Objective: Assess the performance of a crude ethanolic extract, a dichloromethane fraction and a hydroalcoholic residue, which are the basis for chemically and biologically characterizing the husk and seed of Passiflora edulis f. edulis, collected in the region and Colombia with a view to determining potential uses. Methods: Agroindustrial residues of gulupa (peel and seed) were analyzed through a bromatological study; subsequently, they were macerated with ethanol (96%). The crude ethanolic extract was partitioned with dichloromethane, leaving a hydroalcoholic residue. The content of total phenols, the composition of phytophenols (high-performance liquid chromatography-mass spectrometry), the total antioxidant capacity using 3-ethyl benzothiazoline-6-sulfonic acid (ABTS●+) and 2,2-diphenyl-1-pyridyl hydrazyl (DPPH●), the oxygen radical absorbance capacity (ORAC), and the ferric reduction power (FRAP) were determined to the extract, the fraction, and the residue. The evaluation of the inhibitory activity of the angiotensin-converting enzyme inhibitor (ACEI) and the cell viability assay with diphenyl bromide 3- (4,5-dimethylthiazole-2-) il) -2,5-tetrazolium on human leukocytes complemented the characterization. Results: Agroindustrial waste of P. edulis f. edulis, peel and seed, contains as main constituents: Protein (8.49 and 7.29%), fiber (34.2 and 55.7%), phosphorus (1.67 and 3.09), and boron (53.3 and 58.4 mg/kg), respectively. The seed showed 25.5% oil. The crude seed extract exhibited a higher phenolic content (15.34 gEAG/100 g). Likewise, it presented the highest antiradical capacity against ABTS●+ and DPPH● (706.17 and 82.81 trolox equivalent antioxidant capacity [TEAC], respectively) and antioxidant in ORAC and FRAP (142.79 TEAC and 103.63 equivalent ascorbic acid EAA, respectively). The ACEI activity (50% inhibitory concentration 17.62 mg/L) of the crude seed extract was higher than the other samples. No toxicity was found in the samples evaluated at concentrations higher than those of the biological activities manifested. Conclusion: The agroindustrial residues of P. edulis f. edulis (peel and seed) are rich in nutrients, which propose them for use in food matrices. The ethanolic extract from seed showed the highest antioxidant, antiradical, and inhibitory biological activity of the ACEI so that it could be proposed the gulupa seed as a promising phytotherapeutic product associated with its phenolic content, especially its flavonoids. The results obtained allow an added value to the fruit, reducing the chances that its waste contributes to environmental pollution.

AbstractThe aim of this research was to investigate the chemical composition of Coreopsis tinctoria Nutt. fruits extract, to highlight the potential of ultrasound assisted extraction in the fast preparation of extracts rich in polyphenols using different solvents (55%, 78% and 96% hydrous ethanol) and to evaluate the antioxidant potential of formulated extracts. LC-MS/MS was used to characterize the ethanolic extract from Coreopsis tinctoria Nutt. dried fruits. The extract contains different flavonoids (marein, flavanomarein, quercetagetin-7-O-glucoside, okanin aurone, leptosidin, luteolin, apigenin) and phenolic acids (chlorogenic acid, caffeic acid). Several parameters that could affect extraction efficiency were evaluated. Finally, this study focused on determination of plant extracts total phenolic content and their antioxidant capacity. The experimental results allowed the selection of the optimum operating parameters leading to the highest total polyphenolic content, expressed as gallic acid equivalents, and avoiding the degradation of phenolic compounds (ethanol 55%; extraction temperature 323.15 K, extraction time 30 min, liquid/solid ratio 20/1). A good relationship between total phenolic content and antioxidant capacity was obtained.

In the past few years, there has been a renewed interest in studying a wide variety of food products that show beneficial effects on human health.Capsicumis an important agricultural crop, not only because its economic importance, but also for the nutritional values of its pods, mainly due to the fact that they are an excellent source of antioxidant compounds, and also of specific constituents such as the pungent capsaicinoids localized in the placental tissue. This current study was designed to evaluate the antioxidant capacity and total phenolic contents from fruits tissues of twoCapsicum chinenseaccessions, namely, Chak k’an-iik (orange) and MR8H (red), at contrasting maturation stages. Results showed that red immature placental tissue, with a Trolox equivalent antioxidant capacity (TEAC) value of 55.59 μmols TE g−1 FW, exhibited the strongest total antioxidant capacity using both the 2,2-diphenyl-1-picrylhydrazyl (DPPH) and the CUPRAC methods. Placental tissue also had the highest total phenolic content (27 g GAE 100 g−1 FW). The antioxidant capacity ofCapsicumwas directly related to the total amount of phenolic compounds detected. In particular, placentas had high levels of capsaicinoids, which might be the principal responsible for their strong antioxidant activities.

ABSTRACT: Baccharis alnifolia Meyen & Walp. is a native and medicinal, Chilean Altiplano herb that is used as a poultice and also consumed as an infusion to relieve inflammation. This study evaluated the antioxidant and cytotoxic effect of the ethanolic extract of B. alnifolia and their infusion. It was reported that the ethanol extract contains 4.42 mg GAE/g of dry weight and the infusion contains 35.86 mg GAE/L of total polyphenols. Also, it determined the antioxidant capacity using Ferric reducing antioxidant power (FRAP), Oxygen Radical Absorbance Capacity (ORAC) and Trolox equivalent antioxidant capacity (TEAC) assay. Moreover, seven metabolites including between phenolic acids and flavonoids were identified for the first time in both extracts of the medicinal plant Baccharis alnifolia. The UHPLC-DAD chromatograms revealed the majority presence of ferulic acid and luteolin, both could be responsible for the antioxidant and cytotoxic activity. Furthermore, we realized a preliminary screening of cytotoxicity in different tumor cell lines, finding that these extracts have cytotoxic potential on kidney cells.

AbstractObjective: Mint (Mentha spp.) genotypes used as spices are cultivated in Turkey and used for different purposes including herbal tea, spices, the cosmetics industry, and are used in folk medicine. While mint species have been used in traditional practices during humanity’s long history, there is limited research on the comparison of their antioxidant capacity and phenolic contents. This aim of the research is to compare antioxidant capacity and phenolic contents in mint clones to determine superior genotypes for herbal and spice usage and cultivation.Methods: Antioxidant capacity was evaluated by ferric reducing antioxidant power (FRAP) and trolox equivalent antioxidant capacity (TEAC). Total phenolic content in clones were determined by Folin-Ciocalteu method.Results: Maximum total phenolic content [28.27±3.95 μg gallic acid equivalent (GAE)/g dry weight (DW)] and FRAP activity 577.09±46.02 μmol Trolox equiv/g DW were obtained from Clone 13 (M. spicata), while M. piperita clones were higher for TEAC activity (800.02±1.10 μmol Trolox equiv/g DW). The results were first records for M. villoso nervata. FRAP and TEAC activities selected clones were correlated with total phenolic content (r=0.77; 0.73 respectively).Conclusion: According to the results, it can be suggested that Clone 13, Clone 5, (M. spicata) Clone 3 and Clone 8 (M. piperita) are more suitable for use as spice, herbal tea, and antioxidant agents. The clones must be selected for commercial cultivation by the grower and they can be used as spices and herbal teas.

Various beneficial biological activities have been reported for passion fruit (Passiflora edulis), grown in tropical and subtropical regions. However, no study has yet demonstrated its action against the Zika virus (ZIKV) infection. The present study aimed to investigate the anti-ZIKV of the ethanolic passion fruit seed extract (PFSE), from which, liquid chromatography tandem mass spectrometry (LC-MS/MS) analysis allowed to putatively annotate the occurrence of piceatannol, astringin, scirpusin A, scirpusin B, isookanin-7-O-glucoside and naringenin-7-O-glucoside, two quadranguloside derivatives, tyrosine, and phenylalanine. PFSE is well tolerated by cell lines (Vero E6 and HTR-8/SVneo) and tissue explants from the human placenta. With 1- and 24-h treatments, PFSE consistently reduced the viral load and NS1 protein expression of the two strains of ZIKV (MR766 and PE243) in placental explants and cell lines. Thus, PFSE has a promising potential for developing anti-ZIKV treatments, protecting the placenta against infection caused by ZIKV, along with relevant antioxidant potential, represented by ferric reducing antioxidant power (FRAP) of 390.5 Trolox eq per g dry extract and half maximal inhibitory concentration (IC50) for 2,2-diphenyl-1-picrylhydrazyl radical (DPPH•) of 12.1 μg mL–1, and more than 90% protection from lipid peroxidation. Although further studies need to be conducted, PFSE treatment was safe and effective in trophoblast cells and placental explants, thus representing a promising class of all-accessible products based on the reuse of industrial by-products.

Abstract Polyphenolic compounds reportedly produce physiological effects that are beneficial to health. Bee products are particularly rich in polyphenolic compounds. We determined the antioxidant capacity and the phenolic and flavonoid compounds content of 81 samples of honey. We used the Folin-Ciocalteu reagent method to evaluate the total phenolic content. The antioxidant activities were evaluated using in vitro scavenging assays of 2,2-diphenyl-1-picrylhydrazyl (DPPH ) and hydroxyl radicals (OH ), Trolox equivalent antioxidant capacity (TEAC ), and ferric-reducing antioxidant capacity (FRAC ). Total phenolic content ranged from 40.3 to 193.0 mg gallic acid equivalents (GAE )/100 g; the flavonoid content varied from 1.4 to 7.5 mg quercetin equivalents (QE)/100 g. Eucalyptus honeys exhibited significantly higher phenolic content and free radical-scavenging activity than clover honey samples (p<0.05 for all). Principal component analysis explained 73% of the differences observed in antiradical activity with respect to floral origin. Total phenolic content may be more useful than the radical-scavenging assay for detecting antioxidant capacity in honey; it also represents the variable that most appropriately discriminated among these honeys.

Objective: Syzygium cumini L. fruits are reported for several pharmacological applications and winemaking. The present study was conducted to develop and analyze the phytochemical content, and antioxidant properties of lactic acid bacteria mediated fermented S. cumini fruit juice (FSJ).Methods: The pH of FSJ was measured by pH meter, and total acidity was calculated by titration method. The Folin-Ciocalteu colorimetric method was employed to assess the total phenolic content (TPC), and antioxidant properties were determined by 2, 2′-azino-bis-3-ethylbenzthiazoline-6- sulphonic acid, ferric reducing antioxidant power, and ferrous ion-chelating assays.Results: The formula 1 and 2 displayed pH of 3.23, and 3.12, respectively, after 6 months of fermentation. The acidity of F1 and F2 was regularly increased from 0.23 to 0.55 and 0.29 to 0.53 mg lactic acid equivalent per ml sample, respectively. The TPC of FSJ was progressively increased. The F1 and F2 showed an increase in TPC from 0.94 to 1.61 and 1.05 to 1.95 mg Gallic acid equivalent per ml of sample, respectively. The Formula 1 and 2 exhibited antioxidant capacities of 1.90 and 2.06 mg Trolox equivalent antioxidant capacity per ml sample, 0.81 and 0.80 quercetin equivalent antioxidant capacity per ml sample, and 1.67 and 1.82 Vitamin-C equivalents antioxidant capacity per ml sample, respectively. Ferric reducing-antioxidant power of Formula 1 and 2, and Control 1 and 2 were 3.90, 4.29, 2.96, and 3.27 mg Fe2SO4 equivalents per ml of sample, respectively. Whereas, the chelating power of samples was not significantly changed at the end of fermentation.Conclusion: The high-quality FSJ was prepared with enhanced antioxidant capacity and phenolic compounds. The probiotic containing FSJ is an alternative functional food supplement to prevent, cure and manage several degenerative disorders. Further, clinical studies are essential to insight the functional quality of FSJ.

The pressurized liquid extraction (PLE) technique was used, for the first time, to obtain protein extracts with antioxidant activity from side streams (muscle, heads, viscera, skin, and tailfins) of gilthead sea bream (Sparus aurata) in order to give added value to these underutilized matrices. Extraction conditions previously optimized for sea bass (Dicentrarchus labrax) side streams were applied. Protein recovery percentages were 22% (muscle), 33% (heads), 78% (viscera), 24% (skin), and 26% (tailfins), which represented an increase of 1.2–4.5-fold compared to control samples (extraction by stirring). The SDS-PAGE profiles revealed that PLE-assisted extraction influenced protein molecular weight distribution of the obtained extracts. PLE conditions also allowed increasing the antioxidant capacity measured by both Trolox equivalent antioxidant capacity (TEAC; 1.3–2.4 fold) and oxygen radical absorbance capacity (ORAC; 1.9–6.4) assays for all fish extracts. Inductively coupled plasma mass spectrometry (ICP-MS) and high-performance liquid chromatography coupled with electrospray ionization quadrupole time-of-flight mass spectrometry (LC-ESI-qTOF-MS) were used to investigate the presence of toxic metals and mycotoxins in sea bream side streams. The levels of As, Hg, Cd, and Pb were below those established by authorities for fish muscle for human consumption (except for Cd in viscera samples). Through a nontargeted screening approach, no mycotoxins or related metabolites were detected for all sea bream side streams. This study contributes to the research on the valorization of fish processing side streams using environmentally friendly technology.

Sargassum policystum is one of the genus Sargassum from the brown macroalgae group which has an abundance of chemical constituents of fucoidan, fucoxanthin, phenolic acids and alginate which have important biological activities, such as antioxidants and antibacterials. This species thrives in the waters of Kabung Island, Bengkayang, West Kalimantan and has not been exploited so that it tends to act as a marine weed. This study aimed to determine the total phenolic content of the ethanolic extract of S. policystum and its antioxidant and antibacterial activity. Determination of the total content of phenolic compounds using the Folin Ciocalteu method, antioxidant testing using the DPPH method (2,2-diphenyl-1-picrylhydrazyl) and the indicator of sample color change with the addition of these radicals was measured by UV-Vis spectrophotometer. Antibacterial testing was carried out by the test sample diffusion method and the inhibition of the growth of the test bacteria Staphylococcus aureus and Escherichia coli as test bacteria was indicated by the formation of a clear area around the sample wells. The results showed that the phenol content of the ethanolic extract of S. policystum was 79.8 mg/g extract, its antioxidant activity (IC50) was 98.903 ppm, classified as a strong antioxidant, while the comparison antioxidant, Vitamin C, had an IC50 of 6.26 ppm. The inhibitory ability of the ethanol extract against the test bacteria used was indicated by the formation of the largest inhibition diameter on S. aureus of 12.4 mm and E. coli of 12.0 mm.
