Aim: The present study was designed to evaluate the plausible efficacy of ethanolic extract of Rauwolfia serpentina to mitigate copper induced toxicity by investigating cytogenetic biomarkers, i.e., chromosome aberrations (CA) and micronucleus (MN) formation; assessment of biochemical changes in liver enzymes (SGOT, SGPT and ALP) and estimation of protein levels in a freshwater spotted snakehead fish(Channa punctatus). Methodology: The experiment was carried out in six groups, each having 15 specimens for 24, 48, 72, and 96 hr. Group I served as control. Groups II and III were maintained with 3 mg l-1 of Rauwolfia root extract and 0.4 mg l-1 of Cu2+, respectively. Groups IV, V, and VI were simultaneously co-exposed with 0.4 mg l-1 of Cu2+ and three different concentrations of Rauwolfia root extract 1, 2 and 3 mg l-1, respectively. For the genotoxicity assessment, blood and kidney tissues were used. Hepatic impairments were assessed after each exposure period. Results: A significant increase (p<0.05) in chromosomal aberrations, micronuclei frequency, activity of liver enzymes (SGOT, SGPT and ALP) and a decrease in protein level were recorded in Group III in comparison to the control. Groups co-exposed with Cu2+and Rauwolfia serpentine showed a significant (p<0.05) decrease in cytogenetic biomarkers, activity of liver enzymes and an increase in protein levels, as compared to Group III, with respect to control in a dose dependent manner. Interpretation: Thus, result of the present investigation establish the efficacy of R. serpentina root extract against Cu2+ induced toxicity in spotted snakehead (Channa punctatus).

Aim: To explore the efficacy potential of ethanolic root extract of Rauwolfia serpentina against Chromium (VI) toxicity in fish, Channa punctatus. Methodology: Acclimatized fish were divided into six groups, each having 15 specimens. Group I served as a control, while Group II and III fish were exposed to ethanolic root extract of Rauwolfia serpentina (3 mg l-1) and Cr (VI) (96 h LC50/10; 7.68 mg l-1), respectively. Groups IV, V and VI fish were exposed to three different concentrations of ethanolic root extract of Rauwolfia serpentina (1, 2 and 3 mg l-1), simultaneously with 7.68 mg l-1 of Cr (VI). The induction of micronuclei, chromosomal aberrations, protein levels and liver enzymes-SGOT, SGPT and ALP- were assessed in fish of all six groups after designated exposure periods. Results: A significant induction (p<0.05) in chromosomal aberrations, micronuclei frequency and activities of liver enzymes (SGOT, SGPT and ALP) coupled with reduced protein level was recorded in Group III as compared to the control. Whereas, a significant (p<0.05) decrease in the frequency of chromosomal aberrations, micronuclei induction and activities of liver enzymes together with an increase in protein level was observed in Group IV, V and VI with respect to control and Group III. Interpretation: Present investigation evince the ameliorative potential of ethanolic root extract of Rauwolfia serpentina against Cr (VI) induced chromosomal aberrations and micronuclei induction, changes in protein levels and biochemical alterations of liver enzymes in Channa punctatus. Thus would help present work helps in saving the aquatic biodiversity and increasing the production of protein rich food fish.

Irisin is a new myokine that is suspected to influence metabolic syndrome (MetS). However, there is a great controversy with respect to its level in cases of MetS and its correlation with different metabolic parameters. The present study assesses irisin levels in MetS patients and studies its relationship to metabolic and liver functions to evaluate the possible role of the liver in regulation of this level. Sixty subjects were included in this experiment, who were divided into 3 groups: group I (normal control), group II (MetS patients with normal liver enzymes), and group III (MetS with elevated liver enzymes and fatty liver disease). Serum irisin levels showed significant increases in groups II and III compared with group I, and significant increases in group III compared with group II. Also, irisin levels were positively correlated with body mass index, serum triglycerides, homeostatic model assessment of insulin resistance index (HOMA-IR), and liver enzymes. We concluded that serum irisin levels increased in patients with MetS, especially those with elevated liver enzymes, and had a positive correlation with parameters of lipid metabolism and glucose homeostasis with the possibility of hepatic clearance to irisin.

Abstract This study aims to determine the amount of protein content in Aceh cattle beef in the quadriceps muscle (chuck) and to find out the best storage between cold and frozen to protein levels of beef. This study used 10 samples of Aceh cattle beef parts of the quadriceps (chuck) which were divided into 3 treatment groups, group I meat was treated in fresh condition, Group II meat was treated in cold at 8 °C and Group III meat will be treated in frozen at -19 °C. Protein content in meat was analysed by the Independent Samples Test. The results showed that the amount of Aceh beef protein in the quadriceps (chuck) in fresh; 15, 47%, cold 10, 20% and frozen; 9, 97. It was concluded that storing meat in cold and frozen ways is affect the protein content. Frozen meat storage is better to keep protein content than Cold meat storage.

Background: Nephrotoxicity is characterized by raised serum urea and creatinine levels and kidneys being one of the primary organs of drug concentration and excretion are vulnerable to many widely used marketed drugs, including anti-cancer drugs, antibiotics, immunosuppressants, and radio contrast agents, are nephrotoxic. Aim: To determine the nephroprotective effect of ethanolic extract of Carica papaya seeds in Aminoglycoside induced acute nephrotoxicity. Study design: Quasi Experimental study. Methodology: Thirty Sprague Dawley rats were sub divided into 3 groups i.e., I, II and III with 10 rats in each group. Group II and III were administered Aminoglycoside drug; Gentamycin in 80 mg/kg) via intraperitoneal route once daily for 5 consecutive days to induce acute nephrotoxicity. At day 6, nephrotoxicity was confirmed by measuring serum urea and creatinine. Ethanolic extract of Carica papaya seeds (1000 mg/kg) was started once daily through oral route in group III for 5 consecutive days to see the nephroprotective effects of seed extract after causing acute kidney injury. All animals were given standard diet pellets manufactured at NIH. Results: Mean serum urea and creatinine for Group I (Control Group) at day 0 were 24.90 mg/dL ± 1.633 and .750 mg/dL ± .0619 respectively. Mean serum urea and creatinine for Group II (Disease Control Group) was 81.00 mg/dL ± 1.247 and 1.980mg/dL ± .0467 at day 6th. This suggested induction of nephrotoxicity by Gentamycin. Mean serum urea and creatinine for Group III (Ethanolic Extract Treated Group) at 11 day was 72.40mg/dL± .991 and 1.680 mg/dL± .0467 after 5 days treatment with ethanolic extract of Carica papaya seeds. Conclusion: We concluded that ethanolic extract of Carica papaya seeds has significant nephroprotective effects on Aminoglycoside induced acute nephrotoxicity in rats. The present study aimed to investigate the comparative evaluation of pharmacological efficacy between sulfasalazine alone and sulfasalazine in combination with herbal medicine on dextran sodium sulfate- (DSS-) induced UC in mice. Balb/c mice received 5% DSS in drinking water for 7 days to induce colitis. Animals were divided into five groups (n=9): Group I (normal group), Group II (DSS control group), Group III (DSS + sulfasalazine (30 mg/kg)), Group IV (DSS + sulfasalazine (60 mg/kg)), and Group V (DSS + sulfasalazine (30 mg/kg) + Cinnamomi Cortex and Bupleuri Radix mixture (30 mg/kg) (SCB)). Colonic pathological changes were analyzed using hematoxyline/eosin staining. The antioxidant, inflammatory, and apoptotic protein levels were determined using western blotting. SCB supplementation, as well as sulfasalazine, suppressed colonic length and mucosal inflammatory infiltration. In addition, SCB treatment significantly reduced the expression of proinflammatory signaling molecules through suppression of both mitogen-activated protein kinases (MAPK) and nuclear factor-kappa B (NF-κB) signaling pathways and prevented the apoptosis of the colon. Moreover, SCB administration significantly led to the upregulation of antioxidant enzymes including SOD and catalase. Taken together, SCB treatment might offer a better treatment for human UC than sulfasalazine alone or may be useful as an alternative therapeutic strategy against UC, without any evidence of side effects.

PURPOSE: To analyse if the carbonyl proteins measurement could be validated as a method that allows the identification of an intestinal oxidative stress after ischemia and reperfusion injury. METHODS: Twenty-five male Wistar rats (n =21) weighting 200 to 250g were divided into three groups. Group I - control (n = 10). Group II - sham (n = 5) and Group III (n = 10) subjected to 60 minutes of intestinal ischemia and equal period of reperfusion. For this purpose it was clamped the superior mesenteric artery in its distal third. Histological changes and carbonyl protein levels were determined in the samples of all groups. In group III, samples of both normal and reperfused ileal segment were studied. RESULTS: All the reperfused segments showed mucosal and submucosal swelling and inflammatory infiltrate of the lamina propria. Levels of carbonyl protein rose in group III, including in the non-ischemic segments. The sensitivity and specificity of the carbonyl protein tissue levels were respectively 94% and 88%. CONCLUSION: The carbonyl protein method is a useful biologic marker of oxidative stress after the phenomenon of intestinal ischemia and reperfusion in rats. It was also noteworthy that the effects of oxidative stress could be seen far from the locus of the primary injury.

Exposure to relative chemicals has been shown to induce a genotoxic effect that can be observed through formation of micronucleus (MN) in polychromatic erythrocythes (PCE). Rumput Mutiara or Hedyotis corymbosa L. ethanolic extract (HcEE) is known to contain ursolic acid as major compound that possesses antigenotoxic activity on HepG2 cells. This study exerts in vivo approach aiming to evaluate the antigenotoxic effects of HcEE on cyclophosphamide (CP)-induced male Swiss mice. The ursolic acid on HcEE was determined by using thin layer chromatography with silica gel as stationary phase and chloroform-aceton (9:1) as mobile phase. The antigenotoxic activity was carried out by in vivo micronucleus test. Twenty four adult mice were equally divided into seven groups. Group I: control (untreated); group II: Na-CMC 0.5%; group III: CP 50 mg/kg BW; group IV: CP+HcEE 250 mg/kg BW; group V: CP+HcEE 500 mg/kg BW; group VI: CP+HcEE 1000 mg/kg BW; group VII: HcEE 1000 mg/kg BW. HcEE were given for seven days, while CP was administered on the last two days. On the seventh day, the peripheral blood from all mice were collected, smeared, and then stained with Giemsa. The frequencies of MNPCEs and %PCEs were evaluated. Molecular docking was performed to know the interaction between ursolic acid and CYP3A4 by using PLANTS software. There was similar hRF spot between HcEE with ursolic acid standard reference indicated that the extract almost positively contain ursolic acid. HcEE reduced MNPCEs significantly compared to CP group (p<0.05) and combination of CP with HcEE showed reduction of %PCEs (p<0.05). Based on molecular docking analysis, ursolic acid gave lower docking score than CP against CYP3A4 (PDB ID: 2V0M) and similar binding site on amino acid residues Ala 448, Ile 369, Thr 309, and Val 313. All of these data suggest that HcEE perform protective effect against CP-induced genotoxicity.Keywords: Antigenotoxic, Hedyotis corymbosa L., cyclophosphamide, micronucleus, molecular docking

Immunomodulator is a substance that increases or suppresses the immune response through the certain mechanism. The marine sponge Xestospongia sp. has immunomodulatory activity by increasing phagocytic activity. In addition, the phagocytic activity is affected by CD4+ and CD14 cells levels. Thus, this study aims to investigate the effect of Xestopongia sp. extract toward CD4+ and CD14 cells level in model rat. Animals were divided into 4 groups (n=5) and treated for 7 days, as follow: Group I (Ethanolic extract of Xestospongia sp. dose of 300 mg/KgBW); Group II (Ethanolic extract of Xestospongia sp. dose of 400 mg/KgBW); Group III (Phylantus niruri extract); and Group IV (0.5% Na CMC). On day 8, animals were infected with Staphylococcus aureus intraperitoneally. And the blood was collected by cardiac puncture and assayed with ELISA kit CD4 (elabscience®) and ELISA kit CD14 (elabscience®). Ethanolic extract of Xestospongia sp. provided high levels of both CD4+ and CD14 cells (Group II) compared to baseline (Group IV)  (p<0.05). Group I provided similar activity to group III (p>0.05) and Group II provided significant activity with higher levels of CD4+ and CD14 cells compared to group III (p<0.05). In conclusion, both doses of Xestospongia sp. extract provide activity as immunomodulator by increasing CD4+ and CD14 cells levels, yet dose of 400 mg/KgBw provides the higher immunomodulatory activity.

Abstract Background Mucuna pruriens (L.) has been used for the treatment of several ailments in folkloric medicine. The present study therefore investigates the hepatoprotective and nephroprotective potentials of its leaves extract with a view to providing a potent alternative in the management of liver and kidney diseases. Methodology Forty male albino rats were randomly placed into eight groups comprising five animals each. Animals in group I were administered with the distilled water, while groups II and VI were exposed to CCl4 and rifampicin respectively. Animals in groups III and IV were initially exposed CCl4 and treated with 50 and 100 mg/kg bw M. pruriens respectively. Similarly, groups VII and VIII animals were exposed to rifampicin and treated with 50 and 100 mg/kg bw M. pruriens respectively. Animals in group V were treated with 100 mg/kg bw silymarin by oral gavage after an initial exposure to CCl4. Selected biomarkers of liver and kidney damage were determined in the serum and organs homogenate. Liver and kidney slices of experimental animals were also stained for histopathological examination. Results Exposure to CCl4 and rifampicin respectively resulted in marked distortion in lipid profile, inhibition of antioxidant enzymes and a surge in ALT, AST, ALP, urea, uric acid, bilirubin and creatine kinase. Treatment with M. pruriens extract reversed all deranged biochemical and histopathological parameters in a dose-dependent manner. Conclusion Extract of M. pruriens leaves restored deranged biochemical and histopathological parameters in the liver and kidney with similar potency to silymarin. Hence, leaf extract of M. pruriens is a potential hepatoprotective and nephroprotective agent that can be exploited in the management of liver and kidney diseases.
