This study was carried out to screen goat farms for anthelmintic resistance (AR) against oxfendazole (OXF) and to determine contributory factors for its development. For this purpose, Beetal goat farms (n = 18) were randomly selected, with natural mixed gastrointestinal nematodosis infection. In vivo (faecal egg count reduction test) and in vitro (egg hatch assay) tests were used to ascertain the presence of AR while a scorecard was used to determine the role of possible contributory factors for oxfendazole resistance. For in vivo test, the experimental animals were divided into two groups of 10 animals each; one group received OXF treatment, while the other served as control. Pre- and post-treatment coproculture was performed to identify the species and genera of nematodes. Egg hatch assay (EHA) was used to confirm the results of FECRT. Fecal egg count reduction test (FECRT) revealed the development of resistance on six farms and post-treatment larval cultures indicated Haemonchus contortus, Trichostrongylus colubriformis, Cooperia curticei, Teladorsagia circumcincta and Oesophagostomum spp. as dominant species with resistance. Furthermore, EHA confirmed the results of FECRT. Among the presumptive factors for AR, the highest composite score was for rotation of anthelmintics followed by treatment frequency, dose rate and nature of medication. The scorecard for the development of AR, used in this study, may be helpful for the assessment of contributory factors of AR.

The efficacy of Albendazole against trichostrongyle nematode parasites in goats presented for slaughter at the Nsukka municipal  abattoir was evaluated using the In vitro Egg hatch assay (EHA) model. The abattoir was visited once every week for 4 consecutive months during which a total of 240 goats were sampled. Fecal samples were collected per rectum from a minimum of 15 goats on each day of the visit. Egg Hatch Assay was performed on strongyle eggs recovered from pooled faecal sample on each day of sampling with a 2.5% W/V Albendazole. Faecal culture was also set up from the pooled faecal sample on each sampling day to recover and identify the nematode parasites present in the goats. Among the 240 goats sampled, the prevalence of trichostrongylosis as observed by the presence of strongyle eggs was 94.6% (227/240). Faecal culture and larval identification revealed 69.8% of the strongyles as  Haemonchus contortus, while 25.5 and 4.8% were Trichostrongylus colubriformis and Oesophagostomum species respectively. In the EHA, Albendazole had mean LC50 value of 0.16 µg/ml which is slightly in excess of the discriminating dose of 0.1µg/ml as prescribed by the World Association for the Advancement of Veterinary Parasitology (WAAVP) as an indication of anthelmintic resistance. There is therefore an urgent need to screen the nematode parasite population in the Nigeria for the presence Albendazole resistance genes. SummaryThis study determined the prevalence of anthelmintic resistance (AR) in parasitic nematodes on smallholder sheep farms in Lithuania from April to November 2014. Faecal samples were collected from two groups of 10-15 sheep treated with fenbendazole (FBZ) or ivermectin (IVM) on 18 sheep farms. Two samples were collected from each group: on day zero (T1) and 10-14 days after treatment. Faecal egg counts (eggs per gramme, EPG) were determined using a modified McMaster technique. Animals with < 140 EPG on day zero were removed from the analysis. The prevalence of AR was estimated using the in vivo faecal egg count reduction test. AR to FBZ was detected on three of 15 farms where FBZ was used (20 %) and was suspected on one farm (6.7 %). AR to IVM was detected on two of 16 farms where IVM was used (12.5 %). The main species of resistant gastrointestinal nematodes (GINs) identified after treatment were Teladorsagia spp. and Trichostrongylus spp. A questionnaire surveying 71 sheep farmers estimated that 71.8 % of sheep farmers used anthelmintics against GINs. IVM was the most frequently (68.6 %) applied anthelmintic, and 62.7 % of the respondents reported treating their animals twice a year. This study confirmed the presence of AR to GIN infections on sheep farms in Lithuania. Future studies should assess the prevalence of AR to GIN infection using in vitro methods.

SUMMARYA Strain ofHaemonchus contortusshowing moderate levels of resistance to thiabendazole (TBZ) (egg hatch assay LC50 2.1 μM) was subjected, in alternating sequence, to 5 in vitro and 4 in vivo selections with TBZ. This resulted in a significant increase in resistance (egg hatch assay LC50 2.8 μM). The isolates obtained after each combined in vitro and in vivo selection were compared in terms of egg hatch assay, establishment, net egg output, per capita egg output and degree of pathology caused in infected hosts. The increase in resistance was accompanied by a significant increase in larval establishment sheep (22 %) and was associated with more severe pathology, higher egg count and worm recovery 3 months post- infection.

Our research aimed to evaluate the efficacy of anthelmintic treatments in sheep, as well as the resistance occurrence risk for the most commonly used substances. Anthelmintic medication efficacy was evaluated on 30 animals from a private farm, located in Sânmihaiu Almaşului, Sălaj County. In vivo studies were performed by using Fecal Egg Count Reduction Test (FECRT) and testing an albendazole-based (ABZ) product. In vitro, we used Egg Hatch Assay (EHA) and Larval Development Assay (LDA) for albendazole (ABZ), mebendazole (MBZ), fenbendazole (FBZ), thiabendazole (TBZ) and ivermectin (IVM) (only for LDA). FECRT showed that intestinal nematodes developed resistance phenomena against the ABZ-based product, with an extensivity of 80% at seven days post therapy, an egg reduction percentage of 41.89% at seven days post-therapy and 43.9% at 14 days post-therapy. The in vitro EHA highlighted a superior efficacy of TBZ (egg hatch percentage at reference concentration being 51.21) compared to ABZ (71.89%), MBZ (84.46%) and FBZ (79.22%), with a minimum risk of anthelmintic resistance. The LDA test revealed the superior efficacy of FBZ (MIC 0.59 mcg/ml) and IVM (MIC 0.078 mcg/ml), with a minimal risk of inducing parasitic resistance. All in vivo and in vitro tests revealed a limited ABZ efficacy, recommending avoiding the therapy with this substance.

The study was aimed at to investigate the anthelmintic efficacy of aqueous and ethanolic extracts of Cucurbita pepo against gastrointestinal nematodes of sheep by scientifically validated in vitro and in vivo tests. In vitro anthelmintic activity was determined by egg hatch assay, larval migration inhibition assay and larval development assay against eggs and larvae of mixed gastrointestinal nematodes of sheep. The extracts which were found to be effective in in vitro assays were further evaluated by in vivo faecal egg count reduction test against mixed species of sheep gastrointestinal nematodes with 3 different doses (125, 250 and 500 mg kg-1). Haematological and serum parameters were estimated before and after treatment to determine the impact of plant extracts on health of the animal. Aqueous extract of C. pepo demonstrated significant inhibition of egg hatch at 40 and 80 mg/ml. The ED50 value of egg hatch inhibition was 3.51mg/ml. In larval migration inhibition assay, both aqueous and ethanolic extracts of C. pepo demonstrated inhibition of larval migration and the LM50 was 1.75 and 0.32 mg/ml respectively. However, in larval development assay, both aqueous and ethanolic extracts showed only marginal inhibition of larval development at all concentrations tested. In faecal egg count reduction test (FECRT), aqueous extracts of C. pepo at 500 mg/kg caused significant reduction in eggs per gram (96.61 per cent) the reduction being higher than the result obtained with albendazole, the positive control (93.25 per cent). There was a significant difference in EPG reduction between sheep treated with albendazole and C. pepo. Haematological and serum parameters were not affected in both the treated and untreated sheep indicating that the extracts did not cause toxic effects. Based on the results obtained in this study, the aqueous extract of C. pepo possesses anthelmintic activity and could offer an alternative source for the control of gastrointestinal nematodes of sheep.

Abstract Thymol is a monoterpene and acetylation form of this compound can reduce the toxicity and enhance its biological effects. The objective of this study was to evaluate the effect of thymol and thymol acetate (TA) on egg, larva and adult Haemonchus contortus and the cuticular changes, acute toxicity in mice and the efficacy on sheep gastrointestinal nematodes. In vitro tests results were analyzed by analysis of variance (ANOVA) and followed by comparison with Tukey test or Bonferroni. The efficacy of in vivo test was calculated by the BootStreet program. In the egg hatch test (EHT), thymol (0.5 mg/mL) and TA (4 mg/mL) inhibited larval hatching by 98% and 67.1%, respectively. Thymol and TA (8 mg/mL) inhibited 100% of larval development. Thymol and TA (800 µg/mL) reduced the motility of adult worms, by 100% and 83.4%, respectively. Thymol caused cuticular changes in adult worm teguments. In the acute toxicity test, the LD50 of thymol and TA were 1,350.9 mg/kg and 4,144.4 mg/kg, respectively. Thymol and TA reduced sheep egg count per gram of faeces (epg) by 59.8% and 76.2%, respectively. In in vitro tests thymol presented better anthelmintic activity than TA. However TA was less toxic and in in vivo test efficacy was similar.

Abstract The anthelmintic resistance stimulated the search for strategies for controlling gastrointestinal nematodes, including the use of free essential oils or its nanoemulsion. This study evaluated the anthelmintic efficacy of Cymbopogon citratus essential oil (CcEO) and C. citratus essential oil nanoemulsion (CcEOn). Pysicochemical analyses were performed. The in vitro effect was determined using the egg hatch test (EHT) on Haemonchus contortus and in vivo effect was evaluated in sheep infected with gastrointestinal nematodes. The animals were treated with CcEO (500 mg/kg) or CcEOn (450 mg/kg) for the fecal egg count (FEC) and the determination of worm burden. The main component of CcEO was citral. The CcEO content in the nanoemulsion was 20% (v/v), and the mean particle size was 248 nm. In EHT, CcEO and CcEOn (1.25 mg/mL) inhibited larval hatching by 98.4 and 97.1%, respectively. Three animals treated with CcEO died whereas in the group treated with CcEOn one animal died. The FEC and total worm burden of the treated groups did not differ from the negative control (p>0.05). The CcEOn showed efficacy only on H. contortus (p<0.05). In conclusion, nanoencapsulation reduced toxicity and increased efficacy on H. contortus.

The anthelmintic resistance problem is widely recognized in sheep production. Therefore, new methods of control against gastrointestinal nematodes (GIN) need to be integrated. The aim of this work was to assess the toxicity of A. cina 30 CH as a homeopathic product against Haemonchus contortus in in vitro and in vivo assays. A. cina 30 CH was obtained from a commercial laboratory, and confirmation of artemisinin as a key ingredient was performed with mass spectrophotometry. The A. cina 30 CH and the artemisinin pure reagent were used for the inhibition of egg hatching (IEH) and for the inhibition of larval migration of H. contortus L3 (ILM). In addition, three groups of 10 naturally infected lambs with GIN were treated with A. cina 30 CH and albendazole, and 10 were used as control. The parasitic infection was monitored at 0, 7, 14 and 28 d postreatment (PT) to determine the number of eggs per gram  (epg) and  FAMACHA index.  The in vitro data  showed 100 %  IEH and 64.7 % ILM by A. cina 30 CH, and nonlethal activity was observed with the artemisinin pure reagent. The toxicity of A. cina 30 CH against H. contortus in infected lambs was observed after 7 d of infection. Administration of the A. cina 30 CH yielded a 69 % reduction in the epg at 28 d PT, similar to the albendazole (P<0.05). In conclusion, A. cina 30 CH had the ability to IEH and ILM of H. contortus in in vitro assays and reduced the number of eggs of H. contortus, which is the primary parasitic nematode in grazing lambs, thereby reducing infection.

Abstract Introduction The control of gastrointestinal nematodes (GINs) in small ruminants is principally dependent on anthelmintic therapy, which encounters the rising problem of anthelmintic resistance (AR) development. Veterinarians reported anthelmintic failure in several sheep farms in Arbat District, Sulaymaniyah, northern Iraq, which called for a systematic study about the efficacy of three commonly used drugs: albendazole, ivermectin, and levamisole. Material and Methods A faecal egg count reduction test (FECRT) was conducted to elucidate the anthelmintics’ efficacies, with coprocultures to determine the GINs parasitising sheep in the study area. Larval development assays (LDAs) were conducted to determine the drugs’ median inhibitory concentrations (IC50). Results The FECRT revealed that AR was widespread to all three drugs, and ivermectin was the least effective, reducing the faecal egg counts (FECs) by 50.5% to 57.1%. The coprocultures revealed that the GIN genera of sheep in Arbat District were Trichostrongylus, Nematodirus, and Trichuris, and the resistance was mainly due to Trichostrongylus species. The mean IC50 of albendazole, ivermectin, and levamisole were 0.073 ng/mL, 7.97 ng/mL, and 1.43 ng/mL, respectively. Conclusion This study is the first Iraqi report of AR confirmed by both in vivo FECRT and in vitro LDA methods.
