<span class="fontstyle0">Artificial insemination (AI) in cattle using Ongole Cross Breed liquid semen with cauda epidydymal plasma-2 (CEP-2) diluent+0.6% Bovine Serum Albumin (BSA) aimed to improve the genetic quality of livestock with high sperm motility on beef cows. Addition of BSA as extracellular cryoprotectant to support egg yolk function to prevent membrane damage caused by cold shock during cold storage. This research was conducted in Tumpang, Pakis, and Singosari Sub-district, Malang Regency using 38 cows that inseminated using frozen semen of Limousine and Simmental breeds as control (19 cows) and liquid semen with CEP-2 + 0.6% BSA as a treatment (19 cows). The reproductive parameters were Non Return Rate (NRR), Service per Conception (S/C) and Conception Rate (CR). Data were analyzed descriptively and continued by chi-square test. The results showed that insemination using frozen and liquid semen did not affect the success rate of AI in beef cows. The NRR</span><span class="fontstyle0" style="font-size: 7pt;">43-63</span><span class="fontstyle0"> value of frozen was 89.40% and liquid semen was 78.94%; the CR value of frozen semen was 84.21% and liquid semen was 57.89%; and the S/C value of frozen semen was 1.21 and liquid semen was 1.42. The conclusions of the study were the values of NRR</span><span class="fontstyle0" style="font-size: 7pt;">43-63</span><span class="fontstyle0">, CR and S/C of frozen higher than liquid<br />semen.</span>

This study aims to determine the effect of the substitution of bovine serum albumin (BSA) with cattle blood serum and egg white in the diluent of Cauda epididymal Plasma 2 (CEP-2) on sperm quality of Limousin cattle during cooling at 3-5 C. The research material used was rejected Limousin bull sperm (motility of 50-60%) from Artificial Insemination Centre Singosari, Malang. This research was a laboratory experiment using a randomized block design which was composed of six treatments with 10 replications, those were T0 as controls ((90% CEP-2 with BSA + 10% egg yolk); T1 (83.84% CEP-2 + 6.16% cattle blood serum + 10% egg yolk); T2 (81.84% CEP-2 + 8.16% cattle blood serum + 10% egg yolk); dan T3 (90% CEP-2 + 0,4% egg white + 10% egg yolk); T4 (90% CEP-2 + 0.8% egg white + 10% egg yolk); and T5 (90% CEP-2 without BSA + 10% egg yolk). Parameters measured were the percentage of motility, viability, and abnormality of sperms. Results of research after 48 hours of storage showed that the percentage of sperm motility in T0, T1, T2, T3, T4, and T5 were 40.50±5.90, 36±36.16, 34.00±6.58, 40.50±3.69, 38.50±3.37, and 38.50±4.12, respectively, while the percentage of sperms viability were 75.16±4.30, 70.50±2.88, 73.80±2.80, 74.80±3.30, 75.13±3.13, and 74.03±4.13, respectively, and the percentage of sperms abnormality were 10.14±2.34, 10.62±1.34, 11.33±2.00, 10.94±2.82, 10.02±1.95, and 10.78±1.96, respectively. Abstract A sensitive enzyme immunoassay for cephalexin (CEX) was developed using the rabbit antiserum to CEX, Β-D-galactosidase-labeled CEX, and a double-antibody separation method. The immunogen of CEX was prepared by coupling the amino group of CEX to thiol groups introduced into bovine serum albumin by the use of N-(mmaleimidobenzoyloxy) succinimide as a cross-linker. Highly titered antiserum to CEX was produced in rabbits immunized with the immunogen. Enzyme labeling of CEX with Β-D-galactosidase was done by using 7V-(gamma-maleimidobutyryloxy)succinimide as the crosslinker. The limit of detection was 30 ng CEX/mL sample solution. Application of the method to CEX drug residues detected 30 ng/mL in milk, 60 ng/g in egg yolk, and 400 ng/g in hen tissue.

ABSTRAKInseminasi Buatan dengan menggunakan semen cair digunakan untuk daerah yang sulit nitrogen cair dan mempunyai kualitas yang lebih baik dari pada semen beku. Tujuan penelitian ini adalah untuk mengetahui kualitas berbagai bahan pengencer dasar air kelapa penyimpanan dingin 2-5°C.Penelitian ini dilaksanakan di Loka Penelitian Sapi Potong Grati, Pasuruan. Semen yang digunakan berasal dari sapi madura sebanyak dua ekor, berumur 5 dan 3 tahun  dan berat badan yaitu 397 kg dan 360,5 kg. Sapi madura ditampung seminggu 2 kali dengan motilitas > 70% , sedangkan air kelapa yang digunakan adalah air kelapa hijau yang masih muda. Pengenceran semen cair dibagi menjadi 4 yaitu P0 (CEP-3 + 20% kuning telur) sebagai kontrol, P1 (air kelapa hijau +20% kuning telur), P2 (P1 + 0,4% putih telur + 1% fruktosa) dan P3 (P1 + 0,4% putih telur kuning telur +2% fruktosa). Data dianalisis menggunakan uji Pearson’s Chi Square dan Uji Deskriptif. Hasil penelitian menunjukkan motilitas spermatozoa sesuai standar SNI yaitu motilitas> 40% pada pengencer CEP-3 dapat disimpan selama hari ke-8 (40,50±6,43%) sedangkan pada pengencer dasar air kelapa hijau pada P1, P2 dan P3 tidak menunjukkan perbedaan yang nyata (P>0,05) selama disimpan 6 hari yaitu 40,50±10,12%, 38,00±4,22%, 40,00±8,50%. Abnormalitas dari semua perlakuan menunjukan nilai <20%. Viabilitas didapatkan nilai tertinggi pada perlakuan P0(89,58±2,16%) kemudian P1(89,39±3,79%), P2(88,62±4,59%) dan P3(87,93±4,41%).Kata kunci: CEP-3, semen cair, sapi madura, simpan dingin, air kelapa hijau ABSTRACTArtificial Insemination using liquid semen is performed for areas that where liquid nitrogen is difficult to find and havng better quality than frozen semen. Purpose of this research was to investigate the quality on various coconut water base diluents on liquid semen of madura bull during cold storage of 2-5°C. Research was conducted at Laboratory of Reproduction of Grati Beef Cattle Research Station, Pasuruan.Semen that is used comes from two madura bulls aged 5 and 3 years with body weight is 397 kg and 360.5 kg. The semen was collected twice a week with motility> 70%, and the coconut water used is unripe green coconut water. The research treatments were P0 (CEP-3 + 20% egg yolk) as control, P1 (unripe green coconut water + 20% egg yolk), P2 (P1+ 1% fructose + 0.4% egg white) and P3 (P1+ 0.4% egg white + 2% fructose). Data were analyzed using Pearson's Chi Square test and Descriptive Test. The results showed that the motility of spermatozoa was within Indonesian National Standard (SNI) with more than 40% motility in the CEP-3 diluent and it can be stored until the 8th day (40.50 ± 6.43%). The basic diluents of green coconut water at P1, P2 and P3 was not significantly affected (P> 0.05) until 6 days storing with the motility number average are 40.50 ± 10.12%, 38.00 ± 4.22%, 40, 00 ± 8.50%. The abnormality of all treatments was under 20%. Protein foaming-consolidation method is a facile technique for production of porous materials using egg yolk as pore creating agent. Usage of the yolk in the process offers a number of advantages over other proteins such as egg white and bovine serum albumin (BSA). Various materials were successfully fabricated using this technique. The present paper gives a brief review of the preparation and characterization of the materials through protein foaming-consolidation technique for biomedical application.

Polyclonal antibodies from vaccinated mammalian have made a significant contribution to scientific research and diagnosis. The fact that recent technologies allow the production of antibodies in egg yolks laid by hens has led to the development of an alternative to antibody production that is less dangerous to animals. The results of this study showed that it is possible to produce a large amount of IgY antibody to bovine serum albumin (BSA), content of about 8 mg/mL of egg yolk emulsion. The anti-BSA IgY antibodies with high purity and anti-BSA activity.

We recently described an artificial substrate system which could be used to optimize labeling parameters in EM immunocytochemistry (ICC). The system utilizes blocks of glutaraldehyde polymerized bovine serum albumin (BSA) into which an antigen is incorporated by a soaking procedure. The resulting antigen impregnated blocks can then be fixed and embedded as if they are pieces of tissue and the effects of fixation, embedding and other parameters on the ability of incorporated antigen to be immunocyto-chemically labeled can then be assessed. In developing this system further, we discovered that the BSA substrate can also be dried and then sectioned for immunolabeling with or without prior chemical fixation and without exposing the antigen to embedding reagents. The effects of fixation and embedding protocols can thus be evaluated separately.
