The objectives of this study was to determine the half maximum inhibitory concentration (IC50) from four types of Parkia speciosa peel extracts (ethanol, methanol, n-hexane, and aqueous)  through DPPH free radical inhibition. First Parkia’s peel extract made by drying the Parkia’s peel that has been sorted, then crushed and mashed with a blender. Parkia’s powder then macerated for 3 replication using each type of solvent and then solvent evaporation was carried out using a rotary vacuum evaporator. The evaporated extract produced then tested for antioxidant activity using the IC50 method and phytochemical screening was performed to analyze the potential content of functional compounds. The results showed that all types of solvents dissolve alkaloid compounds (except water extract), flavonoids, saponins, tannins, and phenols. IC50 values produced from the four types of petai bark extract using methanol, ethanol, water, and n-hexane solvents sequentially were 76.92; 111; 136; and 201 ppm. Methanol extract had the lowest IC50 value of 76.92 ppm which resulted that the methanol extract of petai skin had a strong (active) antioxidant strength compared to others. Keywords: Antioxidant, Exctract, Parkia’s peel, IC50

Free radicals are molecules that lose electrons in their outer orbitals so that the number of electrons becomes odd and unstable. Free radicals can damage various macromolecule cells including proteins, carbohydrates, fats. Antioxidants are compounds that can donate one or more electrons to free radicals. Avocado leaves contain high bioactive components that can be used as a source of natural antioxidants. Avocado leaves that have been dried, mashed and then shifted with methanol. The extracts were tested by phytochemical screening, including flavonpoid, phenolic, tannin and antioxidant activity using the DPPH (2,2-diphenyl-1-picrylhidrazyl) method. This method is based on the reduction of 2,2-diphenyl-1-picrylhydrazyl (DPPH) against free radicals which causes a change from purple to yellow 2,2-diphenyl-1-picrylhydrazyl) (DPPH). From the research, the IC50 values in avocado leaves were 118.8056 µg / mL and 7,276 µg / mL on ascorbic acid as a positive control. From the IC50 value obtained, it is known that the methanol extract of avocado leaves has strong antioxidant activity to fight free radicals.

The purpose of this study was to investigate the major flavonoids content and bioactivities of Tartary buckwheat sprouts. The crude methanol extract (ME) of Tartary buckwheat sprouts was abundant in flavonoids, and six major flavonoids, including isoorientin, vitexin, isovitexin, rutin, quercetin, and kaemferol were successfully determined from the sprouts by the high-performance liquid chromatography (HPLC) method. Generally, the flavonoid content of buckwheat sprouts was in the order of rutin > quercetin > isovitexin > vitexin> isoorientin > kaemferol. The highest rutin content of the ME and sprout cultures was 89.81 mg/g and 31.50 mg/g, respectively. Antibacterial activity results indicated the ME displayed notable inhibitory activity against the five tested bacteria, and its minimum inhibitory concentration (MIC) values ranged from 0.8 mg/mL to 3.2 mg/mL. Among the six flavonoids, quercetin was the most active compound, which exhibited strong activity against all tested bacteria except for E. coli and S. epidermidis, with its MIC values ranging from 0.2 mg/mL to 0.4 mg/mL. For the antifungal activity assay, the ME of Tartary buckwheat sprouts and four flavonoids could significantly inhibit the spore germination of two pathogenic fungi, and their inhibitory efficiency was concentration dependent. Quercetin was the most active one, which significantly inhibited the spore germination of F. oxysporum f. sp. vasinfectum and F. oxysporum f. sp. cucumerinum, and its median effective inhibitory concentration (IC50) value was 42.36 and 32.85 µg/mL, respectively. The antioxidant activity results showed that quercetin, kaemferol, and rutin displayed excellent antioxidant activity in the DPPH radical scavenging test, and their IC50 value was calculated as 5.60, 16.23, and 27.95 µg/mL, respectively. This is the first report on the antimicrobial activity of the crude extract of Tartary buckwheat sprouts. These results indicated that the methanol extract of Tartary buckwheat sprouts could be used as a potential antimicrobial or antioxidant agent in the future.

Antioxidant is a new hype in naturopathy and at present it has huge demand in cosmetic, nutritional supplementation and pharmacological industry. It reduces cell damages caused by free-radical which are responsible for various ailment like ageing, cancer, coronary heart disease, diabetes mellitus, neurodegenerative disorders, atherosclerosis, cataracts and inflammation. At present any comprehensive data on the antioxidant content and extent of their activity of the indigenous medicinal plants of Bangladesh is not available. Therefore, here antioxidant activity of a popular medicinal plant - Ficus racemosa was determined. Methanolic extract was prepared from the leaf and stem bark of F. racemosa. Their total phenolics and free radical scavenging capacity was determined using folin-ciocalteu reagent (FCR) assay and DPPD-radical scavenging assay respectively. Stem bark and leaves of F. racemosa contains 242.97 and 235.45 mg of GAE / gm of dried extract of phenolics respectively. Current studies also show that F. racemosa barks contain a high antioxidant activity. IC50 value of bark extract was found to be 19?g/ml which is even better that of the standard BHT. Therefore, present studies suggest that F. racemosa bark has a huge potential in terms of commercial use as source of antioxidant. DOI: http://dx.doi.org/10.3329/jles.v8i0.20152 J. Papasan leaves have several properties, including antihyperglycemic, diabetes, antipyretic and antioxidant. Minor metabolite compounds that have the potential as antioxidants from this plant are flavonoids, alkaloids, saponins, and tannins. This study's purpose to determine the antioxidant activity and the flavonoid total contents of Papasan (Coccinia grandis L.) leaf extract based on the differences in polar solvents. The polar solvents used were ethanol, methanol, and water. Extraction used by maceration and dekokta. The antioxidant activity and the flavonoid total contents were established by UV-Vis spectrophotometry according to the decrease in absorbance at a maximum wavelength of 515 nm by the DPPH (2,2-diphenyl-1-picrylhydrazyl) method. Determination of antioxidant activity and the flavonoid total contents were excluded on quercetin comparisons. The values of  IC50 and the flavonoid total contents in the ethanol extract respectively were 287.92 ppm and 49.825 mg QE/g while the IC50 values and total flavonoid levels in the extract of methanol were 73.29 ppm and 50.415 mg QE/g. In the extract of the water, the value of IC50 was 39.80 ppm and the total flavonoid contents were 50.415 mg QE/g. The conclusions of these studies were that the most antioxidant activity was found in water extract, while water and methanol extract had the same and highest levels of total flavonoids.
