THE USE OF GLYCEROL FOR THE CRYOPRESERVATION OF INCONNU’S SPERM

The article studies the possibility to use glycerol as cryoprotectant, instead of dimethylsulfoxide for cryopreservation of sperm of inconnu ( Stenodus leucichthys Gueldenstaedtii, 1772). Investigations were carried out from 2015 to 2016 in the laboratory of the Southern Scientific Center, Russian Academy of Sciences, on the basis of the Astrakhan State Technical University. The material collected on the Alexander sturgeon hatcheries (the Astrakhan region) in the spawning period. Native sperm of 6 male inconnu species was used as a control means. The semen quality was determined in terms of moving activity (life time) of sperm after its activation by water. As the cryoprotectant there were used: base solution - 80%, sucrose - 1.71 g/l, mannite - 0.98 g/l, yolk - 10%, dimethylsulfoxide - 10% and base solution - 87%, sucrose - 1.71% g/l, mannite - 0.98 g/l, yolk - 10%, glycerol - 3 variants: 3; 5 and 10%. In order to provide the most complete penetration of cryoprotectants into the cells there were used electrostimulation of cell membranes. Equilibration time was 5 and 15 minutes. Thawing semen was performed in a water bath at a temperature of 38-40°C. For removing protectors from cells there was chosen a saline solution (0.7% NaCl) as isotonic solution. In tests using dimethylsulfoxide life activity of sex cells was 2 times lower than in tests with glycerol: 78 and 186.2 s at the end of equilibration and 52.3 and 128.9 s after thawing. Sperm showed maximum activity under 5% glycerol concentration during equilibration - 15 min. Concentration of 3% was insufficient, concentration of 10% was excessive, as it suppressed activity of sperm. Egg yolk which coagulated together with glycerol, making difficulty for observing, had to be excluded from the composition of cryoprotectant.

The Central Scientific Library of the Irkutsk Scientific Center SB RAS has an extensive collection of literature on various topics of scientific research of academic institutions of Irkutsk and the Irkutsk region. This library is the successor of the Library of the East Siberian Branch of the USSR Academy of Sciences. With the organization of the Branch its own scientific library was being created. Acquisitions to the library stocks were carried out by its first head A. S. Chumicheva.The first books of the library were books from the exchange stocks of the Library of the Academy of Sciences of the USSR (Leningrad), special libraries’ sector of the Academy of Sciences of the USSR (Moscow); books from other institutions’ libraries all over the country; trophy books from Germany; books from personal libraries of scientists, rare book stores; since 1958 books began to come to hand from the State Public Scientific and Technological Library of the Siberian Branch of the USSR Academy of Sciences (Novosibirsk). The first library accumulations were the source of material that Russian science had in the period of its making on the Irkutsk soil, and an especially stored part of the library stock. There is a currently increasing interest of specialists in rare books’ collections organization, storage and use.A particularly valuable publication is the book, dated 1676, the life-time publication of the French geographer Pierre Duval (1618–1683).In 2019 its ex-libris was studied. They managed to determine the owner of the book and the history of the library. The ex-libris is heraldic, coat of arms belongs to Franciszek Velepolsky (1732–1809), and the book is from the Pinchuv Majorata library.In the library collections there are publications by prominent domestic and foreign scientists of the XIX–XX centuries, including Nobel laureates; books from personal libraries with notes of owners; autographed books, books with dedicatory inscriptions, bookplates. Works of scientific organizations of the XIX–XX centuries, reports of expeditions, and works of scientists about Siberia are of interest.Most of academic journals are presented starting since the first years of their publication.The collection of rare books is being replenished thanks to readers’ donations.

ABSTRAKAir kelapa muda varietas viridisdapat dijadikan pengencer aletrnatif semen cair bagi program IB di daerah minim sarana semen beku. Tujuan penelitian ini untuk menguji pengaruh penggunaan air kelapa muda viridissebagai bahan pengencer terhadap kualitas semen cair kambing Boer setelah didinginkan. Dilaksanakanselama 3 bulan di Laboratorium Fakultas Peternakan UBUnit SumberSekar,Malang. Metodenya yaitu eksperimen. Semen dari  3 pejantan Boer umur 3-5 tahun, dikoleksi seminggu sekali dengan VB. Air kelapa mudaviridis umur 5-7 bulan serta tris aminomethane sebagai kontrol. Didesain menggunakan Rancangan Acak Kelompok (RAK) dengan 2 perlakuan yaitu P0 (tris aminomethane + 10% KT) dan  P1 (air kelapa muda viridis + 10% KT) masing-masing diulang 10 kali. Data dianalisis dengan analisis Ragam (Anova) dengan software Genstat 18. Variabelnya yaitu motilitas individu, viabilitas dan abnormalitas. Hasil penelitian yaitu motilitas individu pada P1bertahan sampai 4 hari (40,5± 24,3%), viabilitas terbaik sampai hari ke-5 (42±24,6%), abnormalitas terendah di hari ke-7(1,31± 0,6). Kesimpulannya, Pengencer air kelapa muda viridis dapat mempertahankan kualitas semen cair kambing Boer selama 4 hari untuk motilitas dan 5 hari untuk viabilitas.Kata Kunci:pengencer, air kelapa, varietas viridisABSTRACTYoung viridis coconut water could be used as an alternative to liquid semen diluent for artificial insemination program in the area with limited facility for frozen semen production. This study evaluated the use of young coconut water as a diluent on liquid semen quality of Boer goat after cold storage. This study was carried out for 3 months at Sumber Sekar Laboratory, Faculty of Animal Husbandry, University of Brawijaya, Malang. The semen was collected from 3 Boer bucks aged at 3 to 5 years old. The semen collection was done once a week with the aid of artificial vagina. The diluents used were young Viridis coconut (5 to 7 months old) and tris aminomethane. The method used was an experiment in a randomized block design with 2 treatments and 10 replicates. The treatments used were T0: tris aminomethane + 10% egg yolk (control) and T1:  young Viridis coconut water + 10% egg yolk. Data were analyzed by analysis of variance using Genstat 18 software. The variables measured were sperm individual motility, viability, and abnormality. The results showed that the sperm individual motility in T1 survived up to 4 days (40.5± 24.3%), the best viability at 5 days (42.0±24.6%),  while the lowest abnormality at 7 days (1.31±0.6). It could be concluded that: 1. Tris aminomethane diluent has higher quality with the storage length up to 9 days, 2. Young Viridis coconut water diluent could preserve liquid semen quality of Boer goat up to 4 days for sperm motility and 5 days for sperm viability.Keywords: diluents, coconut water, viridis variety


Intermittent Lighting Improves the Efficiency of Artificial Insemination in Cage Housed Laying Hens Kavtarashvili A.Sh., Kolokolnikova T.N. Federal Scientific Center “All-Russian Research and Technological Poultry Institute” of Russian Academy of Sciences Omsk Agrarian Scientific Center Summary: The effects of different lighting regimes on the oviposition schedule, productive performance, and reproductive efficiency in cage housed laying hens of layer parental flock (Hisex White-R) were studied; the reasonable regime of artificial insemination (AI) under intermittent lighting is proposed. It was found that intermittent lighting regime 1L:4D:4L:1D:4L:10D compared to the constant lighting significantly alters oviposition schedule: under this regime 82.3% of all daily eggs were laid until 9 am (vs. 66.6% in control). This regime and AI at 10 am improved the productive and reproductive performance compared to control (constant lighting 16L:8D and AI at 12 am): mortality by 1.9%, egg production per initial hen by 3.8%, egg weight by 1.1%, percentage of eggs suitable for incubation by 1.9%, egg fertility by 0.9%, hatchability by 2.3%, hatch of chicks by 2.9%, feed conversion ratio (kg of feed per 10 eggs) by 5.3%, the expenses of electric energy for lighting (per 1000 eggs suitable for incubation) by 54.5%. Its stocks formed the basis of Research library of the Dagestan Scientific Center, Russian Academy of Sciences.

Показана актуальность изучения экологической пластичности древесных растений в связи с необходимостью подбора адаптированного видового состава деревьев и кустарников для защитных лесных насаждений в условиях климатических изменений. Установлено, что экологическая пластичность и пределы толерантности растений связаны со свойством организмов адаптироваться к тому или иному диапазону факторов среды. В статье представлен таксономический состав орехоплодных культур коллекций ФНЦ агроэкологии РАН. Это шесть видов рода Juglans: J. regia, J. mandshurica, J. cinerea, J. rupestris, J. ailanthifolia, J. nigra и три вида рода Corylus: С. avellana L., С. аmericana W., сорта С. pontica C. Koch Президент, Футкурами, Черкесский2. Приведены сведения о положительном опыте культивирования Corylus avellana L. в плантационных насаждениях (1,6 га, посадка рядовая, схема размещения 55 м) в условиях южных черноземов. Цель исследований изучить экологическую пластичность орехоплодных кустарников коллекций ФНЦ агроэкологии РАН. На примере родового комплекса Corylus получены материалы по экологической пластичности орехоплодных кустарников в условиях каштановых (кадастр. 34:36:0000:14:0178), светлокаштановых почв (34:34:000000:122 34:34:060061:10). Установлены уровни изменчивости морфологических признаков ассимиляционного аппарата и плодов С. avellana L., сортов С. pontica C. Koch., которые согласуются с зимо и засухоустойчивостью. Кластерный анализ позволил выявить корреляцию признаков при 5 уровне значимости. Для целей защитного лесоразведения и озеленения засушливых районов рекомендованы Corylus avellana и Черкесский2 с выраженной вариабельностью морфологических признаков, которая указывает на их широкую экологическую валентность и адаптационные возможности в рассматриваемых условиях. The urgency of studying the ecological plasticity of woody plants in connection with the need to select an adapted species composition of trees and shrubs for protective forest plantations in the context of climate change is shown. It is established that the ecological plasticity and tolerance range of plants is associated with the ability of organisms to adapt to a particular range of environmental factors. The article presents the taxonomic composition of nut crops in the collections of FSC Agroecology RAS. Six species of Juglans: J. regia, J. mandshurica, J. cinerea, J. rupestris, J. ailanthifolia, J. nigra and three species of the genus Corylus: С. avellana L., С. аmericana W., varieties of C. pontica C. Koch the President, Futkurami, Circassian2. Data on positive experience of cultivation of Corylus avellana L. in plantation plantings (1,6 hectares, landing ordinary, the scheme of placement of 55 m) in the conditions of southern chernozems are given. The purpose of the research is to study the ecological plasticity of nutbearing shrubs of the collections of the Federal scientific center for Agroecology Russian Academy of Sciences. For example, a generic complex Corylus submissions received on the environmental plasticity of nut bushes in the conditions of brown (the cadaster nubmer 34:36:0000:14:0178), light chestnut soils (34:34:000000:122 34:34:060061:10). The levels of variability of morphological features of the assimilation apparatus and fruits of Corylus avellana L., С. pontica C. Koch varieties were established., which are consistent with winter and drought resistance. Cluster analysis revealed the correlation of features at 5 significance level. Corylus avellana and Circassian2 with a pronounced variability of morphological features, which indicates their broad ecological valence and adaptation capabilities in the conditions under consideration, are recommended for the purposes of protective afforestation and greening of dry areas.

Oil and petroleum products transportation is characterized by a significant cost of electric power. Correct oil and petroleum products accounting and forecasting requires knowledge of many factors. The software for norms of electric power consumption analysis for the planned period was developed at the Ufa Scientific Center of the Russian Academy of Sciences. Based on the principles of the relational data model, a schematic diagram/arrangement for the main oil transportation objects was developed, which allows to hold the initial data and calculated parameters in a structured manner.

AbstractEndoglucanase producing bacteria were isolated from Egyptian soils and the most active bacterial strain was identified as Bacillus subtilis strain Fatma/1. Plackett–Burman statistical design was carried out to assess the effect of seven process variables on endoglucanase production. Carboxymethyl cellulose (CMC), yeast extract and peptone were the most significant variables that enhanced the endoglucanase production and thus were selected for further optimization using face-centered central composite design. The highest yield of endoglucanase (32.37 U/mL) was obtained in run no. 9, using 18 g/L CMC, 8 g/L peptone, 7 g/L yeast extract and 0.1 g/L FeSO4.7H2O. The optimized medium showed about eightfold increase in endoglucanase production compared to the unoptimized medium. The produced crude enzyme was further purified by ammonium sulfate precipitation, then DEAE-Sepharose CL6B column. The purified enzyme was shown to have a molecular weight of 37 kDa. The enzyme showed maximum activity at pH 8.0, temperature of 50 °C, incubation time of 60 min. The half-life time (T1/2) was 139.53 min at 50 °C, while being 82.67 min at 60 °C. Endoglucanase at concentration of 12 U/mL effectively removed 84.61% of biofilm matrix of Pseudomonas aeruginosa with marked reduction in carbohydrate content of the biofilm from 63.4 to 7.9 μg.

В настоящей статье рассматривается проблема изучения музейной коллекции буддийского изобразительного искусства Калмыкии, а также сохранения художественного наследия в условиях глобализации культур. Автор ставит цель создать научно обоснованные информационные ресурсы на базе исследования музейных коллекций и экспонатов. Информация составляет суть музейного дела, в основе которого лежит научно-исследовательская деятельность, объединяющая сферы комплектования, учета и хранения фондов, с одной стороны, с другой коммуникативная, связанная с экспозиционно-выставочной деятельностью. Исходными в работе музея являются не только сохраняемый фонд, но и сведения об экспонатах, собираемые в процессе комплектования фонда и создания каталога музейного собрания, состоящего из коллекций. Основываясь на богатой традиции описания и каталогизации предметов искусства, а также применяя современные технологии и искусствоведческие методы, стало возможным подготовить и издать научный каталог коллекций основного фонда Музея традиционной культуры имени Зая-пандиты Калмыцкого научного центра РАН. This article discusses the problem of studying the museum collection of Buddhist fine art in Kalmykia, as well as preserving the artistic heritage in the context of globalization of cultures. The author aims to create scientifically based information resources that include research on museum collections and exhibits. Information is the essence of museum business, which is based on research activities that combine the fields of acquisition, accounting and storage of funds, on the one hand, and on the other hand, communicative activities related to exposition and exhibition activities. The initial work of the museum is not only the preserved fund, but also information about the exhibits collected in the process of acquiring the fund and creating a museum collection catalog consisting of collections. The Zaya-Pandita Museum of Traditional Culture of the Kalmyk Scientific Center of the Russian Academy of Sciences stores and presents in the educational sphere multidisciplinary and largely unique information about the cultural heritage of the people. Based on a rich tradition of describing and cataloging objects of art, as well as using modern technologies and art criticism methods, it has become possible to prepare and publish a scientific catalog of the collections of the main fund of the Zaya-Pandita Museum of Traditional Culture.
