Although fluoride (F) is well-known to prevent dental caries, changes in cell processes in different tissues have been associated with its excessive exposure. Thus, this study aimed to evaluate the effects of F exposure on biochemical, proteomic, and genotoxic parameters of submandibular glands. Twenty one old rats (n = 30) were allocated into three groups: 60 days administration of drinking water containing 10 mgF/L, 50 mgF/L, or only deionized water (control). The submandibular glands were collected for oxidative biochemistry, protein expression profile, and genotoxic potential analyses. The results showed that both F concentrations increased the levels of thiobarbituric acid–reactive substances (TBARS) and reduced glutathione (GSH) and changed the proteomic profile, mainly regarding the cytoskeleton and cellular activity. Only the exposure to 50 mgF/L induced significant changes in DNA integrity. These findings reinforce the importance of continuous monitoring of F concentration in drinking water and the need for strategies to minimize F intake from other sources to obtain maximum preventive/therapeutic effects and avoid potential adverse effects.

This work aims to evaluate the quality of drinking water in the communities of Baiacu, Sao Francisco do Conde, Santo Amaro and Cachoeira. It was verified that in some places of Santo Amaro and Sao Francisco do Conde, from the microbiological point of view, it is not appropriate for consumption, because it was found that the presence of total coliforms in 50% of the samples and that the chlorine content is below the values indicated by the standards in 75% of the analyzed samples. Thus, the consumption of contaminated water can cause diseases, so it is recommended cleaning of residential reservoirs, maintenance in pipes periodically, as well as the need for continuous monitoring of water quality and the intense participation of public agencies in these actions. Through these actions and the active participation of the population, one can preserve the environment and the lives of all those who transit in the communities of the Bay of All Saints

For waterworks the most fundamental subject is to maintain the safety of drinking water. It has become important every year to monitor the concentrations of trihalomethanes (THM). We developed a new automatic THM analyzer based on fluorometry with membrane separation, and investigated a correlation between the gas Chromatographic method and our THM analyzing method on drinking waters at 10 cities in Japan. The investigation resulted in good correlation (r=0.9807) between both methods. Good results were obtained when continuous measurements of THM in drinking water were performed by the analyzer. Consequently, it was found that the present analyzer could measure THM in drinking waters simply, rapidly, with good precision and could be applied to continuous monitoring of THM.

Abstract Diabetes mellitus is a metabolic disorder characterised by hyperglycemia and oxidative stress. The aim of the present study is to explore the antioxidant effect of Spirulina maxima in rat model along with the histopathological observations. Diabetes was induced by feeding 10% fructose solution orally to Wistar rats (n = 6) for 30 days, analysed for plasma blood glucose and the markers of the oxidative stress [catalase (CAT), superoxide dismutase (SOD), reduced glutathione (GSH) and thiobarbituric acid reactive substances (TBARS)]. These biochemical studies were associated with histopathological examination of liver and kidney sections. The microalga Spirulina maxima being rich in proteins and other essential nutrients is widely used as a food supplement. S. maxima at a dose of 5 and 10% per kg and the metformin (500 mg/kg) as reference drug were given orally for 30 days to the diabetic rats. Diabetic rats showed significant (p < 0.001) elevations in plasma blood glucose, thiobarbituric acid-reactive substances and significant reduction in catalase, superoxide dismutase and reduced glutathione activity. Oral administration of 5 and 10% aqueous extract of S. maxima for 30 days restored not only of blood glucose levels but also markers of oxidative stress. Histopathological observations of tissues manifested that the S. maxima administration had the protective and therapeutic effects against fructose-induced abnormalities in diabetic rats. It is concluded that S. maxima is effective in reinstating the antioxidant activity in addition to its antidiabetic effect in type 2 diabetic rats.

The toxin 6-hydroxydopamine (6-OHDA) is a highly oxidizable dopamine (DA) analog that is widely used for reproducing several cell processes identified in Parkinson’s disease (PD). Due to the close similarity of its neurotoxic mechanism to those of DA, it is suitable as a model for testing the effects of potentially neuroprotective drugs. This study aimed to evaluate the effect of alpha-lipoic acid (LA) on brain oxidative stress (OS) in unilateral intrastriatal (6-OHDA) injected rats. Forty male Wistar rats, four months old (220–260 g), were evaluated. Half of them received LA (35 mg/kg i.p.) from the start to the end of the experiment. On day 2 of the trial, ten LA-supplemented rats and ten non-LA-supplemented rats were subjected to the apomorphine test. Brain homogenates were evaluated for thiobarbituric acid-reactive substances (TBARS) and glutathione peroxidase (GPx) activity. The same evaluation procedures were repeated on day 14 with the remaining animals. An increased TBARS level and decreased GPx activity, suggestive for OS, were recorded in homogenates on day 14 vs. day 2 of the experiment in the 6-OHDA treated rats. The simultaneous application of LA mitigated these changes. Our study demonstrates that the low dose of LA could be of value for decreasing the OS of the neurotoxic 6-OHDA, supporting the need for further studies of the benefit of LA treatment in PD.

Galectins constitute of a soluble mammalian β-galactoside binding lectin family, which play homeostatic roles in the regulation of the cell cycle, and apoptosis, in addition to their inflammatory conditions. Galectin-3 has an important role in the regulation of various inflammatory conditions including endotoxemia, and airway inflammation. Statins, the key precursor inhibitors of 3-hydroxyl-3-methyl coenzyme A (HMG-CoA) reductase, may prevent the progression of inflammation in sepsis after prior statin treatment. Endotoxemia leads to the formation of oxidative stress parameters in proteins, carbohydrates, and DNA. In the present study, we aimed to show the effects of simvastatin on Galectin-3, and glutathione reductase (GR), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), and thiobarbituric acid reactive substances (TBARS) levels in lung tissue of rats which were treated with lipopolysaccharides (LPS) during the early phase of sepsis. Rats were divided into four groups as the control, LPS (20 mg/kg), simvastatin (20 mg/kg), and simvastatin+LPS group. Galectin-3 expression in formalin-fixed paraffin-embedded lung tissue sections was demonstrated by using the immunohistochemistry methods. There were reduced densities, and the decreased number of Galectin-3 immunoreactivities in the simvastatin+LPS group compared with the LPS group in the pneumocytes, and in the bronchial epithelium of lung tissue. In the LPS group, GR, GSH-Px, and SOD were found lower than the levels in simvastatin-treated LPS group (P<0.05, P<0.01, P<0.01 respectively) in the lung tissue. However, TBARS decreased in the simvastatin+LPS group compared with the levels in LPS group (P<0.001). Simvastatin attenuates LPS-induced oxidative acute lung inflammation, oxidative stress, and suppresses LPS-induced Galectin-3 expression in the lung tissue.

Abstract To determine how lipid peroxides and free radical scavengers are changed in the brain of hyper- or hypothyroid rats, we examined the behavior of lipid peroxide and free radical scavengers in the cerebral cortex of aged (1·5 years old) rats that had been made hyper- or hypothyroid by the administration of thyroxine or methimazol for 4 weeks. Concentrations of catalase, Mn-superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX) were increased in hyperthyroid rats compared with euthyroid rats. Concentrations of total SOD, Cu,Zn-SOD and GSH-PX were increased but that of Mn-SOD was decreased in hypothyroid animals. There were no differences among hyperthyroid, hypothyroid and euthyroid rats in the levels of coenzymes 9 or 10. The concentration of lipid peroxides, determined indirectly by the measurement of thiobarbituric acid reactants, was decreased in hyperthyroid rats but not in hypothyroid rats when compared with euthyroid animals. These findings suggest that free radicals and lipid peroxides are scavenged to compensate for the changes induced by hyper- or hypothyroidism. Journal of Endocrinology (1995) 147, 361–365

Six groups, each of six sheep, were fed in pens for 15 months on a ration of chaffed lucerne and wheaten hays. One group was offered rain-water to drink, another group was offered 1.30% sodium chloride, whereas the others were offered one of the following mixtures of sodium chloride, carbonate, and bicarbonate: 1.26 + 0.015 + 0.025, 1.21 + 0.04 + 0.06, 1.12 + 0.08 + 0.13, and 0.95 + 0.161+ 0.25%. The intake of all saline solutions was higher than that of rain-water, ranging from 150% above for 1.30% sodium chloride to 60% above for the highest level of carbonates; the mean daily intakes for the entire experiment by the six groups were 2.6, 6.6, 4.8, 5.7, 5.8, and 4.2 l. respectively. The intake also increased in all groups with temperature, being 40–70% higher in the hottest months than in the coldest months.Weight increase was less from 6 months onward in the group receiving 1.30% sodium chloride, and was less at certain times only in the experiment in the groups receiving 0.04 or 0.10% carbonates, than in that receiving rain-water (control group). There were no differences in weight increase between the control group and the groups receiving the highest concentrations (0.21 and 0.41%) of carbonates The saline drinking waters had no effect on the concentrations of sodium, potassium, calcium, magnesium, or chloride in the blood plasma. The concentration of carbon dioxide was higher, for the last year of the experiment, in the blood of the control group, and, for approximately one-third of the experiment, in that of the group receiving the highest level of carbonates in its drinking water, than in that of any of the groups receiving lower levels of carbonates. There were differences in blood carbon dioxide on one occasion only between the control group and that receiving the highest level of carbonates. None of the solutions used in the experiment had any adverse effect on the general health, food consumption or wool production of the sheep.
