has taken effect in the form of Concrete Polymer Composite: C-PC. Since then (1975) the development of new concrete classes have been ongoing: C-PC = PMC + PCC + PI + PC, where PMC Polymer Modified Concrete (polymer cont. < 1% concrete mass); PCC Polymer Cement Concrete (> 1% concrete mass); PIC Polymer Impregnated Concrete (3-8% concrete mass), PC Polymer Concrete (8-12% concrete mass). Over the time the role of polymers have been extended and it is covered by polymer with additional preposition: polymers on concrete (overlays, coatings, waterproofing and bounding materials). All those polymer composites have been found irreplaceable application in concrete repairing industry. It is enough to say that in ten parts of the European Standards, EN 1504, the category “polymer” can be found 73 times, and that is a proof of the big significance of this material in the repairs and protection of concrete. Just for comparison reason, the term “cement” appears only 59 times in all parts of the EN 1504. Indeed, if repaired concrete is higher class then repairing material should content more polymer. The justification belongs to the adhesion, which is a fundamental challenge for concrete repair. But also short time to exploitation readiness and many others polymer composites advantages are taken into consideration. In the paper the question: how polymers enhance concrete repair performance? is discussed. The repair rules and methods versus polymer repair materials will be considered.

ABSTRACT: Pseudomonas, the main genus of gram-negative microorganisms isolated from milk, is psychrotrophic, biofilm-forming, and thermo-resistant deteriorating enzyme producers. The aim of this study was to quantify Pseudomonas spp. in goat’s and cow’s milk produced in the Paraná state, Brazil, to evaluate the deteriorating activity of the isolates at mesophilic and psychrotrophic conditions and to identify, at the species level, the isolates with alkaline metalloprotease (aprX gene) production potential. Microbiological, biochemical and molecular methods were used for isolating, confirming and identifying of isolates. The mean counts were 1.6 (±6.3)x104 and 0.89(±3)x102 CFU/mL for goat and bovine milk samples, respectively, immediately after milking. Of the Pseudomonas colonies isolated from goat milk (n=60), 91.7% showed proteolytic potential when incubated at 35°C/48 h and 80% at 7°C/10 days, and lipolytic potential was observed in 95% of the isolates incubated in mesophilic and 78.3% at refrigeration conditions. From the isolates of bovine milk (n=20), 35% showed proteolytic activity only when incubated at 35°C/48 h, and lipolytic potential was observed in 25% of the isolates incubated at 7°C/10d and 35°C/48h. It was observed that 83.3% and 25% of the isolates genetically confirmed as Pseudomonas spp. of goat and bovine milk showed the potential for alkaline metalloprotease production, with the species P. azotoformans, P. koreensis, P. gessardii, P. monteilii and P. lurida being the most frequent in goat milk and P. aeruginosa the only species identified in cow milk.

This study aimed to isolate Staphylococcus aureus in refrigerated raw cow milk, and identify the presence of enterotoxin-expression genes, enterotoxin production and adherence ability, and antimicrobial resistance potential of the isolated strains. Fifty raw milk samples obtained in different dairy farms were analyzed for S. aureus and evaluated in the isolates the presence of genes associated with the production of major staphylococcal enterotoxins and biofilm formation. In vitro assays were also performed to evaluate the production of enterotoxins and adherence ability, and the antimicrobial resistance. One half (25/50) of raw milk samples presented coagulase-positive staphylococci and 95.2% of the isolates were confirmed to be S. aureus. Among them, 42.4% were carrying genes for enterotoxins production; however, only one isolate was able to produce enterotoxins. All S. aureus isolates were carrying at least two genes associated with biofilm formation and 95.2% isolates was able to adhere upon the in vitro assay. All isolates demonstrated antimicrobial resistance potential to one or more of the tested antibiotics.

“Plant milks” are water-based beverages, such as, extracts from cereals, pseudo-cereals, oil seeds, legumes or fruits. Plant milk consumption is rising in European and North American markets due to problems related to cow milk allergies, intolerances, but also because of vegan diets and sensitivity to environmental issues. There is no specific regulation for these beverages, therefore their composition can vary considerably, even in the same category. The aim of this study is to characterize the main categories of cereal and pseudo-cereal milks on the market by studying the profile of 8 biogenic amines (histamine, serotonin, spermine, spermidine, putrescine, β-phenylethylamine, cadaverine, tyramine) through a RP-HPLC/FD method with a pre-column derivatization. Biogenic amines are ubiquitous compounds, produced by the decarboxylation of the respective amino acids and they have been proposed as quality and safety markers of different foods and beverages. In the analyzed samples, the total biogenic amines content ranged from a minimum of 1.92 mg/L, to a maximum of 9.27 mg/L. The main biogenic amine found in the samples was histamine. The results show a low content of biogenic amines in all types of analyzed products. This ensures the quality and safety of cereal and pseudo-cereal milk samples.

The environment, including animals and animal products, is colonized by bacterial species that are typical and specific of every different ecological niche. Natural and human-related ecological pressure promotes the selection and expression of genes related to antimicrobial resistance (AMR). These genes might be present in a bacterial consortium but might not necessarily be expressed. Their expression could be induced by the presence of antimicrobial compounds that could originate from a given ecological niche or from human activity. In this work, we applied (meta)proteomics analysis of bacterial compartment of raw milk in order to obtain a method that provides a measurement of circulating AMR involved proteins and gathers information about the whole bacterial composition. Results from milk analysis revealed the presence of 29 proteins/proteoforms linked to AMR. The detection of mainly β-lactamases suggests the possibility of using the milk microbiome as a bioindicator for the investigation of AMR. Moreover, it was possible to achieve a culture-free qualitative and functional analysis of raw milk bacterial consortia.
