Aims: To evaluate the effect of silver nanoparticle, itraconazole and a combination of the two agents on the adherence of Candia albicans, Streptococcus mutans and Enterococcus faecalis on acrylic and silicone tissue conditioners. Materials and Methods: 40 pellets of Group 1 ( GC Soft Liner) and 40 of Group 2 (Sofreliner) were fabricated. The pellets were immersed in a solution of silver nanoparticles or itraconazole or a 1:1 ratio of the combination of the two medicaments. The amount of biofilm formed was assessed by the ELISA reader. Results: A decrease in adherence of Candida albicans was seen in acrylic and silicone-based tissue conditioners when compared to the untreated control samples. Silicone based tissue conditioner showed the least adherence of Candida albicans when treated with 1:1 ratio of silver nanoparticles and itraconazole. There was a statistically significant difference in microbial adhesion between acrylic and silicone tissue conditioner (p<0.001).Pellets treated with silver nanoparticles were studied for adherence of Streptococcus mutans , Candida albicans and Enterococcus faecalis. A statistically significant decrease in all three microbial adherences was observed with this group. (p<0.001) Conclusion: Silver nanoparticles and itraconazole are effective antifungal and antibacterial agents that can potentially decrease microbial colonization of Candia albicans , Streptococcus mutans and Enterococcus faecalis on tissue conditioners. Clinical implications: Silver nanoparticles and itraconazole when used will decrease biofilm formation that will not only improve the overall health of the oral cavity but also prevent denture stomatitis and diseases associated with poor oral hygiene protocols. Objetivos: Evaluar in vitro la acción antimicrobiana de las pastas GuedesPinto modificadas con Diprogenta® y con Otosporin®, comparándolas con la pasta Guedes-Pinto convencional. Materiales y métodos: Se evaluó la acción antimicrobiana de las pastas Guedes Pinto contra cepas aisladas de los siguientes microbios: Streptococcus mutans ATCC 25175 Staphylococcus aureus ATCC 25923, Enterococcus faecalis - ATCC 29212, Pseudomonas aeruginosa - ATCC 4116, Candida albicans ATCC 10231. Se empleó la técnica de difusión en agar por el método de pocillo y en triplicado, utilizando el digluconato de clorexidina al 0,12% como control positivo y solución salina como control negativo. Se transfirió 2 mL del inóculo producido de cada microbio a 100mL de agar fundido a 45ºC, dispensando la mezcla en 30 placas de petri. Las placas fueron mantenidas a temperatura ambiente por dos horas y luego incubadas a 35ºC por 24 y 48 horas. La lectura de los resultados fue sometida al Análisis de varianza de un factor y Test de Tukey para comparaciones múltiples. Resultados: La pasta Guedes-Pinto Convencional formó los mayores halos de inhibición para la mayoría de los microbios, con excepción de Candida albicans, donde ninguna de las sustancias produjo efecto y de la Pseudomonas aeruginosa, para la cual la pasta modificada con Diprogenta® obtuvo mejores resultados, sin diferencia estadística entre ellas. La pasta Diprogenta® presentó mejores resultados que la pasta Otosporin® para todos los demás microorganismos y esto ultimo no formó halo de inhibición para Enterococcus faecalis. Conclusión: La utilización de sustitutos de Rifocort® parece tener un potencial antimicrobiano efectivo contra los principales microbios encontrados en los canales radiculares.

Aim and Design: Sealers with prolonged antimicrobial activity are needed by the fact that microorganisms are commonly recovered from teeth with failed endodontic therapy. The aim of this study was to evaluate the duration of antimicrobial activity of ten root canal sealers (Apexit plus, Tubli Seal Xpress, Endoflas FS,Endomethasone, Endomethasone N, AH Plus, Epiphany, EndoRez, Ketac Endo, Roeko Seal) against Enterococcus faecalis and Candida albicans by the agar diffusion test. The zones of inhibition were examined immediately and after 24, 48, 72 hours, 5 and 7 days. Statistical analysis was done using Kruskal - Wallis test and Mann Whitney U test. Results and conclusions: Against Enterococcus faecalis, Endoflas FS showed the largest inhibitory zones immediately and 24 hours after manipulation, whereas, there was no significant difference between Endoflas FS and Endomethasone after 48 hours. Against Candida albicans, Endoflas FS performed better than the other sealers. All the sealers (except AH Plus, Epiphany and Roeko Seal) demonstrated higher antimicrobial action in the first 24 hours after manipulation The antimicrobial action of all the sealers (except AH Plus and Roeko Seal which showed no antimicrobial activity in any studied time and Epiphany which ceased to show any antimicrobial action after 24 hours) decreased significantly with time.

Aim: The aim of this study was to compare the antimicrobial efficacy of Triple antibiotic paste (TAP), Calcium hydroxide and Acacia nilotica (A. nilotica) (Babbula) against Enterococcus faecalis (E. faecalis), Streptococcus mutans (S. mutans) and Candida albicans (C. albicans) as intracanal medicaments. Materials and methods: The triple antibiotic paste mixture of (metronidazole, ciprofloxacin and minocycline), calcium hydroxide, Acacia nilotica (Babbula) were weighed and mixed with polyethylene glycol and agar well diffusion method was employed to study the antimicrobial efficiency against Enterococcus faecalis, Streptococcus mutans and Candida albicans Results: All the intracanal medicaments showed zone of inhibition against Enterococcus faecalis, Streptococcus mutans and Candida albicans. Among all intracanal medicaments used in the study Triple antibiotic paste produced maximum zone of inhibition followed by Babbula and least by Calcium hydroxide. Conclusions: Triple antibiotic paste showed the best antimicrobial efficacy followed by Babbula and least by Calcium hydroxide against Enterococcus faecalis, Streptococcus mutans and Candida albicans. More phytochemical studies need to be done. Further evaluation of concentrated extracts of Acacia nilotica (Babbula) could yield better results.

Introduction: the aim of this study is to evaluate the antimicrobial capacity of silver nanoparticles immobilized in a zinc oxide-eugenol (ZOE) cement against Enterococcus faecalis for potential use in endodontic treatments. Method: experimental in vitro study, performing synthesis of silver nanoparticles (AgNPs) and UV-visible spectroscopy to confirm presence of AgNPs in the prepared solutions. The ZOE mixture was standardized, producing the integrated AgNPs/ZOE material. Scanning electron microscopy (SEM) and Fourier-Transform Infrared Spectroscopy (FTIR) were used to characterize the integrated material; a KirbyBauer assay was finally run to measure the inhibition halos produced by the compound-microorganism interaction. Results: the UV-visible spectroscopy showed presence of AgNPs in the created solution; both SEM and FTIR show that the AgNPs are integrated into the ZOE system, not altering their properties when performed under conditions like those found in the mouth. The Kirby-Bauer assay shows that all samples had inhibition halos. The AgNPs in guava extract had statistically significant differences with the halos of the other samples (p < 0.05). Conclusions: the obtained AgNPs show bactericidal activity against Enterococcus faecalis, as a statistically significant difference was found between the AgNPs suspended in guava extract and the other groups; this will be the starting point for future research

The purpose of this study was to evaluate the antibacterial efficacy of silver Nanoparticles, Nanoparticle calcium hydroxide and calcium hydroxide as intracanal medicament against Enterococcus faecalis bio film formed on root dentinThe Minimal inhibitory concentration test and Anti microbial test were conducted for silver nanoparticle, Nanoparticle calcium hydroxide. Teeth were inoculated with E.faecalis strains for 1month to establish a standard mono species bio film model. Biofilm formation is confirmed using SEM. The intracanal medicaments were placed inside the canal and colony forming units (CFU) were counted before and 7 days later the placement of the medicament.MIC for AGNP is 25µg/200µl and for NCH is 12.5µg/200µl. The Antimicrobial test conducted shows zone of inhibition about 11mm and 14mm for AGNP and NCH respectively at 500µg. The calcium hydroxides do not show any antibacterial activity at this concentration. In antibacterial efficacy test using CFU count, the AGNP and NCH shows 75% and 61% of antibacterial efficacy respectively whereas calcium hydroxide has exhibited just 11% of antibacterial efficacy. Silver Nanoparticles and nanoparticle calcium hydroxide had statistically significant difference in the reduction in the number of colonies when compared to calcium hydroxide by post hoc turkey test and p-value of <0.01 was considered as significant in this study.

Shallot (Allium ascalonicum L.) has been known as traditional medicine. Shallot which has same genus with garlic(Allium sativum L.) contains allicin that is also found in garlic and has been suspected has fungicidal activity toCandida albicans. It is supported by several researches. Therefore, shallot is suspected has antifungal activity too.The aim of this research was to know antifungal activity of shallot’s water extortion againsts Candida albicans invitro. This was a laboratory experimental research which used completely randomized design, with diffusion method.Shallot’s water extortion was devided into three concentrations, there were 50%, 100% and 200%. Ketoconazole 2%was positive control and aquadest was negative control. The result of this research based on analysis of varians(Anova), there was significant difference between several treatments and was confirmed with Duncan New MultipleRange Test (DNMRT) p<0,05, there was significant difference between 100% shallot’s water extortion with othertreatments, but there was no significant difference between 50% shallot’s water extortion with 200% shallot’s. Theconclusion was shallot’s water extortion had antifungal activity againsts Candida albicans with the best concentration100%, but it was lower than ketoconazole 2%.
