Objective: The aim of the present work was designed to study cellular antioxidant properties of selected plant extracts of Caralluma and Boucerosia on cell lines. Materials and Methods: The present study was carried out in four species of Caralluma R.Br. such as Caralluma adscendens (Roxb.) R. Brown var. attenuata (Wight) Grav. & Mayur. (CAA), Caralluma adscendens (Roxb.) R. Brown var. fimbriata (Wall.) Gravely & Mayur. (CAF), Caralluma stalagmifera C.E.C. Fisch. (CS) and Caralluma stalagmifera C.E.C. Fisch. var. longipetala  Karupp. & Pull. (CSL) and as well as two species of Boucerosia Wight & Arn. such as Boucerosia lasiantha Wight. (BL) and Boucerosia umbellata (Haw.) Wight & Arn.  (BU). The antioxidant efficacy was evaluated by ability of antioxidant molecules in the extract to scavenge ROS produced by normal metabolism by cells, observed by the oxidation of 2, 7- Dichloro fluorescein diacetate to dichlorofluorescein, followed by decrease in fluorescence intensity. Results: Cellular antioxidant activity was demonstrated by the inhibitory concentration 50% (IC50) of reactive oxygen species (ROS). The percentage ROS inhibition in Raw 264.7 cells when treated with different concentrations of methanolic extracts showed variations among selected species. The present study describes the in vitro antioxidant activity of methanol extracts of 34 plant species collected in the Colombian coffee-growing eco-region belonging to Euphorbiaceae, Piperaceae and Solanaceae families. The antioxidant properties of extracts were evaluated by determining radical scavenging power measured with a DPPH assay. The methanolic extracts of  Hyeronimia antioquiensis, Mabea montana, and Alchornea grandis species (Euphorbiaceae), presents EC50 values equal to 0.686, 12.35, and 13.01 µg/mL, respectively, showing high antioxidant potential.

Pitaya peel can be used as a raw material for betalains extraction. The aim of this research was to quantify phenolic compounds, antioxidant activity and betalains on pitaya peel. Furthermore, evaluate the betalains stability against various pH conditions and exposure time of heating. The results showed that pitaya peel contains phenolic compounds and presented antioxidant activity. Moreover it showed high concentration of betalains (101.04mg equivalent to betanin. 100g-1) which were stable over a wide pH range (3.2 - 7.0) and were resistant to heating (100oC) up to 10 minutes at pH range from 3.7 to 5.5. Therefore, pitaya peel is a promising source of betalains which can be applied as a natural colorant for food.

AbstractUse of potassium persulfate (K2S208) for oxidation of 7.0 mM ABTS to a stable ABTS radical for antioxidant studies was first reported in 1999. A feature of this popular antioxidant assay has been the requirement of an overnight reaction (6 to 12 h) for the formation of a stable ABTS colored radical. It is now reported that when the concentration of ABTS is lowered to 0.7 mM, complete oxidation to the stable cation radical occurs in 30 min, thus circumventing the necessary overnight step. Using this format, it is now possible to accurately assess antioxidant activity based on the potassium persulfate/ABTS format in less than one hour which includes formation time of a stable ABTS radical. This methodology documented the presence of antioxidant properties of plant extracts used in Traditional Chinese Medicine. The degree of antioxidant activity was directly related to the extraction method. Greater antioxidant activity was associated with butanol extraction. When incorporated into a microtiter plate format, it supported rapid assessment of multiple determinations of dilutions of plant extracts in less than one hour which included time required for formation of a stable ABTS radical. The ease, improved time prerequisites, and minimal reagent needs with the microtiter plate format, makes this design attractive. It would prove of particular interest to individuals engaged in both smaller and high-volume throughput antioxidant assays of food and health products, and other biological fluids and tissues.

Abstract Wood species resistant to biodegradation by fungi contain phenolic compounds with proven fungicidal activity. Saprophytic fungi are able to colonize wood and later begin to break down its polymeric components through metal-dependent reactions. These generate free radicals at the onset of the degradation process. Later on, extracellular enzymes are active that use metallic cofactors. Five lignans were isolated from the methanolic extracts of the Chilean heartwoods (Austrocedrus chilensis, Fitzroya cupressoides, and Pilgerodendron uviferum) belonging to the Cupressaceae: isotaxiresinol, secoisolariciresinol, isolariciresinol, didemethylmatairesinol, and matairesinol. Their structures were determined spectroscopically. The antioxidant activity of the methanolic extracts and the isolated compounds was determined in vitro against the radical 1,1-diphenyl-2-picrylhydrazyl (DPPH) and their iron-chelating capacity was evaluated by the Chrome Azurol S assay. All isolated methanolic extracts and lignans exhibited antioxidant activity against DPPH and Fe-chelating capacity with values greater than 87%. This may be interpreted that the antioxidant properties and metal-chelating capacity of these compounds have a protective effect on the wood of these species when exposed to attacks by saprophytic fungi.

Leaf methanolic extracts of four wild Rose species (Rosa canina L., R. corymbifera Borkh., R. micrantha Borrer ex Sm and R. sempervirens L.) grown in Sicily were analysed in order to evaluate differences among indigenous roses which could be used such as source of bioactive compounds and natural antioxidants. Leaves were harvested during two different periods (June and October) and their morphological characteristics were described. Leaf content of chlorophylls, total anthocyanins, total flavonoids, total polyphenols (Folin-Ciocalteu method) and antioxidant activity (DPPH method) were determined. Results showed that the contents of chlorophylls and carotenoids were higher in leaves of all species harvested in June than in those harvested in October. On the contrary, total anthocyanins of R. micrantha and R. corymbifera leaves were higher in October. Total polyphenol content was higher in leaves of all species were harvested in October. No difference in total flavonoids content was recorded between the two harvest periods but only among the four species. Antioxidant activity was generally high in leaf extracts of all species in both periods and, particularly, in R. micrantha leaves harvested in June. These outcomes seem to support the assessment that leaves harvested from some Sicilian wild roses may be used as promising source of healthy compounds and that their content varies with the species and harvest period.

Background & Objectives: The present investigation is intended to prepare a Poly Herbal Formulation (PHF) with Piper nigrum (fruits), Terminalia paniculata (bark) and Bauhinia purpurea (bark) and assess their antioxidant and glucose-lowering effects utilizing in vitro models. Methods: The individual plant methanolic extracts and PHF are exposed to phytochemical examination and to distinguish the bioactive factors by GC-MS. We assessed the antioxidant properties of individual plant extracts and the PHF by using the DPPH scavenging method, H2O2 scavenging assay, TBARS assay and total antioxidant estimation. Likewise, the anti-diabetic activity was assessed by ɑ-amylase and α-glucosidase enzyme inhibition and glucose diffusion inhibitory techniques. Results: We found that PHF contains a high measure of total phenolics, total flavonoids and tannin compared to individual plant extracts. The GC-MS identified the bioactive components. We also found that PHF had significantly higher antioxidant and glucose-lowering effects than the individual plant concentrates. Conclusion: In conclusion, it could be reasoned that due to the nearness of antioxidant components, the PHF has good potential in the administration of hyperglycemia, diabetes and the related state of oxidative stress. This study shows that PHF is superior to individual plant extracts, supporting the conventional PHF concept.

Objective: To evaluate cytotoxic and antioxidant activity of the plant Bauhinia racemosa root by in-vitro cell line analysis. Methods: MCF-7 cell lines were used for the study to find the inhibitory concentration of the extract. DPPH and NO scavenging assays were performed to study the antioxidant nature of the extract. Results and Conclusion: In MCF-7 cell line cytotoxicity effect was observed in tested sample concentrations in 48 hours treatment, it also revealed that increased concentration of drug shown increased cytotoxicity over the MCF-7 cell lines. DPPH and NO scavenging Assay revealed a concentration dependent radical scavenging capacity of the tested sample concentrations of Bauhinia racemosa root spectrophotometrically.

Tragopogon porrifolius, commonly referred to as white salsify, is an edible herb used in folk medicine to treat cancer. Samples of Tragopogon porrifolius plant grown wild in Palestine were extracted with different solvents: water, 80% ethanol, and 100% ethanol. The extracts were analyzed for their total phenolic content (TPC), total flavonoid content (TFC), and antioxidant activity (AA). Four different antioxidant assays were used to evaluate AA of the extracts: two measures the reducing power of the extracts (ferric reducing antioxidant power (FRAP) and cupric reducing antioxidant power (CUPRAC)), while two other assays measure the scavenging ability of the extracts (2,2-azino-di-(3-ethylbenzothialozine-sulphonic acid (ABTS)) and 2,2-diphenyl-1-picrylhydrazyl (DPPH)). Anticancer activity of the plant extracts were also tested on HOS and KHOS osteosarcoma cell lines. The results revealed that the polarity of the extraction solvent affects the TPC, TFC, and AA. It was found that both TPC and AA are highest for plant extracted with 80% ethanol, followed by water, and finally with 100% ethanol. TFC however was the highest in the following order: 80% ethanol > 100% ethanol > water. The plant extracts showed anticancer activities against KHOS cancer cell lines; they reduced total cell count and induced cell death in a drastic manner.
