A cultivo da banana ‘Pacovan’ ainda é realizada operando com métodos tradicionais no estado do Amazonas, onde ainda existem registros de produção da fruta com doenças causadas por fungos e por bactérias. Porém, outros métodos de propagação, como a micropropagação in vitro, podem ser desenvolvidos e aperfeiçoados, para que ocorra a elevação da taxa de multiplicação em curto espaço de tempo além de aperfeiçoar, significantemente, a taxa de qualidade da produção de mudas. Entretanto, os índices de contaminação ainda são um dos maiores problemas desta técnica. Este trabalho teve por objetivo avaliar o melhoramento no processo de assepsia analisando diferentes tipos de concentrações de álcool, hipoclorito de sódio e Tween 20 e diferentes variações de tempo, durante o processo de assepsia do explante meristemático, visando identificar o processo mais eficiente de descontaminação. O experimento foi planejado em delineamento experimental inteiramente casualizado para a análise de regressão linear, onde foram constituídos por três tratamentos e vinte repetições, sendo cada repetição representada por cinco explantes em diferentes experimentos. Os dados foram submetidos à análise de variância (ANOVA), seguido de testes de média Tukey 5% de probabilidade. Os resultados permitiram concluir que, com elevado tempo do explante submerso no hipoclorito de sódio há eficácia contra fungos e bactérias. Porém, elevadas concentrações de , altas concentrações se tornam tóxicas aos explantes. Nos experimentos, o menor índice de contaminação dos tratamentos testados foi o E3, permitindo o desenvolvimento normal dos explantes e possibilitando que seja utilizado para o controle de contaminações na micropropagação de banana ‘Pacovan’ in vitro.

Sterilization procedure was standardized for grand naine cultivar of banana using various sterilants in combinations and alone. The observations were recorded regularly with respect to presence of fungal, bacteria as well as percentages of culture establishment. Results indicated that a treatment combination No. 3 [Lactic acid (0.15 %) + Tween-20 (0.1 %) + 0.8 % Commercial bleach (30 Min.) followed by Sodium chlorite (0.3 %) (20 min)] gave the highest percentage of aseptic culture establishment in vitro condition. The present study also showed that, single step aseptic inoculation was unable to control endophytic contaminants while sequential treatments were good enough to reduce microbial load as well as increase culture survival. AbstractTwenty-five banana varieties of section Eumusa (AA-group) and seven of the section Australimusa (Fe'i-group) from Papua New Guinea were evaluated for resistance to Radopholus similis, Pratylenchus coffeae and Meloidogyne spp. The host plant responses were compared with the susceptible reference cvs Grande Naine and Cavendish 901. In vitro propagated plants were transferred to the glasshouse in loamy sand and inoculated with approximately 1000 migratory endoparasitic nematodes at 4 weeks after planting. Reproduction of R. similis and P.coffeae in the roots was determined at 8 or 10 weeks, respectively, after inoculation. Reproduction of Meloidogyne spp. was determined 8 weeks after inoculation with 3300 to 5000 eggs. No resistance to R. similis was found in the diploid varieties. The Fe'i variety Rimina and possibly Menei were resistant to R. similis. All varieties tested were susceptible to P.coffeae and Meloidogyne spp. Tests de résistance de bananiers Eumusa et Australimusa (Musa spp.) envers les nématodes endoparasites migrateurs et galligènes - Vingt-cinq variétés de bananier de la section Eumusa (groupe AA) et sept de la section Australimusa (group Fe'i) provenant de Papouasie-Nouvelle Guinée ont été testées pour leur résistance envers Radopholus similis, Pratylenchus coffeae et Meloidogyne spp. Les résponses de ces variétés ont été comparées à celles des cultivars sensibles de référence Grande Naine et Cavendish 901. Des vitroplants ont été mis en place en serre sur un sol argilo-sableux et inoculés 4 semaines après plantation avec environ 1000 R. similis ou P.coffeae dont la reproduction a été déterminée 8 et 10 semaines, respectivement, après inoculation. La reproduction de Meloidogyne spp. l'a été 8 semaines après inoculation avec 3300 à 5000 oeufs. Aucune résistance à R. similis n'a été observée chez les variétés diploïdes. Les variétés du groupe Fe'i Rimina et Menei se sont montrées résistantes à R. similis, avec un certain doute dans le cas de la dernière. Toutes les variétés testées sont sensibles à P.coffeae et Meloidogyne spp.

Chitosan nanogel-coated polycaprolactone (PCL) fiber mat-based implant prototypes with tailored release of bone morphogenic protein 2 (BMP-2) are a promising approach to achieve implant-mediated bone regeneration. In order to ensure reliable in vitro release results, the robustness of a commercially available ELISA for E. coli-derived BMP-2 and the parallel determination of BMP-2 recovery using a quantitative biological activity assay were investigated within a common release setup, with special reference to solubility and matrix effects. Without bovine serum albumin and Tween 20 as solubilizing additives to release media buffed at physiological pH, BMP‑2 recoveries after release were notably reduced. In contrast, the addition of chitosan to release samples caused an excessive recovery. A possible explanation for these effects is the reversible aggregation tendency of BMP-2, which might be influenced by an interaction with chitosan. The interfering effects highlighted in this study are of great importance for bio-assay-based BMP-2 quantification, especially in the context of pharmaceutical release experiments.

The sugarcane aphid, Melanaphis sacchari Zehntner (Hemiptera: Aphididae), is the main pest of sorghum, Sorghum bicolor L. Moench (Poaceae), in Mexico. To control this insect, farmers currently use synthetic chemical insecticides, which are toxic to humans and biodiversity. However, natural products are a promising potential source of safer alternative means to control different agricultural pests. The main objective of this study was to evaluate the insecticidal effect of contact by fumigation of pure molecules of four commercial fatty acids (palmitic, stearic, pentadecanoic and linoleic acids), the phytosterol ß -sitosterol, and the flavonoid rutin. The results showed that fatty acids were the most effective against M. sacchari ; the highest mortality rate (85%) was produced by linoleic acid and the LC 50 was 1,181 ppm, followed by stearic and palmitic acids with mortality percentages of 74 and 63%, respectively, at a concentration of 2,500 ppm at 72 h. The positive control, imidacloprid, had 100% mortality in 24 h and the tween 20 negative control exhibited 4% mortality in 72 h. Our results show that commercial fatty acids are effective against adults of M. sacchari , and can be considered an environmentally friendly alternative to the frequent use of synthetic chemical insecticides.

With a previous observation of Escherichia coli growth cessation along with temperature variation within three different bacteriological culture media (nutrient agar, Luria-Bertani agar and minimal agar), current investigation further depicted on the possible growth dynamics of Escherichia coli (SUBE01) and Salmonella (SUBS01) growth and viability upon supplementation of different carbon sources (dextrose, sucrose, lactose, glycerol and tween 20) at 37°C under the aeration of 100 rpm. Viability of the tested bacterial species was assessed through the enumeration of the colony forming unit (cfu) appeared upon prescribed incubation for 12-24 hours on different agar plates consisting of the above mentioned carbon sources. Besides, to inspect the cellular phenotypic changes, morphological observations were conducted under the light microscope. Variations in bacterial growth (either growth acceleration or cessation) were further noticed through the spot tests on the agar plates. Considerable shortfalls in the culturable cells of E. coli and Salmonella spp. were noted in the minimal media separately consisting of sucrose, lactose, glycerol or tween 20 while an opposite impact of accelerated growth was noticed in the media supplied with dextrose. The data revealed a hierarchy of consequence of carbon sources as nutrient generators whereby the favourable bacterial growth and survival order of the carbon sources was estimated as dextrose > glycerol > lactose > tween 20 > sucrose.Bangladesh J Microbiol, Volume 32, Number 1-2,June-Dec 2015, pp 39-44

No intuito de elevar as taxas de sobrevivência durante a etapa de aclimatização e posterior plantio a campo, avaliou-se o enraizamento in vitro de bananeira cv. Pacovan, em diferentes concentrações de sais MS e de sacarose. Utilizou-se DIC, esquema fatorial (6x2x3), com seis meios de cultura [sendo três concentrações de nutrientes do meio MS (100%; 50% de macronutrientes; e 50% dos sais macro e micronutrientes), e duas concentrações de sacarose (1,5/3,0%)], dois fotoperíodos (12/16 h) e três tempos de cultivo (21, 28 ou 35 dias) e seis repetições/tratamento. Analisaram-se: altura da planta, número de folhas/planta, massas frescas e secas das partes aérea e radicular. Para altura da planta, massa fresca da parte aérea e radicular, o meio MS 50% dos sais + sacarose (1,5%) com fotoperíodo de 16 h e tempo de cultivo de 35 dias foi satisfatório. Para massa seca da parte aérea foi MS 50% de sais + sacarose (3%), e para massa seca da parte radicular, MS 100% + sacarose (3%) (em 12hs/28 dias e 16hs/21 dias). Para o alongamento/enraizamento in vitro da bananeira cv. Pacovan sugere-se MS 50% de sais (macro e micronutrientes), redução ou manutenção de sacarose (1,5 ou 3%) em 16h/35 dias de cultivo.Palavra-chave: Musa spp., propagação in vitro, sistema radicular. CHANGES IN CULTURE MEDIUM, PHOTOPERIOD AND TIME OF CULTIVATION AFFECT THE IN VITRO ELONGATION AND ROOTING OF BANANA CV. PACOVAN ABSTRACT:In order to achieve high rates of survival during the acclimatization and later planting in the field, was evaluated the in vitro of banana cv. Pacovan plants under different concentrations of sucrose and MS basal salt mixture. The experiment was assembled in a DIC, in 6x2x3, six different culture media [three different MS salt mixture concentrations (100%; 50% of macronutrients; and 50% of macro/micronutrients) and two sucrose concentrations (1.5/3%)], two photoperiods (12/16 hours) and three cultivation times (21, 28 or 35 days). Each treatment was composed by 6 replicates. Plant height, number of leaves/plant, fresh and dry weight of roots and shoots, were analyzed. Satisfactory results for plant height and shoot and root fresh biomass were observed in MS with macro/micronutrients (50%) + sucrose (3%), 16 hours/35 days. The highest values of shoot dry weight were observed in MS with macro/micronutrients (50%) + sucrose (3%); the highest root dry weight was achieved with MS 100% + sucrose (3%) (12hs/28 and 16hs/21 days). The suggested medium for the in vitro elongation and rooting stage of banana cv. Pacovan is the MS with 50% of salts (macro and micronutrients), reduction or maintenance of sucrose (1.5 or 3%) in 16h/35 days of cultivation.Keywords: Musa spp., in vitro propagation, root system. DOI:

A research  to study the sterilization   method  and application   of Kinetin  and IAA to induce the Durian  young  leaf (Durio zibethinus) in MS  medium   was conducted in Balai Benih Induk Hortikultura in Salaman  Magelang  district  of Central  Java  started  on September  until December 2003. The Laboratory experiment   was arranged  in two phases,  which were  the optimation  phase of sterilization   and  induction   phase.  At  the  first  phase,  the  sterilization method  used  was  the modification   of Mulya  (2001) method.  The modification   use of sterilant,  vitamin  C antioxidant, Alcohol  70 %, Benlate, Agrept,  Tween-20  and Betadine  were done to obtain  effectiveness   of the sterilization.  Explants  planted  then in MS medium  for two weeks. Contamination   time, percentage of contamination   and viabilitas  (percentage of living explants)  were observed  then.  At the second phase,  the treatments were arranged  in a 3 x 3 factorial  completely   randomized   design  (CRD)  to observed  the influence  of Kinetin  and IAA combination.   The concentration   of Kinetin  observed were 2, 4, and 6 mg/I, where  as the IAA concentration   were 0.5,  1.0, and  1.5 mg/I. All treatments were  repeated  three  times,  with three samples  on each  replication.   The percentage   of browning explants, percentage  of contaminated   explants,  site of  contamination   and percentage of explants live were observed  at the end of incubation. The results  showed that sterilization  of Durian young leaves explants  with 1  g/l deterjent  for 15 minutes  then by 2 g/l Benlate  and Agrept  for 10 minutes,  then by 1  g/200 mg Vitamin C, then by Alcohol  70 % for 1  minute, then by 20% Clorox,  then by 2 drip of Tween-20  for 10 minute and then by Betadine  decreased  the contamination down to 50 %, and this kind of sterilization  was relatively better than  the other  kinds.  Application   of growth  regulators   were  not  able  to induce  explants growth,  but stimulated  callus formation  at the cutting surface though,  in the application  of Kinetin 4 mg/1 + IAA 0,5 mg/I, Kinetin 4 mg/1 + IAA  1,5 mg/1, Kinetin  6 mg/I+  IAA 0,5  mg/1 and Kinetin 6 mg/l+IAA   1,0 mg/I.

<p><em>Geranium chilloense </em>Willd. Ex Kunth es conocida con el nombre de geranio de los chillos, es una planta ornamental, nativa de Los Andes que se la encuentra de manera silvestre en las quebradas del Distrito Metropolitano de Quito y forma parte de la historia de la flora nativa de Quito ya que ha sido descrita desde la expedición de Alexander Humboldt y Aimé Bonpland en 1802. </p><p> </p><p>El presente estudio presenta un protocolo de propagación <em>in vitro</em> de <em>Geranium chilloense</em> Willd. ex Kunth, para la obtención de plantas completas. Para la germinación <em>in </em>vitro, se evaluaron dos escarificaciones, tres protocolos de desinfección y tres concentraciones de sales MS. La escarificación recomendada consiste en tres días de hidratación, seguida de una inmersión de dos horas en una dilución de ácido acético (10%), ya que permitió obtener un porcentaje de germinación del 31%. Para la desinfección se seleccionó el protocolo P3 con etanol al 70% (1 min), hipoclorito de sodio al 20% v/v (1,2 p.a) más tween (20 min), al obtener baja contaminación fúngica (3%). Mientras que, para la introducción al medio de cultivo, se recomiendan las sales MS reducidas al 25% de su concentración, ya que resultó en mayor porcentaje de germinación (41%). Para la multiplicación se evaluaron concentraciones e interacciones de reguladores de crecimiento, se recomienda el uso del tratamiento T2 (AIA 0,5 ppm), ya que logró el mayor número de brotes por explante (3,88). Finalmente, en la fase adaptación al sustrato se evaluó turba y fiobra de coco, resultando la turba estéril el sustrato que presentó los mayores porcentajes para la variable prendimiento (81%).</p>
