The experiment was conducted in a well-known private livestock research and development farm in Bangladesh to find out the variation of semen quality parameter among three generations (F1, F2 and F3) of Holstein Friesian upgraded breeding bulls. A total of 312 ejaculates were collected from 6 upgraded breeding bullsthrought the experimental year. The recorded data were summarized using Microsoft Excel 2010 and analyzed using GraphPad Prism 5 softwere. Out of the 312 ejaculates, 273 (87.50%) were found to be creamy in color followed by 26 (8.33%) and 13 (4.17%) as yellowish and watery, respectively. Generation had significant (P<0.05) effect on ejaculate volume, consistancy, mass activity, sperm concentration, initial and post freezing motility. The highest (7.389±0.19ml) and the lowest (5.156±0.13ml) volume of semen were found in third (F3) and first generation (F1), respectively. The mass activity ranges from 3.74±0.04 to 4.30±0.05. Sperm concentration and pH varied insignificantly (p>0.05) but initial motility and post freezing motility had the significant differences among the three generations. Initial motility ranges from 75.87±0.32 to 78.40±0.38 percent and the post freezing motility ranges from 50.38±0.41 to 52.16±0.43 percent. It could be concluded that most of the semen quality parameters were influenced by generation and freezing. The present study was carried out on 80 ejaculates, 20 each from four healthy Jaffrabadi breeding bulls to see the relationship betweensexual behavior and semen quality parameters. The results depicted that the overall mean values of libido score, mating ability score andsexual libido score between bulls. The overall color of semen ranged from thin white to very thick creamy. The overall values for semen volume(ml), pH, sperm concentration (million/mL), mass activity, individual sperm motility (%) and live spermatozoa (%) were 2.99 ± 0.12, 6.80 ± 0.01, 1439.68 ± 48.46, 3.94 ± 0.03, 89.51 ± 0.21 and 95.00 ± 0.19, respectively. The values for semen volume, sperm concentration,mass activity (%) and live sperm (%) differed significantly (p less than 0.05) between bulls. There were significant positive correlations (p LESS THAN 0.01)of libido with mating ability, sexual behavior score and ejaculate volume of semen (r = 0.37 to 0.87), while a negative correlation wasfound with sperm concentration (-0.30). Mating ability was significantly (p less than 0.01) correlated with sexual behavior score and ejaculatevolume (0.78, 0.67), and sexual behavior was correlated with semen volume (0.84). Sperm concentration was significantly (p less than 0.05 correlated with mass activity (0.29), while individual sperm motility was positively (p less than 0.05) correlated with live sperm percentage (0.25).Correlations of libido, reflected the importance of sexual behavior of Jaffrabadi bulls in predicting their future utility as a proficient breeder under AI program.

Semen was collected from four rams, using artificial vagina and viability%, motility% and plasma membrane integrity% were measured. Fresh ejaculates (n = 32) were separated by modified swim-up separation using modified human tubal fluid medium. Four fractions of supernatant were collected at 15-minute intervals. The mean volume, mass activity, concentration, motility%, viability%, normal morphology and membrane integrity% (HOST +ve) of fresh semen were 1.0 ± 0.14, 4.1 ± 0.1 × 109 spermatozoa/ml, 85.0 ± 1.3, 89.4 ± 1.0, 85.5 ± 0.7, 84.7 ± 0.5 respectively. There was no significant (P>0.05) difference in fresh semen quality parameters between rams. The motility%, viability% and HOST +ve % of first, second, third and fourth fractions were 53.4 ± 0.5, 68.2 ± 0.3, 74.8 ± 0.3 and 65.5 ± 0.4; 55.5 ± 0.4, 66.2 ± 0.4, 74.5 ± 0.3 and 73.6 ± 0.3 and 66.7 ± 0.5, 66.8 ± 0.5, 65.2 ± 0.4 and 74.7 ± 0.5 respectively. The motility%, viability% and membrane integrity% of separated semen samples differed significantly (P<0.05) between four fractions. The mean motility% and viability% were significantly higher (P<0.05) in third fraction (74.8 ± 0.3%), whereas the mean HOST +ve% was significantly higher (P<0.05) in fourth fraction (74.7 ± 0.5). All quality parameters of separated spermatozoa were significantly (P<0.05) lower than that of fresh semen. Objective: To determine the effect of age and frequency of collection on quality parameters of Jersey bull semen at National Livestock Breeding Center (NLBC), Pokhara. Materials and Methodology: Nine Jersey bulls were selected randomly from NLBC. To obtain the effect of age, six bulls of three different age interval (3-4, 5-7 and 8-9 years) were selected randomly. Remaining three bulls were used to determine the effect of frequency of collection on semen quality. Studies were conducted for three months (Nov/Dec to Dec/Jan, 2016). Semen quality was analyzed using CASA system and SMILE software. ANOVA and paired t- tests were applied to find out level of significance. All the statistical analyses were done using SPSS 17.0 Results: Sperm concentration, post- thaw motility, live sperm, and abnormal sperm were significantly affected by the age of the bulls. Semen quality was found superior in the bulls belonging to 5-7 years of age intervals. Significantly higher value for semen volume, sperm concentration and post- thaw motility was obtained on second collection (P<0.05). However, volume initial motility, pre-filling motility, live sperm and abnormal sperm percentage did not differ significantly among different age intervals and between the frequencies of collection. Conclusion: Different quality parameters of semen like volume, concentration, initial motility, pre-filling motility, post thaw motility, live and normal spermatozoa percentages were higher in 5-7 years old bulls and in second collection of semen. Sperm concentration (0.551 ±0.004 to 1.084 ± 0.014x109), post- thaw motility (42.44±0.11 to 51.72±0.03 %), live sperm (45.62 ± 0.13 to 51.23±0.15 %), and abnormal sperm (11.57±0.12 to 19.59±0.24%) were significantly affected by the age of the bulls. Semen quality was found superior in the bulls belonging to 5-7 years of age intervals. On the other hand, volume (3.98±0.09 to 6.72±0.06 ml), initial motility (72.68±0.02 to 78.64±0.15 %) and pre-filling motility (71.34±0.17 to 77.62±0.02 %) did not differ significantly among different age intervals. Significantly, higher value for semen volume (3.98±0.12 to 6.02±0.14 ml), sperm concentration (0.572±0.119 to 1.095±0.015) and post- thaw motility (40.34±0.21 to 48.54±0.06% was obtained on second collection. However, initial motility (70.58±0.06 to 75.64±0.13 %), prefilling motility (68.11±0.12 to 71.62±0.02 %), live sperm (53.27±0.54 to 56.17±0.02 %) and abnormal sperm (12.31±0.12 to 14.3±0.47 %) did not differ significantly between the frequencies of collection. Different quality parameters of semen like volume, concentration, initial motility, pre-filling motility, post thaw motility, live and normal spermatozoa percentage were higher in 5-7 years old bulls and in second collection of semen.

The present study was conducted to characterize the semen quality parameters in roosters of three genetic groups viz. Rhode Island Red (RIR), Punjab Red and RIR X local cross. The roosters for each genetic group were also evaluated for fertility rate. There was no significant difference in color, pH and abnormalities among different genetic groups. RIR X local cross had significant higher (p less than 0.01) ejaculate volume (0.51±0.02 ml vs. 0.27±0.01 and 0.30±0.01ml), sperm concentration (2804.70±114.61 vs. 1826.10±77.66 and 2133.50±81.18X106 sperms/ml), sperm motility % (66.60±2.12 vs. 55.38±1.82 and 51.95±1.84), sperm viability % (72.43±2.06 vs. 64.67±1.88 and 51.25±1.89) and HOST % (64.59±2.61 vs. 58.55±1.17 and 47.09±2.11) than RIR and Punjab Red respectively. Interestingly, RIR X local cross had significant lower (p less than 0.01) partially damaged sperm % (36.96±2.81 vs. 41.84±1.78 and 48.98±1.89) and fully damaged sperm % (14.15±3.39 vs. 26.27±1.93 and 31.25±2.46) than RIR and Punjab Red respectively indicating desirable performance. The fertility rate for RIR X local cross was also found to be higher than Punjab Red and RIR respectively. Sperm traits were significantly positively correlated among groups with high correlation between fertility rate and sperm traits. It can be concluded that the selection of roosters on the basis of sperm function tests for artificial insemination may improve fertility rate and thereby may reduce maintenance cost of large number of cocks.

Repeatability for seminal traits and their phenotypic relationships with testes measurements and performance traits in Black Bengal bucks were estimated from a total of 116 repeated observations on 15 young Black Bengal bucks. Performance traits included age, body condition and body weight of bucks. Testes measurements were testes length, breadth and volume, and scrotal circumference, while seminal traits were ejaculate volume, semen density, mass activity, sperm motility, sperm concentration, total sperm per ejaculation and percent of normal sperm. High positive correlations (ranging from 0.81 to 0.90) were found between body weight and testes measurements. Semen volume and percent of normal sperm were positively correlated with age, body condition and body weight of bucks (ranging from 0.24 to 0.60). The testes measurements were strongly and positively correlated with semen volume and total sperm per ejaculation (ranging from 0.53 to 0.61), while the correlations between testes measurement and percent of normal sperm were moderate (ranging from 0.34 to 0.44). Among seminal traits, strong correlations were found between semen volume and total sperm per ejaculation (0.81) and, between mass activity and sperm motility (0.82). Repeatability of ejaculate volume was higher (r = 0.78) which indicated that selection or culling for semen ejaculate volume could be practiced from single or few observations. It could be concluded that bucks’ age, body weight and body condition along with testes volume should be considered as selection criteria for improving semen quality and semen production of breeding bucks. Abstract. Horses are seasonal breeders with a natural breeding season beginning in spring and extending through midsummer. In this study, quantitative and qualitative parameters of chilled stallion semen were compared between fertile and subfertile stallions and between the breeding and the non-breeding season. Semen quality parameters compared included ejaculate volume, sperm concentration, total sperm number, sperm morphology, and computer-assisted semen analysis (CASA)-derived sperm movement characteristics obtained from two groups of warmblood stallions (n=8; four fertile stallions and four subfertile stallions), which differ in the seasonal pregnancy rate 80 %–90 % (fertile) vs. 40 %–60 % (subfertile). A total of 64 ejaculates were collected from the stallions (n=8; four in the breeding season and four in the non-breeding season of each stallion). No significant differences in the semen quality parameters between the fertile and the subfertile stallions in the non-breeding season were observed. However, in the breeding season the proportion of morphologically normal sperm, total motility, progressive motility, average path velocity (VAP), and curvilinear velocity (VCL) were significantly higher in the fertile group (P<0.05) when compared with the subfertile group. In addition, a significant seasonal variation in the proportion of morphological normal sperm was found in the fertile group between the breeding and the non-breeding season (P<0.05). Moreover, significant seasonal variations (P<0.05) in CASA parameters of mean VAP, straight line velocity (VSL), and beat-cross frequency (BCF) were observed in the fertile and the subfertile stallions, which tended to be lower in the non-breeding season. In conclusion, differences between the fertile and the subfertile stallions were observed only in the breeding season, and a few of CASA-derived parameters seemed to be significantly lower during the non-breeding season in both the fertile and the subfertile stallions.

This study is designed to evaluate the effect of using parsley seeds (PAS) at different levels as a natural on semen quality and reduce the effect of lipidperoxidation of bucks. Forty New Zealand rabbit bucks 8 month old were divided into four groups (each 10 rabbit bucks). The groups were divided as follows: Control diet (free from PAS), T1: Control diet + 5g/kg PAS, T2: Control diet + 10g/kg PAS; T3: Control diet + 15g/kg PAS. Semen sample were collected twice a week; from each rabbit buck by artificial vagina. The first semen collection was used to evaluate semen quality parameters. However, the second semen collection was used for analysis semen plasma. Results revealed that feeding diets containing different levels of PAS significantly (P<0.05) increased semen volume, mass activity, individual motility, PSV, sperm concentration, total sperm and seminal plasma activity of ALP enzyme and significant (P<0.05) decrease in dead sperm and seminal plasma concentrations of total lipids, triglycerides, cholesterol and MDA and seminal plasma activities of AST and ALT enzymes and TAC as compared with the control group. In conclusion, dietary supplementation with different levels of PAS, especially at the level of 15 g/kg (T3) resulted in significant improvement in semen quality, antioxidant status and reproductive traits of rabbit bucks.

SUMMARYOn the College of Agriculture farm in Abu-Ghraib 147 ejaculates were collected from three docked and four normal Awassi rams born during November 1962. The work covered a period from 1 April 1967 to 31 March 1968.Nine semen characters, namely, volume, mass activity, individual motility, sperm concentration, sperm number, pH, methylene blue reduction time and the percentages of abnormal and dead sperms, were studied. The effect of season and docking on these characters was investigated.Seasonal variation was observed in all traits studied. On the whole, semen quality was poorest during winter and best during summer. The effect of docking was more pronounced on sperm concentration, sperm number and percentage of abnormal sperms. Docking appeared to increase sperm production and reduce sperm abnormality.

Four bulls of mixed breeds (Sahiwal × Zebu, Sindhi × Zebu, Jersey × Zebu and Holstein-Friesian × Zebu )were examined physically and their semen was evaluated in three seasons: I (Rainy): May - November, II (Winter): December - February, III (Summer): March- April. The semen volume, sperm motility, sperm concentration and percentage of dead sperm ranged from 4.1 to 7.6 ml, 56.6 to 76%, 1010.3 to 1290.7 × 106/ml and 18.4 to 24.7%, respectively. All these attributes varied significantly (p<0.05) between breeds and seasons. The variations of semen quality between breeds were not significant (p>0.05).
