Caulerpa racemosa adalah salah satu spesies alga hijau yang dapat dimanfaatkan sebagai bahan baku krim sup. Nilai tambah krim sup tersebut adalah adanya kandungan klorofil sebagai senyawa bioaktif dan sebagai pewarna alami. Permasalahn yang terjadi adalah klorofil tidak tahan terhadap panas pada saat pengolahan. Vacuum drying oven menjadi solusi untuk mengurangi kerusakan klorofil akibat pemanasan. Tujuan penelitian ini adalah mengetahui pengaruh suhu terhadap kandungan klorofil pada proses pengeringan krim sup yang terbuat dari Caulerpa racemosa dengan menggunakan vacuum drying oven. Tahapan penelitian ini meliputi, pembuatan krim sup, pengeringan, ekstraksi klorofil, dan analisis klorofil. Hasil penelitian ini diperoleh empat variasi suhu yakni 60, 70,80, dan 90ºC. Kandungan klorofil paling tinggi pada pengeringan dengan suhu 60ºC, dengan komposisi klorofil a (2,0786 µg/mL), klorofil b (1,8742 µg/mL), dan total klorofil (3,9528 µg/mL). Paparan panas menyebabkan klorofil menjadi tidak stabil. Berdasarkan data yang diperoleh dapat disimpulkan suhu optimal dalam mengeringkan krim sup dengan vacuum drying oven adalah 60ºC.

Direct electron microscopic study of biological molecules has been hampered by such factors as radiation damage, lack of contrast and vacuum drying. In certain cases, however, the difficulties may be overcome by using redundent structural information from repeating units and by various specimen preservation methods. With bilayers of phospholipids in which both the solid and fluid phases co-exist, the ordering of the hydrocarbon chains may be utilized to form diffraction contrast images. Domains of different molecular packings may be recgnizable by placing properly chosen filters in the diffraction plane. These domains would correspond to those observed by freeze fracture, if certain distinctive undulating patterns are associated with certain molecular packing, as suggested by X-ray diffraction studies. By using an environmental stage, we were able to directly observe these domains in bilayers of mixed phospholipids at various temperatures at which their phases change from misible to inmissible states.

Short lifetime or total absence of electron diffraction of ordered biological specimens is an indication that the specimen undergoes extensive molecular structural damage in the electron microscope. The specimen damage is due to the interaction of the electron beam (40-100 kV) with the specimen and the total removal of water from the structure by vacuum drying. The lower percentage of inelastic scattering at 1 MeV makes it possible to minimize the beam damage to the specimen. The elimination of vacuum drying by modification of the electron microscope is expected to allow more meaningful investigations of biological specimens at 100 kV until 1 MeV electron microscopes become more readily available. One modification, two-film microchambers, has been explored for both biological and non-biological studies.

<p align="justify"><strong>ABSTRACT</strong><strong></strong></p><p align="justify"><strong>Background</strong>: High fat diet increase the absorption of lipid in the intestinum, that can lead to increase LDL cholesterol level in the blood. Sea grapes extract (<em>Caulerpa racemosa</em>) contains antioxidant polyphenolic group that can reduce MTP and ACAT-2 in the body that can decrease LDL cholesterol level in the blood.The purpose of this study is to know the effect of sea grapes extract  on decreasing LDL cholesterol of white male Wistar rats (<em>Rattus norvegicus</em>) fed with high fat diet.</p><p align="justify"><strong>Method</strong>:  24 white male Wistar rats, that divided into 3 groups: 1) group of rats fed with standard diet for 28 days; 2) group of rats fed with high fat diet for 28 days; 3) group of rats fed with high fat diet for 28 days and given 10 gram/kg body weight/day of sea grapes extract on 15<sup>th</sup>-28<sup>th</sup> days. Then the blood LDL cholesterol level measured on the 29<sup>th</sup> day.</p><p align="justify"><strong>Result :</strong> One-Way ANOVA Test showed there was significant difference (p=0.004) of LDL level between the group of rats fed with standard diet (12.37 mg/dl) compared to group of rats fed with high fat diet (17.87 mg/dl). There was significant difference (p=0.001) of LDL level between the group of rats fed with high fat diet (17.87 mg/dl) compared to group of rats fed with high fat diet and sea grapes extract (10.12 mg/dl).</p><p align="justify"><strong>Conclusion: </strong>high fat diet significantly increase blood LDL cholesterol level and sea grapes extract (<em>Caulerpa racemosa</em>) significantly decrease blood LDL cholesterol level.</p><p align="justify"> </p><p align="justify"><strong>Keywords :</strong>Sea grapes extract, LDL cholesterol, high fat diet</p>

Alga is a marine resource of potential to fisheries and marine sector. It has an important economic value to promote the economy in Indonesia. Nowdays, algae have been used as a relatively high value fisheries commodity since it has been used for food, industrial, pharmaceutical and cosmetic raw materials. This important potential needs to be supported with understanding of its biology and ecology, so that its utilization could increase the livelihood of the coastal villagers. This study was aimed at inventorying and identifying the members of genus Caulerpa found in North Minahasa Regency waters and studying some biological and ecological aspects of the algae in the area. Resuls showed that there were 7 species recorded, Caulerpa racemosa, C. racemosa var. macrophysa, C. sertularioides, C. taxifolia, C. serrulata,C. lentillifera and C. peltata. Ecologically, the environmental parameters, such as water temperature, salinity, pH, dissolved oxygen, turbidity, were in tolerable ranges for algal growth. Potensi yang cukup penting ini harus ditunjang dengan ilmu pengetahuan tentang biologi dan ekologi dari alga laut, sehingga pemanfaatannya dapat meningkatkan taraf hidup masyarakat pesisir. Penelitian tentang kajian bioekologi alga makro genus Caulerpa di perairan Minahasa Utara ini dilaksanakan dan diharapkan dapat memberikan informasi ilmiah tentang bioekologi alga makro genus Caulerpa, sehingga dapat dimanfaatkan untuk pengembangan pemanfaatan bagi kepentingan masyarakat pesisir khususnya dan industri alga makro umumnya. Penelitian ini bertujuan untuk menginventarisasi dan mengidentifikasi alga makro genus Caulerpa di perairan Kabupaten Minahasa Utara, dan mengkaji aspek bioekologinya. Hasil penelitian menunjukkan bahwa ditemukan 7 spesies, yaitu Caulerpa racemosa, C. racemosa var. macrophysa, C. sertularioides, C. taxifolia, C. serrulata, C. lentillifera dan C. peltata. Parameter lingkungan seperti suhu, salinitas, pH, oksigen terlarut, tingkat kecerahan air berada pada kisaran yang dapat ditolerir untuk pertumbuhan alga makro, sedangkan substrat juga mendukung pertumbuhan alga makro ini©

During vacuum drying of used nuclear fuel (UNF) canisters, helium pressure is reduced to as low as 67 Pa to promote evaporation and removal of remaining water after draining process. At such low pressure, and considering the dimensions of the system, helium is mildly rarefied, which induces a thermal-resistance temperature-jump at gas–solid interfaces that contributes to the increase of cladding temperature. It is important to maintain the temperature of the cladding below roughly 400 °C to avoid radial hydride formation, which may cause cladding embrittlement during transportation and long-term storage. Direct Simulation Monte Carlo (DSMC) method is an accurate method to predict heat transfer and temperature under rarefied condition. However, it is not convenient for complex geometry like a UNF canister. Computational Fluid Dynamics (CFD) simulations are more convenient to apply but their accuracy for rarefied condition are not well established. This work seeks to validate the use of CFD simulations to model heat transfer through rarefied gas in simple two-dimensional geometry by comparing the results to the more accurate DSMC method. The geometry consists of a circular fuel rod centered inside a square cross-section enclosure filled with rarefied helium. The validated CFD model will be used later to accurately estimate the temperature of an UNF canister subjected to vacuum drying condition.

The present study aimed to evaluate the effect of Jerusalem artichoke processing methods and drying methods (freeze drying, sublimation drying, vacuum drying) on the basic physicochemical parameters, profiles and contents of sugars and polyphenolic compounds, and health-promoting properties (antioxidant activity, inhibition of the activities of α-amylase, α-glucosidase, and pancreatic lipase) of the produced purée. A total of 25 polyphenolic compounds belonging to hydroxycinnamic phenolic acids (LC-PDA-MS-QTof) were detected in Jerusalem artichoke purée. Their average content in the raw material was at 820 mg/100 g dm (UPLC-PDA-FL) and was 2.7 times higher than in the cooked material. The chemical composition and the health-promoting value of the purées were affected by the drying method, with the most beneficial values of the evaluated parameters obtained upon freeze drying. Vacuum drying could offer an alternative to freeze drying, as both methods ensured relatively comparable values of the assessed parameters.
