Free radical that are released in cigarette smoke, believed to play an important role in male infertility. The free radicalproduces Reactive Oxygen Species (ROS) that is found increased in male smokers. ROS is one of the free radicalscomponent can be neutralized by vitamin C and E as an antioxidant. This study using those agents to the fifteen minuteshalf piece cigarette to expose mice and count their sperm as the fertilility parameter. The research design used was posttest-only control group with twenty mice were divided into five groups. A group of no smoke exposed was categorizedas a baseline control, the exposed groups were consisted of one group without antioxidant, one group with vitamin Ctreatment, a group of vitamin E treatment and at last the combination of vitamin C and E. All groups were treatedfor fourteen days. The results were analyzed using oneway ANOVA with p<0.05. It is obviously proven that thecombination of vitamin C and vitamin E treatment on the usual dose gave the highest sperm count compared to othergroups.

Abstract: Light is an initiator of free radical oxygen formation inter alia reactive oxygen species (ROS) that increases the oxidative stress. Free radicals can be controlled and prevents by the antioxidant. Vitamin C is an antioxidant that can neutralize the free radical. This study aimed to obtain the effects of vitamin C on spermatozoa quality of wistar rats after light exposure. Subjects were male rats (Rattus norvegicus) which were exposed to light and then divided into three groups: control group (without vitamin C administration), treatment group 1 (treated with vitamin C 1.8 mg/day/rat), and treatment group 2 (treated with of vitamin C 3.6 mg/day/rat) for 50 days. The results showed a significant improvement in the quality of wistar spermatozoa (concentration, motility, and morphology) after treated with vitamin C and exposed to light.Keywords: vitamin C, light, quality of spermatozoa, ROS (reactive oxygen species).  Abstrak: Cahaya merupakan inisiator pembentukan radikal bebas reactive oxygen species (ROS) yang akan meningkatkan stres oksidatif. Radikal bebas dapat dikendalikan dan dicegah oleh antioksidan, salah satunya ialah vitamin C. Penelitian ini bertujuan untuk mengetahui efek pemberian vitamin C terhadap kualitas spermatozoa tikus wistar setelah pemaparan cahaya. Subyek penelitian ialah tikus wistar jantan (Rattus norvegicus) yang diberi paparan cahaya kemudian dibagi menjadi 3 kelompok, yaitu: kelompok kontrol (tanpa pemberian vitamin C), kelompok perlakuan 1 (pemberian vitamin C 1,8 mg/hari/ekor), dan kelompok perlakuan 2 (pemberian vitamin C 3,6 mg/hari/ekor). Hasil penelitian memperlihatkan peningkatan kualitas spermatozoa tikus wistar (Rattus norvegicus) secara bermakna setelah pemberian vitamin C pada tikus wistar (Rattus norvegicus) yang telah diberi pemaparan cahaya.Kata kunci: vitamin C, cahaya, kualitas spermatozoa, ROS

BACKGROUND Infertility is defined as the inability to conceive after at 1 year of regular unprotected intercourse. Male contributes to almost half of infertility cases and in almost 30 % of cases, no definite aetiology is identified, and hence, male infertility is labelled idiopathic in these cases. Oxidative energy production mechanisms are almost always accompanied by reactive oxygen species (ROS), generation whose too much concentrations can lead to extensive protein damage and cytoskeletal modifications and inhibit cellular mechanisms. A number of laboratory techniques have been developed to evaluate oxidative stress by measuring ROS level in the semen. In recent times antioxidant supplements have been proposed as useful agents to increase the scavenging capacity of seminal plasma, controversy still surrounds their actual clinical utility. METHODS 34 male patients were included in this study. Reactive oxygen species detection was done by Flowcytometry using dichloroflurosecindiacetate (DCFH-DA). RESULTS The ROS in the patient group was found to be significantly higher 29.821 (5.6300 than the control group 22.162 (1.6331 having p value < 0.001). The ROS (29.821 ± 5.6300) was found to be significantly reduced after 3 months of antioxidant therapy which got reduced to 19.893 ± 4.2299 respectively. CONCLUSIONS Our study demonstrates that infertile men have significantly higher level of ROS (as measured by flowcytometry) & lower sperm count (oligospermia), decreased progressive & total motility and increased immotile sperms as compared to healthy fertile men. This study further proves that antioxidant therapy based on a combination of carnitine, zinc, coq10, lycopene and vitamin C & E for 3 months is associated with a decrease of ROS as measured by flowcytometry & a variable degree of improvement in above mentioned semen parameters. Background: The addition of antioxidant substances to a cryoprotective solution can increase its protective capacity, shielding spermatozoa from the oxidative stress caused by the seminal cryopreservation process. However, there is no record of a seminal cryopreservation protocol of tambaqui (Colossoma macropomum) using antioxidants as a supplement to the cryoprotective solution. Thus, the objective of this study was to evaluate the effects of adding vitamin C, vitamin E, cysteine, and/or taurine to the seminal cryopreservation of tambaqui.Materials, Methods & Results: Pools of semen (n = 10) were diluted in cryoprotective solutions supplemented with: vitamin C (T1), vitamin E (T2), vitamin C + vitamin E (T3), cysteine (T4), taurine (T5), and taurine + cysteine (T6). The control treatment (T7) was not supplemented. Diluted semen was loaded in 0.5 mL straws, frozen in a dry-shipper, stored in a cryogenic cylinder, and then thawed in a water bath (45ºC for eight seconds). The quality of fresh and cryopreserved semen was evaluated by measuring total motility, progressive motility, curvilinear velocity (VCL), straight line velocity (VSL), average path velocity (VAP), linearity, and straightness using a computerized system of sperm analysis. Sperm membrane integrity parameters were analyzed using eosin-nigrosin staining, sperm morphology was assessed using pink bengal staining, and motility duration was measured by a digital timer. Data were analyzed using the statistical program SAS (2002) and the results were expressed as means ± standard error of the mean. The results showed that, in general, there was no significant increase in seminal quality when antioxidants were added to the cryoprotective solution. The T5 treatment promoted an increase (P < 0.05) in progressive motility when compared to T1 (6.33 ± 1.14% and 2.98 ± 0.88%, respectively). However, it did not differ significantly (P > 0.05) from the other treatments. Treatments T2 and T5 presented the highest values of VCL (34.74 ± 2.58 and 33.60 ± 1.81 μm.s-1, respectively). These were higher (P < 0.05) than T1 (26.31 ± 1.64 μm.s-1) but not different (P > 0.05) from the T7 control (30.87 ± 1.49 μm.s-1). The VSL and VAP results showed that T1 presented the lowest velocity (9.89 ± 1.75 and 15.06 ± 1.92 μm.s-1, respectively) compared to the other treatments (P < 0.05) that did not differ from each other. Combining the two vitamins (T3) or the two amino acids (T6) was not advantageous in relation to the use of only one of these antioxidants.Discussion: The present study reports, for the first time, results of the addition of antioxidants to the tambaqui seminal freezing medium. The addition of taurine and vitamin E, although not significantly different from the control treatment, resulted in a tendency to increase sperm kinetics. This effect may be due to the action of taurine as a regulator of Ca2+ transporters, which is necessary to trigger sperm activation, and to the ability of vitamin E to scavenge reactive oxygen species produced during lipid peroxidation. On the other hand, the reduced sperm quality observed when vitamin C was used may have been related to the toxicity caused by a high concentration of this vitamin. In addition, once the safe dose of antioxidants has been exceeded, the normal physiological functions of reactive oxygen species can be inhibited. Thus, it is concluded that the use of vitamin E and taurine promotes promising results of curvilinear velocity after thawing of sperm. Therefore, these treatments are recommended, as well as more tests to determine their optimal concentrations.

Reproductive tract inflammation is considered an important cause of male infertility. Increased leukocytes in semen can produce many reactive oxygen species (ROS), which affect sperm function. The aim of this study is to identify the main source of ROS in seminal plasma and to assess the effect of ROS on leukocytes. Semen samples ( n = 20) with leukocyte concentration >1 × 106 were collected from a male infertility clinic. This study mainly compares the sperm function parameters of the normal group and the semen white blood cell group >1 × 106. The results identified that ROS in semen was closely related to sperm function parameters, and CD45+ leucocytes were the main source of ROS. Compared with the control group, the concentration of IL-2, IL-4, IL-6, IFN-γ, and TNF-α was higher in the experimental group. Leukocytes in semen may regulate the secretion of ROS through the mammalian target of rapamycin (mTOR) pathway. A considerable amount of ROS can upregulate the expression of IL-6 in leukocytes via the nuclear factor kappa-B (NF-kB) pathway.

Asap rokok diketahui meningkatkan Reactive Oxygen Species (ROS). ROS dapat merusak pembuluh darah dan mengubah reaktivitas trombosit sehingga mengganggu sistem hemostasis. Vitamin C sebagai antioksidan esensial bagi tubuh terbukti dapat mengurangi kerusakan oksidatif akibat ROS. Tujuan penelitian ini adalah membuktikan potensi vitamin C dalam menghambat gangguan hemostasis melalui pengamatan waktu perdarahan mencit yang dipapar asap rokok. Penelitian Randomized Pre Test-Post Test Control Group Design ini dilakukan pada 21 ekor mencit jantan yang dibagi menjadi tiga kelompok. Pemberian perlakuan pada hari ke-1 hingga ke-14, yaitu P1 (kontrol negatif), P2 (paparan asap rokok 10 menit perhari), dan P3 (vitamin C 0,4 mg/gBB/hari + paparan asap rokok 10 menit perhari). Pengukuran waktu perdarahan metode tail bleeding pada hari ke-0 dan ke-15. Analisis data menggunakan uji t-paired, one-way Anova, dan post-hoc LSD. Hasil penelitian menunjukkan rerata waktu perdarahan P1 mengalami perubahan yang tidak bermakna dari 60,16 ± 3,27 menjadi 57,61 ± 4,88 detik. P2 mengalami perubahan yang bermakna dari 59,34 ± 6,93 menjadi 38,85 ± 3,43 detik. P3 mengalami perubahan yang bermaknadari 59,36 ± 3,07 menjadi 51.85 ± 3,45 detik. Terdapat perbedaan rerata waktu perdarahan setelah perlakuan yang bermakna signifikan (p = 0,000) antara P2 dengan P3. Penelitian ini berhasil membuktikan potensi vitamin C dalam mencegah pemendekan waktu perdarahan lebih lanjut sebagai indikator gangguan hemostasis akibat paparan asap rokok.

Introduction. In our days, smoking is one of the most important risk factors for inflammatory diseases. There is a lot of scientific data, devoted to research the effects of smoking on periodontal status. However, some aspects such as the status of free-radical oxidation, is left without proper attention.The aim of the study – to investigate experimentally the indices of free radical oxidation in periodontal tissues on the background of smoking.Methods of the research. Experimental studies were performed on 20 adult male albino rats weighing 160-180 g. Animals were divided into 2 groups: control (10 animals) and experimental (10 animals). Modeling of passive smoking was carried out by placing rats in a specially designed chamber in which tobacco smoke was distributed. The duration of the experiment was 21 days. The concentration of 8-isoprostane was determined by ELISA method using a set of "8-isoprostane ELISA» company «USBiological» (USA). Quantitative assessment of the content of metabolites of nitrogen (II) oxide was carried out by determining their amount. The content of reduced glutathione, superoxide dismutase and catalase activity in periodontal tissue homogenate were determined by conventional methods.Results and Discussion. Concentration of 8-isoprostane in supernatant of periodontal homogenate in rats of control group was (3.94±0.18) pg/ml. In experimental animals this index was significantly increased in 2.9 times. This indicates that despite the action of tobacco smoke in periodontal tissues oxidative stress develops, characterized by the increasing intensity of the production of reactive oxygen species. On 22 day of the experiment, we have observed a significant decrease in superoxide dismutase activity in periodontal tissue homogenate by 40.9 % and catalase activity in 2.4 times compared to the control group. It is known that SH-groups (such as cysteine and methionine fragments of protein molecules) are most easily oxidized by reactive oxygen species to form a reversible and irreversible modifications. On 22 day of the experiment, we have observed a significant decrease in glutathione content in periodontal tissue homogenate by 24.4 % compared to the control group. The content of NO metabolites in supernatant of periodontal homogenate in rats of experimental group was significantly decreased in 2.1 times. The powerful factors that inactivate NO, are free radicals, among them – superoxide radical. Its interaction with NO leads to formation of peroxynitrite that as a strong oxidant has high degree of cytotoxicity.Conclusions. In rats on the background of tobacco smoke in periodontal tissues significantly increases the generation of reactive oxygen species, reduces superoxide dismutase, and catalase activity and glutathione content, indicating a marked imbalance of free radical homeostasis. On the background of tobacco smoke in periodontal tissues occurs inhibition of nitroxydergic processes through significant reduction of nitric oxide metabolites content, which reduces the protective antibacterial potential.

 Abstrak  Timbal merupakan salah satu bahan pencemar udara  yang berpengaruh pada kesehatan organ reproduksi pria. Polusi udara oleh logam berat ini menyebabkan terbentuknya Reactive Oxygen Species dan penurunan simpanan antioksidan tubuh. Penelitian ini bertujuan mengetahui potensi ekstrak etanol bawang dayak untuk mempertahankan diameter tubulus seminiferus pada mencit yang diinduksi timbal asetat. Jenis penelitian ini adalah eksperimental murni laboratorik (true experimental) dengan rancangan penelitian Posttest Only Control Group Design. Subyek penelitian adalah 30 mencit (Mus musculus) jantan strain Balb/C yang diinduksi timbal asetat 0,75mg/KgBB/hari dan dibagi menjadi 5 kelompok perlakuan, yaitu kelompok kontrol negatif (K0), kelompok kontrol positif (K1), dan kelompok perlakuan dengan pemberian ekstrak bawang 30mg/KgBB/hari (K2), 60mg/KgBB/hari (K3), 120mg/KgBB/hari (K4). Pada hari ke 39, hewan coba dikorbankan untuk diukur diameter tubulus seminiferusnya. Hasil penelitian menunjukkan tidak ada perbedaan bermakna diameter tubulus seminiferus kelompok kontrol dan perlakuan. Kesimpulan penelitian ini ekstrak etanol bawang dayak tidak mempengaruhi diameter tubulus seminiferus mencit yang di induksi timbal asetat Kata kunci : ekstrak etanol bawang dayak, diameter tubulus seminiferus , timbal asetat

<p>Radiasi sinar X merupakan salah satu radiasi pengion yang menyebabkan terbentuknya radikal bebas sehingga merusak sel spermatogonia. Penggunaan vitamin C dan E sebagai antioksidan untuk melindungi sel tubuh. Penelitian ini bertujuan mengetahui pengaruh pemberian vitamin C dan E terhadap jumlah sel spermatogonia yang dipapar radiasi sinar X. Metode penelitian eksperimental dengan rancangan <em>post test only control group design</em> pada 35 ekor mencit (<em>Mus musculus</em>) dibagi menjadi 5 kelompok secara acak yaitu KP tanpa paparan radiasi dan pemberian vitamin, KN dengan paparan radiasi sinar X 25 mGy/hari tanpa pemberian vitamin, KP 1 diberikan vitamin C 0,26 mg/hari, KP 2 diberikan vitamin E 0,208 mg/hari, KP 3 diberikan kombinasi vitamin C 0,26 mg/hari dan vitamin E 0,208 mg/hari setiap sebelum pemaparan radiasi sinar X 25 mGy/hari selama 4 hari dengan waktu penelitian 21 hari. Pada hari ke 22 sampel diterminasi diamati melalui mikroskop. Jumlah sel spermatogonia dianalisis <em>One Way Anova </em>kemudian uji <em>post Hoc LSD. </em>Hasil rerata jumlah spermatogonia pada KP, KN, KP1, KP2 dan KP3 yaitu 24,24; 13,28; 19,36; 19,60 dan 23,32. Hasil uji <em>One Way Anova </em>diperoleh nilai p sebesar 0,000 (p&lt;0,05) artinya terdapat perbedaan rerata jumlah sel spermatogonia yang bermakna antara kelima kelompok. Hasil uji <em>post Hoc LSD</em> menunjukkan perbedaan rerata sel spermatogonia antar dua kelompok ditunjukkan oleh hampir semua pasangan kelompok (p&lt;0,05); kecuali untuk perbandingan rerata sel spermatogonia antara KP1 dan KP2 (p&gt;0,05). Terdapat pengaruh pemberian vitamin C dan E terhadap jumlah sel spermatogonia mencit yang dipapar radiasi sinar X.</p>
