The mangrove plant - Avicennia officinalis L has been widely used for the treatment of various diseases such as anti-inflammatory, anticancer, antioxidant therapy. In this study, we optimize the extraction process using the easily obtainable solvents with different polarities toward obtaining high total phenolic- and flavonoid- contents from Avicennia officinalis L's leaves. The influences of solvents (i.e., methanol, ethanol, ethyl acetate, acetone, dichloromethane, and chloroform) in the extraction process on total phenolic-, flavonoid- contents and their antioxidant activity were investigated. Avicennia officinalis L was extracted using six solvents. The total phenolic and flavonoid contents were determined by the phytochemical screening method. The antioxidant activity of extracted phenolics and flavonoids was studied by using 2,2 diphenyl 1 picrylhydrazy (DPPH) radical scavenging assay and 2,2 azinobis (3 ethylbenzothiazoline 6 sulfonate) (ABTS+) radical scavenging assay. As a result, the highest extraction yield was obtained using methanol (10.56%). In addition, the extract obtained using acetone solvent exhibited the highest antioxidant activity in the DPPH assay. Meanwhile, the extract obtained using ethyl acetate solvent showed the highest antioxidant activity in the ABTS+ assay. The acetone- extract also exhibited high phenolic content (100.64 mg GAE/g) and flavonoid content (95.44 mg QCE/g). This study confirms that, besides commonly used solvents such as methanol, ethanol, etc., acetone is also a good solvent for extracting polyphenols from the mangrove plant - Avicennia officinalis L. for antioxidants.

Objective: The present study was undertaken to determine antioxidant activity and total phenolic and flavonoid content of ethanol, methanol, ethyl acetate, and aqueous extracts of the leaves of Laurus nobilis L.Methods: Antioxidant properties were measured using three tests: Free radical scavenging activity against 2,2-diphenyl picrylhydrazyl (DPPH), reduction of molybdate, and reducing (Fe3+/Fe2+) power. Total phenolic and flavonoid content was measured by Folin–Ciocalteu and rutin reagent, respectively.Results: The results showed that both the ethanol and water extracts had greater antioxidant activity than ethyl acetate. Aqueous extract exhibited a higher DPPH radical scavenging and reducing molybdate 545.83±5.89 mg equivalent to ascorbic acid/g dry extract. The strong antioxidant activity of water extract was probably due to its high content of phenols. Furthermore, ethanol and water extracts showed higher total flavonoid content with the values: 153.33±3.59 and 127.25±2.60 mg equivalent rutin/g dry extract, respectively.Conclusion: Our results suggested a potent and excellent antioxidant activity of L. nobilis L. extracts of Morocco. Further research has to be done to isolate the pure bioactive compound and could be considered as a potential source of biomolecules for pharmaceutical and food industry.

Purpose: Milk thistle (Silybum marianum (L.) Gaertn) has been used for centuries as a natural remedy for liver and gall bladder / biliary tract diseases. Silymarin is the purified and standardized extract from milk thistle seeds. The purpose of the present study is to present a comparative analysis of the total phenolic substances content and the antioxidant effect of silymarin and three different types of Silybum marianum extracts (i.e. ethanol extract, water extract and lyophilized extract). Methods: Total phenolic and flavonoid content of extracts was analysed and the antioxidant activity was assessed by determining their free radical scavenging ability by DPPH method. Results: Total phenolic content is the highest in silymarin– 134.12±3.22 mg GAE/g, followed by lyophilized water extract – 51.74±2.10 mg GAE/g. Silymarin has also been found to have the highest antioxidant activity among all extract samples (338.03 VCEAC µmol /g), followed by S.marianum lyophilised extract (123.05 VCEAC µmol/g). Conclusions: A strong correlation between total phenolic and flavonoid content in the extracts and their antioxidant activity has been found.

Ziziphora is a plant used in Turkish and Iran traditional medicine for its antibacterial activity, sedative and stomach soothing properties. Although the chemical profile of the essential oil of different Ziziphora species is well documented, data regarding plant extracts are incomplete. In this study extracts from Ziziphora taurica subsp. cleonioides were obtained using ethyl acetate, methanol and water and the chemical profile of the aerial part of the plant was elucidated. Among the compounds identified, rosmarinic acid was the most abundant (3375.67 ± 38.02 μg/mL), at the extract of methanol, followed by chlorogenic acid (3225.10 ± 16.44). Enzyme inhibition activity against α-amylase and tyrosinase was also estimated. The ethyl acetate extract showed the highest α-amylase activity (1.95 ± 0.04 mg/mL), while the best anti-tyrosinase activity was calculated for the methanolic extract (1.25 ± 0.01 mg/mL). In addition, total phenolic, flavonoid content and antioxidant activity were evaluated. According to our results, bioactivity of the plant is of great interest, nonetheless, at the same time, it is strongly depended on the solvent used during the extraction process. Our data suggest that the plant under study may be an important source to consider against metabolic, skin pigmentation and oxidative stress related disorders.

Ocimum sanctum, commonly known as Tulasi in Nepal, is a pharmacologically important plant due to its active constituents. In this work, extraction was carried out in hexane, chloroform and methanol solvents and their phytochemical screening was performed. Total phenolic and flavonoid contents in the plant were measured by Folin-Ciocalteu colorimetric method and aluminum chloride colorimetric method respectively. Antioxidant activity of the extracts was evaluated using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay. Alkaloids, flavonoids, tannins, glycosides, polyphenols, terpenoids, tannins and steroids are mainly found in the extracts. Based on the result obtained, the plant posses a significant amount of total phenolic and total flavonoid content. Both phenolic and flavonoid contents were highest in methanol extract, followed by chloroform and hexane extract. Antioxidant activity of the extracts as ascorbic acid standard (IC50 value = 41.34 µg/mL) was in the order of methanol extract (IC50 value = 47.73 µg/mL) > chloroform extract (IC50 value = 79.46 µg/mL) > hexane extract (IC50 value = 94.68 µg/mL). The extent of the antioxidant activity of the plant is following the number of total phenolics and flavonoids present in it.

Objective: Corn cob is a part of plant Zea mays that have a phenolic and flavonoid compounds. This research aims to evaluate total phenolic and flavonoid content, free radical scavenging activity and tyrosinase inhibiton of corn cob (Zea mays) extract. Methods: Corn cob were determined in Poltekkes Kemenkes Surakarta. Meanwhile, corn cob were extracted using maceration method applying 70% ethanol solvent. This extract were analyzed through the total fenolic and flavonoids content tests apply spectrofotometric UV Vis, antioxidant activity using radical scavenging test 2,2-difenill-1-pikrilhidrazil (DPPH) and tyrosinase inhibition test applying in vitro tyrosinase enzyme inhibition. Results: Total phenolic content of the corn cob extract were 1.76 %b/b EAG, while the total of flavonoids content were 0.42%b/b EK. The antioxidant activity using DPPH method test of corn cob extract were values IC50 38.57 µg/ml. Tyrosinase inhibition of corn cob extract were values IC50 919.78µg/ml. Conclusion: Corn cob extract had antioxidant activity and tyrosinase inhibition.

  Objective: The aim of the present study is to determine the phytochemical screening, antioxidant activity and α-amylase inhibitory activity of the crude hexane, ethyl acetate and ethanolic stem bark extract of Pisonia grandis.Methods: The evaluation of antioxidant and antimicrobial activity, total phenolic, and flavonoid content were assessed using 2,2-diphenyl-1- picrylhydrazyl, Folin–Ciocalteu’s reagent, and aluminum chloride assay, respectively. The antidiabetic activity was assessed for porcine pancreatic α-amylase for the stem bark of P. grandis. Results: Phytochemical screening confirmed the presence of phenolic, flavonoids, tannins, saponins, terpenoids, and steroids in all the three extracts. The antioxidant activity showed 148.2 μg/ml, total phenolic content (gallic acid equivalent), 0.0665±0.0002 mg/g, flavonoid content (quercetin equivalent), 0.6061±0.1817 mg/g, and inhibitory concentration 50% values were found to be 40.42 μg/ml and showed better in ethyl acetate extract. The antidiabetic activity exhibited mimic action with insulin due to the presence of pinnatol in the stem bark and leaves of P. grandis. Conclusion: P. grandis stem bark crude ethyl acetate extract showed strong antioxidant activity, high phenolic, and flavonoid content. The antimicrobial activity was studied in both Gram-positive and Gram-negative strains against ampicillin and rifampicin as reference drugs. Antidiabetic activity shows effective result by α-amylase inhibitory activity.

Objective: The present study qualitative phytoconstituents examine the total phenol, total flavonoid content (TFC), and antioxidant efficiencies traditionally used plant, Meyna spinosa. Methods: Chemical profiling, estimation of total phenolic content (TPC), TFC, and antioxidant activity of ethanol extracts of M. spinosa have performed by applying standard protocols. Antioxidant activity of leaf and stem was evaluated by 1, 1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assay. TPC and TFC of the plant were assessed using Folin–Ciocalteu colorimetric and aluminum colorimetric assay, respectively. Results: The findings of the study exhibit that ethanol extract of M. spinosa is proved to be the presence of phytoconstituents (7/9) such as alkaloids, terpenoids, saponins, tannins, phytosterols, flavonoids, and phenolic compounds, while carbohydrate, fixed oils, and fats are unavailable. In addition, phenolic compositions of ethanol extract of leaf and stem; 93.21±2.93 and 54.33±0.69 mg gallic acid equivalents/g extract, respectively; TFCs of leaf and stem have recorded as 61.55±1.21 and 37.55±1.28 mg quercetin equivalents/g extract, respectively. Antioxidant efficiency of both leaf and stem is tested using DPPH radical scavenging assay as IC50 20.68±0.32 and 50.99±0.56 μg/ml, respectively. Conclusion: From the above results, it has concluded that the ethanol extract of the M. spinosa leaves and stems seizes rich phytoconstituents which can be applied in food technology, drug industries, ethnopharmacological fields, etc., for the development of healthiness and to battle against negative health consequences.

Background: Ocimum sanctum and Ocimum kilimandscharicum are cultivated in Indian subcontinent both for the religious and medicinal properties. Traditionally, the leaves have been reported for their enormous therapeutic potentials but the roots which are otherwise considered as a waste part have not been explored for their pharmacological activity. Methods: Total phenolic content, free radical scavenging and ferric reducing antioxidant potential of various extracts from Ocimum sanctum and Ocimum kilimandscharicum were assessed and compared. In vitro antioxidant activity was estimated spectrophotometrically and the most potent ethyl acetate extract was chemically characterized by developing the chemical fingerprint and quantifying the probable constituents with the help of HPLC/LC-MS techniques. Results: The ethyl acetate extract of both the species exhibited significant free radical scavenging potential and also reduced the ferric ions. It was observed that ethyl acetate extract have superior ferric reducing potential than other tested extracts, which were evidenced by high ferrous sulphate equivalent value of 77.05 ± 1.54 and 80.98 ± 0.80 at 100 µg/ml for O. sanctum and O. kilimandscharium respectively. The ferric reducing capacity of ethyl acetate extract for both the species was also evidenced by an elevated optical density of 1.64 ± 0.12 and 2.14 ± 0.08. Ocimum sanctum exhibited better antioxidant capacity (11.31 ± 0.20 AScE) as compared to Ocimum kilimandscharium (9.08 ± 0.27 AScE). The total phenolic and flavonoid content were estimated by spectrophotometric method and tentatively characterized by HPLC/LC-MS profiling which revealed the presence of rosmarinic acid, caffeic acid along with its derivatives such as caffeoyl-dihydroxyphenyllactoyl-tartaric acid. Conclusion: The ethyl acetate extract of both the species being rich in phenolic and flavonoid contents exhibited potent antioxidant activity. The presence of flavonoid in ethyl acetate extracts further co-relates the antioxidative properties of roots extracts.  To the best of our knowledge and understanding this is the first report of comparative chemical profiling by RP-HPLC/LC-MS and antioxidant potential of roots of two Ocimum species. Objectives: The objective of this study was to estimate the total phenolic and flavonoid contents and to explore the antioxidant potential of various leaf extracts (chloroform, ethyl acetate, and ethanol) of Canthium coromandelicum, which is considered traditionally as an important medicinal plant.Methods: Antioxidant properties of the extracts were assessed using 1, 1- diphenyl -2- picrylhydrazyl and hydrogen peroxide radical scavenging assays for ascorbic acid equivalents. The total phenolic and flavonoid contents were also investigated to determine their correlation with the antioxidant activity of the leaf extracts and expressed in Gallic acid and quercetin equivalents, respectively.Results: The results showed that the content of total phenols and flavonoids was found to be high in ethyl acetate extract which was recorded as 61.02±1.30 mg Gallic acid equivalent (GAE/g) and 81.72±0.61 QE/g, respectively. Compared to other extracts, ethyl acetate leaf extract was found to possess high antioxidant activity at p<0.05 level, with high percentage of inhibition at 100 μg/ml concentration (82.70%) toward hydrogen peroxide radical scavenging with IC50 value 62.94 μg/ml. Statistically, two-tailed Pearson’s correlation showed strong positive correlations between hydrogenperoxide radical scavenging activity and total phenolic contents (TPC) (r=1.000) at p<0.05 level.Conclusion: The results obtained in this study clearly signifies that the ethyl acetate leaf extract of C. coromandelicum has high content of total phenols which are correlated to its antioxidant activity and thus has the potential to use as a source of natural antioxidants and can be explored as a therapeutic agent in free radical induced diseases.Keywords: Canthium coromandelicum, In-vitro antioxidant activity, Ethyl acetate extract, Pearson’s correlation, 1, 1- diphenyl -2- picrylhydrazyl, Hydrogen peroxide, Nitric oxide, Superoxide.

Dracocephalum moldavica L. has been used as both treatment of coronary heart disorders and food supplements. In the present study, determination of total phenolic and flavonoid content in methanol, ethanol and methanol/ethanol extracts of leaf was studied in Iranian Moldavian balm for the first time. Antioxidant activity of the studied plant was also measured. The methanol extract had the highest phenolic and flavonoid content, anthocyanin, DPPH and H2O2 radical scavenging activity. The ethanol extract showed the least amount of all. The methanol/ethanol extract showed the highest amount in two oxides including nitric and superoxide radical scavenging activities; it also showed the highest Ferric Reducing Ability Power. The obtained chromatograms of the plant using High Performance Liquid Chromatography showed that the highest and the lowest found phenolic compounds were caffeic acid and vanilic acid, respectively. The results show that this plant is a suitable natural antioxidant to reduce the oxidative stress in human being.
