  Objective: Tamarind (Tamarindus indica) has been used as a medical plant for treating many human and animal diseases and widely used as a traditional herbal medicine for the treatment of snake bites. The objective of the study is to investigate whether tamarind seed extract (TSE) has neutralization activity on an adverse histological reaction against venoms of the King Cobra.Methods: A total of 20 healthy mature male mice were randomly divided into 4 groups with 5 mice in each. The control group was injected with 1 ml of normal saline. The second group was injected subcutaneously with a single dose of 24.96 μg/20 g King Cobra venom (KCV) solution. The third group was injected with the same dose of KCV solution and 10 mg/20 g of TSE. The fourth group was injected with the same dose of KCV solution and 15 mg/20 g TSE solution. The animals were sacrificed after 24 hrs of injection of the solution. Fragments of muscle, kidney, and liver were fixed in 10% neutral buffered formalin and processed for light microscopical studies.Results: The result showed that TSE reduced the histopathological changes induced by the KCV in the muscles, livers, and kidneys, and the improvement was proportional to the applied dose of the TSE indicating that TSE prevents adverse histological changes in the muscle, liver, and kidney.Conclusion: The present study demonstrated that TSE reduced the histopathological changes in the muscle, liver, and kidney induced by KCV in mice.

Introduction: The objective of the present study is to investigate protective effects of the tamarind seed extract on the adverse histological reaction of venoms of the King Cobra. Methods: Twenty healthy, mature male mice were randomly divided into 4 groups with 5 mice in each. The control group (C) was injected with 1 ml of normal saline. The second group (V) was injected subcutaneously with a single dose of 24.96µg/20g King Cobra venom (KCV) solution. The third group (TV1) was injected with the same dose of KCV solution and 10mg/20g of Tamarind seed extract (TSE). The fourth group (TV2) was injected with the same dose of KCV solution and 15mg/20g TSE solution. The animals were sacrificed after 24 hours of injection of the solution. Fragments of muscle, kidney and liver were fixed in 10% buffered formalin, and processed for light microscopical studies. Results: Mice of group V, treated with KCV solution showed severe degeneration, disorganization and myonecrosis in the muscle fibers. The liver revealed degenerative changes in most of the hepatic lobules and dilatation in the central veins and the sinusoids. The kidney revealed shrunken glomeruli, dilatation of Bowman capsule and degenerative changes in the renal tubules. Treatment with TSE reduced the histopathological changes induced by the King Cobra venom in the muscles, livers and kidneys and the improvement were proportional to the applied dose of the TSE. Conclusions: The observation is indicating that tamarind seed extract prevents adverse histological changes in the muscle, liver and kidney.

Abstract The study was aimed to evaluate the therapeutic effects of Zizyphus oxyphyla leaves methanolic (ZOX-LME), on serum liver, kidney and hematology along with other serum parameters in Carbon tetrachloride (CCl4) intoxicated rabbits. Experimental animals were divided into five groups, six rabbits in each. These were: group NC (normal control), group, TC (toxic control) and group ST i.e. silymarine administered group at dose rate (50) mg/kg body weight (BW). Group ET1 and group ET2 treated with (ZOX-LME) at dose 200 mg/kg BW and 400 mg/kg BW. CCl4 administration caused significant (P> 0.05) impairment in serum liver enzymes, blood factors and other serum indices. Treatment with (ZOX-LME) significantly (P<0.05) reduced and normalized the levels of serum alanine transaminase (ALT) aspartate transaminase (AST) and alkaline phosphatase (ALP) and hematological indices. Also significant (P< 0.05) reduction was observed in creatinine, urea, uric acid, blood urea nitrogen (BUN), and albumin and glucose concentrations. The altered levels of lipid profile and serum electrolytes (Ca, Mg, Cl, Na, K, and P) were significantly (P<0.05) change toward normal levels with (ZOX-LME) feeding. In addition (ZOX-LME) ingestion caused significant improvement in GSH, GST and CAT levels, while reducing the TBARS levels, exhibited antioxidant capacity. Also (ZOX-LME) showed increase inhibition against percent scavenging of 2, 2-diphenile-1-picrylehydrazyle (DPPH) free radical. Significant (P<0.05) normalizing effects were observed with high dose 400 mg/kg BW of (ZOX-LME and were equivalent to silymarine administered groups. The histological study of liver supported the hepatoprotective and renal curative activity of (ZOX-LME).

The aim of this study was to investigate the dose-related effects of fenitrothion (FNT) on the liver and kidney. The study was conducted on 8-week-old male Wistar rats that were divided into four groups (three experimental groups and one control group) and were treated orally with different doses (25, 50, 100 mg/kg) of FNT for 28 consecutive days. After treatment, the rats were anesthetized with ether and liver and kidney samples were taken for histological studies. The results showed that the histopathological changes in the liver were mainly represented by parenchymatous degeneration of hepatocytes with mild necrosis, leukocytic infiltration in the portal area, severe congestion, and hemorrhage. These changes were dose dependent. Marked tubular dilation, hydropic degeneration in tubular epithelium, moderate congestion, and hemorrhage in the cortical and medulla part of the kidney were recorded. Histopathologic examination of the liver and kidney indicated a significant injury only in rats receiving 100 mg/kg FNT.

Abstract Considering that the involvement of reactive oxygen species (ROS) has been implicated in the toxicity of various pesticides, this study was designed to study the ameliorative effect of Vitamin E (100 mg/kg body weight) on mice (25 - 30 mg) treated with diazinon (32.5 or 16.25 mg/kg body weight) organophosphate insecticide for 14 days. Subchronic DZN exposure and the protective effects of vitamins E (vitE) were evaluated for their effects on haematological indices, the enzymes concerning liver damage [plasma alanine aminotransferase (ALT), aspartate aminotaransferase (AST), alkaline phosphatise (AIP), and some parameters of kidney function (urea and creatinine) in mice. Additionally, the histopathological changes in liver and kidney tissue were examined. The high dose of diazinon (DZNH) decreased the body weight significantly at the end of experiment. Additionally, the liver and kidney were examines for histopathological changes. The high dose of diazinon decreased body weight significantly. Moreover, there was a statistically significant decrease in haemoglobin (Hb), red blood cell (RBC) and hematocrit (Hct) in diazinon-treated mice compared to controls. This decrease was partially remedied in the diazinon-treated group that also received vitE. Damage in the liver and kidney tissues was also evident as elevated plasma ALT, AST, ALP, urea and creatinine. VitE partially counteracts the toxic effect of DZN and repairs tissue damage in the liver and kidney, especially when supplemented to 1/4 LD50 intoxicated animals. Histopathological changes in liver and kidney were observed only in 32.5 mg/kg DZN given group. These results suggest that the effects of DZN are dose dependent. No pathological findings were observed in vitE + DZN treated groups. According to the present study, we conclude that vitE can reduce the detrimental impacts of diazinon on haematological indicies, as well as liver and kidney function.

Abstract Objectives Age-related bone loss occurs in both men and women, and is the leading cause of elderly disability. An age-related bone loss model was established in mice to investigate the protective effects of curcumin and broccoli seed extract. Methods 20 young (6-month-old) male C57BL/6 J mice after administration with vehicle once daily were sacrificed at four time points: day 0, 30, 60 and 90. 8 out of the 109 aged (18-month-old) male C57BL/6 J mice were sacrificed at the beginning and used as control. The rest of these aged mice were then randomly divided into four groups: one group served as control (vehicle), the other three groups were administrated with curcumin (CMN) and/or broccoli seed extract (BSE) by oral gavage. Mice in each group were sacrificed at four time points: day 0, 30, 60 and 90. L2 vertebrae of mice were fixed with paraformaldehyde and scanned with a Scanco Medical μCT40 scanner. Quantitative analyses of bone volume (BV), tissue volume (TV), trabecular number (Tb.N), trabecular thickness (Tb.Th) and trabecular spacing (Tb.Sp) were performed with the Scanco Medical's software. Results BV/TV of the young mice group were significantly higher compared to the aged mice group at all four time points. Similarly, Tb.N and Tb.Th were also higher in the young mice group compared to the aged mice group. In contrast, Tb.Sp was lower in the young mice group. When comparing different groups in the aged mice, we found that mice administered with CMN had a higher BV/TV value compared to the mice in the control group at all three time points. Such a difference is significant by day 30. The mice administered with combined CMN and BSE also showed significant increase in BV/TV on day 30. For Tb.N, both mice administered with either CMN or BSE had higher values at all three time points. But no obvious difference in Tb.N was found for mice administered with combined CMN and BSE. For Tb.Th, both mice administered with CMN and with combined CMN and BSE had higher values compared to the control. For Tb.Sp, both mice administered with either CMN or BSE had lower values compared to the control. Conclusions This study showed that curcumin could slow down bone loss in the mouse model. There is no obvious positive effect with broccoli seed extract or with curcumin and broccoli seed extract combined. The curcumin used in this study may shed light on the alleviation of bone loss in humans. Funding Sources Nu Skin Enterprises.

This study was conducted to compare between two types of opportunistic fungi (Aspergillus fumigatus and Penicillium chrysogenum) in concerning their pathogenicity after intraperitoneal inoculation of mice. A total of twenty four male albino mice were used in this study which divided equally into 3 groups, The first and second groups were inoculated with 0.2ml of 1x 107spores/ml of A. fumigatus and P. chrysogenum  intraperitonially respectively, while the third group was inoculated with normal saline which served as control group. All animals were monitored for 2 weeks after infection. The blood samples were collected by heart puncture after 18 days post infection to isolate of serum that used for biochemical analysis of liver and kidney functions. After that, all animals were sacrificed. Some internal organs of infected groups (liver, kidney, intestine, heart, spleen and lung) were taken to study the histopathological changes. It was found that there was severe histopathological changes in studied organs of infected mice particularly liver, kidney, spleen and intestine which corresponding with significant variation (p<0.01) in enzyme activities of liver and kidney like (Alanine Aminotransferase (ALT), Urea and Creatinine). Also, It was found that P. chrysogenum had more impact on these enzymes (15.65 ± 0.78, 135.23 ± 8.75 and 0.928 ± 0.02 respectively) than A. fumigatus (21.70 ± 1.04, 57.91 ± 5.99 and 0.587 ± 0.03 respectively). Therefore, the present study indicated that fungi present in the environment can induce severe inflammation reach to tissue damage in most vital internal organs So, further studies should be performed to determine the specific virulence factors and active components, which are responsible for pathogenesis of A. fumigatus and P. chrysogenum in spite of the fact that P. chrysogenum can produce antibiotic.

Introduction: Feeding habits and environmental factors may rival genetic susceptibility as etiological factors related to various cancers. Humans are continuously exposed to many synthetic food additives, one of which is sodium nitrite (NaNO2). There is a direct correlation between increases in consumption of nitrite-treated products and incidence of tissue damage, hepatotoxicity, nephrotoxicity and some types of cancer. The objective of this study was to investigate the protective effects of Trigonella foenum-graecum (TFG) on NaNO2-induced hepatotoxicity and nephrotoxicity. Methods: Forty rats were randomly assigned (10 per group) to control (physiological saline solution), TFG (150 mg/kg/day), NaNO2 (80 mg/kg/day), and NaNO2+TFG (80 mg/kg/day + 150 mg/kg/day) groups. This group was offered TFG seed extract two hours before NaNO2. At the end of three months, the rats were decapitated, and blood, kidney and liver tissues were removed. Results: Three months of oral administration of NaNO2 increased serum aspartate aminotransferase (AST), alanine aminotransferase (ALT), urea, creatinine, and pro-inflammatory cytokine levels in the liver and kidney tissues [except for liver Interleukin 1 alpha (IL-1a)] of rats. Serum AST, ALT, urea, creatinine, liver IL-6, and kidney tumor necrosis factor-a (TNF-a), IL-6, IL-1a levels significantly decreased in the NaNO2+TFG group compared to the NaNO2 group. Pathological examinations, it was determined show that exogenously administered TFG could alleviate the effects of NaNO2 hepatotoxicity and nephrotoxicity. Conclusions: Our results suggest that exogenous TFG mitigates NaNO2-administration induced hepatotoxicity and nephrotoxicity. TFG extract exerted antioxidative and anti-inflammatory effects, and played a significant role in preventing hepatic and renal damage induced by chronic NaNO2 administration.

Background: Medicinal plants are of great importance to health of individual and communities. About 80% of the population in Uganda relies on traditional medicine because western-trained medical personnel are limited especially in villages. Most Ugandans use Hymenoxys odorato for medicinal purposes e.g. to treat colds, fever, coughs, anti-helminthes, locally used as tea, anti-allergy and also as an anti-venom to relieve snake bites. Method: A group of 25 male wistar rats of 150 g&ndash;210 g were kept for 14 days while being fed and treated with the extract. At 14th day, anesthesia was given and blood samples collected by cardiac puncture for hematological and biochemical investigations. Serum was analyzed for Alkaline Phosphatase, Aspartate Transaminase and Alanine Transaminase while whole blood was used for complete blood count. The liver and kidney were removed and placed in 10% formalin to prepare for histology staining using haematoxylin and eosin technique. Results: The extract elevated hepatic biomarker enzymes i.e. ALP, ALT and AST. The increase was found to be significantly different (P &gt; 0.05) at 400 and 500 mg/kg doses as compared to the control group. Histological sections of the liver showed distortion of liver cytoarchitecture, steatosis, necrosis of hepatocytes and congestion of the sinusoids at high doses 300, 400 and 500 mg/kg body weight. In the sections of the kidney, there was mild distortion of the integrity of the kidney with glomerular hypercellularity at high doses (400 and 500 mg/kg per body weight). Conclusion: Hymenoxys odorato aqueous extract has toxic effects on the liver and kidney of wistar rats. The effects were observed to be in a dose dependent manner.

Restraint stress may be associated with elevated free radicals, and thus, chronic exposure to oxidative stress may cause tissue damage. Several studies have reported that carvacrol (CAR) has a protective effect against oxidative stress. The present study was designed to investigate the protective effects of CAR on restraint stress induced oxidative stress damage in the brain, liver, and kidney. For chronic restraint stress, rats were kept in the restrainers for 6 h every day, for 21 consecutive days. The animals received systemic administrations of CAR daily for 21 days. To evaluate the changes of the oxidative stress parameters following restraint stress, the levels of malondialdehyde (MDA), reduced glutathione (GSH), superoxide dismutase (SOD), glutathione peroxidase (GPx), glutathione reductase (GR), and catalase (CAT) activities were measured in the brain, liver, and kidney. In the stressed animals that received vehicle, the MDA level was significantly higher (P<0.001) and the levels of GSH and antioxidant enzymes were significantly lower than the nonstressed animals (P<0.001). CAR ameliorated the changes in the stressed animals as compared with the control group (P<0.001). This study indicates that CAR can prevent restraint stress induced oxidative damage.
