GROWTH ABILITY OF Jatropha curcas L. EXPLANTS ON THE IN VITRO MEDIA CONTAINING IBA AND BAResearch on the growth ability of Jatropha curcas L. shoots and callus in solid and liquid media have been conducted. Explants were planted in the initiation MS medium. After ten weeks, the explants were subcultured into solid and liquid media containing combination of IBA and BA treatments. The number of combinations was 12 treatments, each with 6 replications. Observation was conducted from the first week after subculturing upto the fourth week. Parameters of observation were the percentage of explant forming shoots, the number of shoots, height, number of leaves, weight, color, and form of callus. The results showed that the explant which was subcultured in liquid media had higher growth rate than those subcultured in solid media. Treatment of 1 ppm IBA + 0.5 ppm BA gave a good result on the growth of shoots on solid and liquid media. For callus formation, treatment of 2 ppm IBA + 1 ppm BA gave the best result.Keywords: Callus, Jatropha, IBA and BA, solid and liquid media, hormone ABSTRAKPenelitian terhadap kemampuan tumbuh kalus dan tunas tanaman jarak pagar (Jatropha curcas L.) telah dilakukan pada media padat dan cair. Eksplan diinisiasi pada media MS dan setelah 10 minggu dipindahkan ke media padat dan cair yang mengandung perlakuan kombinasi hormon IBA dan BA. Jumlah kombinasi sebanyak 12 perlakuan dan setiap perlakuan dibuat 6 ulangan. Pengamatan dilakukan dari minggu pertama subkultur hingga minggu keempat. Peubah yang diamati adalah persentase eksplan yang membentuk tunas, jumlah dan tinggi tunas, terbentuknya daun pada tunas, perbedaan berat, bentuk, dan warna kalus. Hasil yang diperoleh menunjukkan bahwa eksplan yang disubkultur pada media cair memiliki laju pertumbuhan yang lebih tinggi daripada media padat. Perlakuan IBA 1 ppm + BA 0,5 ppm menghasilkan pertumbuhan tunas yang paling tinggi pada media padat dan cair. Pembentukan kalus yang terbaik diperoleh pada perlakuan IBA 2 ppm + BA 1 ppm.Kata kunci: Kalus, Jatropha, IBA dan BA, media padat dan cair, hormon

The present investigation was conducted for mass multiplication of Jatropha curcas L. through axillary bud culture. For this nodal segment from 3-5 months old nursery grown plants were used as explants for axillary bud culture. The sterilization treatment involving dipping explants in 0.1 per cent HgCl2 solution for 5 minutes resulted in minimum contamination and maximum establishment of nodal explants. The treatment MS medium supplemented with 1.0 mg/L BAP and 1.0 mg/L IAA was the best for culture establishment, shoot proliferation and multiplication of the axillary buds which exhibited highest value in each parameter like establishment (76.1%), number of days taken for shoot initiation (3.1 days), length of longest shoot (6.8 cm), number of leaves on main shoot (7.1) and number of shoots per explant (6.3). Among different treatments for root initiation, half MS media fortified with 1 mg/L IBA, 3 mg/ L NAA and 0.25 g AC gave best result in maximum number of rooting percentage (60) with minimum time taken for root initiation (13.3 days), produced maximum number of roots per shoots (5.1) and length of longest root (4.9 cm) when established shoots were treated with it. Such produced plantlets showed nearly cent per cent survival after hardening and acclimatization. It showed that explants surface sterilized with 0.1 per cent HgCl2 solution for 5 minutes inoculated in MS medium supplemented with 1.0 mg/L BAP and 1.0 mg/L IAA and half MS media fortified with 1 mg/L IBA, 3 mg/L NAA and 0.25 g AC were best in shoot establishment and root development respectively for mass multiplication of J. curcas L. through axillary bud culture.

This experiment was performed at the Tissue Culture Laboratory of the Horticulture Dept. of the Faculty of Agriculture at Assiut Univ., Egypt. After several attempts to determine the proper stage of buds for collection of pollen, we determined that the tetrad stage was most suitable. The pollen was cultured on either MS or B5 liquid or solid media (7% agar). Both media were used as basic salts or supplemented with growth regulators. The four growth substances were BA, NAA, K, and 2,4-D. Each growth substance was added to the medium separately as follow: BA, NAA at 15, 10, or 5 ppm; K at 0.1, 1, 2, or 5 ppm; and 2,4-D at 0.5, 1, or 5 ppm. The solidified medium was superior to the liquid medium at all the treatments that were used for callus formation. Using B5 medium did not result in any callus. The highest value of callus formation was obtained when MS medium supplemented with BA at 5 ppm. Moreover, the callus that was grown on the MS medium that had BA at 5 or 10 ppm developed a merstim tip. The control treatment produced calluses but did not develop any meristem tips. This process can be used to develop haploid plants.

ABSTRACT The banana crop stands out as an activity of great social and economic importance in Brazil, which occupies the fifth place in world production. Synthetic seed production is becoming promising for a micropropagation and in vitro conservation. The aim of the study was to analyze the conversion and growth of ‘Prata-anã’ banana’s microshoots clone Gorutuba from synthetic seed in MS medium and vermiculite, different substrates and concentrations of BAP (6-benzylaminopurine) associated with ANA (acetic naphthalene acid) in the constitution of its capsule were tested. The microshoots were immersed in the sodium alginate matrix (3%) and dripped in a solution of CaCl2.2H2O (100 mM) for complexation and then in KNO3 solution (100 mM) to decomplex. The experimental design was completely randomized in a 2 x 5 factorial design (substrate x BAP concentrations), containing different substrates (MS culture medium and vermiculite) and BAP concentrations (2.22, 4.44, 6.66, 8.88 and 13.32 µmol L-1) associated with NAA (naphthalene acetic acid) 0.54 µmol L-1, totaling 10 treatments, with 4 replicates, and that each replicate containing 5 seeds. The evaluations of conversion, number of leaves, leaf length, leaf height, number of roots, root length and oxidation were performed at 30 and 60 days.The use of the MS medium provided better growth results in relation to vermiculite as substrate, in which the different BAP concentrations did not differ from each other. It was found that, in MS culture medium, BAP concentrations above 8.88 µmol L-1 in the capsule composition are not indicated for microshoots growth.

Abstract In vitro cultivation is a technique with wide application for micropropagation. However, each species has specific mineral needs for this type of cultivation. The objective was to assess the impacts of the saline solution culture medium on the performance of the photosynthetic apparatus and growth of Bromelia antiacantha during in vitro cultivation, and thus to elucidate the mitigation of the nutritional imbalance that can interfere in the electron transport in the plants. Plants were cultivated in a salt concentration gradient of MS medium (0%, 25%, 50%, 75% or 100%). The growth traits and fluorescence a chlorophyll were analyzed. Intermediate concentrations of MS medium resulted in plants with a larger number of leaves and longer root length. The OJIP curves and results of the JIP test showed that the plants grown without MS salts presented less efficient photosystem II (PSII), as indicated by the performance index [Pi(total)]. In contrast, the intermediate concentrations (MS 25% and 50%) had a positive effect on the performance of the photosynthetic apparatus. The MS 25% medium can be used for in vitro cultivation of B. antiacantha, enabling the development of plants with suitable physiological qualities for planting in the field.

Somatic embryogenesis from callus induced in epicotyl and hypocotyl segments can be viable native species in order to better -benefit ratio costs, and rates of clonal multiplication. In this sense, two trials were established to induce callus and adventitious buds on hypocotyl and epicotyl segments of cumaru bean seedlings germinated in vitro in different concentrations and combinations of growth regulators. At first, we used the MS medium supplementwith ANA (0.0, 1.5 mg.L-1) and TDZ (0.0, 4.0 and 8.0 mg.L-1) distributed in factorial 2 x 3 x 2 (x auxin cytokinin x explant) with eight replications. In the second, it was used the WPM medium supplemented with BAP (2.0 mg L-1) and plus 2,4-D (2.0 and 4.0 mg L-1) in a factorial 2 x 2 (auxin x explant) with 15 repetitions each. They were evaluating callus formation and the average number of adventitious shoots during the period of 90 days. The results indicated that the highest average for callus formation was observed when the explants were subjected to concentrations of 8.0 mg L-1 TDZ combined with 1.5 mg L-1 ANA in MS medium. For the formation of buds, the WPM medium plus 2.0 mg L-1 2,4-D in the second experiment, induced higher number of shoots, being significant the use of auxin, and its interaction with the type of explant.

A research  to study the sterilization   method  and application   of Kinetin  and IAA to induce the Durian  young  leaf (Durio zibethinus) in MS  medium   was conducted in Balai Benih Induk Hortikultura in Salaman  Magelang  district  of Central  Java  started  on September  until December 2003. The Laboratory experiment   was arranged  in two phases,  which were  the optimation  phase of sterilization   and  induction   phase.  At  the  first  phase,  the  sterilization method  used  was  the modification   of Mulya  (2001) method.  The modification   use of sterilant,  vitamin  C antioxidant, Alcohol  70 %, Benlate, Agrept,  Tween-20  and Betadine  were done to obtain  effectiveness   of the sterilization.  Explants  planted  then in MS medium  for two weeks. Contamination   time, percentage of contamination   and viabilitas  (percentage of living explants)  were observed  then.  At the second phase,  the treatments were arranged  in a 3 x 3 factorial  completely   randomized   design  (CRD)  to observed  the influence  of Kinetin  and IAA combination.   The concentration   of Kinetin  observed were 2, 4, and 6 mg/I, where  as the IAA concentration   were 0.5,  1.0, and  1.5 mg/I. All treatments were  repeated  three  times,  with three samples  on each  replication.   The percentage   of browning explants, percentage  of contaminated   explants,  site of  contamination   and percentage of explants live were observed  at the end of incubation. The results  showed that sterilization  of Durian young leaves explants  with 1  g/l deterjent  for 15 minutes  then by 2 g/l Benlate  and Agrept  for 10 minutes,  then by 1  g/200 mg Vitamin C, then by Alcohol  70 % for 1  minute, then by 20% Clorox,  then by 2 drip of Tween-20  for 10 minute and then by Betadine  decreased  the contamination down to 50 %, and this kind of sterilization  was relatively better than  the other  kinds.  Application   of growth  regulators   were  not  able  to induce  explants growth,  but stimulated  callus formation  at the cutting surface though,  in the application  of Kinetin 4 mg/1 + IAA 0,5 mg/I, Kinetin 4 mg/1 + IAA  1,5 mg/1, Kinetin  6 mg/I+  IAA 0,5  mg/1 and Kinetin 6 mg/l+IAA   1,0 mg/I.

One of the problem of Eurycoma longifolia Jack propagation was low germination percentage due to recalcitrant seeds and slow growth of seedling from cutting propagation. To overcome this problem is required propagation of Eurycoma longifolia via in vitro culture. The objective of this research was to know the effect of Auxin (2,4-D and NAA) and Cytokines (BAP and Kinetin)  on Eurycoma longifolia callus induction via leaf and petiole explants. In this study, we used plant growth regulator of 2,4 D, NAA, BAP and Kinetin in several levels.  The observed variables were appearing callus time, callus color and callus texture. The results of this study showed that MS medium supplemented with 1 ppm NAA+ 1 ppm BAP was able to induce callus formation in leaf explant for 6 months after culture. While MS medium supplemented with 1 ppm 2,4-D, 1 ppm BAP, combination of 2,4-D and Kinetin and combination of 2,4-D and BAP can induce callus formation from petiole. All the callus formation has yellow color and yellow brown color. The petiole explant that is grown in MS medium supplemented with 1 ppm BAP induced of callus in short time (18 days after culture).

Baru (Dipteryx alata Vog.) is a species with great economic and environmental potential; it has popular acceptance, besides being a very productive species. Alternative propagation methods are important for species maintenance and exploration. Thus, micropropagation emerged as an alternative technique, providing genetic stability and the production of a large number of seedlings. The aim of the present investigation was to develop a callus induction protocol for in vitro baru explants. The tested explants were nodal, internodal and foliar segments. The explants were disinfected for 30 seconds in 70% alcohol (v/v) and 2 minutes in sodium hypochlorite (1.25% active chlorine). This was followed by triple washing. The inoculation was carried out in test tubes containing 15 mL MS medium (30 g L-1 sucrose, 6 g L-1 agar and 100 mg L-1 ascorbic acid) supplemented with 2.0 mg L-1 naphthalene acetic acid (NAA). The solution also contained 0.0, 2.5 or 5.0 mg L-1 of 6-benzylaminopurine (BAP) with the pH adjusted to 5.8. In the incubation phase, the explants were cultured for seven days in the dark and then subjected to a photoperiod of 16 hours (43 µmol m-2 s-1) at 25 ± 2 °C. The treatments were studied with 2.5, 5.0, 7.5 or 10.0 mg L-1 BAP additions to the MS. Callus formation, contamination and oxidation evaluations were undertaken. The results obtained when using 2.0 mg L-1 NAA concluded that such a treatment should be used to induce callogenesis from nodal explants, while for the tested baru leaf explants, the best results for callus formation were given by the combination of 2.0 mg L-1 NAA with 2.5 mg L-1 of BAP to.

The experiment was conducted to investigate the combined effect of different plant growth regulators and charcoal supplementation in MS medium on growth and development of plantlets regenerated from protocorm like bodies (PLBs) of hybrid orchid. The combination of BAP + NAA, BAP + IAA, BAP + IBA, and IAA + IBA at different concentrations with charcoal supplementation was studied. The result revealed that the use of different growth regulators had significant effect on different parameters studied. The maximum weight of PLBs (5.123 g) was obtained from the combination of BAP + IBA at 1.0 mg/l each. The highest shoot height (3.239 cm) and maximum number of rooted plantlets (4.473) was obtained from 1.0 mg/l each of BAP + NAA combination. The maximum number of leaves (3.490) and the maximum length of leaves (1.946 cm) were obtained from 1.0 mg/l each of BAP + IBA and the highest leaf width (1.166 cm) was obtained from 0.5 mg/l BAP +1.0 mg/l IBA combination. The highest root length was obtained from 0.5 mg/l each of BAP + IAA and the maximum number of regenerated plantlets (20) was obtained from 0.5 mg/l IAA + 1.0 mg/l IBA combination. However, the maximum fresh weight of single shoot (0.219 g) and the maximum number of roots per plantlet (6.300) was obtained from 1.0 mg/I each of IAA + IBA combination.
