<p class="Abstract">The present study was aimed to evaluate the methanolic extract and subsequent solvents soluble fractions of <em>Sterculia </em>diversifolia bark for cytotoxic, anti-cancer and immunomodulatory activities. Phytochemical investigation confirmed the presence of alkaloids, flavonoids, saponins, glycosides, etc. In the cytotoxic activity, n-hexane showed potent activity (LD<sub>50</sub>: 7.0 µg/mL) followed by dichloromethane fraction (LD<sub>50</sub>: 16.2 µg/mL). In the anti-cancer activity, dichloromethane fraction showed potent activity (IC<sub>50</sub>: 5.9 µg/mL) followed by ethyl acetate fraction (IC<sub>50</sub>: 9.5 µg/mL). While in the immunomodulatory assay, ethyl acetate fraction showed a very significant activity (IC<sub>50</sub>: 21.0 µg/mL) followed by dicloromethane and n-butanol fractions (IC<sub>50</sub>: 25.0 and 25.3 µg/mL respectively). Hence, it is clear that <em>S. </em>diversifolia has anti-cancer and immunomodulatory agents.</p>

Muscadine grape is rich in stilbenes, which include resveratrol, piceid, viniferin, pterostilbene, etc. Resveratrol has been extensively studied for its biological activities; however, the synergistic effect of stilbene compounds in berry extracts is poorly understood. The aim of this study was to evaluate the anti-cancer activity of stilbene-rich muscadine berry extract and pure resveratrol. Stilbenes were extracted from ripened berries of muscadine grape cultivars, Pineapple, and Southern Home. HPLC analysis was performed to determine quantity of stilbenes. The extracts were tested for their cytotoxic activity against A549 (lung carcinoma cells), triple negative breast cancer (HCC-1806) and HepG2 (human liver cancer) cells. The stilbene-rich extracts of the muscadine berry extracts showed cytotoxic activity against all of the cells tested. The extracts at 1 μg/mL induced death in 50–80% of cells by 72 h of treatment. About 50 μg/mL of resveratrol was required to induce a similar response in the cells. Further, modulation of genes involved in tumor progression and suppression was significantly (p < 0.0005) higher with the HepG2 cells treated with stilbene-rich berry extracts than the pure resveratrol. This shows that the synergistic activity of stilbenes present in muscadine grape berries have more potent anti-cancer activity than the resveratrol alone.

Phytochemical investigation of the ethyl acetate extract of the stem of Dendrosicyos socotrana Balf. f. resulted in the isolation of a new pentacyclic cucurbitane glycoside Socotroside, in addition to the three known cucurbitacins, dihydrocucurbitacin D, dihydrocucurbitacin F and cucurbitacin G. The structures of the isolated compounds were established on the basis of their spectral data. The isolated cucurbitacin aglycones showed marked cytotoxic activity.

AbstractPeople of north-eastern states of India consume raw areca-nut (RAN) and lime which could lead to oral, esophageal and gastric cancers. However, the incidence of these cancers are significantly lesser in those who consume pieces of Potentilla fulgens root along with RAN. Since evaluation of anticancer role, if any, of P. fulgens on RAN-mediated genetic alterations in human is difficult because of other compounding factors, this study was undertaken in mice to focus on gastric carcinogenesis since ad libitum administration of RAN extract with lime in drinking water induced stomach cancer due to greater exposure of its lining. A total of 160 mice were used at different time points and either methanol extract of P. fulgens roots (PRE) or mixture of four compounds of ethyl-acetate fraction (EA-mixture) was mixed with mice feed. Histological studies revealed that RAN + lime induced cancer in all the mice and interestingly only 20% developed cancer when PRE/EA-mixture was provided along with RAN + lime. Higher frequency of precocious anaphase and over expression of p53 and Securin genes were significantly reduced by PRE/EA-mixture. Thus PRE/EA-mixture mitigates the RAN-induced tumor-initiating process in stomach by maintaining expression of tumor suppressor and check-point genes under control.

A phytochemical investigation of an ethyl acetate fraction of Mahonia bealei trunks led to the isolation of three new and four known compounds. By means of NMR and MS spectroscopic methods, the structure of the new compounds was elucidated as bishexadecyl epiphyllate (1), 3-(4-hydroxy-3,5-dimethoxyphenyl)-3-ethoxy-2-sulfopropane-1-ol (2), and 6α-acetoxystigmastan-3β-ol (3).

Calligonum comosum (Polygonaceae), an Egyptian desert plant, was extracted and fractionated using petroleum ether, methylene chloride, and ethyl acetate. The total methanolic extract and other fractions were tested for their anticancer activity using Ehrlich ascites, brine shrimp and antioxidant assays. Ethyl acetate fraction proved to be the most active in all assays. Eight compounds were isolated, purified, and identified from this fraction as (+)- catechin (1), dehydrodicatechin A (2), kaempferol-3-O-rhamnopyranoside (3), quercitrin (quercetin-3-O-rhamnopyranoside) (4), β-sitosterol-3-O-glucoside (5), isoquercitrin (quercetin- 3-O-glucopyranoside) (6), kaempferol-3-O-glucuronide (7), and mequilianin (quercetin-3- O-glucuronide) (8). All isolated compounds were tested for their cytotoxicity and antioxidant activity. Compound 2 showed the best cytotoxic and antioxidant activity.

The genusOcotea(Lauraceae) is distributed mainly in tropical and subtropical regions. Some species of this genus asO. puberulaandO. quixoshave been described in the literature, showing antibacterial activity. AndOcotea macrophyllashowed anti-inflammatory activity with inhibition of COX-1, COX-2, and LOX-5. The purpose of this study was the phytochemical investigation of the plant speciesOcotea notatafrom Restinga Jurubatiba National Park, Macaé, RJ, Brazil, and the search for antimycobacterial fractions and compounds. The crude extract was evaluated for antimycobacterial activity and presented95.75±2.53% of growth inhibition at 100 µg/mL. Then, it was subjected to a liquid-liquid partition and subsequently was chemically investigated by HPLC, revealing the major presence of flavonoids. In this process the partition fractions hexane, ethyl acetate, and butanol are shown to be promising in the antimycobacterial assay. In addition, ethyl acetate fraction was chromatographed and afforded two flavonoids identified by MS and NMR as afzelin and isoquercitrin. The isolated flavonoids afzelin and isoquercitrin were evaluated for their antimycobacterial activity and for their ability to inhibit NO production by macrophages stimulated by LPS; both flavonoids isoquercitrin (Acet22) and afzelin (Acet32) were able to inhibit the production of NO by macrophages. The calculated IC50of Acet22 and Acet32 was 1.03 and 0.85 µg/mL, respectively.
