Presence of Salmonella in organic farming may lead to contamination in fresh produce. This study was designed to detect Salmonella contamination in organic vegetable farm situated in Serdang and to evaluate the antibiotic susceptibility profiles of the isolates. A total of 460 samples of brinjal, cucumber, ladies’ fingers and soil were collected and examined for the presence of Salmonella. The obtained isolates were identified and confirmed by biochemical characterization and serotyping. Antibiotic sensitivity profiles of the isolates were determined by using agar disk diffusion method. Salmonella spp. was detected in brinjal (1.7%, n=2), cucumber (1.7%, n=2), ladies’ fingers (2.5%, n=3) and soil (5.0%, n=5) samples. The prevalent serovars were Salmonella enterica ser. Senftenberg, S. enterica ser. Weltevreden and S. enterica ser. Corvallis. All strains were resistance to penicillin and vancomycin, with multiple antibiotic resistance (MAR) index between 0.21 and 0.36, demonstrated here as multi-drug resistant (MDR) Salmonella. The result highlighted that organic vegetables constituted potential sources of Salmonella, informing continuous monitoring and tightened surveillance are necessary to ensure food safety.

Microbial contamination of vended foods are of public health importance due to the potential of becoming a reservoir of foodborne pathogens and resistant strains of bacteria. This study looked at the presence of pathogenic bacteria in a popular Ready-To-Eat (RTE) traditional food, Fufu in Ghana. Sixty (60) Fufu samples were obtained from various food joints categorized as Opened, Semi-closed and Closed or Restaurants. Samples were processed and analyzed using standard bacteriological methods. The susceptibility profiles of the isolates were obtained by using the Kirby-Bauer disk diffusion method with the EUCAST guidelines with the five antibiotics. Prevalence of E. coli was 85% and Salmonella species was 68%. Microbial count of isolated E. coli ranged from 0 to 3×106 cfu/ml. There were no significant differences (p>0.05) among the different modes of operations. Fufu samples from Opened, Semi-closed and Closed joints were respectively contaminated with E. coli and Salmonella species as follows: 92%, 76%; 80%, 60% and 80%, 65%. The Salmonella species showed highest resistance to erythromycin (58.5%) and E. coli species were commonly resistant to Ceftazidime (88.2%) and Ceftriaxone (94.1%). All isolates were susceptible to nitrofurantoin. Multidrug resistance was detected among 27.5% of E. coli strains and 14.6% of Salmonella species. Fufu from the different eating joints in the Tamale Metropolis were substantially contaminated with multidrug resistant pathogens. The study recommends surveillance studies of resistant pathogens in foods, increased education and training of food vendors on sanitation, food handling and safety practices in the region.

The risk of foodborne diseases as well as the dissemination of antibiotic resistant bacteria increases with the consumption of street-vended food and beverages. This study investigated the prevalence of Salmonella spp. and Citrobacter spp. in street-vended beverages sold in Chow Kit, Kuala Lumpur, Malaysia. The Kirby-Bauer disk diffusion method was used to identify the antibiotic resistance profile of Salmonella spp. and Citrobacter spp. isolates towards 11 selected antibiotics. Six beverage samples were found positive for presumptive Salmonella spp. and Citrobacter spp. Upon confirmation via Microgen kit and PCR biochemical testing methods, only one isolate was confirmed to be Salmonella enterica serovar Derby while the other isolates were identified as Citrobacter spp. (n= 12; 2 isolates from each positive beverage sample). The antibiogram test showed that 58.3%, 16.7%, and 8.3% of the strains tested were resistance to tetracycline, cephalexin, and ampicillin respectively, while all isolates were fully resistant toward penicillin and erythromycin. The isolate with the highest MAR index (0.45) was S231, with resistance to five of the tested antibiotics (penicillin, erythromycin, tetracycline, cephalexin, and ampicillin). Seven isolates had a MAR index of 0.27 and were resistant to three antibiotics, while the remaining four isolates had the lowest MAR index (0.18) and were resistant to only two antibiotics. This study shows that street-vended beverages have a high risk of spreading antibiotic-resistant bacteria to the public and that Citrobacter spp. should be considered as emerging multidrug-resistant bacteria in the food production system.

ABSTRACT: The aim of the present study was to investigate the presence of Salmonella spp. in samples collected from beef meat at three points of the slaughter line (after skinning, washing and cooling) at three slaughterhouses in Brazil that export meat. Detection was based on ISO 6579:2002 and confirmed by PCR and qPCR. The isolates were typified using slide agglutination tests and PFGE. The antibiotic sensitivity profile was determined using the disk diffusion method. Contamination was detected in only one slaughterhouse. The overall frequency of contamination by Salmonella spp. was 6.7% of carcasses (6/90) and 2.6% of carcass surface samples (7/270). All isolates were confirmed by PCR and qPCR. The serological analysis and the PFGE showed a single profile: Typhimurium. The strains demonstrated 100% susceptibility to ampicillin, cefotaxime, ciprofloxacin, chloramphenicol, gentamicin and tetracycline. Positive carcasses after cooling pose a direct risk to consumers, since the meat is considered ready to be marketed after this process.

Streptococcus equi subsp. zooepidemicus (S. zooepidemicus), is a β-hemolytic Streptococcus belonging to the Lancefield group C; it is a rare human pathogen, but in horses, it is frequently associated with endometritis. This study aimed to isolate S. zooepidemicus strains, associated with bacterial endometritis in mares, and to define their antimicrobial resistance profile. Twenty-three isolates were recovered from one hundred ninety-six equine uterine swabs (11.7%). Bacterial identification was carried out by Api 20 Strep and confirmed by matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS), while antimicrobial susceptibility testing was performed by disk diffusion method on Muller Hinton agar plates. The antibiotic resistance profiles of the isolates revealed a high percentage of resistance to amikacin (95.6%), ampicillin (73.9%) and tetracycline (69.6%), while ceftiofur and ceftriaxone were highly effective with 82.6% and 78.3% of the isolates inhibited, respectively. An intriguing value of resistance to penicillin (34.8%), which represents the first-choice antibiotic in equine S. zooepidemicus infections, was observed. Furthermore, a high prevalence of multidrug-resistant strains (82.6%) was recorded. Continuous surveillance of this potential zoonotic pathogen and an appropriate antimicrobial stewardship program with the promotion of correct use of antimicrobials, after a proper diagnosis, are needed to allow an effective therapy.

The presence of Salmonella spp. in marine animals is a consequence of contamination from terrestrial sources (human activities and animals). Bacteria present in marine environments, including Salmonella spp., can be antibiotic resistant or harbor resistance genes. In this study, Salmonella spp. detection was performed on 176 marine animals stranded in the Sicilian coasts (south Italy). Antibiotic susceptibility, by disk diffusion method and MIC determination, and antibiotic resistance genes, by molecular methods (PCR) of the Salmonella spp. strains, were evaluated. We isolated Salmonella spp. in three animals, though no pathological signs were detected. Our results showed a low prevalence of Salmonella spp. (1.7%) and a low incidence of phenotypic resistance in three Salmonella spp. strains isolated. Indeed, of the three strains, only Salmonella subsp. enterica serovar Typhimurium from S. coeruleoalba and M. mobular showed phenotypic resistance: the first to ampicillin, tetracycline, and sulphamethoxazole, while the latter only to sulphamethoxazole. However, all strains harbored resistance genes (blaTEM, blaOXA, tet(A), tet(D), tet(E), sulI, and sulII). Although the low prevalence of Salmonella spp. found in this study does not represent a relevant health issue, our data contribute to the collection of information on the spread of ARGs, elements involved in antibiotic resistance, now considered a zoonosis in a One Health approach.

BackgroundEscherichia coli is an important aetiological agent of bovine mastitis worldwide.MethodsIn this study, 82 E. coli from bovine mastitis milk samples from 49 farms were analysed for their genetic diversity using phylogenetic grouping and multilocus sequence typing. The isolates were examined by PCR for a selection of virulence factors (VFs). Antimicrobial susceptibility profiles were assessed using the disk diffusion method.ResultsThe most prevalent phylogroups were group B1 (41.5 per cent of the isolates) and group A (30.5 per cent). A variety of 35 different sequence types (STs) were identified, including ST1125 (11 per cent), ST58 (9.8 per cent), ST10 (8.5 per cent) and ST88 (7.3 per cent). Aggregate VF scores (the number of unique VFs detected for each isolate) ranged from 1 to 3 for 63.4 per cent of the isolates and were at least 4 for 12.2 per cent. For 24.4 per cent of the isolates, the score was 0. The three most frequent VFs were traT, fyuA and iutA. The majority (72 per cent) of the isolates harboured traT. The majority (68.3 per cent) of the isolates were fully susceptible to all antimicrobials tested, with 22 per cent resistant to ampicillin and 14.6 per cent to tetracycline. Resistance rates were low for gentamicin (3.7 per cent), amoxicillin/clavulanic acid (2.4 per cent) and ceftiofur (1.2 per cent), respectively.ConclusionAmong the study’s sample population, E. coli strains were genotypically diverse, even in cows from the same farm, although some STs occurred more frequently than others. Susceptibility to clinically relevant compounds remained high.

The presence of Salmonella in food products and emergence of antibiotic resistance are the major challenges facing public health policies. A total of 2749 crocodile meat samples obtained from the Central Veterinary Laboratories in Zimbabwe were screened for Salmonella specieswere collected from three Zimbabwean commercial farms between the year 2012 and 2019 for a retrospective observational study to determine the prevalence and magnitude of antibiotics resistant Salmonella species in crocodile meat. The isolation of Salmonella was in accordance with the ISO 6579:2002 and the antibiotic susceptibility testing was carried out based on Clinical and Laboratory Standard Institute’s recommendations by means of the Kirby-Bauer disk diffusion method. SILAB Database was used to determine the prevalence of Salmonella species. Prevalence was stratified by year and farms. Twenty Salmonella isolates were identified using biochemical tests, and 15 were confirmed by polymerase chain reaction (PCR). Antimicrobial susceptibility profiles of the confirmed Salmonella isolates were examined using 14 antibiotics. The overall prevalence of Salmonella species in crocodile meat samples was 0.5%. The prevalence of Salmonella species ranged from 0.04% to 0.44% in the crocodile meat samples and annual prevalence ranged from 0.01% to 1%. The highest prevalence of Salmonella (4.4%) was recorded in the year 2012. Salmonella isolates from one of the three tested farms were resistant to Erythromycin (73.33%), Ampicillin (80%), and Penicillin G (100%). Generally, Salmonella isolates displayed lower resistance to Cefepime, Ceftriaxone, Amikacin, Tetracycline, Ertapenem, Florfenicol, and Erythromycin (0-53.33%) whereas all Salmonella isolates showed susceptibility to Cefepime, Ceftriaxone, Ertapenem, and Florfenicol. Although the study indicates low prevalence of Salmonella species in crocodile meat, there is a need for strict implementation of Hazard Analysis Critical Control Point (HACCP) to reduce contamination rates in meat and its products

Salmonella infections remain a global challenge. The culture method is the gold standard for the detection of genus Salmonella. Application of Polymerase Chain Reaction (PCR) has become an effective tool for the detection of virulence and antimicrobial resistance genes. This study investigated the prevalence of Salmonella by culture and detection of invA gene and blaCTX-M and blaCTX-M-3 gene markers by PCR. A total of 612 blood samples were collected from hospitalized febrile patients between March 2020 and April 2021. The samples were cultured, isolates identified by standard method with Analytical Profile Index (API 20-E) kits and were subjected to in-vitro antimicrobial susceptibility test (AST) using disk diffusion method. Extended-spectrum beta-lactamase (ESBL) detection was carried out by double-disc synergy test. Detection of invA gene and antibiotic-resistant genes makers was done by qPCR. A total of 24 Salmonella isolates were identified given a prevalence of 3.9% Salmonella-associated bacteraemia. Children within 1-10 years with persistent pyrexia of unknown origin (PUO) accounted for 50% of the Salmonella isolated with a mean age of 5.299 years. Specifically, 75% (18/24) Salmonella isolates and their corresponding samples of positive Salmonella culture were positive for the invA gene. The AST results indicated 100% Salmonella isolates developed resistance to ceftazidime, cefotaxime , augmentin, ampicillin, ertapenem, and doripenem. None of drug resistant-Salmonella isolates expressed ESBL enzyme phenotypically. Seven resistance patterns were observed, and the pattern CAZ-CTX-OFL-AUG-NIT-AMP-ETR-DOR was the most encountered pattern. Twelve (50%) Salmonella isolates harbored the blaCTX-M and blaCTX-M-3 genes and were mostly from children. The study has added to the growing knowledge on the suitability of the invA gene primer set as a PCR target for the detection of Salmonella. It also revealed a paradigm shift in the occurrence of invasive Salmonella harboring blaCTX-M and blaCTX-M-3 genes in PUO cases. There is a need for judicious use of cephalosporin and carbapenem antibiotics to preserve their efficacies.

Houseflies (Musca domestica) are well-known mechanical vectors for spreading multidrug-resistant bacteria. Fish sold in open markets are exposed to houseflies. The present study investigated the prevalence and antibiotypes of multidrug-resistant (MDR) Salmonella spp. in houseflies captured from a fish market. Direct interviews with fish vendors and consumers were also performed to draw their perceptions about the role of flies in spreading antibiotic-resistant bacteria. A total of 60 houseflies were captured from a local fish market in Bangladesh. The presence of Salmonella spp. was confirmed using PCR method. Antibiogram was determined by the disk diffusion method, followed by the detection of tetA, tetB, and qnrA resistance genes by PCR. From the interview, it was found that most of the consumers and vendors were not aware of antibiotic resistance, but reported that flies can carry pathogens. Salmonella spp. were identified from the surface of 34 (56.7%) houseflies, of which 31 (91.2%) were found to be MDR. This study revealed 25 antibiotypes among the isolated Salmonella spp. All tested isolates were found to be resistant to tetracycline. tetA and tetB were detected in 100% and 47.1% of the isolates, respectively. Among the 10 isolates phenotypically found resistant to ciprofloxacin, six (60%) were found to be positive for qnrA gene. As far as we know, this is the first study from Bangladesh to report and describe the molecular detection of multidrug-resistant Salmonella spp. in houseflies in a fish market facility. The occurrence of a high level of MDR Salmonella in houseflies in the fish market is of great public health concerns.

Genus Mentha presents group of plants which are the most studied in family Lamiaceae. Aboveground parts are used for different purposes in pharmacy, food industry or confectionery. Most important is natural product extracted from leaves - essential oil (EO). The aim of presented experiment was to demonstrate different chemotype and compare antibacterial activity of two Mentha species EO. Plant samples were obtained from various environments – from Slovakia and from Italy. Dominant compounds were determined by GC/MS. The results showed high amount of menthol and menthone in tested Slovak peppermint EO. On the other hand, carvone and 1,8-cineole were determinate as dominant compounds in Italian spearmint EO. The antimicrobial activity of the EO was investigated by disc diffusion and broth micro dilution methods. EO was evaluated for their antibacterial activity against 7 microorganisms: Enterobacter cloacae, Salmonella spp., Klebsiella pneumoniae, Escherichia coli, Staphylococcus aureus, Streptococcus pyogenes. The determination results of antibacterial activity by agar disk diffusion method ranged from 7 to 14 mm of the growth inhibition zone. MIC of tested mint EO varied from 0.625 to 2.5 μg/mL. In addition, both EO showed relatively the same antibacterial activity against the selected Gram-negative bacteria. However, there is a variation in the antibacterial activity against Gram-positive bacteria.
