The article substantiates the formulation of experimental work on the study of adaptive features of antioxidant factors in the body's defensive reactions to physical activity. The influence of physical activity on the oxidative situation in muscles is discussed; the results of studies of LPO intensity, (dynamics of the level of malondialdehyde), the activity of enzymes involved in antioxidant protection (glutathione – S – transferases, catalases, and super oxidants) in blood plasma and erythrocytes of wrestlers during annual training macrocycle are described. It was determined that the level of malondialdialdehyde significantly exceeds the control throughout the year. MDA concentration reaches a maximum at the end of the competition period and is not restored during the rest period. The activity of superoxide dismutase and catalase is significantly reduced and does not return to control values during the year. The mentioned changes confirm chronic decompensation in the antioxidant defense system. A leading enzyme that protects against active forms of oxygen in the blood plasma and red blood cells of athletes is glutathione - S - transferase, compensatory increase in activity that reaches 630% relative to the level of the control group

Background. The problem of gastric cancer remains unresolved throughout the world, while chronic atrophic gastritis (CAG) increases the likelihood of its development by 15 times. In the Russian Federation, the incidence of gastric cancer (GC) is among the highest, with it prevailing among males. One of the leading mechanisms in molecular pathology of membranes is lipid peroxidation (LPO). The severity of oxidative membrane damage depends on concomitant diseases, contributing to emergence and progression of pathological processes and development of cancer. Currently, the problem of LPO is unsolved in biological systems.The aim of this study was to investigate the state of LPO and antioxidant defense system in CAG and GC. Materials and methods. The parameters were studied in 45 patients with CAG and 50 patients with GC. The control group included 50 practically healthy volunteers without gastrointestinal complaints, who did not have changes in the gastric mucosa according to the fibroesophagogastroduodenoscopy (FEGDS) findings.Results. In patients with CAG, an increase in malondialdehyde, superoxide dismutase, catalase, glutathione S-transferase, and glutathione peroxidase was found in the blood plasma compared with the control group. In patients with CAG, lipid peroxidation was activated, and the malondialdehyde level increased by 3.5 times relative to normal values. At the same time, the body fought against oxidative stress by increasing the activity of antioxidant enzymes, such as superoxide dismutase, catalase, glutathione S-transferase, and glutathione peroxidase. All patients with GC showed pronounced oxidative stress in the blood plasma in the form of a 45-fold increase in malondialdehyde. The activity of the main antioxidant enzyme superoxide dismutase was reduced in GC. Catalase was activated, which indicated pronounced oxidative stress, significant damage to blood vessels, and massive cell death. Glutathione-related enzymes (glutathione S-transferase and glutathione peroxidase) and the antioxidant protein ceruloplasmin were activated, which also indicated significant oxidative stress and severe intoxication in patients with GC.Conclusion. Depending on the stage and type of cancer, an in-depth study of lipid peroxidation and factors of the antioxidant defense system can be used to correct therapy and prevent cancer and can serve as markers of progression and prognosis in gastric cancer.

AbstractThe aim of the study was to analyze the activity of antioxidant enzymes (glutathione S-transferase, catalase, superoxide dismutase) and the concentration of malondialdehyde in order to determine the role of detoxification mechanisms in prostate cancer. The activities of superoxide dismutase (SOD), catalase (CAT), and glutathione S-transferase (GST) were measured using ready-made kits; lipid peroxidation intensity was determined by the thiobarbituric acid method. Superoxide dismutase was the only enzyme among antioxidant and detoxification enzymes for which a statistically significant difference in activity was found between the studied groups (1.4 U·ml−1 in patients vs. 1.6 U·ml−1 in control). No statistically significant differences were found for GST, CAT or the concentration of MDA between the group of men with prostate cancer and the control group. The lower SOD activity in men with prostate cancer may be due to a deficiency in their antioxidant defense system.

Glutathione S-transferases (GSTs) are enzymes that included, in a more range of detoxifying reactions by conjugation of glutathione, to electrophilic material. Polymorphisms n the genes that responsible of GSTs affect, the function of the GSTs. GSTs play an active role in protection of cell against oxidative stress mechanism. Polymorphisms of GSTP1 at codon 105 amino acids forms GSTP1 important site for bind of hydrophobic electrophiles and the substitution of Ile/Val affect substrate specially catalytic activity of the enzyme and may correlate with reach to different diseases in human like diabetes mellitus type2 disease. Correlation between these polymorphisms and changes in the parameters file of diabetic patients has also been found, therefore, the results variation considerably among the studies; therefore, these control study was designed to leading to detecting know, as there are no studies on this performed in the people of Iraq. The polymerase chain reaction-restriction fragment length polymorphism was used to study GSTP1genetic polymorphism in 60 T2DM patients and 50 healthy individuals. Our results showed that presence of the GSTP1 heterozygous mutant allele Ile/Val was more common in subjects with T2DM than in the control group (40.00% and 32.00%, respectively; p = 0.01), as well as the found of the homozygous mutant of GSTP1 allele Val/Val was common in T2DM patient and not found in the control group (3.33% and 0.00%, respectively; p = 0.001).GSTP1 genotypes do not have an effect on blood lipids after infection with diabetes mellitus. Agarose gels used to determined genotypes according to the bands were that appeared in electrophoresis of gel.

In this study, goldfish (Carassius auratus) were exposed to 0 (control group) and 0.81 mg/L TCCA for four consecutive days. The liver transcriptome, the molecular indices of oxidative stress, and gills histopathology were investigated. Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis indicated that energy metabolism-related pathways such as glycolysis/gluconeogenesis were significantly enriched, suggesting their perturbation in the liver of goldfish. Additionally, TCCA exposure also caused pathological damage in gills, which compromised physiological function and decreased oxygen intake capacity of gills, thus leading to the enhancement of anaerobic metabolism. This finding was confirmed by the significant upregulation of lactate dehydrogenase in the liver of goldfish. Moreover, many phase I and phase II metabolic enzymes might be activated to alleviate TCCA-induced toxicity in goldfish, and glutathione S-transferases (GSTs) and cytochrome P450s (CYPs) play a crucial role in the metabolism of TCCA in the liver of goldfish. Furthermore, the antioxidant enzyme analysis showed that TCCA exposure induced oxidative damage in the liver and partially impaired the antioxidant defense system of goldfish, evidenced by decreased superoxide dismutase (SOD) and catalase (CAT), and increased malondialdehyde (MDA) level. In summary, this study will improve our understanding of the molecular mechanisms of the TCCA-induced toxicity in goldfish.

Several biomarker enzymes such as catalase (CAT) and glutathione S-transferase (GST) can be used to measure oxidative stress in animals caused by exposure to xenobiotics. The objective of the present study was to characterize different points of the Capivari (CP1 and CP2), Paraguaçu (PG1 and PG2) and Subaé (SB1 and SB2) Rivers, state of Bahia, in relation to the presence of xenobiotics, using CAT and GST as bioindicators in M. jelskii. The water-sampling sites were considered urban or rural and in all of them signs of environmental degradation were observed. Therefore, acute exposure tests (96h) were performed with water samples collected during the dry and rainy seasons. Results showed that the activity of CAT and GST in prawns exposed to water from CP1 and CP2 were very similar, while those exposed to water from PG1, PG2, SB1 and SB2 formed distinct groups of data. Significant increase in the activity of at least one of the analyzed enzymes in each sampling site was observed, when compared to animals in the control group. This demonstrated a possible oxidative stress in M. jelskii caused by the presence of xenobiotics in the water (e.g., domestic sewage, pesticides, oil, and heavy metals). Enzymatic activities were higher in animals from experiments carried out in the rainy season, except for the CAT activity of animals exposed to water from Subaé River. This study demonstrated the potential of M. jelskii as bioindicator and contributed to the knowledge of aspects of the antioxidant defense system of this species.

The GSTP1 gene encodes glutathione S-transferase P1, which is a member of the glutathione S-transferases (GSTs), a family of enzymes playing an important role in detoxification and in the antioxidant defense system. There is some evidence indicating that GSTP1 c.313A>G polymorphism may be beneficial for exercise performance. Therefore, we decided to verify the association between the frequency of GSTP1 c.313A>G variants, physical performance, and athletes' status in two cohorts: in a group of Russian athletes ( n = 507) and in an independent population of Polish athletes ( n = 510) in a replication study. The initial association study conducted with the Russian athletes revealed that the frequency of the minor G allele was significantly higher in all athletes than in controls; that was confirmed in the replication study of Polish athletes. In the combined cohort, the differences between athletes ( n = 1017) and controls ( n = 1246) were even more pronounced (32.7 vs 25.0%, P < 0.0001). Our findings emphasize that the G allele of the GSTP1 gene c.313A>G single nucleotide polymorphism is associated with improved endurance performance. These observations could support the hypothesis that the GSTP1 G allele may improve exercise performance by better elimination of exercise-induced ROS.

Hodgkin?s Lymphoma (HL) is a heterogeneous malignant disease of lymph node. The glutathione S-transferases (GSTs) have an important role in the detoxification of a wide variety of toxins and carcinogens. Studies have been indicated that genetic variation in the GST gene family may lead to susceptibility in HL. Hereby, we investigated the association of GSTT1 and GSTM1 null genotypes with HL in the Iranian population. This case-control study consisted of 76 patients suffering from HL and 120 healthy individuals as a control group. Genomic DNA was extracted and genotyping of GSTT1 and GSTM1 genes for the identification of their null genotypes was carried out using multiplex PCR method. Our findings indicated that GSTM1 null genotype is associated with risk of developing HL in our population (P=0.025; OR=2.00; 95%CI=1.110- 3.602); however, no association was found for GSTT1 null genotype. Our study also showed that the GSTM1 null genotype increased the risk of disease in the individuals younger than 45 years, and it had a positive association with low ESR. GSTM1 null genotype may have the key role in increasing the risk of HL in the Iranian population.

Children with obesity have high risk to have abnormal cholesterol rate. Obesity and high cholesterol rate can cause cardiovascular disease at a later time. Children have normal rate of cholesterol if the cholesterol rate in the blood is <170 mg/dL, the threshold category between 170-199 mg/dL, and high category is >200 mg/dL. Soy Milk and avocado juice are the ways of non pharmacological care that can be applied to reduce cholesterol rate. This study aims to compare Soy Milk and avocado juice giving toward cholesterol rate in children with obesity in State Elementary School 1 and 2 of Katerban, Central Java Province, Indonesia. The study used quasi experiment design with non-equivalent control group framework. Samples of the study were 30 children taken by use purposive sampling. Soy Milk and avocado juice effective to reduce cholesterol level in obesity children (p value=0,000, p<0,05), but neither soy milk and avocado juice there’re no one that more effective to decrease cholesterol level (p value=0,902, p>0,05). 60% of respondent were male student age 11 years (36,7%). Father education were high. Soya milk and avocado juice are able to reduce cholesterol rate. Parents must give attention to children’s dietary intake to reduce cholesterol and obesity, also motivate them to do physical activity.

Background: Diabetes mellitus (DM) is a common metabolic disease worldwide and has many complications. The vascular events are the major complication of DM that have an important effect on mortality and disability. The physical activity (PA) enhances the vascular function by several pathways. The aim of this study was to evaluation of the relationship between PA and vascular diseases in patients with DM.Materials and Methods: This research was performed as the case-control study that was extracted from a prospective epidemiological research study in Iran (PERSIAN). The patients with type 2 DM more than six months defined as case group and the non-DM subjects in control group with ratio 1:2, and both groups were matched in the term of age and sex. The MET score was used to evaluate the level of PA and blood glucose, lipid profile, body mass index, overweight, dyslipidemia, glomerular filtration rate, myocardial infarction (MI), unstable angina, and stroke.Results: Overall, 1242 patients with DM were extracted, and 2484 non-diabetic subjects were investigated. In the case group, 355(28.6 %) and 887(71.4%) were men and women, respectively, the and 710 (28.6%) men and 1774(71.4%) women in control group. The mean MET score was 30 and 40.97 in the DM and non-DM groups, respectively (P˂0.001). The frequency of MI, stroke, and cardiac ischemia were 44 (3.5%), 37 (3%), and 267 (21.5%), respectively in DM group, and 54 (2.2%), 43 (1.7%), and 389 (15.7%), respectively in non-DM group.Conclusion: The incidence of vascular events associated with PA level in patients with DM and adherence to regular PA reduce the vascular events and DM complications.
