This study explores the effect of baclofen on the malignant phenotype of glioblastoma (GBM) and the growth of xenograft tumors and investigates the related mechanisms, aiming to reveal the effect of baclofen on the occurrence and development of GBM. The development of new therapeutic drugs for GBM lays a theoretical and experimental foundation. Research results show that baclofen could inhibit GBM cell proliferation and migration and promote GBM cell apoptosis; baclofen dose- and time-dependently could induce GBM cell YAP phosphorylation. YAP participated in the effect of baclofen on GBM cell proliferation and migration inhibition. Baclofen induced YAP phosphorylation in GBM cells through the GABABR2-Gs-Lats1/2 signaling pathway. Baclofen could inhibit the expression of survivin and Bcl2. Baclofen inhibits subcutaneous tumors by inducing YAP phosphorylation in vivo.

Cigarette smoking is associated with impairment of repair mechanisms necessary for vascular endothelium homeostasis. Reducing the exposure to smoke toxicants may result in the mitigation of the harmful effect on the endothelium and cardiovascular disease development. Previous investigations performed by the tobacco industries evaluated in vitro the effect of electronic cigarette (e-cig) compared to cigarette smoke demonstrating a significant reduction in endothelial cell migration inhibition following e-cig aerosol exposure. In the present study, we replicated one of these studies, evaluating the effects of cigarette smoke on endothelial cell migration compared to e-cig and heated tobacco products. We used a multi-center approach (ring-study) to verify the robustness and reliability of the results obtained in the replicated study. Consistently with the original study, we observed a substantial reduction of the effects of e-cig and tobacco heated products on endothelial cell migration compared to cigarette smoke. In conclusion, our study further confirms the importance of e-cig and tobacco heated products as a possible harm reduction strategy for cardiovascular diseases development in smokers.

In this study we investigate the migration inhibition of Gypenosides (Gyp) and its combined effects with 5-fluorouracil (5-FU) on human colon cancer SW-620 cells, hoping to explore more potential clinical use of Gyp. Our data implied Gyp could significantly inhibit the migration potential of SW-620 cells including down-regulating matrix Metalloproteinases expression and decreasing cells adhesion ability. What’s more, evidence showed cells treated with Gyp exerted serious microfilament network collapse as well as a significant decline in the number of microvilli. A significant migration inhibitory effect was seen in Gyp groups along with the decline of cell adhesion. Further, the combination studies suggested Gyp could synergistically enhance the antitumor effect of 5-FU in SW-620 cells through the apoptosis way. The present study indicated Gyp could prevent cell migration and further enhance the cell killing effect of 5-FU on human colon cancer SW-620 cells.

At present allergic diseases are detected in 30% people and their frequency is only growing. A significant role in allergic pathology occurrence belongs to ambient air contamination and chemicals being introduced not only into children’s bodies, but their parents’ ones as well since pollutants can act as allergens and sensitizing agents. Our research goal was to examine influence exerted by parents’ pre-gestation exposure to chemicals on sensitization among teenagers living in an area where ambient air was contaminated. We examined overall immunoglobulin E contents and leukocytes migration inhibition test with formaldehyde and sodium nitrite in 115 teenagers whose parents worked under adverse working conditions at chemical and petrochemical enterprises and in 244 schoolchildren whose parents didn’t have any occupational contacts with chemicals. Each group was divided into sub-groups depending on inhalation chemical burden on schoolchildren’s bodies caused by ambient air contamination and contaminated air indoors (with hazard index (HI) for immune disorders being lower than 2 and HI≥2). The research allowed establishing that teenagers whose parents had worked at chemical and petrochemical enterprises during a pre-gestation period had elevated IgE contents more frequently as well as changes in leukocytes migration inhibition test with formaldehyde; it indicated there was sensitization to this chemical. Parents’ occupational contacts with chemicals led to an increase in relative risks of elevated igE contents and 2.5 times higher sensitization among schoolchildren with HI<2. Risk that sensitization to formaldehyde might occur was equal to 2.3 among senior schoolchildren with HI≥2 whose parents worked at chemical enterprises.

Background: Colorectal cancer is the second leading cause of mortality and the most prevalent cancer worldwide. Most patients, who come with late-stage, have ineffective treatments and some side effects in chemotherapy. Aaptos suberitoides has potential anti-cancer effects due to its bioactive compounds such as aptamine. This study aimed to evaluate the migration inhibition effect of Aaptos suberitoides fraction in HCT-116 cell line.Methods: This study was an experimental study. Aaptos suberitoides specimen was taken in Tinjil Island and fractionated with ethyl acetate. HCT-116 cell line was added with Aaptos suberitoides fraction and cellular migration activity was observed in 48 hours of which the scratch assay was performed. The gap closure area was determined with ImageJ software.Results: The data showed that a low concentration of Aaptos suberitoides fraction inhibited migration activity in HCT-116 cell line as follow; 1 and 5 mg/L Aaptos suberitoides fraction inhibit 3-4 % cancer cell migration in 24 hours, and 10-11% inhibition in 48 hours, respectively. However, 10 mg/L fraction concentration only inhibited 7-14% of the migration effect.Conclusion: Aaptos suberitoides fraction suggests insignificant migration inhibition in colorectal cancer cells and only inhibits less than 15 % HCT-116 cell line.

Background: Long non-coding RNAs (lncRNA) have been identified as novel molecular regulators in cancers. LncRNA ADAMTS9-AS2 can mediate the occurrence and development of cancer through various ways such as regulating miRNAs, activating the classical signaling pathways in cancer, and so on, which have been studied by many scholars. In this review, we summarize the molecular mechanisms of ADAMTS9-AS2 in different human cancers. Methods: Through a systematic search of PubMed, lncRNA ADAMTS9-AS2 mediated molecular mechanisms in cancer are summarized inductively. Results: ADAMTS9-AS2 aberrantly expression in different cancers is closely related to cancer proliferation, invasion, migration, inhibition of apoptosis. The involvement of ADAMTS9-AS2 in DNA methylation, mediating PI3K / Akt / mTOR signaling pathways, regulating miRNAs and proteins, and such shows its significant potential as a therapeutic cancer target. Conclusion: LncRNA ADAMTS9-AS2 can become a promising biomolecular marker and a therapeutic target for human cancer.
