The Influence of Kenikir (Cosmos caudatus Kunth) Ethanol Extract Concentrations on Peel off Gel Formula and its Anti-Aging Effect

ABSTRAK Doksorubisin adalah agen kemoterapi yang mempunyai efek samping hepatotoksik dengan cara pembentukan radikal bebas. Daun kenikir (Cosmos caudatus Kunth.) memiliki potensi untuk mencegah efek hepatotoksik akibat penggunaan doksorubisin. Penelitian bertujian untuk membuktikan potensi Ekstrak Etanol Daun Kenikir (EEDK) sebagai hepatoprotektor pada tikus yang diinduksi doksorubisin. Penelitian ini merupakan penelitian eksperimental menggunakan randomized matched post only control group design. Sebanyak 25 ekor tikus galur wistar dibagi menjadi 5 kelompok secara acak. Kelompok I, control normal diberi asupan aquadest, kelompok II kontrol hepatotoksik diberikan doksorubisin dosis 5 mg/KgBB. Kelompok III, IV dan V merupakan kelompok perlakuan EEDK dengan dosis berturut-turut 250,500, dan 1000 mg/kgBB dengan doksorubisin dosis 5 mg/kgBB. Pemberian doksorubisin dilakukan pada hari ke-1, ke-5,ke-9 dan hari ke-13 secara intraperitonial (ip) dan pemberian EEDK dilakukan 1 kali sehari selama 14 hari secara peroral (po). SEluruh hewan uji diambil darahnya pada hari ke-15. Kadar SGOT dan SGPT dievaluasi untuk melihat adanya kerusakan hati. Data kadar SGOT dianalisis dengan one way anova, sedangkan data kadar SGPT dianalisis menggunakan Kruskall-wallis. Hasil penelitian menunjukkan bahwa doksorubisin dapat menyebabkan hepatotoksik ditandai dengan peningkatan kadar SGOT dan SGPT. Ekstrak etanol daun kenikir mampu memberikan efek hepatoprotektor yang signifikan (p≤0,05) pada tikus yang diinduksi doksorubisin ditunjukkan dengan penurunan kadar SGOT dan SGPT. Kata Kunci: doksorubisin, daun kenikir, hepatoprotektor, SGOT, SGPT  ABSTRACT Doxorubicin is a chemotherapeutic agent has hepatotoxic side effects through the formation of free radicals. Marigold leaves (Cosmos caudatus Kunth.) has the potential to prevent hepatotoxic effects caused by the use of doxorubicin. This study aims to prove the potential of Ethanol Extract of Marigold Leaves (EEDK) as a hepatoprotector in doxorubicin-induced rats. This study was an experimental study with a randomized matched post test only control group design. 25 wistar male rats were divided into 5 groups randomly. Group I as normal control was given aquadest intake, group II as hepatotoxic control was given doxorubicin dose 5 mg/Kg BW, group III, IV and V was EEDK treatment group dose 250, 500 and 1000 mg/Kg BW and doxorubicin dose 5 mg/Kg BW. Doxorubicin was given on day 1, 5, 9 and 13intraperitoneal (i.p) and EEDK was given once a day for 14 days orally (p.o). All blood samples were taken on the 15th day. SGOT and SGPT levels were evaluated to see liver damage. SGOT level data were analyzed statistically by One Way Anova, while SGPT level data were analyzed using Kruskall-Wallis. The results showed that doxorubicin-induced hepatotoxity characterized by increased SGOT and SGPT levels. Ethanol extract of marigold leaves can provide hepatoprotector effect significantly (p≤0.05) in doxorubicin-induced rats indicate by decreased SGOT and SGPT levels. Keywords: doxorubicin, marigold leaves, hepatoprotector, SGOT, SGPT

Aims: To evaluate the application potential of Xylaria striata Pat. in anti-aging field. Methodology: Extracting the fruit body of X. striata by ultrasonic-assisted extraction method. In this study, Drosophila melanogaster was used as an anti-aging organism material. After feeding with different concentrations of extract, the natural survival time, survival time under oxidative stress and survival time under UV irradiation of D. melanogaster were all measured. In addition, the in vivo activity of SOD、CAT and MAD, protein concentration and body weight were determined to evaluate the anti-aging effect of ethanol extract from X. striata. Results: The results showed that the ethanol extract of X. striata could extend the lifespan of D. melanogaster under both irradiation and oxidative stress condition. And the ethanol extract could enhance the activity of CAT in D. melanogaster, especially at concentration of 50 μM, and the content of MAD in D. melanogaster were decreased significantly. Conclusion: This study clarified the anti-aging activity of X. striata in D. melanogaster and  it would provide some theoretical basis for its further development and utilization in anti-aging drugs and health food.

BACKGROUND: The product development of an anti-aging dosage for human skin is a very interesting issue because every human wants to be looked young as long as possible. A studies and development of plant that contain of antioxidant properties need to be studied further. AIM: The aim of this research was to formulate and evaluate the physical and anti-aging effect of the Peel-Off Gel Mask from Red Bean Ethanol Extract (PGMRB). METHODS: The Extract Ethanol of Red Bean was extracted by maceration methods with ethanol 96%. The Peel-off base was consisted of Polyvinyl alcohol, glycerin and carbomer 940. The gel formulation was made in 3 formulas (F1, F2, and F3) consisted with 1%, 3% and 5% ethanol extract, respectively. The blank formula (F0) was also prepared as a control formula. The evaluation of gel formulation included organoleptic test, pH determination, stability test, homogeneity test, peeling time, irritation test and anti-aging effect test included moisture level, skin pore size, the evenness test and dark spot test by using skin analyzer. RESULTS: All of the formula showed a homogen, pH around 6, peeling time less than 20 minutes, no irritation and stable for 12 weeks storage at room temperature. The treatment with PGMRB demonstrated an improvement on the moisture level, pore size, evenness and the number of black spots with F3 containing 5% extract as the best formula. CONCLUSION: The ethanol extract of red bean can be formulated become a peel-off gel mask and having the anti-aging effect.

<p>Chronic dyslipidemia, especially in hypercholesterolemia, can directly disrupt endothelial cell function through increased formation of oxidized LDL which will stimulate an inflammatory response including interleukin 6 (IL-6) release. For this reason, it is necessary to have anti-inflammation that can reduce the effects of hypercholesterolemia on blood vessels. Polyphenols have anti-inflammatory effects and kenikir leaves are vegetables that are rich in polyphenols. This study aims to prove the effect of kenikir leaf (<em>Cosmos caudatus</em>) ethanol extract on IL-6 levels of serum of male Wistar rats fed high cholesterol. The method used is a true experimental study with a Post-test Only Control Group Design. Samples using 24 male rats were divided into 4 groups DS, DTK, Ss, and Cc. DTK, Ss and Cc groups were given high cholesterol feed for 20 weeks. In the 17th to 20th weeks the Ss group was given additional simvastatin and the Cc group was given an additional extract of <em>Cosmos caudatus</em> leaf. Whereas the DS group was only given standard feed for 20 weeks. After 20 weeks all rats in all four groups were examined serum GSH and IL-6 levels. The results showed the highest GSH serum levels average in Cc with p-value=0,016 but not different from Ss. IL-6 serum levels in DTK were higher than those of the other three groups and were significantly different with p-value=0,035. The difference occurs between DTK and DS, Ss and Cc, not different between DS, Ss and Cc. The ethanol extract of kenikir leaf influences the serum IL-6 levels of male Wistar rats fed high cholesterol.</p>

ABSTRACT From the ethanol extract of 100 full term placentae approximately 200 μg of an α-ketolic Kober chromogen have been isolated in a »free« form. The evidence obtained indicates that this α-ketolic Kober chromogen is identical with 16-oxooestradiol-17β.
