This study is aimed at examining total polyphenol, flavonoid, gallotannin and condensed tannins content in acetone, chloroform, ethyl acetate and petroleum ether extracts of Halacsya sendtneri (Boiss.) D?rfl., their antimicrobial and antioxidant activities, as well as identifying and quantifying the phenolic components. The antioxidant activity is consistent with the results of total quantity of phenolic compound. The results showed that the acetone extract of plant species Halascya sendtneri (Boiss.) D?rfl. possessed the highest antioxidant activity. IC50 values were determined: 9.45?1.55 ?g/mL for DPPH free radical scavenging activity, 13.46?1.68 ?g/mL for inhibitory activity against lipid peroxidation, 59.11?0.83 ?g/mL for hydroxyl radical scavenging activity and 27.91?0.88 ?g/mL for ferrous ion chelating ability. The antimicrobial activity was tested using broth dilution procedure for determination of the minimum inhibitory concentration (MIC). The MICs were determined for 8 selected indicator strains. All of the extracts showed strong to moderate strong antimicrobial activity. The phenolic composition of Halacsya sendtneri extracts was determined by the HPLC method. The dominant phenolic compound in acetone, chloroform and ethyl acetate extract is rosmarinic acid. Ethyl acetate extract was also abundant in p-hydroxybenzoic acid and ferulic acid. The main compounds in petrol ether extract were chlorogenic acid and quercetin.

Using natural products as antioxidant agents has been beneficial to replace synthetic products. Efforts have been made to profile the antioxidant capacities of natural resources, such as medicinal plants. The polyphenol content of Himalayan rhubarb, Rheum emodi wall, was measured and the antioxidant activity was determined using DPPH and ABTS+ assay, and the oxidative stress was assessed using SOD enzymatic assay. Five different solvent fractions, n-hexane, n-butanol, ethyl acetate, dichloromethane, and water, were used for screening the antioxidant capacity in effort to determine the optimum extraction solvent. The total phenolic contents for R. emodi fractions ranged from 27.76 to 209.21 mg of gallic acid equivalents (GAE)/g of dry weight. DPPH and ABTS+ assay results are presented into IC50 values, ranged from 21.52 to 2448.79 μg/mL and 90.25 to 1718.05 μg/mL, respectively. The ethyl acetate fraction had the highest antioxidant activity among other fractions. Also, n-butanol and water fractions showed significantly lower IC50 values than the positive control in DPPH radical scavenging activity. The IC50 values of SOD assay of fractions ranged from 2.31 to 64.78 μg/mL. A similar result was observed with ethyl acetate fraction showing the highest SOD radical scavenging activity. The study suggests that the ethyl acetate fraction of R. emodi possess the strongest antioxidant activity, thus the most efficient in extracting antioxidant contents. Moreover, a highly significant correlation was shown between total polyphenol content and antioxidant activity screening assays. The compounds related to the antioxidant activity of R. emodi were identified to myricitrin, myricetin 3-galloyl rhamnoside, and myricetin, which have not been reported in studies about R. emodi before.

Antioxidant activities of wampee peel extracts using five different solvents (ethanol, hexane, ethyl acetate, butanol and water) were determined by using in-vitro antioxidant models including total antioxidant capability, 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity, reducing power, and superoxide scavenging activity. Ethyl acetate fraction (EAF) exhibited the highest antioxidant activity compared to other fractions, even higher than synthetic antioxidant butylated hydroxyl toluene (BHT). In addition, the EAF exhibited strong anticancer activities against human gastric carcinoma (SGC-7901), human hepatocellular liver carcinoma (HepG-2) and human lung adenocarcinoma (A-549) cancer cell lines, higher than cisplatin, a conventional anticancer drug. The total phenolic content of wampee fraction was positively correlated with the antioxidant activity. This is the first report on the antioxidant and anticancer activities of the wampee peel extract. Thus, wampee peel can be used potentially as a readily accessible source of natural antioxidants and a possible pharmaceutical supplement.

The foods from plants were known to ensure against degenerative diseases and maturing because of their antioxidant activitycredited to their high content. Information on antioxidant activity of Indian medicinal plant is abundant. To the best of our knowledge, biological properties have not been accounted in the literature for this species of . As a point, this is the first results to assess the anti-oxidant activity of the plant which belongs to the family . The antioxidant activity of Methanol, , Ethyl acetate and Aqueous extracts of E. was determined using the DPPH free radical scavenging activity, ABTS radical scavenging activity and reducing power assay. The DPPH scavenging activity showed higher activity observed in extract (63%) of E. than (54%), (44%) and aqueous (30%). the ABTS assay inhibition in extract (58%) than (43%), (38%) and aqueous (32%) extracts. The reducing power assay of different extracts was increased in extract (54%) than (40%), (34%) and aqueous (28%) extracts. Overall, the and ethyl acetate extract had higher antioxidant properties than other extract. However, in this study, extracts exhibit great potential for antioxidant activity and may be useful for their nutritional and medicinal functions.

Bioassay guided fractionation of ethyl acetate soluble fraction of methanolic extract of Bridelia retusa yielded tambulin (1), beta-sitosterol (2), and beta-sitosterol glucoside (3), and their structures were elucidated using modern spectroscopic techniques. Compounds 1-3 were evaluated for their antioxidant activity and compound 1 was found to be potent antioxidant. The DPPH assay showed that the compound 1 has potent antioxidant activity with IC50 166.15±1.92 SEM [muM] and the radical scavenging activity was found to be 86.03% where as the standard butylated hydroxy toluene (BHT) has IC50 value 128.83±2.1 SEM [muM]. The radical scavenging activity of standard butylated hydroxy toluene was about 86% .The total phenolic content in ethyl acetate soluble fraction was found to be 147.20±1.5 mg GAE/gm and the total flavonoid content was found to be 16.64±0.00 mg QE/gm.Journal of Institute of Science and Technology, 2015, 20(1): 97-101

The methanolic extract of Artemisia incisa Pamp. was dissolved in distilled water and successively partitioned with n-hexane, chloroform, ethyl acetate and n-butanol. The antioxidant potential of all these fractions and remaining aqueous fraction was evaluated by four methods, i.e., the scavenging activity of the 1,1-diphenyl-2-picrylhydrazyl radical (DPPH), the total antioxidant activity, the ferric reducing antioxidant power (FRAP) assay and the ferric thiocyanate assay. In addition, the total phenolics was determined. The obtained results revealed that among the studied fractions the ethyl acetate soluble fraction showed the most potent DPPH-radical scavenging activity with an IC50 value of 5.3 ? 0.71 ?g mL-1, which is even more effective than the standard antioxidant butylated hydroxytoluene (BHT) (IC50 value 0f 12.1? 0.92 ?g mL-1). The ethyl acetate fraction also showed the highest FRAP value (3677.13 ? 27.1 ?g TE mL-1), inhibition of lipid peroxidation (60.93 ? 0.84 % at 500 ?g mL-1) and total phenolic content (95.5 ? 0.05 ?g GAE g-1) as compared to other fractions. However, the remaining aqueous fraction was found to posses the highest antioxidant activity of all the fractions.

Background: The study was conducted to identify the bacterial strain associated with marine sponge Hyrtiosaff. erectus collected from the Red Sea coastal water and to assess the utilization of their secondary metabolites for human benefit as antioxidant, anti-Alzheimer, anti-viral, anticancer and anti-inflammatory agent. Methods: After biochemical identification of Pesudomance sp. bacterial strain, the total polyphenol contents, cytotoxic, antioxidant, anti-Alzheimer, anti-viral, anticancer and anti-inflammatory activity of the Pesudomance sp. ethyl acetate extract were investigated by applying different biochemical assays. Polyphenol contents were investigated using spectrophotometric techniques. Antioxidant activity was determined by 1,1-diphenyl-2-picrylhydrazyl radical (DPPH), and 2,2/-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) ABTS radical scavenging activity assays. The cytotoxic effects were investigated by using the human cancerous cell lines. Results: The anti-Alzheimer, anti-viral, anticancer and anti-inflammatory activities were determined using ELISA. Qualitative phytochemical analysis of the Pesudomance sp. extract demonstrated the presence of a large and diverse group of substances such as alkaloids, carbohydrates, flavonoids, phenols, terpenoids, saponins, and tannins. The strong antioxidant activity of the Pesudomance sp. extract was mainly attributed to the protective role of polyphenols against reactive oxygen. It was also observed that Pesudomance sp. extract possessed significant anti-Alzheimer activity with 94% at 1 mg. The extract showed also high antiviral activity (90%) using reverse transcriptase enzymes inhibition assay. The examination of the anticancer activity by applying two experimental models, i.e., PTK and SHKI cleared out high significant percentages of 76.19 and 83.09 %; respectively. Conclusion: The anti-inflammatory profiling using TNF, COX1, COX2, IL6 also revealed high antiinflammatory activity with different metabolic pathway of 62.70, 75.444, 79.27 and 54.15 %; respectively. The present study concluded that ethyl acetate extract of Pesudomance sp. possessed strong antioxidant, anti-Alzheimer, and anti-viral, anticancer and anti-inflammatory activities. Further studies are required to purify the bioactive compounds.

Yinchenhao (Artemisia capillaris Thunb.) is a pharmaceutical agent that not only shows therapeutic effects against hepatobiliary diseases but also offers various physiological benefits. This study examined the extraction rates of antioxidant extracts from Yinchenhao leaves using different solvents. The extraction rates using different solvents were as follows: ethanol (12.1 ± 0.87%) > water (7.7 ± 0.45%) > n-butanol (1.3 ± 0.16%) ethyl acetate > (1.3 ± 0.14%) > n-hexane (1.1 ± 0.15%). The n-butanol and ethyl acetate extracts showed higher 1,1-Diphenyl-2 picrylhydrazyl radical free radical scavenging activity, 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulphonate) free radical scavenging activity and ferric ion reducing antioxidant activity, while n-hexane extracts showed weak antioxidant activity. In conclusion, Yinchenhao leaf has potential as a natural antioxidant, and n-butanol and ethyl acetate may be effective extraction solvents for studying its antioxidant activities.

The antioxidant and hepatoprotective activities of ethyl acetate extract was carefully investigated by the methods of DPPH radical scavenging activity, Hydroxyl radical scavenging activity, superoxide radical scavenging activity, hydrogen peroxide radical scavenging activity, and its reducing power ability. All thesein vitroantioxidant activities were concentration dependent, which were compared with standard antioxidants such as BHT, α-tocopherol. The hepatoprotective potential ofPodophyllum hexandrumextract was also evaluated in male Wistar rats against carbon tetrachloride- (CCl4-) induced liver damage. Pretreated rats were given ethyl acetate extract at 20, 30, and 50 mg/kg dose prior to CCl4administration (1 mL/kg, 1:1 in olive oil). Rats pretreated withP. hexandrumextract remarkably prevented the elevation of serum AST, ALT, LDH, and liver lipid peroxides in CCl4-treated rats. Hepatic glutathione levels were significantly increased by the treatment with the extract in all the experimental groups. The extract at the tested doses also restored the levels of liver homogenate enzymes (glutathione peroxidase, glutathione reductase, superoxide dismutase, and glutathione-S-transferase) significantly. This study suggests that ethyl acetate extract ofP. hexandrumhas a liver-protective effect against CCl4-induced hepatotoxicity and possessin vitroantioxidant activities.

Objective: This study aims to investigate the potency of matoa as a tyrosinase inhibitor and antioxidant and also to identify the chemical compounds in the most active fraction and an ethanol extract from the leaves and stem bark of matoa.Methods: The extracts were tested for their tyrosinase inhibitory activity by evaluating the formation of L-dopachrome at 490 nm. Antioxidant activity was tested using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) method. The most active extract from liquid-liquid partition analysis was fractionated, and then, the assays were performed.Results: The ethanol extract of leaves and stem bark of matoa showed weak anti-tyrosinase activity (percent inhibition was 24.54±0.22% and 21.93±0.57%, respectively, final concentration 200 μg/mL), but they showed strong DPPH radical scavenging activity (IC50 values were 6.11 μg/mL and 5.47 μg/mL, respectively). The ethyl acetate fraction was the most active fraction with an IC50 value of 5.38 μg/mL. Ethanol extract from the leaves and stem bark of matoa and the ethyl acetate fraction contains flavonoids, tannins, saponins, triterpenoids, and glycosides.Conclusion: Matoa does not have potency as a tyrosinase inhibitor, but it has good antioxidant activity, although still lower than that of quercetin.

ABSTRACT The process of excessive free radicals  in our body plays a major part in the development of chronic and degenerative illness such as cancer, autoimmune disorders, aging, cataract, rheumatoid arthritis, cardiovascular and neurodegenerative diseases. The recently study reported that free radical and oxidants play a dual role as both toxic and beneficial compounds, since they can be either harmful or helpful to the body. Therefore, natural antioxidants can be explored to prevent degenerative diseases and in the present paper we have investigated antioxidant activity of extracts from Nicolaia speciose (Bl.) Horan flower and Muntingia calabura L.  fruit for its free radical scavenging activity using 2,2-diphenyl, 1-picryl hydrazil (DPPH) radical scavenging activity. The results revealed that both ethyl acetate fractions of N.  speciaose and M. calabura have the highest antioxidant activitiy with IC50 29.81 and 14.48 µg/ml respectively.   The antioxidant activity of both ethanolic extracts were more potent than hexan and water fractions with IC50 39.27 and 137.20 µg/ml respectively. The investigation of IC50 values indicated that the antioxidant activity show moderate to very active. Active compounds were identified using thin layer chromatography with FeCl3, AlCl3, 2,4-DNPH and anisaldehide-H2SO4 spray reagents. Chromatogram of ethylacetate fraction showed that spots on the hRf 12 and 56 were thought to contain phenolic compounds with a carbonyl group, while the hRf 37 was suspected flavone compounds with 3-OH group and the hRf 50 was alleged the ortho-dihydroxy flavone or ortho hydroxy and free carbonyl. Chromatogram of ethyl acetate of talok fruits fraction showed that spots on the hRf 19.31 and 44 were suspected flavone compounds with ortho-hydroxy-carbonyl group and or ortho-hydroxy. ABSTRAK Radikal bebas berlebih dalam tubuh dapat memicu tumbuhnya sel kanker, penyumbatan pembuluh jantung, kerusakan oksidatif otak dan penuaan dini. Hal ini menyebabkan penelitian tentang potensi antioksidan terus mengalami peningkatan, terutama antioksidan alami dari tanaman. Bunga kecombrang (Nicolaia speciosa (BI.) Horan) dan buah talok (Muntingia Calabura L.) dilaporkan aktif sebagai antioksidan dan mengandung senyawa aktif dengan tingkat kepolaran yang berbeda. Oleh karena itu, penelitian ini bertujuan untuk menguji aktivitas dengan penangkapan radikal bebas fraksi-fraksi ekstrak bunga kecombrang dan buah talok serta karakterisasi golongan senyawa aktifnya. Hasil fraksinasi ekstrak diuji dengan larutan DPPH 0,4 mM dan diukur dengan spektrofotometer UV-Vis pada panjang gelombang 517 nm. Hasil absorbansi sampel dan kontrol diolah untuk mendapatkan persen penangkapan radikal DPPH dan aktivitas antioksidan dievaluasi melalui nilai IC50. Senyawa aktif diidentifikasi menggunakan kromatografi lapis tipis dengan penampak bercak FeCl3, AlCl3, 2,4-DNPH dan anisaldehid-H2SO4.  Fraksi-fraksi ekstrak bunga kecombrang dan buah talok menunjukkan tingkat kekuatan antioksidan antara sedang sampai sangat aktif. Fraksi etil asetat bunga kecombrang serta buah talok memiliki aktivitas penangkapan radikal bebas tertinggi dengan IC50 sebesar 29,81 µg/ml dan 14,48 µg/ml. Pada fraksi etil asetat ini, buah talok mempunyai potensi aktivitas antioksidan dua kali lipatnya dari bunga kecombrang.  Fraksi air, ekstrak etanol serta fraksi heksan bunga kecombrang memiliki IC50 sebesar 39,27 µg/ml, 44,08 µg/ml, dan 135,36 µg/ml. Sedangkan ekstrak etanol, fraksi air serta fraksi heksan buah talok memiliki IC50 sebesar 137,20 µg/ml, 282,47 µg/ml, dan 2611,70 µg/ml. Hasil KLT fraksi etil asetat buah kecombrang menunjukkan bahwa bercak pada hRf 12 dan 56 mengandung senyawa fenolik dengan gugus karbonil, sedangkan hRf 37 diduga senyawa flavon dengan gugus 3-OH dan pada hRf 50 merupakan flavon dengan o-dihidroksi dan atau o-hidroksi karbonil bebas. Kemudian hasil KLT fraksi etil asetat buah talok menunjukkan bahwa bercak dengan nilai hRf  19, 31, dan 44 merupakan flavon dengan gugus o-hidroksi karbonil dan atau gugus o-dihidroksi. Bercak pada hRf 31 juga mengindikasikan adanya senyawa terpenoid.
