The positive-sense single stranded RNA virus, Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2), resulted in a global pandemic with horrendous health and economic consequences not seen in a century. At a finer scale, immunologically, many of these devastating effects by SARS-CoV-2 can be traced to a “cytokine storm” resulting in the simultaneous activation of Janus Kinases (JAKs) and Signal Transducers and Activators of Transcription (STAT) proteins downstream of the many cytokine receptor families triggered by elevated cytokines found in Coronavirus Disease 2019 (COVID-19). In this report, cytokines found in the storm are discussed in relation to the JAK-STAT pathway in response to SARS-CoV-2 and the lessons learned from RNA viruses and previous Coronaviruses (CoVs). Therapeutic strategies to counteract the SARS-CoV-2 mediated storm are discussed with an emphasis on cell signaling and JAK inhibition.

AbstractThis article addresses the call of the Psychology of Global Crises conference for linkage of academic work with social issues in three parts: First, examples from conference participants with their mix of bold calls for social transformation and realization of limits, a combination that generated few clear paths to achieving them. Second, presentation of Jamesian practical idealism with psychological insights for moving past impediments blocking implementation of ideals. And third, a case study of impacts from the most recent prominent crisis, the global pandemic of 2020, which threatens to exacerbate the many crises that had already been plaguing recent history. The tentacles of COVID’s impact into so many problems, starting with economic impacts from virus spread, present an opportunity to rethink the hope for constant economic growth, often expressed as the American Dream, an outlook that has driven so many of the problems surging toward crises. Jamesian awareness of the construction of ideological differences and encouragement of listening to those in disagreement provide not political solutions, but psychological preludes toward improvements in the face of crises.

Emerging research on the economic consequences of the COVID-19 pandemic draws attention to the labor effects of the crisis in the Global South. Developing countries show high levels of labor informality, where most workers cannot work from home and depend on daily income. In addition, the scarce and late state aid makes it difficult for workers to cope with the economic hardships caused by the pandemic. This research explores the employment trajectories of workers throughout the ongoing pandemic in Chile: a neoliberal country with a strong male breadwinner culture and high levels of income inequality. Using longitudinal non-probabilistic data for Chilean employment, this study finds that men lost their jobs to a lesser extent and returned to the labor market faster than women. Likewise, male workers with family (with a partner and young children) remained employed in a higher proportion than female workers with family, and most of these women shifted from employment into care work. The existing literature already pointed out how economic crises can have adverse effects on progress towards gender equality, and the current economic crisis seems to be no exception. Labor informality and low-skilled jobs were highly related to unemployment during the first months of COVID in Chile. These are important variables in a developing economy such as Chile, where around one-third of the population works under these conditions. This article concludes by reflecting on the importance of addressing the present crisis and future economic recovery with a gender perspective.

Planetary health is a broad multidisciplinary effort that attempts to address what has been described as “Anthropocene Syndrome”—the wicked, interrelated challenges of our time. These include, but are not limited to, grotesque biodiversity losses, climate change, environmental degradation, resource depletion, the global burden of non-communicable diseases (NCDs), health inequalities, social injustices, erosion of wisdom and civility, together with the many structural underpinnings of these grand challenges. The ultimate aim of planetary health is flourishing along every link in the person, place and planet continuum. The events of “2020” have illuminated the consequences of “mass trauma” and how sub-threshold anxiety and/or depressive symptoms erase the rigid lines between mental “health” and mental “disorders”, and unmasked the systemic forms of injustice, discrimination, and oppression that have too often escaped discourse. Here, we query the ways in which post-traumatic growth research might inform the larger planetary health community, especially in the context of a global pandemic, broadening socioeconomic inequalities, a worsening climate crisis, and the rise of political authoritarianism. The available research would suggest that “2020” fulfills the trauma criteria of having a “seismic impact on the assumptive world”, and as such, provides fertile ground for post-traumatic growth. Among the many potential positive changes that might occur in response to trauma, we focus on the value of new awareness, perspective and greater wisdom.

Transcription factors of the STAT (signal transducer and activator of transcription) family are important in signal transduction of cytokines. They are subject to post-translational modification by phosphorylation on tyrosine and serine residues. Recent evidence suggested that STATs are methylated on a conserved arginine residue within the N-terminal region. STAT arginine methylation has been described to be important for STAT function and loss of arginine methylation was discussed to be involved in interferon resistance of cancer cells. Here we provide several independent lines of evidence indicating that the issue of arginine methylation of STATs has to be reassessed. First, we show that treatment of melanoma and fibrosarcoma cells with inhibitors used to suppress methylation (N-methyl-2-deoxyadenosine, adenosine, dl-homocysteine) had profound and rapid effects on phosphorylation of STAT1 and STAT3 but also on p38 and Erk signaling cascades which are known to cross-talk with the Jak/STAT pathway. Second, we show that anti-methylarginine antibodies did not precipitate specifically STAT1 or STAT3. Third, we show that mutation of Arg31 to Lys led to destabilization of STAT1 and STAT3, implicating an important structural role of Arg31. Finally, purified catalytically active protein arginine methyltransferases (PRMT1, -2, -3, -4, and -6) did not methylate STAT proteins, and cotransfection with PRMT1 did not affect STAT1-controlled reporter gene activity. Taken together, our data suggest the absence of arginine methylation of STAT1 and STAT3.

In recognition of the growing challenge that food insecurity has on population health, a multisectoral partership in Nova Scotia has been working since 2001 to address province-wide accessibility to a nutritious diet. The participatory food costing (PFC) model has been at the forefront of provincial and national efforts to address food insecurity; a local foods component was incorporated in 2004. This model has engaged community partners, including those affected by food insecurity, in all stages of the research, thereby building capacity at multiple levels to influence policy change and food systems redesign. By putting principles of participatory action research into practice, dietitians have contributed their technical, research, and facilitation expertise to support capacity building among the partners. The PFC model has provided people experiencing food insecurity with a mechanism for sharing their voices. By valuing different ways of knowing, the model has faciliated muchneeded dialogue on the broad and interrelated determinants of food security and mobilized knowledge that reflects these perspectives. The development of the model is described, as are lessons learned from a decade of highly productive research and knowledge mobilization that have increased stakeholders’ understanding of and involvement in addressing the many facets of food security in Nova Scotia.

ABSTRACTViruses with RNA genomes dominate the eukaryotic virome, reaching enormous diversity in animals and plants. The recent advances of metaviromics prompted us to perform a detailed phylogenomic reconstruction of the evolution of the dramatically expanded global RNA virome. The only universal gene among RNA viruses is the gene encoding the RNA-dependent RNA polymerase (RdRp). We developed an iterative computational procedure that alternates the RdRp phylogenetic tree construction with refinement of the underlying multiple-sequence alignments. The resulting tree encompasses 4,617 RNA virus RdRps and consists of 5 major branches; 2 of the branches include positive-sense RNA viruses, 1 is a mix of positive-sense (+) RNA and double-stranded RNA (dsRNA) viruses, and 2 consist of dsRNA and negative-sense (−) RNA viruses, respectively. This tree topology implies that dsRNA viruses evolved from +RNA viruses on at least two independent occasions, whereas −RNA viruses evolved from dsRNA viruses. Reconstruction of RNA virus evolution using the RdRp tree as the scaffold suggests that the last common ancestors of the major branches of +RNA viruses encoded only the RdRp and a single jelly-roll capsid protein. Subsequent evolution involved independent capture of additional genes, in particular, those encoding distinct RNA helicases, enabling replication of larger RNA genomes and facilitating virus genome expression and virus-host interactions. Phylogenomic analysis reveals extensive gene module exchange among diverse viruses and horizontal virus transfer between distantly related hosts. Although the network of evolutionary relationships within the RNA virome is bound to further expand, the present results call for a thorough reevaluation of the RNA virus taxonomy.IMPORTANCEThe majority of the diverse viruses infecting eukaryotes have RNA genomes, including numerous human, animal, and plant pathogens. Recent advances of metagenomics have led to the discovery of many new groups of RNA viruses in a wide range of hosts. These findings enable a far more complete reconstruction of the evolution of RNA viruses than was attainable previously. This reconstruction reveals the relationships between different Baltimore classes of viruses and indicates extensive transfer of viruses between distantly related hosts, such as plants and animals. These results call for a major revision of the existing taxonomy of RNA viruses.

Introduction. Free flap surgery is a routine procedure in many developed countries with good surgical outcomes. In many developing countries, however, these services are not available. In this paper, we audit free flaps done in a resource constrained hospital in Kenya. Objective. This is a five-year audit of free flaps done in a tertiary hospital in Kenya, between 2009 and 2014. Materials and Methods. This was a prospective study of patients operated on with free flaps between 2009 and 2014. Results. A total of one hundred and thirty-two free flaps in one hundred and twenty patients were performed during the five-year duration. The age range was eight to seventy-two years with a mean of 47.2. All the flaps were done under loupe magnification. The overall flap success rate was eighty-nine percent. Conclusion. Despite the many limitations, free flaps in our setup were successful in the majority of patients operated on. Flap salvage was noted to be low due to infrequent flap monitoring as well as unavailability of theatre space. One therefore has to be meticulous during surgery to reduce any possibilities of reexploration.

I’ve been so thankful to the many professionals who have so generously given their time and shared their expertise for this column. From lessons learned, case studies and differing opinions on RA and its future, it is amazing how various and rich this area of librarianship is—and how rewarding and frustrating! In an effort to continue to provide a broad spectrum of thoughts and ideas, I asked Dr. Louise Spiteri of Dalhousie University to write for this issue. Spiteri recently completed two stages of research examining subject headings and user-generated content and how these connect with RA access points. Jen Pecoskie was Spiteri’s research partner in both studies.—Editor

AbstractDuring the coronavirus global pandemic crisis, we have received information from authentic and inauthentic sources. Fake news, continuous rumors, and prejudiced opinions from digital platforms and social media have the capacity to disrupt social harmony, to stall personal development, and to undermine trust on all levels of human interaction. Despite the wide plurality of perspectives, the diversity of contents, the variety of voices, and the many often-conflicting reasons for publishing, our interactions with information on digital devices are progressively shaping such situations and affecting decisions on all levels. We look at the limitations of existing designs and guidelines in the current paradigm, and we ask to what extent researchers and developers can focus and contribute, through their innovations, to the reduction of uncertainty and cases of misdirection, how they can mitigate tensions between information and humans, and how they can contribute to the maintenance and enhancement of worthy human values. Human-engaged computing (HEC) calls for innate user capacities to be enhanced rather than displaced by digital technologies so that the human factor in interactions is fully exploited and truly efficient symbiotic relationships between humans and devices can be achieved. Under the framework of HEC, we propose 12 research agendas from the theoretical, principled, and practical aspects, in order to develop future synergized interactions between humans and information. The present crisis presents us with a good opportunity to reflect on the need to empower humans in relation to the tools they use and to consider the next paradigm shift for designing information interaction.

As the use of technology has become more prevalent within the educational environment over the past decade, the emergence of the use of virtual manipulatives to support student learning in math has made transitioning to technology-infused math instruction unavoidable. Students in rural areas, however, have tended to receive far less technology-infused instruction due to the many challenges faced by rural schools that can adversely affect academic opportunities and disrupt equity in learning and teaching. In the current paper, we report on a classroom study conducted to examine whether the previously proven effects of concrete manipulatives can carry over into those of virtual manipulatives when teaching math fact fluency in multiplication and explored the potential for virtual manipulatives in rural classrooms from the teacher’s perspective.  Quantitative and qualitative results both indicated a promising potential for usage of virtual manipulatives, with meaningful implications for practitioners. The educational implications for designing and planning effective instruction incorporating virtual manipulatives are discussed.
