The aim of this research to isolate and identify and to test the antioxidant activity of extract and fraction of polyfloral honey originated from Bone regency, South Sulawesi. The results showed that methanol gave results in flavonoid, tannin, saponnin, steroid, and alkaloid with IC50 value of DPPH method of 683,153 µg/mL. DCM extract gave positive results for tannin, steroid and alkaloid with IC50 values of 701,743 µg/mL. n-hexane extract positively contains tannin and alkaloid with IC50 1709,536 µg/Ml, water extract positevely contains tannin, sapponin steroid, alkaloid with IC50 value 1698,345 µg/mL. Pure honey contains all aspects tested with IC50 values 2826,471 µg/mL. This showed extracts and sample have weak antioxidant activity.

ABSTRACT Kratom (Mitragyna speciosa) is one of the herbal plants from Indonesia. Kratom plants have several benefits overcoming diarrhea, increase endurance, and deal with pain. Kratom leaf has several pharmacological effects, one of which is antioxidant. This study aims to determine the antioxidant activity of kratom leaf ethanol extract using the 1.1-Diphenyl-2-Pikrilhidrazil (DPPH) method expressed by IC50 values. Kratom leaves were extracted by the 3x24 hour remaceration method using 70% ethanol solvent, then concentrated with a rotary evaporator at 40 ° C, followed by an oven at a temperature (40-50) ° C to obtain a thick extract. Kratom leaf extract contains alkaloid compounds, flavonoids, tannins,and saponins. The antioxidant activity test was carried out with qualitative and quantitative tests.The qualitative antioxidant test was carried out using the Thin Layer Chromatography (TLC) method with n-hexane: ethyl acetate (7: 3) eluent, followed by DPPH spraying,which showed positive results giving antioxidant activity. The quantitative test for antioxidants was carried out using the 1,1-Diphenyl-2-Picrylhydrazyl (DPPH) method using a  SpectrophotometerUV-Vis with vitamin C as a comparison. The quantitative antioxidant test was carried out using the 1,1-Diphenyl-2-Pikrilhidrazil (DPPH) method with vitamin C as a comparison. The results of antioxidant activity testing of kratom leaf ethanol extract showed an IC50 value of 91.86 ppm,so that it was included as a powerful antioxidant group. Wualae (Etlingera elatior (Jack) R.M Smith) is one of the plants in the Zingiberaceae family, widely used in medicine ranging from rhizomes, fruits, and flowers. Empirically Wualae are usually used by people for treating skin diseases, flavoring food, a natural soap, and in Southeast Sulawesi, it is used to treat typhoid fever. This study aims to examine the antioxidant activity of Wualae (Etlingera elatior) plant. The plant extracts were extracted by maceration method, antioxidant activity test was performed by 2,2-Diphenyl-1-picrylhydrazyl (DPPH) method. The results showed that ethanol extracts of fruits, leaves, stems and rhizomes of Wualae (Etlingera elatior) has the potential antioxidant activity with the IC50 values of 72.518 mg/L, 99.890 mg/L, 52.345 mg/L and 58.638 mg/L, respectively. and Vitamin C has IC50 value of 3,787 mg/L.

This study aimed to investigate phytochemical screening and antioxidant activity of n–hexane, ethyl acetate and ethanol extract from lakoocha leaves. The powdered simplicia was macerated with n–hexane, ethyl acetate and ethanol 96% successively, filtered, then concentrated using rotary evaporator to obtain n–hexane extract, ethyl acetate extract and ethanol extract. Phytochemical screening and antioxidant activity was performed against these extracts. Antioxidant activity was determined by DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging method using ultraviolet-visible spectrophotometer at wavelength of 516 nm after incubated for 60 minutes in dark place. Quercetin was used as positive control. The result of phytochemical screening showed n-hexane extract contains steroid, ethyl acetate extract contain steroid, tannin, glycoside, flavonoid and saponin, whereas ethanol extract contain tannin, glycoside, flavonoid and saponin. The IC50 value of n–hexane, ethyl acetate and ethanol extract was 1062.03±1.42 ppm, 323.18±0.02 ppm and 99.23±0.07 ppm respectively, whereas for quercetin was 2.32±0.01 ppm. This study showed that ethanol extract had antioxidant activity with strong category whereas n-hexane extract and ethyl acetate extract had inactive antioxidant activity with very weak categories.       Keyword: Antioxidant Activity, DPPH, Lakoocha leaf

ABSTRACT Herbal supplements are one of the products that are taken from outside the body derived from a mixture of herbal plant ingredients have antioxidant activity to prevent the presence of free radicals in the body. Testing natural antioxidant activity in herbal supplement products using the DPPH method (1,1-diphenyl-2-picrylhyrazil). Measurement of UV-Visible spectrophotometry absorbance with DPPH absorbance value at a wavelength of 517 nm. Sensory analysis using a hedonic test at the panelist preference level, using four attribute parameters namely taste, aroma, color and overall acceptance. Results Tests of antioxidant activity in herbal supplement products on various combination techniques and concentrations of garlic, ginger, local lemons and honey, resulting in the reduction of DPPH which is characterized by a reduction in the intensity of the color from purple to fade to yellow. Antioxidant testing obtained IC50 values ​​from all herbal supplement samples from various treatment techniques and concentrations showed IC50 values ​​less than 50 found in the treatment of chopped engineering materials, concentration 1 (K1) of 23.97%. While concentrations of 2 (k2) and 3 (k3) as well as in various combination techniques and other concentrations indicate that IC50 values ​​range from 50 ppm to 100 ppm. This shows that herbal supplements have very strong antioxidants (IC50 value <50) found in the treatment of ingredients with chopped techniques with the lowest concentration. whereas herbal supplements with treatment techniques and other concentrations have a strong antioxidant value of IC50 (50-100). The results of sensory analysis indicate that the average panelist had a preference level for herbal supplement products with chopping and extraction treatment techniques. Keywords: Antioxidant, DPPH, Herbal supplements, IC50   ABSTRAK   Suplemen herbal merupakan salah satu produk yang menjadi asupan dari luar tubuh yang berasal dari hasil campuran bahan tanaman herbal yang mempunyai aktivitas antioksidan tinggi untuk mencegah adanya radikal bebas dalam tubuh. Pengujian aktivitas antioksidan alami pada produk suplemen herbal menggunakan metode DPPH (1,1-diphenyl-2-picrylhyrazil). Pengukuran absorbansi secara spektrofotometri-UV Visibel dengan Nilai absorbansi DPPH pada panjang gelombang 517 nm. Analisis sensori dengan menggunakan uji hedonik pada tingkat kesukaan panelis, menggunakan empat parameter atribut yaitu rasa, aroma, warna dan penerimaan keseluruhan. Hasil Pengujian aktivitas antioksidan dalam produk suplemen herbal pada berbagai teknik kombinasi dan konsentrasi bawang putih, jahe, lemon lokal dan madu, menghasilkan peredaman DPPH yang ditandai dengan berkurangnya intensitas warna ungu menjadi pudar sampai kekuningan. Pengujian Antioksidan didapatkan Nilai IC50 dari seluruh sampel suplemen herbal dari berbagai teknik perlakuan dan konsentrasi menunjukkan nilai IC50 kurang dari 50 terdapat pada perlakuan bahan teknik rajangan, konsentrasi 1 (K1) sebesar 23,97 %. Sementara konsentrasi 2 (k2) dan 3 (k3) serta pada berbagai teknik kombinasi dan konsentrasi lainnya menunjukan bahwa nilai IC50  berkisar 50 ppm - 100 ppm. Hal ini menunjukkan bahwa suplemen herbal memiliki antioksidan yang sangat kuat (nilai IC50 <50) terdapat pada perlakuan bahan dengan teknik rajangan dengan konsentrasi yang paling rendah, sedangkan suplemen herbal dengan teknik perlakuan dan konsentrasi lain memiliki nilai antioksidan kuat IC50 (50-100) ppm. Hasil analisis sensori menunjukan bahwa rata-rata panelis memiliki tingkat kesukaan pada produk suplemen herbal dengan teknik perlakuan rajangan dan perlakuan sari. Reactive Oxygen Species (ROS) are produced by humans as a result of cellular metabolism and environmental factors such as pollutants or cigarette smoke. ROS is a very reactive molecule and has the ability to damage cell structure. Oxidative stress is a condition between oxidants and antioxidants that is not balance, pathophysiologically, oxidative stress can trigger the risk of various diseases including hypertension, atherosclerosis, diabetes, heart failure, stroke and other chronic diseases. Oxidative stress can be overcome by intake of antioxidants. The purpose of this study was to determine the antioxidant activity contained in the meat and shells of a knife scallop (Solen sp.) that lives in Bangkalan waters, Madura. The method used in this study is an analysis of antioxidant activity with the DPPH method. Knife scallop has antioxidant activity which is evident from the IC50 values obtained. IC50 values of the shell and meat extract with ethanol solvent were 489.56 ppm and 748.49 ppm. IC50 values of the shell and meat extract with ethyl acetate solvent were 916.43 ppm and 2045.93 ppm. While the IC50 value of the shell and meat extract with chloroform solvent was 119.37 ppm and 1692.80 ppm. Based on IC50 data of knife scallop shell and meat extracts on the 3 types of solvents, it can be concluded that knife scallop’s shell and meat extract with ethanol solvent has the greatest antioxidant activity compared to ethyl acetate and chloroform solvents.

 Jumpai (Glinus Oppoitifolius (L.) Aug. DC) contains flavonoids compounds that can act as antioxidants by donating hydrogen so as to stabilize the lack of electrons in free radicals. This study aims to determine the antioxidant potential of stem, leaf, and flower extracts by looking at the IC50 value. The stems, leaves, and flowers were extracted by maceration using 70% l ethanol solvent. The results of the antioxidant activity test using the DPPH method (2,2-diphenyl-1-picrilhydrazil) showed very strong antioxidant activity with IC50 values of 9.523 µg / ml stem, 32.89 µg / ml leaves, and 23.07 µg / ml flowers with vitamin comparisons. C obtained IC50 value of 1,698 µg / ml. Based on these results, it can be rejected that the stems, leaves, and flowers have antioxidant activity with a very strong category against DPPH free radicals (2,2-diphenyl-1-picrilhydrazil).

The objectives of this study was to determine the half maximum inhibitory concentration (IC50) from four types of Parkia speciosa peel extracts (ethanol, methanol, n-hexane, and aqueous)  through DPPH free radical inhibition. First Parkia’s peel extract made by drying the Parkia’s peel that has been sorted, then crushed and mashed with a blender. Parkia’s powder then macerated for 3 replication using each type of solvent and then solvent evaporation was carried out using a rotary vacuum evaporator. The evaporated extract produced then tested for antioxidant activity using the IC50 method and phytochemical screening was performed to analyze the potential content of functional compounds. The results showed that all types of solvents dissolve alkaloid compounds (except water extract), flavonoids, saponins, tannins, and phenols. IC50 values produced from the four types of petai bark extract using methanol, ethanol, water, and n-hexane solvents sequentially were 76.92; 111; 136; and 201 ppm. Methanol extract had the lowest IC50 value of 76.92 ppm which resulted that the methanol extract of petai skin had a strong (active) antioxidant strength compared to others.Keywords: Antioxidant, Exctract, Parkia’s peel, IC50

Objective: To assess the antioxidant activity from another part of the plant, in this study, leaf extracts in n-hexane were fractionated.Methods: Ten fractions were obtained and tested in vitro for antioxidant activity using two methods, 1,1-diphenyl-2-picrylhydrazyl (DPPH) and ferricreducing antioxidant power (FRAP), to identify the most active fraction.Results: The IC50 of the most active fraction was 36.24 μg/mL using the DPPH method, and the EC50 was 39.54 μg/mL using the FRAP method. Themost active fraction was also shown to contain terpenoids.Conclusion: The most active fraction of an n-hexane extract of the leaves of Gacinia bancana Miq., which was tested by both DPPH and FRAP methodshad antioxidant activities with IC50 and EC50 values of 36.2482 μg/mL and 39.5442 μg/mL, respectively. Phytochemical screening showed that activefraction contains terpenoids.
