Phenotypic and genotypic characterization of Bordetella bronchiseptica strains isolated from pigs in Poland

AbstractA total of 209 Bordetella bronchiseptica (Bbr) strains isolated from pigs were examined. Phenotypic study included: biochemical characterization (motility, catalase, oxidase, urease activity, nitrate reduction and growth on MacConkey agar) and antimicrobial susceptibility (disc diffusion method). Genotypic studies based on detection of three genes encoded virulence factors, such as: flagella (fla), dermonecrotoxin (dnt), and exogenous ferric siderophore receptor (bfrZ), using PCR.Most of the Bbr strains tested had a homogeneous biochemical profile. 97.6% of them provided suitable results in biochemical tests. All Bbr isolates tested showed high resistance to penicillin (100%), linco-spectin (100%) and ceftiofur (97.9%). Over 57% and 43% of Bbr strains were resistant to ampicillin and amoxicillin, respectively. All Bbr isolates showed high sensitivity to most chemotherapeutics used such as enrofloxacin (97.9%), tetracycline (97.9%), oxytetracycline (97.9%), amoxicillin with clavulonic acid (95.8%), florfenicol (90.4%), and gentamicine (77.6%). Over of 94% of Bbr strains were moderately susceptible to norfloxacine.Molecular analysis confirmed that almost all evaluated Bbr strains (94.7%) possessed the fla gene. A lower percentage of isolates had the dnt gene (72.7%) and the lowest percentage of strains (51.7%), had the bfrZ gene.

In this study 100 samples were collected from infected children with acute and chronic tonsillitis who attended to Al-Yarmook Teaching Hospital (ENT consultation clinic) from 5/12/2013 to 1/3/2014. The result of laboratory culture was positive in 67 samples. Depending on their cultural, morphological and biochemical characterization of bacterial isolate of them were identified as (37.31%) belonged to Streptococcus pyogenes and the diagnosis is confirmed by the use of Remel Rapid STR System, (34.32%) belonged to S.parasanguinis, (11.94%) S.mitis, (11.94%) S.oralis and (4.47%) S.thoraltensis . Results confirmed that cup assay gave highest inhibition zone after 24 hrs compare with well diffusion methods for suspension of L.acidophilus gave highest inhibition zone after 48 hrs for incubation, while ahigh inhibition zone revealed for suspension of L.fermentum after 24 hrs incubation. the study included also the measurement of the inhibition activity for bacteriocins produced by L.acidophilus bacteria against pathogenic bacteria on nutrient agar by well diffusion method in which results revealed stability of the bacteriocins produced towards PH which kept its activity with PH 4-6 for 24 hrs, and the highest stability was with PH 4, however it lost a lot of its activity with acidic PH less than 2 and alkaline PH as 8. The treatment of bacteriocins with salts such as Nacl it revealed little effect in inhibition zone within 1 & 2% concetrations. The salt MgSo4 & Kcl showed reduction in the inhibitory activity in the low concentration, however the higher concentration of salt caused great reduction and 5% concentration led to loss of inhibitory activity for bacteriocins completely.

Abstract Four phenylacetaldehyde dehydrogenases (designated as FeaB or StyD) originating from styrene-degrading soil bacteria were biochemically investigated. In this study, we focused on the Michaelis-Menten kinetics towards the presumed native substrate phenylacetaldehyde and the obviously preferred co-substrate NAD+. Furthermore, the substrate specificity on four substituted phenylacetaldehydes and the co-substrate preference were studied. Moreover, these enzymes were characterized with respect to their temperature as well as long-term stability. Since aldehyde dehydrogenases are known to show often dehydrogenase as well as esterase activity, we tested this capacity, too. Almost all results showed clearly different characteristics between the FeaB and StyD enzymes. Furthermore, FeaB from Sphingopyxis fribergensis Kp5.2 turned out to be the most active enzyme with an apparent specific activity of 17.8 ± 2.1 U mg-1. Compared with that, both StyDs showed only activities less than 0.2 U mg-1 except the overwhelming esterase activity of StyD-CWB2 (1.4 ± 0.1 U mg-1). The clustering of both FeaB and StyD enzymes with respect to their characteristics could also be mirrored in the phylogenetic analysis of twelve dehydrogenases originating from different soil bacteria.

In vitro antimicrobial sensitivity of 12 Hungarian isolates and the type strain ATCC 33144 of Actinobaculum suis to different antimicrobial compounds was determined both by the agar dilution and by the disc diffusion method. By agar dilution, MIC50 values in the range of 0.05-3.125µg/ml were determined for penicillin, ampicillin, ceftiofur, doxycycline, tylosin, pleuromutilins, chloramphenicol, florfenicol, enrofloxacin and lincomycin. The MIC50 value of oxytetracycline and spectinomycin was 6.25 and 12.5µg/ml, respectively. For ofloxacin, flumequine, neomycin, streptomycin, gentamicin, nalidixic acid, nitrofurantoin and sulphamethoxazole + trimethoprim MIC50 values were in the range of 25-100µg/ml. With the disc diffusion method, all strains were sensitive to penicillin, cephalosporins examined, chloramphenicol and florfenicol, tetracyclines examined, pleuromutilins, lincomycin and tylosin. Variable sensitivity was observed for fluoroquinolones (flumequine, enrofloxacin, ofloxacin), most of the strains were susceptible to marbofloxacin. Almost all strains were resistant to aminoglycosides but most of them were sensitive to spectinomycin. A strong correlation was determined for disc diffusion and MIC results (Spearman's rho 0.789, p<0001). MIC values of the type strain and MIC50 values of other tested strains did not differ significantly. Few strains showed a partially distinct resistance pattern for erythromycin, lincomycin and ampicillin in both methods.

The study was conducted to determine the frequency of isolation ofSalmonella,CampylobacterandE. coliO157 in the faecal samples of rats trapped across the regional corporations in Trinidad and to assess their resistance to antimicrobial agents. A total of 204 rats were trapped for the detection of selected bacteria. Standard methods were used to isolateSalmonella,CampylobacterandE. coliO157. Characterization ofE. coliwas done on sorbitol MacConkey agar to determine non-sorbitol fermentation, blood agar to determine haemolytic and mucoid colonies and by usingE. coliO157 antiserum to determine O157 strain. The disc diffusion method was used to determine resistance to nine antimicrobial agents. Of the 204 rats, 4 (2.0%), 7 (3.4%) and 171 (83.8%) were positive forSalmonellaspp.,Campylobacterspp. andE. coli, respectively. Of the 171 isolates ofE. colitested 0 (0.0%), 25 (14.6%) and 19 (11.1%) were haemolytic, mucoid and non-sorbitol fermenters, respectively. All isolates were negative for the O157 strain. The frequency of resistance to the 9 antimicrobial agents tested was 75% (3 of 4) forSalmonella, 85.7% (6 of 7) ofCampylobacterspp. and 36.3% (62 of 171) forE. coli(;χ2).

Objectives: To study the bacteriological profile of enteric fever and their antibioticsensitivity pattern to commonly used antibiotics in view of emerging resistance at Civil HospitalKarachi (C.H.K.) and provide a guideline for making a protocol for empirical antibiotic therapywhere culture facilities are not available. Methodology: Design: Descriptive laboratory Basedstudy. Place and Duration of study: The study was conducted from 1st January, 2010 till 30thJune, 2012 at the Central Lab, Civil Hospital Karachi. Patients and Methods: A cross sectionalanalysis was done on a total of 37,805 blood specimens sent for C/S from the entire inpatient andoutpatient departments to the central lab, CHK. All blood cultures were inoculated in thioglycolatebroth and incubated at 37° for 7 days. Positive blood cultures were processed, colonies wereidentified using standard biochemical tests and antibiotic susceptibility was checked by Kirby-Bauer disc diffusion method as per CLSI criteria. Results: Out of these specimens, 430 sampleswere found positive for the salmonella strains, among them 395 (91.86%) were Salmonella typhiand 35 (8.14%) were Salmonella paratyphi A. Salmonella infection was more common in male(55.85%) and more prevalent in children under 10 years of age (34.88%). The resistance patternfor Amoxicillin, Chloramphenicol, Co-trimoxazole, was (29%), (41%) and (5%) respectively;while the resistance was found to be higher in the commonly prescribed drugs belonging to firstand second generation of cephalosporins and flouroquinolones. Conclusion: Amoxicillin andChloramphenicol, the first line of drugs for the treatment of enteric fever are losing their efficacyand most of the organisms have developed resistance. Also, resistance against the second lineof therapy, involving the use of cephalosporins and quinolones is rapidly emerging.

Background: The debranching starch enzymes, isoamylase 1 and 2 are well-conserved enzymes present in almost all the photosynthetic organisms. These enzymes are involved in the crystallization process of starch and are key components which remove misplaced α-1,6 ramifications on the final molecule. Aim: In this work, we performed a functional and structural study of a novel isoamylase from Ostreococcus tauri. Methods: We identified conserved amino acid residues possibly involved in catalysis. We also identified a region at the N-terminal end that resembles a Carbohydrate Binding Domain (CBM), which is more related to the family CBM48, but has no spatial conservation of the residues involved in carbohydrate binding. Results: The cloning, expression and biochemical characterization of this N-terminal region confirmed that it binds to polysaccharides, showing greater capacity for binding to amylopectin rather than total starch or amylose. Conclusion: This module could be a variant of the CBM48 family or it could be classified within a new CBM family.

Rhizospheric bacteria exhibiting antagonistic effects are a good source for the production of antibiotics. The antibiotics produced are naturally bactericidal or bacteriostatic in nature. In the present investigation, thirty-five rhizospheric bacteria were isolated from different soil samples. Agar well diffusion method, streak agar method, disc diffusion method and biochemical tests were performed to screen the ten antibiotic-producing bacteria. Among them, strain JRR34 selected on the basis of primary antagonistic activity was identified as Streptobacillus sp. Media optimisation was done to ensure maximum production of secondary metabolites. Streptobacillus sp. JRR34 showed good inhibitory activity against Escherichia coli, Pseudomonas aeruginosa and Staphylococcus aureus. The ethyl crude extract of Streptobacillus sp. JRR34 rhizobacteria possessing good antagonistic activity against a wide range of pathogenic bacteria can be a vital source of novel antibiotics.

ResumenEl síndrome nefrótico  se diagnóstica con base en cuatro criterios: proteinuria, hipoalbuminemia, edema e hipercolesterolemia. Generalmente se presenta entre los 2 y 10 años de edad. Este trabajo se realizó para determinar la caracterización bioquímica del síndrome nefrótico de la población infantil del Hospital del Niño de Villahermosa, Tabasco, México.Estudio descriptivo, transversal, retrospectivo realizado en el laboratorio clínico del Hospital del Niño Dr. Rodolfo Nieto Padrón de enero 2016 a diciembre 2017. Se obtuvo el rango y promedio de las pruebas bioquímicas de pacientes con síndrome nefrótico.Se analizaron 33 pacientes referidos con  diagnóstico de síndrome nefrótico,  encontrándose una media de albumina de 1.3g/dl, colesterol de 482.6 mg/dl y triglicéridos de 523.2 mg/d y una proteinuria media de 295.9 mg/dll. Palabras clave: Síndrome nefrótico,  Hipoalbuminemia, Hipercolesterolemia.   AbstractThe nephrotic syndrome is diagnosed based on four criteria: proteinuria, hypoalbuminemia, edema and hypercholesterolemia. Almost all regions in Nusa Tenggara Timur East Nusa Tenggara (NTT) Province Indonesia are endemic areas of Haemorragic Septicaemia (HS), which is caused by Pasteurella multocida  (P. multocida ) Serotypes B: 2. The fragment  pfhaB1gene is one of the virulence factors of P. multocida.The objective of this study was to determaine the phylogenetic, homology of P. multocidapfhaB1gene fragment of isolatedfrom Buffalo and Cattle in NTT. The P. multocida isolateswere re-cultured and further microscopic examined the biochemical tests, PCR, sequencing, homology, and phylogenetic relatedness test. P. multocida was observed as gram negative, coccobacilus, no growth on MacConkey Agar, does not produce H2S and gas, nonmotile and indole positive, does not produce urease enzymes, does not use citrate as a carbon source, does not ferment maltose and lactose but it does ferment glucose, sucrose and mannitol. ThepfhaB1gene fragmentfrom buffalo and cattle NTT isolates and also Katha strain vaccine showed DNA band 506 bp. P.multocida isolates from buffalo and cattle in NTT have 91% - 99% score homology with the comparative isolate. The isolate P. multocida from buffalo and cattle in NTT are in one cluster and their phylogenetic relatedness is close to isolates from Iran and India. It is concluded that the  pfhaB1gene fragmentof P. multocida from buffalo and cattle isolates have phylogenetic relatedness close and homolog with the other comparative isolates.    Keywords: Haemorrhagic Septicaemia; Nusa Tenggara Timur; Pasteurella multocida;  pfhaB1gene

Diarrhoea is a self-limiting  disease  caused by various  enteropathogens and antimicrobial  therapy  is  an  important adjunct  to  fluid  therapy  in  the  management of the  cases.  We have studied  the  flood-affected  diarrhoeal  (post-flood diarrhoea) patients  admitted in  Dhaka Medical College Hospital  during  August to September,  2007.  A total  of ll4 faecal  samples  were  collected  from  the  patients of all age  groups,  and  investigated  for  bacterial  enteropathogens. Specimens  were cultured  in  appropriate  media and  identification  of the  organisms  showing  growth  was done  by relevant  biochemical  tests.  In addition, the isolates of Vbrio cholera wer confirmed  using  pollvalent  antisera. Sensitivity of the isolated organisms was done by disc diffusion method.  Among the isolated bacterial enteropathogens, majority was V. cholera (46, 40.3%). Other  isolates  included Escherichia coli (18,  23.0%),  Aeromonas species  (8,10.O%)  and V parahaemolyticus  (5,  7.0%). No pathogenic bacteria were isolated from 27 of the cases.  No Salmonella or Shigella was detected in any case. Majority of the cases  (24, 52.0%)  infected  with V cholerae  were  children  having  less than  L0 years of  age. Most of the isolates of V cholerae (77.7%) were sensitive to Ciprofloxacin.  Almost all of the V cholerae isolates (97.90 ) were resistant to Tetracycline, followed by Co-trimoxazloe (93,5%)  and  Nalidixic  acid (59.2%).  All of the E. coli isolates (100.0%) were sensitive to Ceftriaxone, followed by Ceftazidime (88.8%) and Ciprofloxacin (77.7oh).  Thus it  can  be concluded  that  Ciprofloxacin and Ceftriaxone  can  be considered  as the drugs  ofchoice  for  the  treatment  of  post-flood  diarrhoea  along  with proper  rehydration  therapy.Bangladesh  J Med Microbiol2008;  02 (01):13-17DOI: http://dx.doi.org/10.3329/bjmm.v8i1.21929
