Background: Aggregation of misfolded proteins under stress conditions in the cell might lead to several neurodegenerative disorders. Amyloid-beta (Aβ1-42) peptide, the causative agent of Alzheimer’s disease, has the propensity to fold into β-sheets under stress, forming aggregated amyloid plaques. This is influenced by factors such as pH, temperature, metal ions, mutation of residues, and ionic strength of the solution. There are several studies that have highlighted the importance of ionic strength in affecting the folding and aggregation propensity of Aβ1-42 peptide. Objective: To understand the effect of ionic strength of the solution on the aggregation propensity of Aβ1-42 peptide, using computational approaches. Materials and Methods: In this study, Molecular Dynamics (MD) simulations were performed on Aβ1-42 peptide monomer placed in (i) 0 M, (ii) 0.15 M, and (iii) 0.30 M concentration of NaCl solution. To prepare the input files for the MD simulations, we have used the Amberff99SB force field. The conformational dynamics of Aβ1-42 peptide monomer in different ionic strengths of the solutions were illustrated from the analysis of the corresponding MD trajectory using the CPPtraj tool. Results: From the MD trajectory analysis, we observe that with an increase in the ionic strength of the solution, Aβ1-42 peptide monomer shows a lesser tendency to undergo aggregation. From RMSD and SASA analysis, we noticed that Aβ1-42 peptide monomer undergoes a rapid change in conformation with an increase in the ionic strength of the solution. In addition, from the radius of gyration (Rg) analysis, we observed Aβ1-42 peptide monomer to be more compact at moderate ionic strength of the solution. Aβ1-42 peptide was also found to hold its helical secondary structure at moderate and higher ionic strengths of the solution. The diffusion coefficient of Aβ1-42 peptide monomer was also found to vary with the ionic strength of the solution. We observed a relatively higher diffusion coefficient value for Aβ1-42 peptide at moderate ionic strength of the solution. Conclusion: Our findings from this computational study highlight the marked effect of ionic strength of the solution on the conformational dynamics and aggregation propensity of Aβ1-42 peptide monomer.

Crew survival in a distressed submarine (DISSUB) scenario may be enhanced by the knowledge of the risks of different types of decompression sickness (DCS) should the crew attempt tower escape. Four models were generated through calibration against DCS outcome data from 3,919 pressure exposures, each for the prediction of one of four categories of DCS: neurological, limb pain, respiratory and cutaneous. The calibration data contained details of human, goat, sheep and pig exposures to raised pressure while breathing air or oxygen/nitrogen mixtures. No exposures had substantial staged decompression or cases of suspected pulmonary barotrauma. DCS risk was scaled between species and with body mass. A parameter was introduced to account for the possibility of the occurrence of some symptom types masking others. The calibrated models were used to estimate likelihood of DCS occurrence for each symptom category following submarine tower escape. Escape depth was found to have a marked effect only on predicted rates of neurological DCS. Saturation at raised internal DISSUB pressure prior to escape was found to affect predicted rates of all symptom types. The iso-risk curves presented are offered as guidance to submarine crews and rescue forces in preparation for, or in the event of, a DISSUB scenario.

(1) Spraying of huts with 0·1 per cent. pyrethrum in kerosene, at the rate of 25 c.c. per 1,000 cubic feet, deterred 90 per cent. of A. minimus from entry on the following night. The repellent effect persisted in diminished degree for at least four days.(2) A rather larger quantity of pyrethrum, sprayed in a Freon aerosol, produced a similar but somewhat less marked effect.(3) The repellent effects of Lethane 384 in kerosene were much less marked, and not discernible on the second night.(4) Different species varied greatly in their sensitivity to the repellents.(5) Two types of repellent effects were indicated. Repellent effects proper were the most important, and males were more sensitive to these than females of the same species. There was also a masking effect, to which anthropophilic females were sensitive, which resulted from obscuring of attractive human scents by the chemical odours.

The initial effect of two pulses of heavy cattle grazing on chenopod shrubland plants and invertebrates in northern South Australia was assessed in a two year trial. Both plant and invertebrate communities were structured by subhabitats which were defined by edaphic and water-distribution features. The cover of grasses and the dominant shrubs, Atriplex vesicaria and Maireana astrotricha, declined significantly following grazing but only total vegetation cover was significantly lower than controls one year after grazing. Plant species richness was not affected by the grazing pulses. Abundance of ants increased, but the responses of other invertebrates were inconsistent following grazing. Rainfall had a marked effect upon both plant cover and invertebrate abundance. Key words: chenopod shrublands, pulsed cattle grazing, Atriplex vesicaria, invertebrates, bioindicators

An effect simulating broken cream in whole milk was produced when Bacillus cereus was grown in washed cream at 22° C. A similar but less marked effect was obtained when a concentrated culture filtrate of B. cereus was added to washed cream.Microscopic examination of washed cream to which B. cereus or its concentrated filtrate had been added showed that the fat-globule membranes had been broken down. The hydrolysis of lecithin in washed cream in which B. cereus had grown was demonstrated by qualitative estimation of free choline.Since these results could not be reproduced when a non-lecithinase-producing strain of B. cereus or its concentrated culture filtrate was added to washed cream, it appears that the hydrolysis of the lecithin of the fat-globule membrane is at least partly responsible for the formation of broken cream.

The preparation and biological examination of a number of arylcarbamic esters (arylurethanes) are described. The experiments demonstrate the marked effect of iso propyl phenylcarbamate and some related compounds in very low concentrations upon the germination and seedling growth of cereals. The absence of effect of the same compounds in similar concentrations upon some dicotyledonous plants is noted.

In order to investigate the air injection method on the performance of an airlift. For this purpose an air lift system with a riser 2000 mm long and 80 mm in diameter, was designed and tested. Seven different air injection methods were used at a constant submergence. The experimental results showed a marked effect on the airlift performance when operated with different air injection methods. The arrangement of five nozzles gives the best performance, and the one nozzle is the worst. Although the injection angle has a little effect on the airlift performance, but view the general conclusions as a whole, the best lifting efficiency can be obtained when the angle of the nozzle placed along the tangential direction of pipe wall is equal to 10º at a given air flow rate QG =37m3/h.

The aggregation of huntingtin fragments with expanded polyglutamine repeat regions (HttpolyQ) that cause Huntington’s disease depends on the presence of a prion with an amyloid conformation in yeast. As a result of this relationship, HttpolyQ aggregation indirectly depends on Hsp104 due to its essential role in prion propagation. We find that HttQ103 aggregation is directly affected by Hsp104 with and without the presence of [ RNQ + ] and [ PSI + ] prions. When we inactivate Hsp104 in the presence of prion, yeast have only one or a few large HttQ103 aggregates rather than numerous smaller aggregates. When we inactivate Hsp104 in the absence of prion, there is no significant aggregation of HttQ103; whereas with active Hsp104, HttQ103 aggregates slowly accumulate due to the severing of spontaneously nucleated aggregates by Hsp104. We do not observe either effect with HttQ103P, which has a polyproline-rich region downstream of the polyglutamine region, because HttQ103P does not spontaneously nucleate and Hsp104 does not efficiently sever the prion-nucleated HttQ103P aggregates. Therefore, the only role of Hsp104 in HttQ103P aggregation is to propagate yeast prion. In conclusion, because Hsp104 efficiently severs the HttQ103 aggregates, but not HttQ103P aggregates, it has a marked effect on the aggregation of HttQ103, but not HttQ103P.

Vinculin plays a role in signaling between integrins and the actin cytoskeleton. We reported earlier that F9-derived cells lacking vinculin are less spread, less adhesive, and move two times faster than wild-type F9 cells. Expression of intact vinculin in null cells restored all wild-type characteristics. In contrast, expression of the head (90 kDa) fragment exaggerated mutant characteristics, especially locomotion, which was double that of vinculin null cells. Expression of the tail domain also had a marked effect on locomotion in the opposite direction, reducing it to very low levels. The expression of the head plus tail domains together (no covalent attachment) effected a partial rescue towards wild-type phenotype, thus indicating that reexpressed polypeptides may be in their correct location and are interacting normally. Therefore, we conclude that: (1) the head domain is part of the locomotory force of the cell, modulated by the tail, and driven by the integrin/matrix connection; (2) intact vinculin is required for normal regulation of cell behavior, suggesting that vinculin head-tail interactions control cell adhesion, spreading, lamellipodia formation and locomotion.

The author has found a perfect resemblance to exist between the appearances presented by pyroxylic spirit (Bihydrate of Methyline of Dumas and Peligot) under voltaic agency, and those which he had previously observed in regard to alcohol; a coincidence which was to be expected, from the interesting analogy existing between these two liquids in other respects.The pyroxylic spirit, after three distillations from quicklime, had a specific gravity of 801 at 62° F., and boiled in contact with mercury at 140° F., under a pressure of 29.5 inches. The leading facts observed were the following:—Minute quantities of acid, alkaline, and saline bodies, when dissolved with rectified pyroxylic spirit, favoured the voltaic action, by increasing the conducting power of the liquid. When it held in solution of pure potash, and was submitted to the agency of thirty-six pairs of four-inch plates in a tube with parallel platinum foil poles, hydrogen was given off from the negative pole, while no gas proceeded from the positive pole. A ten thousandth part of potash had a marked effect in promoting this action.
