Seaweed has potential nutrient content such as carotenoids, vitamins, fatty acids, carbohydrates, minerals, and other essential substances. Carotenoids have important biological functions as an antioxidant, and immunostimulatory which can prevent the disease, anti-inflammatory, anti-stress, anti-aging, and protect the skin from the harmful effects of ultraviolet radiation. Seaweed generally consumed as a vegetable by people in Bali, known as the local name Bulung Boni (Caulerpa spp.) and Bulung Sangu (Gracilaria spp.).. So far there has been no report or results of research on the effects of extract ethanol of Bulung Boni (Caulerpa sp.) and Bulung Sangu (Gracilaria sp.) as an antioxidant that can prevent lipid peroxidation which can be seen in decreased level of MDA in liver tissue or blood plasma. Therefore it is necessary to determine of plasmaMDA level of Wistar rat after fed high cholesterol diet treated with extract ethanol of Caulerpa sp. and Gracillaria sp. This experimental study used completely randomized design. Research using total of 24 Wistar rats divided into six sample groups of equal size, all fed with a diet high in cholesterol especially in negative control. The study consisted of negative control group (standard diet), positive control group (high cholesterol diet), high-cholesterol diet with Caulerpa sp. extract dose of 20 mg and 60 mg/100 g, high cholesterol diet with Gracilaria sp. extract dose of 20 mg and 60 mg/100 g body weight rat per day.The study resulted that rats fed high cholesterol diet with treated extract ethanol Caulerpa sp. and Gracilaria sp. with a dose of 20 mg and 60 mg per 100 g body weight rat / day had plasma MDA level significantly lower (p <0.05) compared with rats fed high cholesterol diet without treated with extract of Caulerpa sp. and Gracilaria sp.

Objective: The aim of this study is to evaluate the antidyslipidaemic and cardioprotective effects of aqueous extract of Curcuma longa (AECL) in rats fed a high cholesterol diet (HCD). Methods: Twenty (20) rats were randomly grouped into four groups: A-D of five animals per group. Groups A-C received HCD (2000mg/kg, oral) and carbimazole (60mg/kg, oral) daily for eight weeks. Group A served as negative control. Group B (positive control) was treated with atorvastatin (20mg/kg), while group C served as treatment group and received AECL (400mg/kg) daily for eight weeks. Group D served as normal control and received no treatment. After the administration, biochemical markers of Lipid profiles (total cholesterol (TC), triglycerides (TG) and high density lipoprotein cholesterol (HDL-C)) were assayed using standard methods. The hearts of the rats were harvested for histopathological studies. Results: AECL significantly induced hypolipidaemia and stabilized lipid biochemical markers (p<0.05 or p<0.01); and protected the cardiac muscle fibres from injuries. Conclusion: Tumeric (Curcuma longa) has antidyslipidaemic and cardioprotective effects. Obesity has been linked with several comorbidities like cardiovascular disorders, diabetes, and hypertension. Weight reduction is directly associated with the consumption of green tea. So, with this premise of background, we evaluated the potential of green tea consumption on obesity and serum lipid profiles in both normo- and hyper-cholesterol emic rabbits. Sixty New Zealand white rabbits (age: 6-8 months; weight 1.5-2 kg) were used for this experiment. Rabbits were randomized and equally divided into three different groups. Group I (control group) received 10 ml/kg of distilled water twice daily, Group II received high cholesterol diet twice daily, and Group III received high cholesterol diet along with green tea solution 10 ml/kg, twice daily for 90 days. Weight measurements and serum analysis were conducted on Day 0 and day 90. For the obtained data, SPSS analysis was performed (SPSS 21). A significant increment of body weight was observed in control (1.97 ± 0.12 vs 2.87 ± 0.21, p<0.0001) and high cholesterol groups (1.91 ± 0.23 vs 3.13 ± 0.31, p<0.0001) at Day 90 when compared with Day 0. Though the results were not significant, there was a reduction in the body weight in the green tea group (2.20 ± 1.47 vs 1.72 ± 0.22) at Day 90 when compared with Day 0. Consumption of green tea for 90 days resulted in weight loss and inhibition of the increase in the levels of TC, LDL, and VLDL. So, green tea can act as a natural food supplement for the management of obesity and dyslipidemia.

Effect of garlic (Allium sativum Linn.) extract on reduction of total cholesterol and HDL cholesterol ratio in hypercholesterolemic rats (Rattus norvegicus). Biofarmasi 6: 45-51. Garlic has been widely recognized as herbal medicine for prevention and treatment of cardiovascular disease, atherosclerosis, and hyperlipidemia. The main bioactive compound, allicin, can influence cholesterol metabolism, reduce total cholesterol and increase HDL cholesterol. The purpose of this research was to find out the effect of garlic extract on the reduction of total cholesterol and HDL cholesterol ratio in hypercholesterolemic rats. This research was an experimental research with pre and post-test control group design. The research used male Wistar rats (Rattus norvegicus) with ±3 mounts old and ±200 grams of body weight. The rats were divided into two groups, group I and II; each group consisted of 15 rats. Before the treatment, all rats were induced by a high-cholesterol diet for two weeks, to get a hypercholesterolemic condition. Group I, as a control group, were induced by a high-cholesterol diet, while group II, treatment group, were also induced by a high-cholesterol diet orally by 0.432 grams garlic extract /200 grams body weight/day. The treatment occurred for 25 days. The data were taken before the treatment and after the last treatment, by taking the blood from orbital sine of rats which been fasted for 12 hours, and the level of serum cholesterol total and the level of serum HDL cholesterol were checked from both groups, then the results were analyzed with t-test. Based on the results, it could be concluded that the oral induction of 0.432 grams garlic extract /200 grams body weight/day for 25 days on rats, could not reduce total cholesterol, therefore the serum total cholesterol and HDL cholesterol ratio was not significant (p>0.05). Meanwhile, the decrease of total cholesterol in treatment group and the decrease of HDL cholesterol in both groups were significant (p<0.05).

Background: This research aimed to assess the effect of kombucha tea on reducing cholesterol (HCH) uptake and ameliorating biochemical and physiological side effects of high cholesterol diets.  Objectives: The aim of this research was evaluate the effectiveness of Kombucha tea in reducing cholesterol uptake as well as pathological and immunological side effects  from high cholesterol diets in Rabbit.  Methods: A total of 32 rabbits were assigned into four groups: 1. Control, fed with standard pellets; 2. HCh+St, fed with high cholesterol diet; 3. HCh+Kom, fed with high cholesterol diet and orally received kombucha beverage; and 4. Kom+St, orally received kombucha beverage along with standard diet. Then, the blood Cholesterol (CHO), Triglycerides (TG), Low-density Lipoprotein (LDL), Malondialdehyde (MDA), ferritin, Creatine Phosphokinase (CPK), Interleukin-1 (IL-1), interleukin-6 (IL-6), and liver enzymes (alanine transaminase [ALT], and aspartate transaminase [ AST]), as well as liver histopathological features, were evaluated in all groups. Results: CHO and TG showed the highest level in the blood of the HCh+St group, whereas those gavaged with HCh+Kom displayed no change in TG and LDL. Conversely, the HCh+Kom group showed a significant increase in HDL when compared with HCh+St and Kom+St groups. MDA ferritin and CPK in the Kom+St group was significantly higher than the other groups, followed by HCh+Kom. No significant difference was observed in the values of IL-1 and IL-6 as well as ALT and AST between the experimental groups and the control group. Histopathological observations of the liver tissues revealed foam cells with an acidophilic appearance in the HCh+St group and hyperemia in the sinusoid of HCh+Kom and Kom+St groups. Degradation in pancreatic acini was observed in the HCh+St rabbits.  Conclusion: The data suggest that kombucha consumption is effective in preventing problems caused by high cholesterol diets.

Objective: One of the first clinically detectable changes in the vasculature during atherogenesis is the accumulation of cholesterol within the vessel wall. Hypercholesterolemia is characterized by dysfunctional endothelial-dependent vessel relaxation and impaired NOS3 function. Since DNA methylation at gene promoter regions strongly suppresses gene expression, we postulated that high-fat/high-cholesterol diet suppresses endothelial NOS3 through promoter DNA methylation. Methods: Domestic male pigs were fed control diet (CD) or isocaloric high fat and high cholesterol diet (HC; 12% fat and 1.5% cholesterol) for 2, 4, 8 or 12 weeks prior to tissue collection. Furthermore, to determine the effects of risk factor withdrawal, an additional group of swine received HC for 12 weeks and then CD for 8 weeks; a control group received HC continuously for 20 weeks. Endothelial cells were harvested from common carotid aorta. In parallel in vitro studies, cultured human aortic endothelial cells (HAEC) were treated with human LDL, GW3956 (LXR agonist) and RG108 (DNA methyltransferase [DNMT] inhibitor). In cells from both sources, DNA methylation at the NOS3 promoter was measured using methylation specific pyro sequencing, and endothelial gene expression was measured using RT PCR. Results: HC diet increased plasma cholesterol level from 75 mg/dl on CD to a plateau of about 540 mg/dl within 2 weeks. Endothelial NOS3 expression was significantly reduced (71±9 % of CD) after 4 weeks of HC, a level sustained at subsequent time points. Withdrawal of HC for 8 weeks did not recover NOS3 expression. After 12-week HC, the NOS3 promoter was hypermethylated. Withdrawal of HC did not reverse NOS3 promoter methylation. In vitro treatment of HAEC with human LDL (200 mg/dl total cholesterol) or GW3956 (5μM) suppressed NOS3 mRNA to 50% and 30% respectively, suggesting that LXR/RXR is involved in suppression of NOS3. Nitric oxide production was consistently suppressed by GW3959. Both could be reversed through inhibition of DNMTs by RG108. Conclusions: DNA methylation and LXR/RXR pathway can mediate the HC-suppression of endothelial NOS3. The study identifies novel pharmaceutical targets in treating endothelial dysfunction. Crosstalk between these pathways is under investigation.

Lipoprotein-associated phospholipase A2 (Lp-PLA2) contributes to atherosclerotic plaque instability and subsequent sudden coronary death. Statins are associated with decreased stroke risk and may improve stability of atherosclerotic plaques. The present study investigated the effect of simvastatin on expression of Lp-PLA2 levels in atherosclerotic plaques and on Lp-PLA2 activity in atherosclerotic aortas. Rabbits were a fed chow (control group) or a high-cholesterol diet (atherosclerosis group) for 12 weeks. An additional group on the high-cholesterol diet received simvastatin (5 mg/kg per day) for the last 4 weeks (simvastatin group). Lp-PLA2 activity in plasma and atherosclerotic aortas was significantly higher in the atherosclerosis group than in the control group and, consistent with this, abundant Lp-PLA2 protein was detected in plaques in the atherosclerosis group. Simvastatin significantly reduced Lp-PLA2 activity in plasma and aorta tissue, and reduced Lp-PLA2 protein level in atherosclerotic plaques. Whereas there was no significant difference in total atherosclerotic lesion area between simvastatin and atherosclerosis groups, simvastatin significantly reduced macrophage content, lipid retention and the intima/media ratio but increased the content of smooth muscle cells in atherosclerotic lesions. Thus, statin treatment markedly reduced Lp-PLA2 in both plasma and atherosclerotic plaques. This was associated with attenuation of the local inflammatory response and improved plaque stability.

Dyslipidemia is an abnormal lipid metabolism which may cause fat degeneration on hepatocytes cells and elevated triglyceride serum level. Dyslipidemia can be prevented by the consumption of high antioxidant food. Noni(Morinda citrifolia) contains many antioxidant such as flavanoid, kuersetin, tannin, and saponin, which are able to prohibit the elevation of ROS.This research is aimed to find out the effect of noni(Morinda citrifolia) extract in reducing the number of hepatocyte’s cells with fat degeneration and decreasing the triglyceride level which is elevated due to high cholesterol diet induction.In this study white rats divided randomly into 4 groups, control group (K-), high cholesterol diet induced group (K+), high cholesterol diet induced with 100 mg/Kg BW noni extract group (P1), high cholesterol diet induced with 200 mg/Kg BW noni extract group (P2). The result shows that noni  extract with dose 100 mg/Kg BW and  200 mg/Kg BWcan reduce the number of hepatocytes cells with fat degeneration (p= 0,026 and p=0,027) and decrease the level of triglyceride serum (p=0,036 and p=0,010).The conclusion is noni extract with dose 100 mg/KgBW reduces effectively  the number of hepatocyte’s cells with fat degeneration and decreases the level of triglyceride serum which increase because of high cholesterol diet.

Background: Hyperlipidemia or Dyslipidemia is the major cause of atherosclerosis1 and associated conditions. Low levels of high-density-lipoprotein cholesterol (HDL-C) are the major causes of increased atherogenic risk 1. Aggressive cholesterol reduction in patients with atherosclerotic disease is now the standard of care2. In addition to life style modification, patients with risk factors need lipid lowering drug therapy. The drugs available now do not reduce LDL oxidation, and oxidative stress associated with hyperlipidemia. In recent years, antioxidants have been subjected to epidemiological studies4 that have related their consumption to a reduction in the incidence of oxidative damage related diseases.Methods: Hypercholesterolemia was induced in rats by administration of high cholesterol diet for 30 days in standard rat chow diet. Rats were divided into four groups of six each. Group-I and II with intake of normal diet and High cholesterol diet respectively. Group III and IV are given high cholesterol diet along with Lutein 50mg/kg and Atorvastatin 5mg/kg orally once daily respectively. At the end of 30 days animals were subjected to overnight fasting. Blood samples were drawn by retro-orbital puncture for biochemical analysis. The animals were sacrificed after thiopentone injection and liver and aorta were dissected out and processed for histopathological study and biochemical analysis.Results: Lutein treated group showed even more significant reduction in TBARS levels than the normal control group and Atorvastatin treated group. The efficacy of Lutein in slowing down the atherosclerosis and fatty infiltration of liver is proved in this study.Conclusions: Hence the present study had shown significant hypolipidemic, antiatherogenic and antioxidant effect of Luetin in Hyperlipidemic rats.

Including herbal medicine, complementary and alternative medicine (CAM) is popular worldwide. The traditional Chinese medicine xinkeshu has been widely used to treat coronary heart disease in China. This study was designed to investigate the protective effect and probable mechanism of xinkeshu tablet to atherosclerotic myocardial ischemia rabbit. Rabbits were divided into four groups (n=12each) and fed with different diet for 12 weeks: Control (standard diet), Model (high-cholesterol diet), XKS (high-cholesterol diet with 184.8 mg/kg/d xinkeshu), and Atorvastatin (high-cholesterol diet with 5.0 mg/kg/d atorvastatin). Plasma lipoprotein, ECG, endothelium-dependent vessel relaxation, histomorphological study, and expressions of eNOS and VCAM-1 on coronary arteries were assessed. The findings showed that, similar to atorvastatin, xinkeshu presented significant effects on rescuing endothelium-dependent vessel relaxation, inhibiting atherosclerotic progress, preventing myocardial ischemia, and changing eNOS and VCAM-1 expression. However, xinkeshu showed no lipoprotein lowering effect in hypercholesterolemia rabbits. The results of the present study indicated that xinkeshu exerted potent antiatherogenic and anti-ischemic properties on atherosclerotic myocardial ischemia rabbit. An endothelial protecting effect may be involved in the mechanism other than antihyperlipidemic effect.

Background : The 1st and 2nd degree hemorrhoids is managed non-operatively with the anti-inflammatory and plebothropic drugs. Graptophyllum pictum extract (GPE) has already been used widely in Indonesia to treat hemorrhoid with good result, however, the mechanism is not supported by the molecular research.Objective : This study is intended to study the phlebothropic effects of GPE by measuring the degree of edema and extravassal leucocytes of experimental wistar hemorrhoid.Methods : Experimental study with Randomized Controlled Trial Post-test only design in male wistar rats, weight around 200 gr, induced for the development of a disease-like condition of hemorrhoids by 6% croton oil on the anus for 3 days. Random allocation was performed to divide the 14 wistar rats in 2 groups. Group I as control got normal saline solution, while group II was treated with GPE 100mg/kgbw, started on day 4th for 5 consecutive days. On 9th day blood was extracted from retroorbital fossa and anus was resected up to 2 cm from anal verge and weighted. The degree of anal edema was measured by rectoanal coefficient, that is rasio between anal weight (miligrams) and body weight (grams).Results : Until the end of study, all wistar rat were still alive. The mean (±SD) of body weight   of control group was 173.84 ­(±13.37) and the treatment group was 171.70  (±13.10), and there was no significant differences (p = 0.833). The mean (±SD) of rectoanal coefficient in the treatment group was 2.46 (±0.41) and it was significantly lower than control group  (3.13 ± 0.85) (p = 0.029). The mean (±SD) of extravassal leukocytes in the treatment group was 900.14(±48.09) and it was significantly lower than the control (1003.28 ± 99.30) (p = 0.042).Conclusions : Graptophyllum pictum extract at doses of 100 mg/Kgbw have phlebotonic effect  in decreased edema and extravassal leukocytes.
